During the evolution organisms are subjected to the continuous impact of environmental factors. In recent years an increasing number of studies have focused on the physicochemical limits of life on Earth such as temperature, pressure, drought, salt content, pH, heavy metals, etc. Extreme environmental conditions disrupt the most important interactions that support the function and structure of biomolecules. For this reason, organisms inhabiting extreme habitats have recently become of particularly great interest. Although filamentous fungi are an important part of the polar ecosystem, information about their distribution and diversity, as well as their adaptation mechanisms, is insufficient. In the present study, the fungal strain Penicillium griseofulvum isolated from an Antarctic soil sample was used as a study model. The fungal cellular response against short term exposure to low temperature was observed. Our results clearly showed that short-term low temperature exposure caused oxidative stress in fungal cells and resulted in enhanced level of oxidative damaged proteins, accumulation of reserve carbohydrates and increased activity of the antioxidant enzyme defence. Ultrastructural changes in cell morphology were analysed. Different pattern of cell pathology provoked by the application of two stress temperatures was detected. Overall, this study aimed to observe the survival strategy of filamentous fungi in extremely cold habitats, and to acquire new knowledge about the relationship between low temperature and oxidative stress.

Opis:
Real-time cell electronic sensing (RT-CES) based on impedance measurements is an emerging technology for analyzing the status of cells in vitro. It allows label-free, real time monitoring of the biological status of cells. The present study was designed to assess dynamic data on the cell processes during equine herpesvirus type 1 (EHV-1) infection of ED (equine dermal) cells and primary murine neuronal cell culture. We have demonstrated that the xCELLigence system with dynamic monitoring can be used as a rapid diagnostic tool both to analyze cellular behavior and to investigate the effect of viral infection.

W ramach pracy otrzymano włókninę zbudowaną z włókien o różnych średnicach i wielkości, której mikrostruktura posiada biomimetyczny charakter, tzn. składa się z włókien o średnicach zbliżonych do średnic włókien występujących w tkankach. Badania biologiczne wykazały, że włókna o niskich średnicach są gorzej tolerowane przez tkanki.
A three dimensional fibrous material, made from fibres differing in diameters and porosity, has been designed and prepared. These materials will constitute a 3D scaffold containing fibrous components mimicking the structure of natural tissue. The biological studies indicate that the fibres with bigger diameter allow for more intense and quick regeneration of surrounding tissue.

Opis:
The influence of surface (nano)roughness of soft elastomeric materials containing nanocrystalline TiO2 on tissue cell behaviour after implantation tests was investigated. Addition of small amount (0.2wt.%) of TiO2into polymer matrix changed surface roughness giving material of well developed lamellar morphology and slightly diminished contact angle (wettability). This lamellar (fibrilar) morphology had strong influence on cell response after implantation into soft tissue indi-cating the absence of eosynophiles in tissue around implant.

Opis:
Objective. The aim of the study was determination of the level in expression genes associated with cell stress response in a patient with PTSD. Case study. A 57-year-old PTSD patient, A.P., was the subject of the study. Blood samples were taken from the ulnar vein to assess the relative expression of the tested genes in leukocytes via a quantitative reverse transcription polymerase chain reaction (qRT-PCR). Results. A considerably decreased level in the expression HSF-1, NF-kB, Hif-1 and IL6 m-RNA was observed in the patient when compared to the control group. A low expression in HSPA1A, HSPB1, IL10 and CRP m-RNA compared with the control group was also noted. Conclusion. It is possible that the lower level of transcriptional factors: HSF-1, NF-kB and Hif-1 as well as IL6 m-RNA may be associated with the effect of the PTSD patient’s treatment.

Opis:
The multixenobiotic resistance (closely related to multidrug resistance) system controls transport across the plasma membrane as a defense against toxic molecules. Multixenobiotic resistance system consists of an efflux pump, ABCB1 (also named P-glycoprotein, P-gp), and/or a molecule of the ABCC family (also named multiple resistance associated protein, MRP). ABCB1 is able to increase efflux of many low-molecular foreign molecules. Measuring system induction may be used as a biomarker of cell/organism exposure to foreign substances. Various established cell lines were tested for constitutive and induced multixenobiotic resistance proteins by Western blotting immunodetection. The pumping function was indirectly assayed with Rhodamine B by visualization of cell fluorescence in the presence of verapamil. Changes in ABC proteins were measured by flow cytometry after exposition to various perfluorinated carboxylic acids. MCF7 and HeLa cells were found to contain the highest constitutive level of both ABCB1 and ABCC1. HEK293 exhibited much less ABCB1 and no activity of pumping out Rhodamine B. The pumping activity was found to be related to the amount of the cell-type specific 170 kDa ABCB1 protein. An 8-day exposure to 10-4 M perfluorononanoic acid resulted in about 2-2.5-fold increase of ABCB1 level. That was confirmed also for short times by flow cytometry of cells exposed to perfluorinated acids and its natural congeners. Both ABCB1- and ABCC1-related fluorescence increased along with the carbon chain in acids from C6 up to C9 and decreased for C10. Measuring of multixenobiotic resistance changes in vitro induced by chemicals may be a convenient test for screening for their potential toxicity.

The analysis of retinal ganglion cell responses to electrical stimulation is invaluable way to understand how the retina reacts and develops. Artificial visual prosthetics have made progress to some extent. Although there are several studies about retinal ganglion cell responses in the literature, experimental findings and information about responses to special stimulation patterns for developing high-resolution visual prosthetics are quite limited. In this study, rabbit retinal tissue was stimulated by biphasic, monophasic and ramp function current pulses with various amplitudes and pulse widths in a set of in vitro experiments. Rabbit retinal ganglion cell responses and spike activities are comparatively analyzed based on the applied electrical stimulation parameters. We conclude that biphasic current pulses provide a lower stimulation threshold of 10 μA with shorter pulse widths than those obtained by monophasic stimulation. Additionally, despite the fact that ramp function stimulation patterns have remarkable potential for eliciting spike formation with low stimulation thresholds, biphasic stimulation is charge-balanced and more convenient for high-resolution visual prosthesis systems.

Gut-derived adenocarcinoma Caco-2 cells were treated with sodium butyrate (NaB) at physiologically relevant concentrations. We characterized its effects on proliferation, differentiation, apoptosis, adhesion to the solid support and interleukin-8 secretion. Differentiation was determined by brush border alkaline phosphatase activity. Apoptosis was assessed by acridine orange and Hoechst stains. Differentiation and apoptosis were analyzed in both adherent and floating cell populations. The transformed Caco-2 cells did not retain their malignant phenotype in the presence of NaB. They appeared to undergo a change in the phenotype induced by NaB, as indicated by reduced proliferation, enhanced differentiation, stimulation of apoptosis leading to decreased viability of cells, and stimulation of interleukin-8 secretion. Considering all the above facts and data, we postulate that Caco-2 cells cultured in NaB supplemented medium could regain the phenotypic characteristics of the phenotype of the parent cell from which originated the Caco-2 line.

Since it is known that various cell lines may ex-press different behaviours on the scaffolds surface, a comprehensive analysis using various cellular mo-dels is needed to evaluate the biomedical potential of developed biomaterials under in vitro conditions. Thus, the aim of this work was to fabricate bone scaffolds composed of a chitosan-agarose matrix reinforced with nanohydroxyapatite and compare the biological response of two cell lines, i.e. mouse calvarial preosteoblasts (MC3T3-E1 Subclone 4) and human foetal osteoblasts (hFOB 1.19). Within this study, the osteoblasts number on the scaffold surface and the osteogenic markers level produced by MC3T3-E1 and hFOB 1.19 cells were determined. Furthermore, changes in calcium and phosphorous ions concentrations in the culture media dedicated for MC3T3-E1 and hFOB 1.19 were estimated after the biomaterial incubation. The obtained results proved that the fabricated biomaterial is characterized by biocompatibility and osteoconductivity since it favours osteoblasts attachment and growth. It also supports the production of osteogenic markers (collagen, bALP, osteocalcin) by MC3T3-E1 and hFOB 1.19 cells. Interestingly, the developed biomaterial exhibits different ion reactivity values in the two culture media dedicated for the mentioned cell lines. It was also revealed that mouse and human osteoblasts differ in the cellular response to the fabricated scaffold. Thus, the use of at least two various cellular models is recommended to carry out a reliable biological characterization of the novel biomaterial. These results demonstrate that the tested bone scaffold is a promising biomaterial for bone regeneration applications, however further biological and physicochemical experiments are essential to fully assess its biomedical potential.
