Opis:
Interleukin–1 is one of the most potent proinflammatory cytokines involved in many physiologically important processes both beneficial and pathological. It appears in two different forms: IL-1 and IL-1b synthesized mainly by macrophages and monocytes. IL-1 remains inside the cells and is released during acute inflammatory diseases accompanied by cell lysis. IL-1b becomes biologically active after specific cleavage and is secreted to fulfill its multitude functions. This cytokine is involved in multiple gene up-regulation, higher expression of many cytokine receptors or induction of nitric oxide synthesis. Interleukin-1 is responsible for symptoms like fever or appetite loss during inflammatory diseases. It is capable of lymphocytes, macrophages or NK cells activation. This cytokine has a chemoattractant activity toward neutrophiles and monocytes and takes part in Th2 response development. Apart from other functions and activities, IL-1 is associated with many invasive illnesses such as parasitic infections. This cytokine influences the outcome of numerous parasitoses since it can limit parasite spread and survival within infected host by cooperation with other components of the immunological system and by the induction of anti-parasitic compounds. However, the proinflammatory activity of IL-1 may prove harmful in certain cases and may be responsible for parasite infection development.

Opis:
Precise understanding of the mechanisms of reciprocal relations between the nervous and the immune systems, has been the subject of numerous studies for the recent two decades. These mechanisms are significant, particularly at the stage of early response to bacterial, parasite, or viral infections. They are also essential from the medical point of view, as they may help in the development of the new methods of treatment of infectious diseases, and also may provide better methods to neutralize possible side effects of the therapy. As it is commonly understood, both forms of IL-1 ( and b), play an important role as a signaling molecules in these mechanisms. Regardless of the route of administration, they cause to the activation of the brain neurotransmitters, and the hypothalamo-pituitary-adrenal-axis (HPA). The HPA response induced by activity of the immune system is a normal, physiological phenomenon with essential meaning. It gives the negative feedback where glucocorticoids, released from the adrenal cortex, inhibit activity of the immune system, and by this reduce the probability of the over-stimulation of this system and its selfaggression. Therefore, precise recognition of the mechanism which is the indicator of influence of cytokines on the brain and also leads to initiate that response has a significant scientific and practical meaning. Also, the two mechanisms are probably the most important, and under appropriate conditions could complement each other. These are enzymatic and neural ways by which immune system influences the brain. The former predicts, that Il-1 influences the tissue, stimulating them to the synthesis, via the cyclooxygenases (COX) activation, and release molecules such as prostaglandines (especially PGE2), which have the ability to penetrate the brain barrier. The latter assumes that IL-1, directly or indirectly, can influence the peripheral nerves (the most important is probably the vagus nerve), which afferent sensory endings subsequently send signals to the brain. The results of many experiments (including the authors’ own) indicate that both mentioned above ways take part in the mechanism of the communication between the immune and the nervous systems. These point out that signaling via the vagus nerve and via COX may contribute to the effects of IL-1 administration on animals behavior, and the activation of HPA axis and brain neurochemistry. However, the vagal pathway and the COX-system appear to substitute each other during infections or pathogens invasion. The experimantal data also suggest that these two mechanisms by which IL-1 can affect the brain are probably the most important ones, by which the immune system influences the nervous system.

Opis:
Interleukin 1 (IL-1) and interleukin 6 (IL-6) are typical examples of multfunctonal pro-infammatory cytokines involved in the regulaton of the immune response, hematopoiesis, and infammaton. Both peripheral and intraventricular administraton of these cytokines causes acute phase symptoms, e.g. fever, actvaton of the hypothalamic-pituitary-adrenal axis and psychological depression. The amygdala belongs to the structures of the limbic system involved in the regulaton of the immune response. Increased actvity of immune system may lead to changes in the role of amygdala, medial prefrontal cortex, anterior cingulate cortex or insula. The aim of the study was to present the mutual interactons between the amygdala and pro-infammatory cytokines such as interleukin-1β (IL-1 beta) and interleukin 6 (IL-6). Most of the data included in this review comes from animal studies.

Opis:
IL-1 family of cytokines, is a group of several proteins which are involved in regulation of immune response. It has been shown that members of that family , especially: IL-1α, IL -1β, IL-18 and IL-33 act as mediators in nociceptive transmission and play a crucial role in pathogenesis of multiple diseases. The discovery of IL-1Ra, naturally occurring antagonist of I L-1 receptor (IL-1RI i IL-1RII) and IL-18 binding protein has led to many studies involving use of recombinant receptor antagonist in pain therapy or in treatment of such a diseases as: rheumatoid arthritis, multiple sclerosis, skin diseases or diabetes.
Rodzina Interleukiny-1 to rodzina kilkunastu, należących do cytokin, cząsteczek białkowych zaangażowanych głównie w regulację odpowiedzi odpornościowej. Udokumentowano, że członkowie tej rodziny, w szczególności: IL-1α i -1β, IL-18 oraz IL-33 pełnią istotne funkcje w mediowaniu transmisjii nocyceptywnej oraz w patogenezie wielu chorób. Odkrycie naturalnie występującego antagonisty (IL-1 Ra) receptorów z rodziny IL-1 (IL-1RI i IL-1RII) oraz białka wiążącego IL-18, doprowadziło do rozpoczęcia szeregu badań nad zastosowaniem zrekombinowanych antagonistów w leczeniu bólu czy zapobieganiu postępu choroby w takich schorzeniach jak, m.in.: reumatoidalne zapalenie stawów, stwardnienie rozsiane, choroby skóry czy cukrzyca.

An average of 4.9 tapeworms were discovered on day 7 of a low-abundance H. diminuta infection of rats of race WAG alb. After 11 months, the mean was only relatively slightly tower at 3.6. These means represent 97.1 and 71.4% of the 5 cysticercoids supplied. In turn, 7 days and 11 months after rats were supplied with 110 cysticercoids, the respective percentages were 85.1 and 56.0. All 7- and 12-day tapeworms from the low-abundance and crowded infrapopulations were characterized by the presence of a terminal proglottid of lingulate shape, in which the excretory canals joined. In contrast, older (48-day and 11-month-old) worms showed typical apolysis of gravid proglottids. There were no reports of the destrobilation of tapeworms, and the relatively large number of tapeworms persisting 11 months into the infection is particularly noteworthy. The results point to the lack of any rapid rejection of tapeworms of the kind characterized in many other studies on H. diminuta.

Background. During the last few decades, adiposity has become a relatively common phenomenon worldwide. The available data on the effects of pro-inflammatory factors in both depression and adiposity has been attracting great attention. Aim. We sought to assess the prevalence of -889C>T IL-1α, -31T>C and -511C>T IL-1β, -330T>G IL-2 and -174G>C IL-6 genes and their association with adiposity and depression in Polish subjects. Methods. A cohort study was conducted in 2013/2014, covering a sample of 297 individuals (217 female and 80 male). Anthropometric data was handled using the BIA analysis method, while for genotyping PCR-RFLP techniques were used. Results. A positive correlation between depression and anthropometric parameters: adipose tissue (in kg) and adipose tissue (in %) (R=0.135 and p=0.02, R=0.114 and p<0.05, respectively) was found. No association between studied polymorphisms and depression was observed. Conclusion. Although it was not possible to demonstrate any influence of the studied polymorphisms as the genetic modulator of depression, authors believe that the presented data are noticeable and may provide the basis for future studies on larger groups.

In this study plasma concentrations of IL-1α and IL-8 in 29 female dogs with malignant mammary gland tumours (19 without metastasis and 10 with metastasis) and in 10 healthy control animals were determined. Concentrations of IL-1α and IL-8 were analysed using a specific canine ELISA assay. Mean plasma concentrations of IL-1α and IL-8 were significantly higher (p<0.05) in female dogs with both non-metastatic and metastatic malignant tumours compared to the healthy animals. The concentrations of both tested cytokines were significantly increased (p<0.05) in the dogs with metastasis. In female dogs with mammary carcinomas, the plasma concentration of IL-1α was significantly higher (p<0.05) in the animals with grade 3 tumours compared to the dogs with grade 1 tumours. The concentration of IL-8 was significantly higher (p<0.05) in the dogs with grade 3 tumours compared to that found in the animals with grade 1 and grade 2 tumours. A moderate correlation (r=0.433) was found between IL-1α and IL-8 concentrations in the female dogs. These findings suggest that increased malignancy and invasiveness of canine mammary tumours is associated with an increased production of IL-1α and IL-8 in the tumour microenvironment, which, in turn, leads to an increase in their circulating levels. This may indicate that circulating levels of the cytokines investigated could be considered as diagnostic and prognostic markers in canine malignant mammary tumours. However, further studies in this fields are needed.

Variable position of genital ducts and pores (PGP) was found in 23.3% of five months old tapeworms obtained as a result of administering to rats a dose of 6 cysticercoids proceeding from the third generation of inbred Hymenolepis diminuta - WMS il1. In tapeworms coming from generations no 18 and 32 PGP changes were reported only in 7.5 month, with only 5.9% of strobilas in the 32 generation group being characterized by this feature. These results indicate a time shift with respect to the appearance of PGP changes in tapeworms proceeding from latter inbred generations to a further period of tapeworm invasion in rats. The number of PGP changes is positively correlated with the number of type 2p1a proglottids (two testes on the poral and one on the aporal side). The average number of type Op3a proglottids increases with the number of generations. In 5-month old tapeworms of unilateral PGP coming from generation no 3, 18 and 32 it amounts to 5.9%, 6.6% and 8.1 %, respectively.

Crowded infrapopulations of H. diminuta obtained from the WMS i11 inbred line resembled those of the potentially non-inbred WMS strain in not showing either the abrupt reductions in the numbers of worms or the destrobilation described by other autors. Denser populations of both types of tapeworm have a greather abundance of 4 - testis proglottids of the lp3a type (having 1 testis on the poral side and three on the aporal), and fewer of type 0p3a. Changes in the positioning of genital ducts and pores show a marked positive correlation with the abundance of type 2pla proglottids. Strobilae of H. diminuta WMS i11 contain a relatively greather number of type 0p3a proglottids and fewer of type 1p3a, than those of the WMS „strain"; something which is probably linked with the respective selection of the maternal Tapeworms.

Opis:
Growth arrest and DNA damage-inducible (GADD) 45 proteins are regulators of cell death and survival. The proinflammatory cytokine IL-1β strongly increases the level of the transcript encoding GADD45α in rat insulin-producing INS-1E cells. The activation of Gadd45α gene is clearly dependent on JNK and NF-κB activation and the synthesis of the secondary mediator nitric oxide (NO). Interestingly, the observed twelve-fold increase in the GADD45α-coding transcript level is not followed by increased expression of GADD45α at the protein level. An analysis of IL-1β toxicity in INS-1E cells overexpressing GADD45α revealed no correlation between the GADD45α protein level and the sensitivity to IL-1β toxicity. These findings suggest that the potential engagement of GADD45α in IL-1β toxicity towards beta cells is limited to the effects induced by the basal expression level of this protein.
