Opis:
Cytoplasmic male sterility (CMS) is a maternally inherited trait in which a plant is unable to produce functional pollen. It occurs in many plants, including carrots. The expression of this trait depends on environmental conditions, especially temperature – therefore, DNA markers become an excellent tool for selecting carrot genotypes with this feature. In this study, we used seventeen DNA markers available in the literature, linked with various mitochondrial DNA sequences that are associated with the CMS trait. Their amplification was tested on male sterile and male fertile plants. Only five markers – A321/392, I214/401/435, K751, N795, O790 – correctly identified the tested genotypes. They can be used as selection tools in carrot breeding programs.

Opis:
Barley scald, caused by Rhynchosporium commune is one of the most prevalent diseases in barley (Hordeum vulgare L.) worldwide. The primary loss from scald is reduced yield, which can exceed 25% in dry areas. In our earlier studies, we developed a low-resolution linkage map for recombinant inbred lines of the cross Tadmor/WI2291. Quantitative trait loci (QTLs) for scald were localized on chromosomes 2H and 3H flanked by Simple Sequence Repeat (SSR) markers HVM54 and Bmac0093b on 2H and HVLTPP8, HVM62 and Bmag0006 on 3H. These chromosome 3H markers were found to be located close to the Rrs1 − R. commune resistance gene(s) on chromosome 3H. In this study, 10 homozygous resistant and 10 homozygous susceptible plants each from the F7 population of Tadmor/ Sel160, a panel of 23 barley varieties used routinely in the International Centre for Agricultural Research in the Dry Areas (ICARDA) breeding program and three populations were used for scald resistance screening using 25 DNA markers that are located very close to scald resistance gene(s) on barley chromosomes. Only five of those markers clearly discriminated co-dominantly between resistant and susceptible plants. These markers, Ebmac0871- SSR, HVS3-SCAR, Bmag0006-SSR, reside on different arms of barley chromosome 3H. Ebmac871 is localized on the short arm of 3H and HVS3 and Bmag0006 are localized on the long arm of 3H. This result indicates that the scald resistance genes which they tag are probably close to the centromeric region of this chromosome. Scald resistance from several sources map to the proximal region of the long arm of chromosome 3H, forming the complex Rrs1 locus. The availability of highly polymorphic markers for the discrimination of breeding material would be extremely useful for barley breeders to select for the trait at the DNA level rather than relying on phenotypic expression and infection reaction.

Opis:
All tree breeders cope with the same challenge of the very long time interval of a single breeding cycle. What is more, trees are long−lived, with desirable breeding traits expressing late during their life cycle. Increasing problems with climate change, globalization or economic growth have forced us to accelerate tree breeding and improve selection precision, both of which can be achieved by genomic selection (GS). The idea of GS was introduced nearly 20 years ago as an extension of marker−assisted selection (MAS) in order to advance breeding technologies using genetic markers. Unlike MAS, which exploits only a set of marker−trait associations, GS relies on a high number of genetic markers that are spread throughout the entire length of the genome. All markers effects are assessed simultaneously in order to build a precise model that allows prediction of genetic estimated breeding value of a particular individual using genetic data only. GS has already revolutionized dairy cattle breeding resulting in remarkable improvements across multiple traits and is becoming more and more common in crop production. We now know that genetic architecture of quantitative traits is complex, but recent advances in genomics have made it possible to deal with this problem in an unprecedented way. There are certain concerns regarding GS in forest tree species that include genotype−environment (G×E) interaction and the usefulness of the predictive model built up by GS in the next generation of trees. Nevertheless, experimental results obtained so far have shown that the genetic gain per unit time as well as selection precision can be substantially increased. Here we present the basic principles of GS for forest tree species, giving examples of studies carried out so far and discussing problems and future possibilities that GS may soon open up for forest tree breeders.

Opis:
The efficiency of breeding new potato cultivars may be increased by pre-breeding that is by developing parental lines, which have new traits, not present in the genetic pool available for breeders or have new combinations of genes, or have improved level of economically important traits. The use of parental lines in commercial breeding programs is expected to ensure the biological progress in the newly created cultivars of potato. At the beginning, the development of parental lines in Młochów Research Center of Plant Breeding and Acclimatization Institute – Nation-al Research Institute (IHAR-PIB) was focused on resistance to viruses and later on resistance to late blight and other pathogens. The procedures of selecting resistant parental lines are described. These procedures were initially based on purely phenotypic tests for resistance, which lately were supplemented with marker assisted selection (MAS) apply-ing molecular markers linked with some resistance genes.
