Opis:
Methylglyoxal is an endogenous electrophile produced in Escherichia coli by the enzyme methylglyoxal synthase to limit the accumulation of phosphorylated sugars. In enteric bacteria methylglyoxal is detoxified by the glutathione-dependent glyoxalase I/II system, by glyoxalase III, and by aldehyde reductase and alcohol dehydrogenase. Here we demonstrate that glyoxalase III is a stationary-phase enzyme. Its activity reached a maximum at the entry into the stationary phase and remained high for at least 20 h. An rpoS- mutant displayed normal glyoxalase I and II activities but was unable to induce glyoxalase III in stationary phase. It thus appears that glyoxalase III is regulated by rpoS and might be important for survival of non-growing E. coli cultures.

Opis:
A semi-empirical heterogeneous adsorption model for the accurate prediction of analyte retention in a column chromatography with binary mobile phase on polar bonded adsorbent surface was proposed. The performance of the proposed equation was compared with two retention models reported in the literature. All models were verified for different LC systems by means of four criteria: the sum of squared differences between the experimental and theoretical data, approximation of standard deviation, Fisher test and F-test ratio.

Opis:
Sound propagation from the vehicles moving on the city roundabout, with taking into account the wind is investigated. Solution of the problem for one moving sound source is found by means of the integral Fourier transforms extended over space variables and time. Inverse transforms are calculated approximately, using a stationary phase method and iterative technique. The solution for a general problem is obtained as a superposition of many partial solutions. The numerical analysis of noise characteristics is performed for the three-way Korfanty roundabout case in Łódz.

Opis:
Spontaneous mutations arise not only in exponentially growing bacteria but also in non-dividing or slowly dividing stationary-phase cells. In the latter case mutations are called adaptive or stationary-phase mutations. High spontaneous mutability has been observed in temperature sensitive Escherichia coli dnaQ49 strain deficient in 3'→5' proofreading activity assured by the ε subunit of the main replicative polymerase, Pol III. The aim of this study was to evaluate the effects of the dnaQ49 mutation and deletion of the umuDC operon encoding polymerase V (Pol V) on spontaneous mutagenesis in growing and stationary-phase E. coli cells. Using the argE3OC →Arg+ reversion system in the AB1157 strain, we found that the level of growth-dependent and stationary-phase Arg+ revertants was significantly increased in the dnaQ49 mutant at the non-permissive temperature of 37°C. At this temperature, in contrast to cultures grown at 28°C, SOS functions were dramatically increased. Deletion of the umuDC operon in the dnaQ49 strain led to a 10-fold decrease in the level of Arg+ revertants in cultures grown at 37°C and only to a 2-fold decrease in cultures grown at 28°C. Furthermore, in stationary-phase cultures Pol V influenced spontaneous mutagenesis to a much lesser extent than in growing cultures. Our results indicate that the level of Pol III desintegration, dependent on the temperature of incubation, is more critical for spontaneous mutagenesis in stationary-phase dnaQ49 cells than the presence or absence of Pol V.

The sound radiation from vehicles travelling on the city roads with T junction was considered. The wind effect on acoustic field was taken into account. The solution of this problem was found with the help of the integral Fourier transforms and stationary phase method as the superposition of solutions for the cases of vehicles moving along the straight roads and roads with right-angle bend. As an example, the numerical analysis of traffic noise characteristics was carried out for the T junction city road on one of streets in the town of Łódź (Poland).

Opis:
A two-dimensional Cauchy Poisson problem for water with a porous bottom generated by an axisymmetric initial surface disturbance is investigated here. The problem is formulated as an initial value problem for the velocity potential describing the motion in the fluid. The Laplace and Hankel transform techniques have been used in the mathematical analysis to obtain the form of the free surface in terms of a multiple infinite integral. This integral is then evaluated asymptotically by the method of stationary phase. The asymptotic form of the free surface is depicted graphically in a number of figures for different values of the porosity parameter and for different types of initial disturbances.
