Twenty apparently healthy buffaloes were withdrawn of feed and water for 48 hours. Buffaloes were administered with fluids and were subjected to endoscopy every 12 hours. Olympus™ [GIF V70] flexible video endoscope was passed through the ventral nasal meatus, the pharynx, oesophagus and then into the reticulo-rumen in physically restrained buffaloes. The entire reticulum and part of the rumen could be visualized, when the animals were off feed and water for at least 48 hours and evacuations of rumen contents were done even after 48 hours of starvation to visualize the rumen in six buffaloes. The reticulum appeared light brown to pink coloured with honeycomb shape and the rumen appeared smooth, shiny pink, with numerous papillae throughout its surface. The procedure was well tolerated by all the buffaloes and satisfactory reticular and ruminal images could be obtained including biopsy.

Opis:
The effects of feeding, fasting, and re-feeding on the ruminal profile of growing cattle were studied. Ruminal fluid and urine samples were obtained from 12 crossbred steers weighing approximately 300 kg during the following periods: 11 h of normal feeding (postprandial period), 48 consecutive hours of fasting, and followed by 48 h of re-feeding. Fasting promotes changes in the ruminal profile, such as an increase in ruminal pH, reduction in the number of rumen protozoa and bacteria, and decrease in the urinary excretion of allantoin; however, it does not change the urinary uric acid excretion rate. The overall mean ruminal pH was higher during fasting (7.53±0.27) in comparison to those at normal feeding (6.72±0.25) and re-feeding (6.62±0.31) (p<0.05). During re-feeding, the ruminal profile returned to normal, except for the protozoa count, which despite a slight increase only after 48 h of re-feeding, did not recover to baseline values.

Results of the earlier studies suggested an involvement of ciliates Eudiplodinium maggii in the digestion and metabolism of chitin in the rumen. In the presented paper we described the results on the preliminary identification and characterization of chitinolytic enzymes of this ciliate as well as the method of their purification. The protozoal crude enzyme preparation was used as source of enzymes, whereas the molecular filtration on Sephadex G-150 (single step method) or separation of protein by precipitation with ammonium sulfate followed by molecular filtration (two step method) were applied to purify the identified enzymes. The identification studies resulted in the detection of endochitinase, exochitinase and N-acetyl-D-glucosaminidase. The highest activity of identified enzymes were obtained in 4.0 - 4.5 pH and at 45 - 50 °C. Results of the comparative study on purification procedures showed that the single-step method enabled us to obtain enzymes of higher purity and higher activity than the two-step purification method.

The model is based on the idea of the Baker Transformation, but exhibits some different phenomena: the transformation does not mix points at all in some parts of the phase space (and under some conditions mixes them strongly in other parts), as observed in ruminant animals.

The addition of the inoculant can accelerate the fermentation process and improve the quality of the feed material in fermentation. This study aimed to isolate, test and select the rumen microorganisms that have high ability to degrade the fiber and improve the quality of materials used in inoculants to ferment agricultural wastes as alternative feed. Microbial isolate was extracted from cattle rumen fluid. The isolates type of fungus were grown on Potato Dextrose Agar (PDA) while the bacteria was grown on Nutrient Agar (NA) medium. Microbes of fungi were tested and selected based on their ability to produce ligninase, cellulase and hemicellulase enzymes. Bacteria were grown on MRS broth media as a confirmation for LAB colonies, then tested for gram staining, catalase test, growth on different NaCl levels (4 and 6.5%) and growth at various pH (3, 4, and 5). The results showed that five fungal isolates had a growth diameter of 0.1–0.37 mm on lignin, cellulose and hemicellulose media after seven days. Eleven bacterial isolates were indicated as lactic acid bacteria; almost all isolates could grow on NaCl 6.5%, but only four bacterial isolates could grow on NaCl 4% and five bacterial isolates could grow at pH 3. In conclusion, three isolates of fungi have a remarkable ability to degrade lignin. Five isolates of lactic acid bacteria have a comprehensive ability as a preservative in fermentation, so eight microbial isolates from rumen fluid can be used as inoculants in fermenting the feed material from agro-industrial waste.

Adequate nutrition of ruminants and physiologically correct assessment of the nutritional value of feed rations need additional knowledge about the quantitative transformation of the main components of any particular feed in diff erent parts of the digestive tract. The study of the principles of substrate energy use is necessary in each particular case to defi ne the actual need in substrates, which determines the effi ciency of their use for implementing biosynthesis and physiological functions. The determination of the infl uence of the energy supply of the diet of young cattle aged 7-12 months on the effi ciency of energy use in the organism. Effi ciency of the use of metabolizable energy of diets for calves with the increased level of energy nutritional value by 5.0% (MEC is equal to 9.6 MJ/kg) promoted the increase of energy deposition and weight gain synthesis by 10.04%, weight gain energy by 19.50%, the effi ciency of the use of metabolizable energy for weight gain by 3.81%.
