Opis:
For several decades an increase of platinum in the environment was recorded. This is connected with the release of platinum from automobile catalytic converters and pharmaceutical industries. In the conducted experiments the effect of various forms and concentrations of platinum on the germination and roots growth of different plants (oat, garden cress and tomato) was investigated. Two separate experiments were performed with lower (experiment 1st: 1, 5 and 10 mg/L) and higher (experiment 2nd: 10, 20 and 40 mg/L) [Pt(NH3)4](NO3)2 or PtCl4 dosages. There was no significant effect of dose and form of platinum on the germination of seeds (experiment 1st and 2nd). The effect of platinum containing substances dose on root growth of young seedlings was observed during experiment, dependent on the species of tested plants. The root length of oat plants was similar in all treatments in experiment 1st. The shortest tomato roots were observed in the case of a 10 mg/L of PtCl4, it was statistically shorter than the control plants (treated with water) as well. The roots of the tomato treated with [Pt(NH3)4](NO3)2 were the same length as observed for the control sample. The garden cress root growth was not affected by [Pt(NH3)4](NO3)2, but a solution containing Pt4+ stimulated the root growth. The roots of tomato and oat treated with [Pt(NH3)4](NO3)2 had similar length as the control plants in contrast to the far shorter roots treated with PtCl4 (experiment 2nd). Moreover, the treatment with [Pt(NH3)4](NO3)2 stimulated the growth of garden cress roots compared to the control, while only the lowest dose of PtCl4 stimulated the root growth.

Opis:
The root system architecture (RSA) displays complex morphological characteristics because of diverse root growth behaviors. Recent studies have revealed that swarming behavior among roots is particularly important for RSA to adapt to environmental stimuli. However, few models are proposed to simulate RSA based on swarming behavior of roots. To analyze plasticity of RSA affected by swarming behavior, we propose viewing it as a swarm of single roots. A swarming behavior model is proposed by considering repulsion, alignment, and preference of individual single roots. Then, the swarming behavior model is integrated into a simple and generic RSA model (called ArchiSimple). Lastly, characteristics of RSA affected by swarming behavior model and non-swarming behavior model are compared and analyzed under three different virtual soil sets. The characteristics of RSA (such as primary root length, lateral root length, lateral roots, and resource uptake) are significantly promoted by swarming behavior. Root system distributions can also be greatly affected by swarming behavior. These results show that root foraging and exploration in soil can be regarded as collective behavior of individual single root.

Opis:
The aim of this study was to investigate the phytotoxic effects of seven metal ions (Cd(II), Cr(VI), Cu(II), Hg(II), Ni(II), Pb(II) and Zn(II)) on length of roots of five rapeseed (Brassica napus L. subsp. napus) cultivars registered in Slovakia (Atlantic, Baldur, Californium, Oponent and Verona). The phytotoxic effect of metals was evaluated using IC50 values. The studied metal ions inhibited germination and root growth of rapeseed seedlings. In general, the toxicity of metal ions decreased in the following order Cu > Cr >Hg > Cd > Pb > Ni > Zn. Atlantic, Baldur and Californium were more sensitive to Cd than to Ni, for Oponent and Verona higher toxicity exhibited Ni. From the studied rapeseed cultivars Atlantic and Californium were found to be most sensitive to tested metals. On the other hand, high tolerance to metal treatment was determined for Baldur. Czech cultivar Opponent showed high tolerance to Cd, Cr, Cu and Pb, but it was sensitive to Hg and Ni. The above-mentioned results confirmed differences in the metal tolerance of tested rapeseed cultivars.

Opis:
The growth and saponin accumulation were measured in two lines of transgenic hairy roots of Platycodon grandiflorum, Pl 6 and Pl 17, cultured for 8 weeks in 250-ml shake flasks containing 50 ml of hormone-free woody plant medium supplemented with 40 g/l sucrose and in the Pl 17 line cultured for 12 weeks in a 5-l mist bioreactor containing 1.5 l of the same medium. With both methods, the growth of transgenic hairy roots was assessed as both fresh and dry weight and the biomass growth was correlated with the conductivity and sucrose uptake. The accumulation of saponins was measured and compared with that in roots derived from the field cultivation. The saponin concentrations were significantly higher in the two hairy root lines cultured in shake flasks [6.92 g/100 g d.w. (g%) and 5.82 g% in Pl 6 and Pl 17, respectively] and the line cultured in the bioreactor (5.93 g%) than in the roots derived from the field cultivation (4.02 g%). The results suggest that cultures of P. grandiflorum hairy roots may be a valuable source for obtaining saponins.

Opis:
Effects of 62 isolates (of 41 taxa) of fungi on growth of 1-year-old oak (Quercus robur) seedlings were studied in an inoculation experiment. The fungi were isolated from roots of 80–96-year-old Q. robur that had been subjected to periodic flooding and had symptoms of oak decline. The fungal genera included Alternaria, Aspergillus, Calonectria, Chaetomium, Cladosporium, Clonostachys, Corynespora, Cylindrocarpon, Dicyma, Geotrichum, Ilyonectria, Isaria, Metarhizium, Oidiodendron, Ophiostoma, Pezicula, Phialocephala, Phialophora, Pyrenochaeta, Sporendocladia, Sporothrix, Thelonectria, Trichoderma and Trimmatostroma. Mycelial colonies of fungi growing in potato-dextrose broth were used for soil inoculation. Plant growth was assessed 2 years after inoculation, when the plants were 3 years old. Stem lengths, and dry weights of stems, roots and leaves were measured. Stem growth was inhibited by 31 isolates (50%) and root growth by 12 isolates (19%). Stem growth was stimulated by two isolates (3%) and root growth by 17 isolates (27%). The overall ratio of inhibitors to stimulants was 2.1. The proportion of taxa that inhibited stem growth was 16 times greater than that which promoted stem growth. The proportion of taxa that promoted root growth was only 1.5 times greater than that which inhibited root growth. The structure of the fungal communities in periodically flooded oak forests suggests that they are more likely to inhibit than to promote vigour in oaks.

While each individual root responds to such environmental factors as light or gravity the question arises how these reactions subordinate to the whole root system, which is supposed to maintain its primary functions. Data presented here confirm that in cucumber the gravity and light modulate the growth direction of the lateral roots subunits of the system. Another important factor affecting behavior of lateral roots is an orientation of the main root. These facts all together suggest that the root system functions as an integrated entity, capable of adapting its architecture to changing environmental conditions. Its flexibility, based on unknown signaling network, guarantees optimal functioning of the system.

Opis:
Prepenetration and penetration phenomena after inoculation of roots of Pinus sylvestris seedlings grown in vitro with the P-, S- and F-group isolates of Heterobasidion annosum were observed using scanning electron microscop. There were no differences in the behaviour of hyphae and in the appearence of mycelia formed by the three IS-group isolates. Four types of the root penetration by hyphae were observed. In the first, the entrace of hypha into root was achieved throught tiny pore formed in walls of the cortical cells. In the second, swellings resembling appressoria were formed by the hyphal tips at the points of contact with the root surface. In the third, the hyphae penetrated cells through holes in eroded areas of roots, and in the fourth by the natural crevices at points of the cortical cell junctions. It is concluded that the different frequencies of the four types of penetration observed after inoculation with isolates of the three IS group should be carefully explained in term of their different pathogenic and saprobic capabilities on pine.
