Opis:
IntroductionAging stands for the changes in the human body, which are recognized as natural processes that concern every person and are related to the passage of time. Despite ‘normalness’ of the process, a tendency is emerging to treat aging as a pathology requiring proper diagnostics. Consequently, an ‘anti-aging’ medicine has been established - defined as study of preventing and slowing down the process of aging.Aim of the studyThe aim of this study is to summarize and present the current knowledge, based on the overview of writing on the properties and effectiveness of the use of PRP in anti-aging therapy.  Description of knowledge Autologous platelet-rich plasma is a concentrate of human blood platelets suspended in a small amount of plasma, wherein concentration of thrombocytes is higher than initial concentration. For autologous plasma to have high healing properties, platelet levels should be at least 1 million in 5 ml of plasma. The basic condition for initiating repair and regenerative processes in vivo is the formation of a blood clot. Also, by degranulating, platelets release many growth factors, such as PDGF, TGF-β, EGF, FGF, VEGF and others, which are involved not only in blood coagulation, but also in immune response and tissue healing. The large variety of PRP-obtaining methods and the range of concentration levels of growth factors in PRP may lead to ambiguity in the literature, as to the extent to which formulation was actually used in a variety of preclinical and clinical studies. Some of those studies are presented in this article. Summary Platelet-rich treatments are part of the growing popularity of regenerative medicine, which is beginning to be recognized as the medicine of the future. However, without proper control methods or optimization of numerous variables (e.g. concentration or ratio of growth factors) during PRP preparation, it is not known which ingredients are needed and preferred, and which are harmful for tissue regeneration. However, numerous studies show PRP injection as a promising method of rejuvenating the skin, therefore platelet-rich treatments are an important part of the growing popularity of regenerative medicine.

chronic lower-leg venous ulcer
autologous platelet-rich plasma
Opis:
Wound healing is a complex pathophysiological process, in which platelets play a crucial role. Platelet alpha-granules release growth factors to the wound bed; the factors are necessary in the healing process. In chronic wounds, such as poorly healing lower-leg ulcers of venous origin, there is decreased activity of multiple growth factors, so the concept of exogenous delivery of such factors seems a logical strategy. Platelet-rich plasma therapy in patients with lower-leg ulcers of venous origin combined with conventional treatment methods (previously ineffective in these patients) seems, based on our observation, an important adjunct leading to recovery. The aim of the study was to present an original method of autologous platelet-rich plasma application through the creation of a sort of “biological chamber” containing a concentrate of growth factors. Material and methods. The described therapy was implemented in 10 patients, who had been ineffectively treated for more than one year in the outpatient setting. Patients with exacerbation of inflammatory process, signs of wound infection and ankle brachial pressure index < 0.8 were excluded from the study. After the application of platelet-rich plasma, further treatment was continued with the use of moist therapy and compression therapy according to a uniform regimen. Results. Complete healing was achieved within 4-10 weeks from the beginning of the product administration in all patients. Conclusion. The presented method seems technically simple, effective and relatively inexpensive

Opis:
Platelet-rich plasma (PRP) is being increasingly used in the modern medicine as a material stimulating, regenerating and accelerating tissue healing, including muscle injuries in sports. The aim of the present study was to determine the effect of PRP on strength recovery in exercise induced muscle damage (EIMD). A baseline value was established in six healthy male volunteers during maximal voluntary strength isometric contractions (MVC) of the elbow flexors in both arms. This was followed by MVC performance (80% IRM) concentric-eccentric exercise of both elbow flexors until point of exhaustion, and 24 h later the subjects were injected with PRP in the non-dominant arm (PRP-arm). Maximal isometric strength and muscle soreness was compared before and at 24, 48, 72 and 96 h after delayed onset muscle soreness (DOMS) between the arms. The results indicate that the muscle soreness was significantly decreased in PRP administered arm when compared to control-arm, however, the levels for isometric flexors peak torque (PT) and average strength (AS) significantly declined in the bout arm post exercise induced muscle damage. Administration of PRP had no effect on the muscle strength parameters, however, flexor PT values were higher in PRP administered arm compared to the control arm, but this was only achieved on the second day of post-exercise induced muscle damage.

Opis:
The aim of this study was to determine the activity of proacrosin and acrosin in spermatozoa originating from the sperm-rich fractions (SRF) and whole ejaculates (WE) of dog semen. In addition, experiments were conducted to determine the activity of antitrypsin inhibitors in the fluids of different ejaculate fractions and whole seminal plasma. Ejaculates were collected from five dogs of mixed breed and one Beagle dog (aged from 2 to 9 years). In the SRF, it was confirmed that the activity of the free acrosin form was predominant (acrosin / proacrosin; 2.38 ± 0.22 / 1.05 ± 0.08 mIU / 106 spermatozoa). On the other hand, spermatozoa originating from the WE exhibited significantly higher (p<0.05) proacrosin activity (proacrosin / acrosin; 2.19 ± 0.19 /1.30 ± 0.11 mIU / 106 spermatozoa). Furthermore, acrosin inhibitor activity was lower in the fluids of the pre-sperm fraction (0.09 ± 0.006 IU / cm3), whereas it was higher in the fluids of the post-sperm fraction (0.11 ± 0.007 IU / cm3). Using PAGE analysis, the antitrypsin activity of the enzyme was represented by the presence of one electrophoretic band in the fluids of the pre-sperm and post-sperm fractions and whole seminal plasma. Furthermore, two electrophoretic bands were detected in the fluids of the SRF. The findings of this study indicate that specific proteinase inhibitors present in the individual ejaculate fractions of dog semen may act by stabilizing the sperm acrosin system.
