Prokaryotic organisms are exposed in the course of evolution to various impacts, resulting often in drastic changes of their genome size. Depending on circumstances, the same lineage may diverge into species having substantially reduced genomes, or such whose genomes have undergone considerable enlargement. Genome reduction is a consequence of obligate intracellular lifestyle rendering numerous genes expendable. Another consequence of intracellular lifestyle is reduction of effective population size and limited possibility of gene acquirement via lateral transfer. This causes a state of relaxed selection resulting in accumulation of mildly deleterious mutations that can not be corrected by recombination with the wild type copy. Thus, gene loss is usually irreversible. Additionally, constant environment of the eukaryotic cell renders that some bacterial genes involved in DNA repair are expandable. The loss of these genes is a probable cause of mutational bias resulting in a high A+T content. While causes of genome reduction are rather indisputable, those resulting in genome expansion seem to be less obvious. Presumably, the genome enlargement is an indirect consequence of adaptation to changing environmental conditions and requires the acquisition and integration of numerous genes. It seems that the need for a great number of capabilities is common among soil bacteria irrespective of their phylogenetic relationship. However, this would not be possible if soil bacteria lacked indigenous abilities to exchange and accumulate genetic information. The latter are considerably facilitated when housekeeping genes are physically separated from adaptive loci which are useful only in certain circumstances.

The field experiment (microplots) was carried out in 2005-2007 in the experimental site belonging to the University of Podlasie in Siedlce. Nitrogen 15N at 10.3 at. % enrichment was applied in a form of (15NH4)2S04 at the amount of 1.66g per 1 m2 in early spring. In parallel to goats rue (Galega orientalis Lam.) cultivation, also plant that did not show the ability of biological N2 reduction (spring barley (Hordeum sativum) was grown, and it was also fertilized with 15N in the form of (15NH4)2SC>4 at 10.3 at. % enrichment. The quantitative abilities of biological nitrogen reduction for Rhizobium galegae cultures living together by goat's rue (Galega orientalis Lam.) was determined by means of isotope dilution method. The abundance of at. % 15N was determined by spectroscopic method on the spectrophotometer NOI - 6E, then the amount of nitrogen transferred from the air due to biological N2 reduction process, was calculated. The yield of as a sum from three cuts of dry matter of tested plant in subseąuent study years was [kg o m-2]: 2005 - 1.092; 2006 - 0.831; 2007 - 0.509. The quantity of biologically reduced nitrogen reached up the mean value for three experimental years at the level of 28.863 g N o m2 but in each year as follows [g N o m-2]: 2005 - 37.603; 2006 - 26.080; 2007 - 22.906 during the whole vegetation period.
Doświadczenie polowe mikropoletkowe przeprowadzono w latach 2005-2007 na polu należącym do Akademii Podlaskiej w Siedlcach. Azot 15N o wzbogaceniu 10,3 at. % stosowano w formie 15(NH4)2SO4 w ilości 1,66 g na 1 m2 wczesną wiosną. Równolegle z uprawą rutwicy wschodniej (Galega orientalis Lam.) uprawiano roślinę niemającą zdolności biologicznej redukcji N2 (jęczmień jary - Hordeum sativum), którą również nawożono 15N w formie 15(NH4)S04 o wzbogaceniu 10,3 at. %. Ilościowe możliwości biologicznej redukcji azotu przez kultury bakterii Rhizobiimt galegae współżyjące z rutwicą wschodnią (Galega orientalis Lam.) określono po zastosowaniu metody izotopowego rozcieńczenia. Na spektrofotometrze NOI - 6E oznaczono at. % 15N, a następnie obliczono ilość azotu pochodzącego z powietrza w wyniku biologicznej redukcji N2. Sumaryczny plon suchej masy rośliny testowej w kolejnych latach badań wynosił [kg o m-2]: 2005 - 1,092; 2006 - 0,831; 2007 - 0,509. Ilość biologicznie zredukowanego azotu osiągnęła średnią wartość z trzech lat badań na poziomie 28,863 g N o m-2, a w kolejnych latach kształtowała się następująco [g N o m-2]: 2005 - 37,603; 2006 - 26,080; 2007 - 22,906 w ciągu okresu wegetacyjnego.

Due to inadequate efforts to reinforce nitrogen fixation capability of bean via symbiosis with rhizobia, improvement of bean productivity is still highly dependent on chemical fer- tilization. An advanced understanding of agro-ecosystem-bean-Rhizobium interaction is required to improve symbiosis efficiency. Thus, seasonal development of rhizobial nodu- lation was characterized according to 20 agro-ecological properties for 122 commercial bean fields. Principal component analysis identified soil texture as a major descriptor of agrosystem-bean-disease-Rhizobium interaction. Nonparametric correlation analysis indi- cated significant associations of root nodulation with bean class, fungicidal treatment of seed and soil, Fusarium root rot index, planting date and depth, soil texture, clay and sand content. Ordinal regression analysis demonstrated that rhizobial nodulation was improved by applying initial drought, heavier soil textures with greater organic matter and neutral pH, using herbicides and manure, growing white beans, irrigating every 7–9 days, later sowing in June, reducing disease and weed, shallower seeding, sowing beans after alfalfa, avoiding fungicidal treatment of seed and soil, and omitting urea application. This large- scale study provided novel information on a comprehensive number of agronomic prac- tices as potential tools for improving bean-Rhizobium symbiosis for sustainable legume production systems.

The aim of the study was to determine the health status of seeds and the quality of seedlings of pea of Tarchalska variety after the use of Nitragina vaccine and a non-commercial bacterial vaccine IUNG. It was observed that in 2010 treatment with vaccines reduced the amount of seeds with pathogenic changes. Fungi belonging to 11 species were isolated from the pea seeds. Alternaria alternata species and Penicillium genus were the most abundant.
Celem pracy było określenie zdrowotności nasion oraz jakości siewek grochu siewnego odmiany Tarchalska po zastosowaniu szczepionki Nitragina oraz niekomercyjnej szczepionki bakteryjnej IUNG. Obserwowano, że w 2010 r. zaprawianie szczepionkami wpłynęło na zmniejszenie ilości nasion ze zmianami patogennymi. Z nasion grochu wyizolowano grzyby należące do 11 gatunków. Najliczniej był reprezentowany gatunek Alternaria alternata oraz rodzaj Penicillium.

The establishment of the nitrogen-fixing symbiosis between rhizobia and legumes requires an exchange of signals between the two partners. In response to flavonoids excreted by the host plant, rhizobia synthesize Nod factors (NFs) which elicit, at very low concentrations and in a specific manner, various symbiotic responses on the roots of the legume hosts. NFs from several rhizobial species have been characterized. They all are lipo-chitooligosaccharides, consisting of a backbone of generally four or five glucosamine residues N-acylated at the non-reducing end, and carrying various O-substituents. The N-acyl chain and the other substituents are important determinants of the rhizobial host specificity. A number of nodulation genes which specify the synthesis of NFs have been identified. All rhizobia, in spite of their diversity, possess conserved nodABC genes responsible for the synthesis of the N-acylated oligosaccharide core of NFs, which suggests that these genes are of a monophyletic origin. Other genes, the host specific nod genes, specify the substitutions of NFs. The central role of NFs and nod genes in the Rhizobium-legume symbiosis suggests that these factors could be used as molecular markers to study the evolution of this symbiosis. We have studied a number of NFs which are N-acylated by α,β-unsaturated fatty acids. We found that the ability to synthesize such NFs does not correlate with taxonomic position of the rhizobia. However, all rhizobia that produce NFs such nodulate plants belonging to related tribes of legumes, the Trifolieae, Vicieae, and Galegeae, all of them being members of the so-called galegoid group. This suggests that the ability to recognize the NFs with α,β-unsaturated fatty acids is limited to this group of legumes, and thus might have appeared only once in the course of legume evolution, in the galegoid phylum.

The aim of this work was to examine the effects of A. chroococcum on the prolifera-tion of R. leguminosarum bv. viciae (Rlv) in a solid-carrier inoculant and on symbiotic effectiveness of Rlv with pea plants grown under laboratory and field conditions. In a laboratory experiment it was found that proliferation of both bacterial species, Rlv and A. chroococcum, in the dual-culture inoculants was efficient, and that A. chroococ-cum had no adverse effects on the development of the rhizobia (Rlv) in the solid-carrier inoculant. In a pot experiment the highest number of nodules was detected on roots of pea plants inoculated with the dual-culture inoculant containing Rlv and A. chroococcum, slightly lower numbers on pea roots inoculated with the mono-culture inoculum of Rlv and almost no nodules were found on the roots of pea un-inoculated (control treatment) with the bacteria. In the micro-plot experiment conducted in the years 2011–2012 pre-sowing inoculation of pea seeds with the mono-culture inocu-lant of Rlv or with the mixed inoculant of Rlv and A. chroococcum slightly increased nodule numbers/plant, pod numbers/plant and seed numbers/pod, as compared to the un-inoculated control, but these differences were not reflected in pea seed yields/m2, which were similar in all treatments.

Opis:
Chemical composition of lipopolysaccharide (LPS) isolated from an effective (97) and ineffective (87) strains of R. l. viciae has been determined. LPS preparations from the two strains contained: glucose, galactose, mannose, fucose, arabinose, heptose, glucosamine, galactosamine, quinovosamine, and 3-N-methyl-3,6-dideoxyhexose, as well as glucuronic, galacturonic and 3-deoxyoctulosonic acid. The following fatty acids were identified: 3-OH 14:0, 3-OH 15:0, 3-OH 16:0, 3-OH 18:0 and 27-OH 28:0. The ratio of 3-OH 14:0 to other major fatty acids  in LPS 87 was higher that in LPS 97. SDS/PAGE profiles of LPS indicated that, in lipopolysaccharides, relative content of S form LPS I to that of lower molecular mass (LPS II) was much higher in the effective strain 97 than in 87. All types of polysaccharides exo-, capsular-, lipo, (EPS, CPS, LPS, respectively) examined possessed the ability to bind faba bean lectin. The degree of affinity of the host lectin to LPS 87 was half that to LPS 97. Fatty acids (FA) composition from bacteroids and peribacteroid membrane (PBM) was determined. Palmitic, stearic and hexadecenoic acids were common components found in both strains. There was a high content of unsaturated fatty acids in bacteroids as well as in PBM lipids. The unsaturation index in the PBM formed by strain 87 was lower than in the case of strain 97. Higher ratio of 16:0 to 18:1 fatty acids was characteristic for PMB of the ineffective strain.

There has been an increasing interest in soybean cultivation in hydroponic systems. Because soybean plants can utilize nitrogen derived from biological N2 fixation, the use of Rhizobium inoculated plants may eliminate or decrease the need for mineral nitrogen fertilization in hydroponic systems. Thus, the objective of this study was to establish effective methods for making inoculated soybean transplants for a substrate-based hydroponic system. Inoculated plants were effectively produced by rooting seedling stem cuttings in a medium containing a Rhizobium inoculant. We also investigated the effects of different nitrogen forms and subirrigated nutrient solution concentrations on the growth and root nodule formation of the inoculated plants. The application of a nitrogen-free nutrient solution had minimal positive effects on the growth and nodulation of the inoculated plants. Ammonium-fed plants lacked root nodules regardless of the ammonium concentration. Furthermore, a 50% nutrient solution combining with nitrate or urea as the nitrogen source did not promote root nodulation. Therefore, inoculated plants should be subirrigated with an approximately 25% nutrient solution supplemented with nitrate or urea to induce early growth and nodulation prior to transplantation.

The extracellular polymeric substance (EPS) produced from Rhizobium radiobacter F2, designated as EPSF2 , was investigated as a biosorbent for the removal of Pb(II) and Zn(II) from aqueous solution. The optimum biosorption pH values were 5.0 for Pb(II) and 6.0 for Zn(II). Kinetics study revealed that the biosorption followed pseudo-first-order model well, and the equilibrium data fit the Langmuir model better. The adsorbed metal ions could be effectively desorbed by HCl. Desrobed EPSF2 regained 80% of the initial biosorption capacity after five cycles of biosorption-desorption-elution. These results demonstrated that EPSF2 could be a promising alternative for Pb(II) and Zn(II) removal from aqueous solution.

Six rhizobium (Rhizobium leguminosarum bv. Trifolii TA1, Sinorhizobium meliloti 1021, Mesorhizobium huakuii IFO 15243T, Ochrobactrum lupini LUP 21T, Bradyrhizobium japonicum USDA110 and B. elkanii USDA 76) and two Escherichia coli strains (E. coli ATCC 25922 and E. coli HB 101) were compared in respect to polymyxin B and EDTA resistance, as well as bacterial outer membrane (OM) permeability to a fluorescent hydrophobic agent (N-phenyl-1-naphthylamine - NPN). TEM (Transmission Electron Microscopy) and a microbial test demonstrated that all the rhizobia were much more resistant to polymyxin B in comparison with E. coli strains. EDTA and polymyxin B enhance permeability of B. japonicum and O. lupini OM. Other rhizobia incorporated NPN independently of the presence of membrane-deteriorating agents; however, the level of fluorescence (measured as NPN absorption) was strain dependent.

The paper presents results on the effect of agronomic factors on the content of chlorophyll, the biometric parameters of plants and quality of fenugreek seeds. It has been shown that potassium fertilization and inoculation of fenugreek seeds had no significant effect on the plant’s performance. The only trait influenced by those factors was the seed weight per plant; under higher doses of potassium fertilization K2.5 and K3.75, its value was about 10% higher, but fell down by 19.3% in treatments with inoculated seeds compared to non-inoculated ones. Soil water deficit caused reduction of plant height by 15.5%, number of pods by 18.3%, number of seeds per pod by 20%, weight of seeds by 28%, weight of the plant’s aerial parts by 18.0%, and harvest index by 13.2%. In response to water deficit, the content of chlorophyll decreased significantly from 57.8 SPAD units at 43 DAS to 52.4 and 54.0 SPAD units in the next phases (51 and 58 DAS), when 40% and 80% of pods attained the final length. Increasing doses of potassium caused a significant increase in the crude protein (by 3.2-5.4%) and potassium content (by 7-8%) in seeds of fenugreek grown under water deficit. After seed inoculation, an increase in the protein content was found.

Dependence on chemical fertilizers for future agricultural growth would mean further loss in soil quality. The possibilities of water contamination and unsustainable burden of the fiscal system. The Government of India has been trying to promote an improved practice involving use of biofertilizers along with fertilizers. These inputs have multiple beneficial impacts on the soil and can be relatively cheap and convenient for use. The agricultural chemicals like pesticide and weedicides were being used to save the standing crop from the attack of pests and to boost crop production. At the same time the excess use of chemical fertilizers degraded the soil properties. In this context, the farmers are expecting for the cheapest and eco-friendly alternative for getting higher yield. Biofertilizers is only the alternative way to get more production without harmful to environment. So the effect of different doses of biofertilizer (Rhizobium) on germination studies of Fenu-greek seed germination percentage, seedling growth, fresh weight and dry weight, chlorophyll, protein and soluble sugar content has been studied in the laboratory condition. There parameters increased to application when biofertilizer compared to control.

Nitrogen fixing bacteria have been used for centuries to improve the fertility of agricultural soils. Since the introduction of inorganic nitrogen (N) fertiliser that provides a reliable boost to crop yields whilst reducing land and labour requirements, the use of biological nitrogen fixation has been in decline. Recently, concerns have been expressed about the sustainability of inorganic N fertiliser application, however, there remain doubts about whether N2 fixing bacteria alone can provide agriculture with sufficient fixed N to feed a burgeoning global population. In this paper we review the current state of our knowledge regarding those diazotrophic bacteria that have a role to play in agriculture. We focus on our current areas of research, particularly, the importance of understanding the classification and mechanism of action of N2 fixing bacteria that are used in agricultural soils. We discuss the applications of N2 fixing bacteria that illustrate their potential to provide sustainable N, particularly focussing on Australian and South American agricultural systems where these bacteria are widely exploited to maintain soil fertility. We also identify problems with the use of bacteria as inoculants, including ineffective inoculation due to poor quality preparation, the use of appropriate isolates and issues with sustainability. We review the outlook for biological N fixation highlighting how molecular biology may enable the expression of N fixation in non-leguminous crops.

Opis:
During nodule development on pea roots, apoplast undergoes changes in activity of plant cell wall proteins such as expansins (EXPs). Because the accumulation of EXP protein has been correlated with the growth of various plant organs, we investigated using Western Blot and immunolocalization studies with antibody against PsEXP1, whether this protein was accumulated in the expanding cells of nodule. Immunoblot results indicated the presence of a 30-kDa band specific for pea root nodules. The EXP proteins content rose during growth of pea root nodules. Expansin(s) protein was localized in nodule apoplast as well as in the infection thread walls. The enhanced amount of expansin-like proteins in meristematic part of nodule, root and shoot was shown. The localization of this protein in the meristematic cell walls can be related to the loosening of plant cell wall before cell enlargement. Both, plant cell enlargement and infection thread growth require activity of expansin(s). Possible involvement of EXPs in the process of pea root nodule development is also discussed.
