This paper presents the theoretical study about carbon nanotube substrates for tissue engineering and its applications. Because the replacement of bone tissue with artificial tissue can violate the remodeling process completely, the artificial material should not only consist of the same material properties, but also exhibit other characteristics which are equally important and need to be taken into consideration. These are above all the mechanosensation. Besides replacing natural tissue, the nanostructured scaffolds presented in the paper can help the tissue growth by stimulating this process. The developed trabecular bone remodeling simulation method responsible for the nanostructured scaffold behavior is implemented here. Thus, the nanostructured bone-like scaffolds reflect the remodeling capability of the biological system, not only due to their application as replacement of natural tissue, but also due to their effects in the field of mechanosensation.

Inflammation plays an important role in the development of many obesity-related diseases. This study aimed to investigate the effect of ezetimibe on inflammation and myocardial remodeling in obese rats. A rat model of obesity was established, and myocardial damage was examined by transmission electron microscopy and Masson staining. Twenty obese rats were divided into two groups (n=10): obese group and ezetimibe group. Ten SD rats were used as controls. Western blot was performed to monitor the expression of P-p38MAPK and interleukin (IL)-6. Immunohistochemical staining was used to monitor the expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1. In the obese rats group, we observed increased inflammatory factors and myocardial hypertrophy. In contrast, the ezetimibe group exhibited decreased expression of inflammatory factors and an improvement in myocardial remodeling compared to the obese group. Mechanistically, we found that ezetimibe decreased P-p38MAPK, IL-6, intercellular adhesion molecule-1, and vascular cell adhesion molecule-1 levels in the hearts of the obese rats. Taken together, these results indicate that ezetimibe may improve myocardial remodeling in obese rats by inhibiting inflammation.

CRS is a process involving a number of adverse changes in the mucosa of the paranasal sinuses and nasal polyps, e.g. increased fibroblast proliferation, angiogenesis, increased formation of fibrous tissue (subepithelial fibrosis) and tissue destruction. There are biomarkers whose levels can be increased in chronic inflammation of the paranasal sinuses: peripheral blood eosinophilia, IgE immunoglobulin, cytokines – IL-4, IL-5, IL-13, IL-25, IL-33, periostin, P-glycoprotein, CXCL-12, CXCL-13, INF-Υ, TNFα, TGFβ1, albumins, eotaxin. These biomarkers are not pathognomonic for CRS. The concentration of biomarkers is also increased in bronchial asthma and atopic dermatitis. The TGFβ, in particular the β1 subunit, was identified as the main factor involved in the remodelling of tissue stroma. In conjunction with continuous improvement of tissue testing methods, it is advisable to search for new factors that will more accurately allow the assessment of tissue remodelling in the chronic processes of paranasal sinuses.

Opis:
Physical effort can elicit differential adaptive changes in the tissues of trained versus untrained rats. Proteolytic activity in the extracellular matrix could be engaged in such adaptation due to its influence on the elasticity of tissues. The effects were investigated of a single physical effort on the activity of elastase, cathepsin K, and plasmin in the skeletal muscles, heart muscles, and aortas of untrained (UT, n=30) and trained (T, n=30) rats. T rats underwent 6 weeks of endurance training. After the last training session, T and UT rats were divided randomly into 3 subgroups. Ten rats from each group (Tpre, n=10) and (UTpre, n=10) were sacrificed. The other 20 rats from each group performed 60 min. of aerobic exercise and were sacrificed immediately post exercise (T0h, n=10; UT0h, n=10) or 3h later (T3h, n=10; UT3h, n=10). Enzyme activity was measured fluorometrically. Cathepsin K and plasmin activity increased in the soleus muscles of UT0h versus UTpre, plasmin activity increased also in UT3h versus UTpre. Elastase, cathepsin K and plasmin activity increased in the heart muscles of T0h and T3h versus Tpre. No aortic differences were observed. Thus, a single bout of physical effort elicited different responses in tissues of T versus UT rats. Increased proteolytic enzyme levels in muscles could influence tissue remodeling. Unchanged aortic cathepsin K levels may help prevent aortic remodeling and neointima formation.

Morphogenetic theories investigate the mechanisms of creation and regulation of definite biological forms in living organisms. The incredible diversity of shapes and sizes is generated through a barely unknown coordination of biochemical processes occurring at molecular levels. Such a crosstalk not only defines the rules of a robust scheme of matter differentiation, but it also has the capacity to adapt with respect to some variations of the environmental conditions. In this work, we propose a continuum model of growth and mass transport for biological materials during morphogenetic processes. Using the theory of configurational forces, we define the thermomechanical bases for understanding how both the mechanical and the biochemical states can orchestrate growth. The model is successfully applied to describe the morphogen-driven growth control in the imaginal wing disc of Drosophila melanogaster.

The functional adaptation of bone is a process of bone tissue remodeling induced by variable in time mechanical demands that the skeleton has to satisfy. It is a very complex but highly organized process composed of events at micro-level (molecular and cellular) but having effects in macro-scale (variation of bone internal structure and external shape). Mathematical models of this phenomenon proposed in the literature represent formulas postulated on the basis of the results of medical observations or laboratory investigations and describe locally the evolution of a material in space and time. In the present paper a use is made of the hypothesis of optimal response of bone, proposed earlier by the author, what enables derivation (instead of postulation) the remodeling rules from a very general and global assumption. It turns out that such a formulation has many similarities to engineering optimal control problems. The link between the postulated local adaptation rules and those derived from the global assumption is also discussed.
Funkcjonalna adaptacja kości jest procesem polegającym na przebudowie tkanki kostnej wywołanej zmieniającymi się w czasie wymaganiami mechanicznymi, jakie musi spełniać szkielet kostny. Proces ten jest niezwykle złożony, ale doskonale zorganizowany i składa się z szeregu zjawisk zachodzących na poziomie mikro (molekularnym i komórkowym) lecz mających efekt na poziomie makro (zmiana zewnętrznego kształtu kości oraz jej struktury wewnętrznej). Matematyczne modele tego zjawiska, postulowane w oparciu o obserwacje medyczne i badania laboratoryjne, opisują lokalną ewolucję materiału w czasie i przestrzeni. W tej pracy zastosowano hipotezę optymalnej odpowiedzi kości zaproponowaną wcześniej przez autora w celu wyprowadzenia (zamiast postulowania) związków rządzących przebudową kości w oparciu o bardzo ogólne założenia. Okazuje się, że takie sformułowanie ma wiele wspólnego z zagadnieniami optymalnego sterowania. W pracy zaprezentowano przykład zastosowania omawianego podejścia oraz przeprowadzono krótką dyskusję na temat związków między postulowanymi modelami i wyprowadzonymi w oparciu o przyjętą hipotezę.

Continuous bone remodeling consists in simultaneous resorption of tissues and synthesis of a new matrix. If, due to variable external or internal conditions, the equilibrium is disrupted, significant rearrangments of the micro-structure and bone shape are possible. Many mathematical and computational models of this adaptation phenomenon can be assigned one of two categories; namely, theoretical models originating from the theory of adaptive elasticity and computational models making use of the optimization theory. In the present paper the approach based on the hypothesis of optimal response of a bone is proposed. It enables derivation of various adaptation laws associated with extremum of the objective functional under a set of appropriate constraints and makes a bridge between the aforementioned categories. In order to illustrate possible application of the proposed general approach the specific formulation is presented and mathematical relations governing the adaptation process are derived. Four numerical examples illustrating some of possible applications of the presented relations are included.
Na przebudowę kości mają zasadniczy wpływ dwa procesy: resorpcja tkanek oraz synteza nowej matrycy. W stanie ustalonym są one w równowadze, lecz gdy na skutek zmiennych warunków zewnętrznych któryś z nich zaczyna przeważać może nastąpić nawet znaczna zmiana struktury wewnętrznej i zewnętrznego kształu kości. W literaturze poświęconej problemowi modelowania zjawiska adaptacji kości można wyróżnić dwa charakterystyczne podejścia, jedno oparte na teorii adaptacyjnej sprężystości i drugie wykorzystujące matematyczne metody optymalizacji. W niniejszej pracy zaproponowano nowe sformułowanie wykorzystujące hipotezę optymalnej reakcji układu. Łączy ono w sobie wiele zalet obu wspomnianych metod. W celu zilustrowania ogólnej idei wyprowadzono konkretne, proste prawo adaptacji. Przedstawiono też kilka przykładów numerycznych ilustrujących niektóre z możliwych zastosowań omawianych związków teoretycznych.

The Swc4p protein, encoded by an essential gene, is shared by two chromatin-remodeling complexes in Saccharomyces cerevisiae cells: NuA4 (nucleosome acetyltransferase of H4) and SWR1. The SWR1 complex catalyzes ATP-dependent exchange of the nucleosomal histone H2A for H2AZ (Htz1p). The activity of NuA4 is responsible mainly for the acetylation of the H4 histone but also for the acetylation of H2A and H2AZ. In this work we investigated the role of the Swc4p protein. Using random mutagenesis we isolated a collection of swc4 mutants and showed that the essential function of Swc4p resides in its N-terminal part, within the first 269 amino acids of the 476-amino acid-long protein. We also demonstrated that Swc4p is able to accommodate numerous mutations without losing its functionality under standard growth conditions. However, when swc4 mutants were exposed to methyl methanesulfonate (MMS), hydroxyurea or benomyl, severe growth deficiencies appeared, pointing to an involvement of Swc4p in many chromatin-based processes. The mutants' phenotypes did not result from an impairment of histone acetylation, as in the mutant which bears the shortest isolated variant of truncated Swc4p, the level of overall H4 acetylation was unchanged.

The dynamic bone remodeling process is a computationally challenging research area that struggles to understand the actual mechanisms. It has been observed that a mechanical stimulus in the bone greatly affects the remodeling process. A 3D finite element model of a femur is created and a probabilistic analysis is performed on the model. The probabilistic analysis measures the sensitivities of various parameters related to the material properties, geometric properties, and the three load cases defined as Single Leg Stance, Abduction, and Adduction. The sensitivity of each parameter is based on the calculated maximum mechanical stimulus and analyzed at various values of probabilities ranging from 0.001 to 0.999. The analysis showed that the parameters associated with the Single Leg Stance load case had the highest sensitivity with a probability of 0.99 and the angle of the force applied to the joint of the proximal femur had the overall highest sensitivity.

In this paper, the effect of initial density distribution upon the predicted density via numerical simulations of bone remodeling was evaluated. The main purpose was to correlate the numerical results with clinical data according to which the initial bone quantity is an essential factor for long term survival of dental implants. Methods: Two-strain energy density-based bone remodeling theories were employed, one which accounts for overload resorption and the second one, which does not. The remodeling parameters were derived from the mechanostat theory. Bone remodeling around an osseointegrated dental implant was simulated based on finite element method using a generic mandible plane model. A variable time step was introduced to increase the speed of the remodeling simulations by keeping the truncation errors small. The simulations were performed for several initial density distributions correlated with values from clinical classifications of bone quality. For each density value, the occlusal load was defined in two ways so that to consider normal and overload mastication forces, respectively. Results: The results showed that the initial density distribution influences the predictions of bone remodeling simulations. For the analyzed model, the remodeling algorithm predicted overload resorption only in the case of low initial density, which can be associated with low bone quality, which, from clinical perspective, may probably lead to implant loss. Conclusions: The paper demonstrated that when simulating bone remodeling around dental implants using finite element method, it is important to account for initial density distribution in correlation with the bone quantity.

Bone remodeling is an integrated process of resorption and osteogenesis. Such processes are performed on cyclical basis, as regulated by specifi c bone cells, including osteoclasts, osteoblasts and osteocytes. Not long ago it was claimed that bone remodeling involves only osteoclasts, conditioning bone resorption and osteoblasts, responsible for osteogenesis. Recent studies have shown however, that the major regulatory part in bone remodeling is taken by osteocytes. Such cells regulate the activity of osteoclasts and osteoblasts, infl uencing the RANK/RANKL/OPG pathway as well as the signalling canonical pathway Wint/-catenine. The role of osteocytes in regulation of RANK/RANKL/OPG pathway is basically associated with regulation of RANKL secretion by osteoblasts, while in regulation of Wnt/ /-catenine, with secretion of sclerostin, which inhibits osteogenesis by blocking activation, proliferation and diff erentiation of osteoblasts from the mesenchymal stem cells and slows down Wnt/-catenine signalling.
Aktywność metaboliczna szkieletu związana jest z ciągłą przebudową tkanki kostnej. Mechanizm ten jest niezbędny do przystosowania szkieletu do warunków zewnętrznych i obciążeń mechanicznych oraz zapewnia równowagę mineralną. Prawidłowy przebieg procesu przebudowy kości zależy od aktywności komórek kostnych, do których należą osteoblasty, osteoklasty i osteocyty. Osteoblasty uczestniczą w kościotworzeniu oraz resorpcji kości. Wydzielają cytokiny i czynniki wzrostu, dzięki czemu pełnią rolę regulacyjną procesu osteoklastogenezy i resorpcji kości przez wpływ na szlak regulacyjny RANK/RANKL/OPG. Osteocyty z kolei regulują osteoblastogenezę, proces apoptozy osteoblastów oraz funkcję szlaku RANK/RANKL/OPG przez wpływ na kanoniczny szlak sygnalizacyjny Wnt/-katenina. Osteocyty wydzielają również sklerostynę, działającą antagonistycznie w stosunku do szlaku kanonicznego WNT/–katenina. Sklerostyna hamuje proces kościotworzenia. Szlak Wnt/–katenina może być potencjalnym celem terapii, prowadzącym do zwiększenia masy kostnej, natomiast sklerostyna – jako inhibitor tego szlaku – stanowi nowy obiecujący cel w badaniach nad terapią anaboliczną schorzeń tkanki kostnej.

This study aimed at the simulation of bone tissue remodeling within a bone defect with the utilization of the finite element method (FEM), enabling - via elaborated application - objective evaluation of orthodontic forces which positively influence periodontium in vivo. The initial position of each bracket on the passive archwire was registered, and then a geometrical and discretemodel of the appliance was created automatically. Assessment of the dental scans obtained using cone beam computed tomography (CBCT) allowed evaluation of the range of bracket displacement: from the initial position to the final one achieved on the active archwire. Those displacements established terminal conditions in the finite element analysis, enabling calculation of orthodontic force levels. An individual design of a tooth with periodontal ligaments and the periodontal defect subsequently loaded with the determined forces allowed simulation of bone remodeling according to Carters adaptation process. Mainly, the bone apposition processes took place in the central part of the periodontal defect, in proximity of the alveolar ridge. However, FEM application in the analysis of bone tissue regeneration within bone defects enables precise evaluation of the achieved changes, therefore allows determination of orthodontic forces positively influencing periodontium in vivo.
