Opis:
Gypsophila aretioides, a cushion form evergreen plant, is a high potential wild species ideal for the use in rock garden, or as a ground cover in sunny dry areas. This plant has the competence to be developed as a new ornamental species. The purpose of this experiment was to provide an efficient micropropagation protocol for G. aretioides in order to facilitate the availability of this species for further studies of domestication. The influence of various concentrations of 6-benzylaminopurine (BAP) and thidiazuron (TDZ) was investigated for multiplication stage. TDZ at low concentration of 0.05 mg dm−3 resulted in the maximum shoot (9.7) and leaf (42.3) number. The shoots were best rooted on MS medium containing 0.6 mg dm–3 indolebutyric acid (IBA) with 7.8 roots per shoot. Despite achievement of a successful protocol for in vitro multiplication and root induction of Gypsophila, low survival rate was obtained when rooted explants were exposed to ex vitro conditions. This is an important issue, which requires particular consideration and further studies. The possible reasons contributing to the low acclimatization rate of this species are being discussed.

Opis:
Grafting plays an important role in rhododendron propagation. Successful graft union depends on selection of a suitable stock, grafting season, and method. In this study, three methods (whip, cleft, and side grafting) were compared in two seasons. The stocks were mainly cuttings of R. catawbiense-Hybridum, R. ‘Cunningham's White', but also R. pachytrichum and R. brachycarpum. Scions were taken from several cultivars deriving from R. catawbiense. To shorten the production period, scions were grafted on unrooted stocks, and the results are encouraging. The best stock was R. ‘Cunningham's White', especially when side grafting was applied in spring.

Isatis concstricta Davis (an endemic plant of Turkey) suffers from low propagation rates under natural conditions and is threatened due to fast unplanned urbanisation. The study compared the effects of variants of BA + NAA on shoot regeneration on leaf and petiole explants excised from one week old in vitro regenerated seedlings. MS medium containing 1 mg L–1 BA + 1 mg L–1 NAA induced maximum proliferation on petiole and leaf explants with 13.33 and 12.75 shoots per explant repectively. However, leaf explant induced shoots were sturdy and healthier compared to petiole explant induced shoots. These shoots were rooted on MS medium containing 0.5 mg L–1 IBA and the plants were acclimatized in peat moss and sand (v/v). They grew to flowering under ex vitro conditions. This system of regeneration is advantageous for conventional propagation and the results will help in establishment of a powerful and meaningful micropropagation system for I. constricta.

Lachenalia is a poorly known genus of ornamental bulbous plants from South Africa with a huge floricultural potential. The paper discusses in vivo multiplication with leaf cuttings of Lachenalia cv. ‘Namakwa’, ‘Romaud’, ‘Ronina’ and ‘Rupert’. Mature leaves were collected from non-flowering donor plants. Each leaf was halved to make proximal and distal leaf cuttings. ‘Ronina’ distal leaf cuttings produced the lowest number of bulblets (5.7 per cutting), while ‘Namakwa’ proximal leaf cuttings turned out the most productive (10.5 bulblets per cutting). The bulblets produced by cv. ‘Namakwa’ distal leaf cuttings were the lightest (0.3 g per bulblet), and those formed on cv. ‘Ronina’ proximal leaf cuttings were the heaviest (0.9 g per bulblet). In general, the weight of individual bulblets ranged from 0.1 to 1.2 g. Irrespective of the cutting type, the leaves of cv. ‘Namakwa’ produced the greatest number of bulblets, and the leaves of ‘Ronina’ yielded the heaviest bulblets. With no dependence on their genotype, proximal leaf cuttings were advantageous in terms of total number of generated bulblets and their quality (weight, diameter).

Ceratophyllum demersum L. is a medical plant of the family Ceratophyllaceae that has been traditionally used for the treatment of diseases such as ulcer, diarrhoea, wounds and fever since ancient times. This study presents antioxidant activities of water and methanol extracts of in vitro propagated C. demersum. Shoot tip explants of C. demersum for in vitro plant regeneration were cultured on liquid Murashige and Skoog (MS) medium containing 0.25–1.25 mg/L 6-Benzylaminopurine (BAP) and 0.10 mg/L Thidiazuron (TDZ) combinations for eight weeks. Maximum number of plantlets per explant (110.67) was obtained on MS medium supplemented with 0.75 mg/L BAP + 0.10 mg/L TDZ. In order to determine antioxidant activities of C. demersum, antioxidant contents (β-carotene, flavonoid, lycopene and total phenols), metal chelating and reducing power capacities of methanol and water extracts obtained from C. demersum were investigated. It was detected that water extract was more effective in all activities. Total phenols (76.55 μg/mg) in water extract were measured as the highest antioxidant component. Based on the water extracts data, as IC50 value for chelating on ferrous ions was 9.24 mg/mL, EC50 value for reducing power activity was 8.23 mg/mL. It was also determined that the metal chelating and reducing power activities of the both extracts are increased depending on the concentration. Therefore, all these data suggest that both tested extracts of in vitro propagated C. demersum would exert beneficial effects by virtue of their antioxidant properties and may be utilized as a promising source of therapeutics.

Different nutrient media (MS [Murashige and Skoog 1962]; QL [Quoirin and Lepoivre 1977] and WPM [Lloyd and McCown 1980]); plant growth regulators BA (benzil adenin), GA3 (gibberellic acid), IBA (indole-3-butyric-acid), NAA (naftalen acedic acid); and sucrose concentrations were studied to determine the in vitro culture effects on healthier and faster seedling development from mature lentisk (Pistacia lentiscus L.) seeds. After 28 days of culture, the percentage of germinated seeds was the highest (70%) in the full-strength MS medium. The cytokinin BA was superior to other tested treatments in terms of its ability to promote germination of lentisk seeds. When tested at different concentrations, sucrose gave the best results obtained at concentrations of 1–4%, whereas high concentrations (6 and 8%) mainly decreased germination rate and there was no a regular pattern for elongation of the aerial parts of plants. With this described protocol, on average 76.67% seeds germinated 4 weeks after culture. Developed seedlings were satisfactorily acclimatized in sterilized peat, soil and perlite containing compost, with high percentage survival viability was obtained 9 months after transfer to in vivo conditions (93.33%). The results obtained showed that the enriched full-strength MS medium supplemented with 1 mg L–1 BA and 3% sucrose induced homogeneous and healthy seedling development in a period of 4 to 8 weeks of culture.

Opis:
Autumn daffodil (Sternbergia lutea) is a rare and endangered bulbous plant with yellow flowers grown naturally in Iran and unfortunately for different reasons this plant is at the risk of eradication. Natural multiplication rate of S. lutea is low, thus propagation by chipping method is one inexpensive and simple way to overcome this problem. This research was therefore performed to investigate the propagation method in two experiments. In the first experiment, the effects of some plant growth regulators (PGRs) on induction and regeneration of bulblets from bulb chips (propagules) were evaluated. The propagules were prepared by chipping method and treated with indole butyric acid (IBA), gibberellic acid (GA3), benzyl adenine (BA), kinetin (Kin) and cycocel (CCC) at 100 and 200 mg L–1 concentrations for 2 h and then incubated for two months at 20 ±1°C. At the end of incubation, the results showed the highest percentage of concurrent bulblet + root regeneration (CBRR), number and fresh weight of bulblets related to 100 mg L–1 GA3. The highest percentage of only bulblet regeneration (OBR), only root regeneration (ORR) and non-regeneration were obtained at 200 mg L–1 IBA, 100 mg L–1 Kin and control (distillated water: DW) treatments, respectively. In the second experiment, the bulblets that had rooted from each treatment of the first experiment were planted and the produced bulbs were compared. The results indicated that the highest percentage of bulblet sprouting, bulb number and diameter were found at 100 mg L–1 GA3. Therefore, the application of GA3 with mentioned concentration is appropriate for vegetative propagation of this plant by bulb chipping.

Experimental studies were conducted in 2015–2017 in the Department of Dendrology, Pomology and Nursery of Poznan University of Life Sciences. The aim of the research was to study the usefulness of three rootstocks: Larix decidua, Pinus armandii, Cedrus deodara and two grafting methods: side grafting and split grafting for propagation purposes of Cedrus deodara ‘Karl Fuchs’. In addition, it was evaluated how the rootstock and the age of cultivar shoots influenced the level of chloroplast pigments in the needles. A higher percentage of grafts success was obtained on Pinus armandii and Cedrus deodara using the side grafting method. Grafted scions of ‘Karl Fuchs’ produced the greatest number of lateral buds and the longest lateral increments of growth on Cedrus deodara root stock. Side grafting significantly improved the percentage of plants that restarted their growth in the second year of cultivation. The largest number of buds on two-year-old shoots were counted on plants grafted on Cedrus deodara using split grafting method. The rootstocks used in this experiment had a slight effect on the content of chloroplast pigments in the needles. Plants grafted on Pinus armandii had the highest level of chlorophyll B, and those grafted on Larix decidua – chlorophyll A/B ratio. The content of chloroplast pigments in the needles depended on the age of shoots. The highest level of chlorophyll A, chlorophyll B and carotenoids was observed when needles were collected from the last year’s growth.

Yucca elephantipes is an important commercial ornamental pot plant, excellent for growing in flats, patios or winter gardens. Traditional vegetative propagation of the most decorative yuccas is complicated due to a very low rate of propagation, so in vitro culture is an alternative method for commercial propagation of these plants. The influence of BA (0.4, 2.2, 4.4, 11.1, 22.2 µM) and TDZ (0.5, 2.3, 4.5, 11.4, 22.7 µM) on shoot multiplication of Yucca elephantipes Regel on Murashige and Skoog (MS) medium was studied. Explants cultured on medium without growth substances were used as a control. The two types of explants used in the experiment: shoot tips and nodal segments of shoots, were obtained from aseptically grown shoot clusters. When comparing regeneration capability of 2 types of Yucca elephantipes explants, it was found that more newly formed shoots and roots were obtained from nodes. The highest formation of shoots was obtained from nodes on MS medium supplemented with 4.5 µM TDZ or 11.1 and 22.2 µM BA (6.5, 6.0, 5.8, respectively). The shoots regenerated from nodes showed best elongation. On the control medium and on the media with the lowest level of BA or TDZ, their average length was 31.0–37.8 mm. The growth regulator-free medium and the media with a low level of BA were the most effective in inducing roots.
Juka słoniostopa (Yucca elephantipes) jest cenną rośliną pojemnikową do mieszkań, patio, a także do ogrodów zimowych. Tradycyjna metoda rozmnażania wegetatywnego większości gatunków ozdobnych juki jest mało wydajna, stąd zastosowanie metody kultur in vitro stwarza duże możliwości zwiększenia współczynnika mnożenia przy produkcji tych roślin na skalę masową. Badano wpływ BA (0,4; 2,2; 4,4; 11,1; 22,2 µM) i TDZ (0,5; 2,3; 4,5; 11,4; 22,7 µM) na namnażanie pędów Yucca elephantipes Regel na pożywce Murashige’a i Skooga (MS). Eksplantaty rosnące na pożywce bez regulatorów wzrostu stanowiły kontrolę. Pobrano dwa rodzaje eksplantatów: wierzchołki pędów i odcinki węzłowe, które pochodziły z kultur sterylnych. Porównując zdolności regeneracyjne eksplantatów Yucca elephantipes, stwierdzono, że więcej nowych pędów i korzeni regenerowało z odcinków węzłowych. Najlepszą regenerację pędów uzyskano z odcinków węzłowych na pożywkach z dodatkiem 4,5 µM TDZ lub 11,1 i 22,2 µM BA (6,5; 6,0; 5,8, odpowiednio). Pędy uzyskane z węzłów wykazywały lepszy wzrost elongacyjny. Na pożywce kontrolnej oraz w obecności niższych stężeń BA lub TDZ ich średnia długość wynosiła 31,0-37,8 mm. Najsilniejszą indukcję korzeni obserwowano na pożywce kontrolnej oraz przy dodatku BA w niskim stężeniu.

From past to the present, cultural, medicinal, landscape and culinary use of junipers has contrib- uted to it being one of the most widely distributed woody plants in the world. Therefore, it is so important that finding appropriate production methods to obtain quality seedlings in line with the usage area. In the present study, it was tried to determine the best conditions and applications on propagation by cuttings of three juniper taxa including Juniperus communis L. ‘Hibernica’, Juniperus chinensis L. ‘Stricta’, and Juniperus chinensis L. ‘Stricta Variegata’. For this purpose, two different greenhouse treatments (Greenhouse-1 with the air temperature at 20±2°C, rooting table temperature at 25±2°C and humidity level at 70±2%; Green- house-2 with the air temperature at 20±2°C, rooting table temperature at 20±2°C and humidity level at 70±2%) and different auxin applications (Indole-3-Butyric Acid, Indole-3-Acetic Acid and α-Naphthalene Acetic Acid at concentrations of 3000 and 5000 ppm) were designed. The highest rooting percentages were generally achieved in Greenhouse-1. IAA 5000 ppm treatment for J. communis ‘Hibernica’ (93.33%) and J. chinensis ‘Stricta’ (66.67%, also in Greenhouse-2) and IBA 3000 ppm treatment for J. chinensis ‘Stricta Var- iegata’ (60.00%) showed the best rooting results. This study, in which the best production conditions are investigated in some juniper taxa, is a guiding feature for the producers to ensure the sustainable utilization of junipers.

The present report illustrates an improved and reproducible micropropagation system for economically valuable bamboo species Pseudoxytenanthera stocksii using nodal segments. Direct organogenesis from nodal segments was accomplished by culturing on Murashige and Skoog (MS) medium supplemented with 6.0 mg/L 6-benzylaminopurine (BAP). On this medium combination 9.0±0.25 shoots per explant were induced. Shoot multiplication was subjective to the combination of auxins and cytokinins concentrations used and number of repeated transfer of mother explants or subculturing of in vitro regenerated shoot clumps on fresh medium. Maximum 41.9±1.00 shoots were observed on MS medium supplemented with 4.0 mg/L of BAP and 0.25 mg/L of naphthalene acetic acid (NAA). Half strength MS medium containing various concentrations of auxins was used for in vitro root formation. Successful rooting with maximum response was achieved on half strength MS medium + 50% sucrose and 1.0 mg/L IBA (24.3±0.27 roots). The rooted plantlets were planted the soilrite and hardened in the greenhouse for 6-8 weeks. Completely acclimatized plantlets exhibited 96% survivability. The present propagation protocol could satisfy the demand of P. stocksii and could be explored for the commercial propagation of this valuable manga bamboo.

Opis:
The hop (Humulus lupulus L.) is one of the most infected plants by viruses. Preparation of apical meristems in combination with heat therapy was successfuly used for obtaining of virus-free hop plants in various countries in the world. In our experiments we prepared the meristems both before and after the heat therapy. Hop plants regenerated from such meristems were free not only from the thermolabile ILAR-viruses, but in the great number also from the thermostable CARLA - viruses. The method of virus elimination and micropropagation of virus-free plants in vitro, described in this paper, enables to obtain a large number of virus-free plantlets in a short time.
Chmiel (Humulus lupulus L.) należy do roślin uprawnych w największym stopniu porażonych przez wirusy. Wirusy występujące w chmielu mogą powodować znaczne obniżenie masy plonu ( 4-38%) i zawartości alfa-kwasów w szyszkach (18-30%). Stosowane do tej pory metody uwalniania chmielu od wirusów, polegające na regeneracji roślin z merystemów wierzchołkowych albo na regeneracji roślin z merystemów w połączeniu z terapią wysokotemperaturową, pozwalały na skuteczne wyeliminowanie z chmielu wirusów termolabilnych zwłaszcza z grupy ILAR, natomiast wirusy termostabilne z grupy CARLA najczęściej pozostawały w zainfekowanych roślinach. Zmodyfikowana metoda uwalniania chmielu od wirusów różni się od dawniej stosowanych tym, że możliwie najmniejsze merystemy (0,2-0,3 mm) wypreparowywano zarówno przed terapią termiczną, jak również po terapii wysokotemperaturowej. Cięcie oddzielające merystem od reszty regenerującej rośliny przeprowadzano w strefie elongacyjnej (powstającej w trakcie wzrostu w wysokiej temperaturze), która hamuje replikację wirusów i najczęściej jest od tych patogenów wolna. Zastosowana metoda charakteryzowała się dużą śmiertelnością merystemów podczas terapii termicznej (około 70%) i regeneracji roślin, ale była skuteczna w eliminowaniu również termostabilnych wirusów z grupy CARLA w około 80-90%. Opracowane zostały także pożywki do hodowli chmielu in vitro dla różnych etapów hodowli tej rośliny. Skład pożywek został maksymalnie uproszczony (całkowicie usunięto składniki termolabilne z pożywek do masowego klonowania roślin) w celu przyspieszenia i ułatwienia prac związanych z ich przygotowaniem. W wyniku przeprowadzonych prac uwolniono od wirusów następujące odmiany chmielu: Lubelska, Lomik, Izabella, Marynka i Northern Brewer. W roku 1995 na terenie Wielkopolski założone zostały pierwsze plantacje produkcyjne z wyprodukowanych sadzonek chmielu, wolnych od wirusów.
