Opis:
A study was conducted to determine the Production of cellulase and pectinase enzyme by using Plant growth promoting rhizobacteria like Pseudomonas fluorescence and Bacillus subtilis. These to micro organism are isolated by serial dilution method. One gram of soil sample was diluted in to 10 ml of sterile distilled water and 1 ml of sample solution was serially diluted in 9ml of sterile water up to 10 dilution. Each sample from dilution 10-5 and 10-6 were taken and streaked in to KB and NA medium and incubate at 24 hrs. After 24 hrs Pseudomonas fluorescence and Bacillus subtilis was observed in the medium of KB and NA medium. Both the culture was sub cultured and maintain in the same for the further work. CMCase medium was prepared and sterilized by autoclave for 121 ºC for 15 minutes after sterilization these medium contain petriplate was streaked by bacteria and incubates for 48h after incubation period a clear halo zone was produced by these bacteria among these bacteria Pseudomonas fluorescence are able to produce high amount of cellulose compare to Bacillus subtilis. Pectin agar medium was prepared and sterilized by autoclave for 121 ºC for 15 minutes after sterilization these medium contain petriplate was streaked by bacteria incubates for 48h after incubation period a clear halo zone was produced by these bacteria, among these bacteria Pseudomonas fluorescence are able to produce high amount of Pectinase compare to Bacillus subtilis. Plant growth promoting rhizobacteria (PGPR) are beneficial bacteria that colonize plant roots and enhance plant growth by a wide variety of mechanisms.

Opis:
Terry fabrics are exposed to frequent and intensive washing and must therefore be resistant to water, alkali, washing agents and rubbing. Washing agents frequently contain cellulase enzymes. The influence of two kinds of cellulase enzymes on the properties of a 100% cotton terry fabric was investigated. The whole cellulase mixture and an endoglucanaserich cellulase product were used. Fabric strength, resistance to rubbing, wettability, hand feeling, whiteness and other properties were observed. Moreover the influence of different pretreatment processes on the properties of the terry fabric was investigated. The fabric was scoured with alkali or with pectinase enzymes, and bleached with hydrogen peroxide or peracetic acid. It was established that enzymatic scouring and peracetic acid bleaching consume less energy and water but provide fabrics with lower whiteness and wettability, suitable only for further dyeing in dark or medium shades. Cellulases deteriorate the properties of terry fabrics; however, worse are the whole cellulase mixtures.
Tkaniny frotte są intensywnie prane i dlatego muszą być odporne na działanie wody, zasad, środków piorących i tarcie. Środki piorące często zawierają celulazy. Badano wpływ dwóch rodzajów enzymów celulazy na właściwości 100%-bawełnianych tkanin frotte. W badaniach użyto mieszaniny celulaz i endoglukanaz. Badano m.in., wytrzymałość, odporność na ścieranie, zwilżalność, chwyt, stopień bieli. Ponadto badano wpływ różnych procesów obróbki wstępnej na właściwości tkanin. Tkaniny poddano obróbce alkalicznej i enzymatycznej używając pektyny oraz bielenie nadtlenkiem wodoru lub kwasem nadoctowym. Ustalono, że podczas obróbki enzymatycznej i bielenia kwasem nadoctowym zużywa się mniej energii i wody, ale uzyskuje się tkaniny o niższym stopniu bieli i mniejszej zwilżalności, nadające się tylko do dalszego barwienia w odcieniach ciemnych lub średniociemnych. Stwierdzono, że celulazy pogarszają właściwości tkanin frotte, jednakże gorszy wpływ mają mieszaniny celulazowe.

Opis:
An enzyme-assisted modified steam distillation process was adopted to extract turmeric oil from Curcuma longa L. rhizomes. The diastase, xylose, cellulase, pectinase, and lipase enzymes were used for the pre-treatment of fresh turmeric rhizome to obtain a higher yield by rapturing biological cells. The quantitative and qualitative analysis of turmeric oil was performed by GC–MS. The various influencing parameters for the extraction of turmeric oil such as an enzyme, incubation/pre-treatment time, distillation time have been studied in the present work. The obtained turmeric oil by enzymatic pretreatment process is richer in bioactive/medicinal components than in the other traditional methods. The maximum yield was obtained with cellulase enzyme, which is 25–27% higher than the yield obtained by the traditional hydro distillation process. The detailed qualitative and quantitative analyses are also presented. The present method can be considered energy-efficient, effective, economical, and eco-friendly.

Opis:
Stagonospora nodorum produces cell wall degrading enzymes when grown in culture media containing cell wall components. The pathogen grew as well on minimal agar plates containing cellulose, xylan and pectin as glucose, except having sparser mycelia. Four cell wall-degrading enzymes, cellulase, xylanase, pectinase and b-1,3-glucanase were coordinately induced in culture filtrates growing on xyaln and cellulose as substrates. An aggressive isolate (sn26-1) secreted more cell wall-degrading enzymes than the others. Based on isoelectric focusing profiles, six to seven xylanase isozymes were induced by cellulose and xylan. No difference was found in the high (sn26-1) and low (9074) aggressive isolates. Addition of cell wall-degrading enzyme mixtures, not high xylanase alone, to a spore suspension of a low aggressive isolate (9074) caused a limited increase in tissue necrosis. We conclude that the cell wall degrading enzymes play a role in early penetration of the host by the fungus, but they are not important elicitors for disease development.

Opis:
The enzymatic activity (chitinolytic, proteolytic, pectolytic andcellulolytic) of twenty strains of Streptomyces isolated from soil and rhizosphere of Scots pine was analyzed. Most strains produced chitinases, catalyzing the degradation of chitin, the main component of fungal cell walls including fungi pathogenic for plants. This activity was about 4 times higher in the presence of Fusariumoxysporum than Rhizoctonia solani mycelium or chitin flakes. The number of the proteolytic strains was also significant. In general, rhizosphere and soil organisms synthesizedcomparable amounts of these enzymes. Over half of the analyzed Streptomyces strains produced pectolytic enzymes (polygalacturonase, pectin lyase and pectate lyase). This property was more common among rhizosphere than among soil strains. The Streptomyces strains also showedcellulolytic activity (endocellulases, exocellulases) – enzymes decomposing basic components of cell walls of plant and lower fungi (cellulose). The cellulolytic activity was differentiated and depended on the Streptomyces strain. Conclusion of our studies is that Streptomyces are the microorganisms more chitinolytic andproteolytic than pectolytic and cellulolytic.
