The usefulness of mutagenic treatment to enlarge isozymic variability of barley and the use of induced mutants for genetic analysis were evaluated. N-methyl-N-nitroso urea, sodium azide and gamma rays were employed as mutagenic agents. Electrophoretic assays of 3848 M₂ seedlings obtained by chemical mutagenic treatment of the spring barley cultivars Dema, Aramir, Bielik and 3100 M₂ seedlings obtained by physical mutagenic treatment of the cv. Dema revealed 70 isozymic mutants, which represent 30 separate mutants in 25 M₁ plants. Most of mutations (27) were induced by chemical mutagen at polymorphic esterase loci. The occurrence of induced mutants at monomorphic loci, Got2 and Lap2, made it possible to perform genetic analysis of those loci in barley including mapping respective genes within chromosomes.

Effect of EMS (ethyl methane sulphonate) on induced morphological mutants and chromosomal variation in cowpea was studied using five different doses of mutagen along with a control in randomized blocked design with three replications. The morphological mutants there are two types of viable and chlorophyll mutants. Viable mutant contains tall, dwarf, early maturity, late maturity, leaf mutants pod mutant and flower mutants. The frequency of chlorophyll mutant contains albino, xantha and viridis. This concentration can damage or modify important components of plant cells and have been reported to affect the morphology, anatomy, biochemistry and physiology of plants differentially depending on the concentration level. These effects include changes in the cellular structure and metabolism of the plants e.g., dilation of thylakoid membranes, alteration in photosynthesis, modulation of the antioxidative system and accumulation of phenolic compounds. The morphological and chromosomal variation was found to be mutagen sensitive in somatic cells of cowpea. It was found to increase with increasing the concentration of EMS in Cowpea plants. The chemical mutagen like ethyl methane sulphonate induces high frequency of chromosomal changes like anaphasic bridge; anaphasic laggard, anaphasic bridge and clumbing of chromosome were including control plants also observed.

Arbuscular mycorrhizae are endogenous fungi that form symbiotic association with the vast majority of vascular plants. They help to increase the nutrient uptake, especially phosphorus and to exploit a larger area of soil. Also, they improve soil aggregation, contribute to nutrient cycling processes in plants and improve their tolerance of heavy metal contamination or drought, as well as their susceptibility to root pathogens or herbivores. These fungi colonize the root epidermis and outer cortical layers and release bioactive molecules. The transcriptome and the proteome of plants are substantially altered. They have evolved multiple mechanisms that result in improvements in plant resistance to disease and plant growth and productivity, being included in a lot of combination of biofertilizers. In order to study these mechanisms, gamma irradiation was used to create mutants that are defective in their association. Culture under in vitro conditions is presented as method for maintaining the useful mutants in pure line.

The Swc4p protein, encoded by an essential gene, is shared by two chromatin-remodeling complexes in Saccharomyces cerevisiae cells: NuA4 (nucleosome acetyltransferase of H4) and SWR1. The SWR1 complex catalyzes ATP-dependent exchange of the nucleosomal histone H2A for H2AZ (Htz1p). The activity of NuA4 is responsible mainly for the acetylation of the H4 histone but also for the acetylation of H2A and H2AZ. In this work we investigated the role of the Swc4p protein. Using random mutagenesis we isolated a collection of swc4 mutants and showed that the essential function of Swc4p resides in its N-terminal part, within the first 269 amino acids of the 476-amino acid-long protein. We also demonstrated that Swc4p is able to accommodate numerous mutations without losing its functionality under standard growth conditions. However, when swc4 mutants were exposed to methyl methanesulfonate (MMS), hydroxyurea or benomyl, severe growth deficiencies appeared, pointing to an involvement of Swc4p in many chromatin-based processes. The mutants' phenotypes did not result from an impairment of histone acetylation, as in the mutant which bears the shortest isolated variant of truncated Swc4p, the level of overall H4 acetylation was unchanged.

Opis:
Mutation testing – a fault-based technique for software testing – is a computationally expensive approach. One of the powerful methods to improve the performance of mutation without reducing effectiveness is to employ parallel processing, where mutants and tests are executed in parallel. This approach reduces the total time needed to accomplish the mutation analysis. This paper proposes three strategies for parallel execution of mutants on multicore machines using the Parallel Computing Toolbox (PCT) with the Matlab Distributed Computing Server. It aims to demonstrate that the computationally intensive software testing schemes, such as mutation, can be facilitated by using parallel processing. The experiments were carried out on eight different Simulink models. The results represented the efficiency of the proposed approaches in terms of execution time during the testing process.

Opis:
Soluble APRIL (sAPRIL), the active form of a proliferation-inducing ligand (APRIL), is implicated in the proliferation of tumor cells. Suppressing APRIL function has been considered as a potential strategy for the therapy of APRIL-associated tumors. In the present study, we generated human sAPRIL and its two mutants, Gln187-D-sAPRIL (Gln187 deleted) and Gly187-sAPRIL (Gln187 replaced by Gly). In vitro experiments showed that the two mutants had similar specific binding capacity to lung carcinoma A549 cells compared to the wild-type sAPRIL, and both, especially Gly187-sAPRIL, exhibited significant antagonistic effect on sAPRIL-induced tumor cell proliferation in a dose-dependent manner, which might be predominantly mediated by blocking sAPRIL-induced MEK and ERK phosphorylation but not p38MAPK or JNK signaling. In vivo experiments with nude mice bearing A549 cell-derived xenograft tumor showed that only the Gly187-sAPRIL mutant could significantly suppress the tumor growth. These results suggest that Gln187 may be a crucial amino acid in APRIL-mediated tumor cell proliferation via the MEK-ERK signaling pathway and that the sAPRIL mutants may serve as novel potential antagonists of APRIL for the therapy of APRIL-associated cancers.

Seeds of Lathyrus sativus cv. Derek and Krab were used as biological material for induced mutagenesis. Three mutant lines were obtained from seeds of grass pea cv. Derek and 15 lines from mutagenised seeds of cv. Krab. Twelve ethanol-soluble carbohydrates were identified in the seeds. We have selected grass pea mutant lines with high oligosaccharides content (lines D4, K56, K25, and K7) and lines with low raffinose family oligosaccharides (RFO) content (lines K12, K29 and K13). Mutations changing the levels of RFO have not affected the contents of galactosyl cyclitols.

The Chinese hamster cell mutant V-C8 is defective in the Brca2 gene (Kraakman-van der Zwet et al., 2002, Cell Biol.; 22: 669). Here we report that V-C8 cells were 10-fold more sensitive to camptothecin, an inhibitor of topoisomerase I, than the parental V79 cells. The level of the relaxation activity of topoisomerase I in nuclear extracts was also lower (4-fold) in V-C8 than V79 cells, in spite of the fact that the level of the topoisomerase I protein was the same in these cells. The survival of V-C8 cells in the presence of camptothecin, the sensitivity of V-C8 topoisomerase I to camptothecin, and the level of the relaxation activity in V-C8 nuclear extract were almost completely restored by transfection of V-C8 cells with the murine Brca2 gene or by the transfer of human chromosome 13 providing the BRCA2 gene. These results indicate that the observed changes in the topoisomerase I activity in V-C8 are due to the defective function of the Brca2 gene.

Chemical mutagenesis is an efficient tool used in mutation breeding programme for improving various vital characteristics in floricultural crop, like Dianthus. In this study, ethyl methane sulphonate (EMS), sodium azide (SA) and colchicine (COL) with three different concentrations (0.1%, 0.4% and 0.7%) were used to analyse their effect on seed germination behaviour, survivability and pollen sterility in both first (M1) and second (M2) mutant generations. It was noted that increase in the dose of EMS and SA, germination percentage and survivability were decreased; whereas colchicine doses were proportional to increase germination percentage at seedling stage, but they were not survived till maturity. In M1 and M2, higher lethality over control (44.3 and 32.89, respectively) was shown by 0.7% of SA and EMS, respectively. Pollen sterility was also increased with increasing mutagenic doses. The maximum pollen sterility was 71.8% and 61.1% for 0.7% COL in M1 and M2, respectively. So, the effect of chemical mutagenesis on biological parameters with SA (0.7%) treatment in M1 and EMS (0.7%) treatment in M2 were much more beneficial as compared to colchicine. For each studied parameter, chemical mutagenesis was higher in M1 than M2. Hence, for the first time in Dianthus, we reported that these mutagens can be used for improving the germination behaviour and the metrical traits in Dianthus cultivar.
