Staphylococcus aureus is responsible for many types of infections related to biofilm presence. As the early diagnostics remains the best option for prevention of biofilm infections, the aim of the work presented was to search for differences in metabolite patterns of S. aureus ATCC6538 biofilm vs. free-swimming S. aureus planktonic forms. For this purpose, Nuclear Magnetic Resonance (NMR) spectroscopy was applied. Data obtained were supported by means of Scanning Electron Microscopy, quantitative cultures and X-ray computed microtomography. Metabolic trends accompanying S. aureus biofilm formation were found using Principal Component Analysis (PCA). Levels of isoleucine, alanine and 2,3-butanediol were significantly higher in biofilm than in planktonic forms, whereas level of osmoprotectant glycine-betaine was significantly higher in planktonic forms of S. aureus. Results obtained may find future application in clinical diagnostics of S. aureus biofilm-related infections.

The glycine receptor belongs to the ligand-gated ion channel superfamily. It is a chloride conducting channel composed of four transmembrane domains. It was previously shown that the second transmembrane domain (M2) of the glycine receptor forms an ion conduction pathway throught lipid bilayers. The amino-acid sequence of the transmembrane segment M2 of the glycine receptor has a high homology to all receptors of the ligand-gated ion channel superfamily. In our report, we have used a synthetic M2 peptide. It was incorporated into a planar membrane of known lipid composition and currents induced by M2 were measured by the Black Lipid Membrane technique. When the planar lipid bilayer was composed of 75% phosphatidylethanolamine and 25% phosphatidylserine, the reversal potential measured in a 150/600 mM KCl (cis/trans) gradient was -19 mV suggesting that the examined pore was preferential to anions, PK/PCl = 0.25. In contrast, when 75% phosphatidylserine and 25% phosphatidylethanolamine was used, the reversal potential was +20 mV and the pore was preferential to cations, PK/PCl = 4.36. Single-channel currents were recorded with two predominant amplitudes corresponding to the main-conductance and sub-conductance states. Both conductance states (about 12 pS and 30 pS) were measured in a symmetric solution of 50 mM KCl. The observed single-channel properties suggest that the selectivity and conductance of the pore formed by the M2 peptide of the glycine receptor depend on the lipid composition of the planar bilayer.

Opis:
A reversible effect of pH on the ionization of amino-acid residues at the active center of choline oxidase was observed near the optimum pH (8). Inactivation of choline oxidase took place in the pH ranges 3-6 and 9-11, in which irreversible changes in the structure occur leading to the enzyme inactivation. The first order rate constants of the enzyme's inactivation at various pH values were estimated for the irreversible changes. The Arrhenius analysis revealed no significant changes in the activation enthalpy, while an increase in the activation entropy reflected an increase in the conformational freedom.

Mutation breeding in crop plants is an effective approach in improvement of crop having narrow genetic base such as soybean. The main objective of the present study is to determine the effect of different doses of gamma irradiation on different morpho-agronomic characteristics. Many physical mutagens have been employed for obtaining useful mutants in various crop species. The role of mutation breeding increases the genetic variability for the desired traits in various crop plants. Soybean (Glycine max (L.) Mrr.) var. Co1. Was treated with physical mutagens like Gamma rays. For inducing mutation of Soybean treated with various concentrations of gamma rays 10, 20, 30, 40, 50, 60, 70, 80, 90, and 100 KR. Agronomic traits and morpho metric characters such as germination percentage, days of first flower, root length, shoot length, seedling survival, number of fruits per plant, fruit length, seed yield per plant, fresh weight per plant, dry weight per plant and 100 seed weight. All parameters were studied in M1, M2, M3 and M4 generations. The results of the present study re- vealed that the increasing level of gamma rays treatment with decreased significantly most of agronomic and morphological traits evaluated in M1 populations. In M2,M3 and M4 populations with significant increase of morphology and yield components in soybean. The yield parameters like plant height, number of cluster per plant, number of seeds per plant and seed yield per plant were recorded the moderated and high mean value in the 50 KR of gamma rays.

Plants of soybean (Glycine max L.) cv. Augusta grew in the soil with addition of Ni in the concentration of 80 (C-80) or 120 mg∙kg-1 dry mass (C-120) or without Ni (C-0), and they were subjected to UV-B (300 nm – 315 nm) dose of 1.8 kJ m-2 d-1 for 28 days. The addition of nickel into soil in the concentration of 120 mg kg-1 (C-120) caused a decrease of the plant height by 35% compared to the control (C-0), and in the presence of UV-B (U-120) – by 43%. The shoot fresh and dry mass in the C-80 and C-120 plants were lower by 33% and 52% than in the C-0. In the presence of Ni, the intensity of net photosynthesis decreased by 55%. UV-B caused an increase of flavonoid content by 25% compared to the control (C-0), and Ni induced a reduction in the content of these compounds from 20% to 40%.

Metal ions can modify plant metabolism and change the level of biologically active components. In the present study, the impact of short-term exposure to strontium on the accumulation of the metal as well as the content of isoflavones in soybean sprouts was investigated. The seeds were germinated in hydroponics with 0, 1, 1.5, 2.5, 5.0, or 10.0 mM of Sr for 72 hours. The content of strontium was assessed using flame atomic absorption spectrometry and the amount of isoflavones was determined with high performance liquid chromatography. dose-dependent accumulation of Sr and a linear correlation between the Sr concentration in the growth medium and the content of the element in the plant samples were observed. The largest changes in the isoflavone content, compared to the control, were noted in soy sprouts germinated in the presence of 5 and 10 mM of strontium. daidzin, genistin, malonyldaidzin, and malonylgenistin were the dominant isoflavones and their content increased by approx. 28, 44, 34, and 47%, respectively, compared to the control. low amounts of aglycones were found; moreover, their content decreased by ca. 19–30%. Our research can be important for obtaining a natural product enhanced with strontium and isoflavones, which contribute to prevention of osteoporosis associated with endog- enous oestrogen deficits

It is demonstrated that amino acids such as glycine, betaine, and lysine may be applied as selective copper leaching agents in the process of hydrochemical oxidation of chalcopyrite at atmospheric pressure and in the temperature range 25÷65 °C. The advantage of the proposed method is its relative environmental friendliness. Solid samples of the initial mineral and leached chalcopyrite residues were analyzed by X-ray diffraction analysis, and Mössbauer and Raman spectroscopy. The results of the atomic absorption analysis showed that copper's extraction efficiency increased when the experiment's duration was raised and when the process temperature was higher. The leaching ability of amino acids was evaluated by the specific leaching rate of chalcopyrite in these reagents. Amino acid leaching was thought to be mainly due to the complexation of copper (II) glycinates, copper (II) betainates and copper (II) lysinates. This reaction can be enhanced by adding hydrogen peroxide to the system as an oxidant. It was determined that glycine was more effective as a leaching agent than betaine and lysine. Based on experimental data, it was assumed that the most possible mechanism is selective copper leaching due to the formation of chalcopyrite-based solid solution Cu1-XFeS2.

The aim of the paper was to present the possibility of applying the novel method (RP-HPLC ion pair method) for the simultaneous dissolution determination of combined tablet dosage form containing acetylsalicylic acid and glycine in pharmaceutical industry. The samples were gradient eluted using a Pursuit XS Ultra C18 column (150x3.0 mm, with a particle size of 2.8µm) with variable composition of mobile phase A (1-heptanesulfonic acid sodium salt aqueous solution (2.8 g/L), pH 2.2 ± 0.05 adjusted with orthophosphoric acid) and phase B (methanol). The detection was carried out at 210 nm with a consist flow rate of 0.4 mL min−1. The method was validated by determining precision (repeatability and intermediate precision), accuracy, specificity, linearity, range, system suitability, robustness and stability in accordance with ICH guidelines. The method was accurate, precise and linear within the range of 0.03 – 0.18 mg mL−1 for acetylsalicylic acid and 0.016 – 0.096 mg mL−1 for glycine. The method is simple, convenient and suitable for analyzing acetylsalicylic acid and glycine in pharmaceutical formulations. The method could also be used for routine assay determination after small modification of sample preparation.

Among the methods applied to ensure optimal pharmaceutical availability of a drug is the incorporation of solid dispersions, i.e. combinations containing a therapeutic substance and a carrier deprived of its pharmacological activity. While manufacturing solid dispersions, special attention must be paid to carriers with a polymeric structure and hydrophilic properties, e.g. polyvinylpyrrolidone (PVP) and also phosphatidylcholine (PC). The aim of this study has been to evaluate the influence of the carriers PVP and PC 45 on pharmacokinetic parameters of Mg2+ absorption from Mg(Lev)2, Mg(LevGly), Mg(LevArg) as well as from solid dispersions containing these salts. The o/w partition coefficient was determined and the log P value calculated for pure salts and for solid dispersions containing the salts during this study. The process of Mg2+ absorption was examined in vitro on a model of the rat’s small intestine. Our analysis of the results indicates that addition of PVP or PC 45 to solid dispersions (containing magnesium levulinate salts) significantly improves the degree of Mg2+ ion absorption. It has been found that addition of PVP and PC 45 to solid dispersions with magnesium levulinate salts significantly influences the rate of Mg2+ absorption from the formulations. Moreover, the results indicate that additional ligand (glycine or arginine) in the structure of magnesium levulinate triggers the effect consisting in depressed lipophilicity for these compounds. Using the PVP or PC 45 carriers for making solid dispersions containing magnesium levulinate and derivatives with glycine or arginine ligands is quite a promising solution for attaining improved pharmaceutical availability of drugs.
Jedną z metod osiągnięcia optymalnej dostępności farmaceutycznej leku jest zastosowanie stałych rozproszeń, układów zawierających substancję leczniczą i nośnik, który jest pozbawiony własnego działania farmakologicznego. Przy wytwarzaniu stałych rozproszeń na szczególną uwagę zasługują nośniki o budowie polimerowej i właściwościach hydrofilowych. Należą do nich m.in. poliwinylopirolidon (PVP), a także fosfatydylocholina. Celem badań była ocena wpływu wybranych nośników: poliwinylopirolidonu (PVP) i roztworu fosfatydylocholiny (PC 45) na poprawę parametrów farmakokinetycznych procesu wchłaniania jonów Mg2+ z soli lewulinianu magnezowego Mg(Lev)2, Mg(LevGly), Mg(LevArg) oraz ze stałych rozproszeń zawierających te sole. W badaniach wyznaczono współczynnik podziału o/w i wyliczono wartość log P dla czystych soli oraz dla stałych rozproszeń zawierających te sole. Badania procesu wchłaniania jonów Mg2+ przeprowadzono metodą in vitro na modelu jelita cienkiego szczura. Wykazano, że dodatek PVP lub PC 45 do stałych rozproszeń zawierających sole lewulinianu magnezowego znacząco wpływa na poprawę stopnia wchłaniania. Na podstawie tych badań stwierdzono, że dodatek PVP lub PC 45 do stałych rozproszeń z solami lewulinianu magnezowego istotnie (p=0.01) wpływa na szybkość procesu wchłaniania jonów Mg2+ z otrzymanych formulacji. Ponadto wyniki badań wskazują, że dodatkowy ligand (glicyna, arginina) w strukturze cząsteczki lewulinianu magnezowego wywołuje efekt zmniejszenia lipofilowości dla tych związków. Zastosowanie nośników PVP lub PC 45 do wytwarzania stałych rozproszeń zawierających lewulinian magnezowy i pochodne z ligandami glicyny lub argininy jest obiecujące w poprawie dostępności farmaceutycznej.

Two experiments were conducted during 2010 and 2011 main cropping seasons under different farmer fields at Pawe District of Northwestern Ethiopia in order to evaluate different commercially produced microbial inoculants, referred to as commercial rhizobial strains, on soybean yield. The first experiment was conducted with the aim of evaluating four commercial rhizobial strains (Legume fix, TSBF-531, TSBF-442 and MAR-1495) on soybean yield, while the second experiment conducted with the aim of evaluating and selecting the best rhizobial strain from the two rhizobial strains (Legume fix and MAR-1495). Non-inoculated soybean with and without the recommended rate (RR) of nitrogen (N) fertilizer treatments also included in the experiments. Treatments were arranged in a single replicate per field. Recommended rate of phosphorous (P) fertilizer was applied uniformly to all treatments. The two experiments analysis of variance indicated that there was significant difference (P < 0.05) among treatments in number of nodules, nodule weight, biomass yield and grain yield of soybean. Accordingly, the first experiment result indicated that the highest number of nodules, nodule weight, biomass yield and grain yield were recorded by using rhizobial strains of legume fix and MAR-1495. The second experiment result revealed that rhizobial strain MAR-1495 resulted consistent and highest number of nodules, nodule weight, biomass yield and grain yield of soybean. The increment in grain yield over the un-inoculated control, legume fix and full RR of N fertilized treatments recorded by MAR-1495 was 25.5, 9.7 and 9.9%, respectively. Moreover, inoculation of soybean seeds with rhizobial strain MAR-1495 also resulted to higher protein content in soybean plant tissue. Therefore, this study recommends use of the most suitable commercial rhizobial strain MAR-1495 for the study area to inoculate seeds before sowing for the production of soybean crop.

Soybean (Glycine max (L.) Merrill) is an exotic crop to Ethiopia. Hence, inoculation with effective Bradyrhizobia spp. is crucial to improve its productivity. This study was therefore conducted in West Gojam Zone of Amhara Region in 2015 and 2016 aimed at evaluating the effect of bradyrhizobia strains (MAR-1495, SB-12 and TAL-379) with and without phosphorus fertilizer (23 kg P2O5 ha-1) on root nodulation and yield of soybean, and on residual soil nitrogen. The treatments were laid in randomized complete block design including a satellite control treatment with three replications. Effect of the use of rhizobia strains and P fertilizer on nodulation, yield and yield components of soybean was found statistically significant (P<0.05). The highest grain and dry matter yields of 2.7 and 6.4 t ha-1, respectively, were obtained from MAR-1495+P which was statistically at par with the yields obtained from MAR-1495 alone. A grain yield advantage of 30.8% over the control treatment was found from the use of MAR-1495 alone. Similarly, the maximum number of effective nodules (14.9) per plant was recorded from MAR-1495+P which was statistically at par with the number of effective nodules (11.6) per plant counted from MAR-1495 inoculated treatment. The highest residual soil N exceeding the control treatment by 0.029% (0.29 g total N per kg soil) was obtained from MAR-1495 alone. Therefore, inoculation with the strain MAR-1495 was found the most effective for improved soybean production in the study district and similar agroecologies.

Hydrate complexes of magnesium aspartate, magnesium aspartate glycinate, magnesium aspartate arginate have been synthesized and characterized by the elemental and spectral analysis. The formulae of the magnesium complexes were Mg(C4H6O4N) • 4H2O, Mg(C6H10O5N2) • 5H2O, and Mg(C10H19O5N5) • 7H2O. The main thermal process: dehydration of the complexes, was studied using the thermoanalytical methods: differential thermal analysis (DTA), thermogravimetric analysis (TG), and differential scanning calorimemtry (DSC) measurements. The influence of additional ligands (glycinate or arginate) introduced to the structure of magnesium aspartate on the physicochemical properties of the newly synthesized compounds was analysed.
Metodą analizy elementarnej i spektralnej scharakteryzowano syntezowane hydraty kompleksów asparaginianu magnezowego, glicynianoasparaginianu magnezowego, arginiano-asparaginianu magnezowego, Kompleksy magnezowe o wzorach : Mg(C4H6O4N) • 4H2O, Mg(C6H10O5N2) • 5H2O, and Mg(C10H19O5N5) • 7H2O. Zastosowano metody termoanalityczne: różnicową termiczną analizę (DTA), termograwimetryczną analizę (TG), skaningową kalorymetrię różnicową (DSC) do badania głównego procesu termicznego: dehydratacji badanych kompleksów. Potwierdzono wpływ wprowadzonych do struktury asparaginianu magnezowego dodatkowych ligandów glicyny lub argininy na fizykochemiczne właściwości nowo syntezowanych związków.

The purpose of the studies conducted in the years 1996-1998 was to determine the quantitative and qualitative composition of bacterial and fungal communities in rhizosphere of soybean cultivated in monoculture and non-rhizosphere soil. Besides, the proportion of bacteria and fungi, which were distinguished by their antagonistic effect towards soil-borne pathogens was established. A microbiological analysis of 1g of dry weight of soil from rhizosphere of soybean resulted in 3.21 x 10⁶ to 8.70 x 10⁶ bacterial colonies and from 70.51 x 10³ to 123.74 x 10³ fungal colonies. In the case of non-rhizosphere soil, 3.50 x 10⁶ to 4.75 x 10⁶ bacterial colonies and 16.16 x 10³ to 51.38 x 10³ fungal colonies were obtained. Besides, soybean cultivation in monoculture had a negative effect on the number of antagonistic isolates of bacteria (Bacillus spp., Pseudomonas spp.) and fungi (Gliocladium spp., Penicillium spp., Trichoderma spp.). Smaller numbers of antagonistic bacteria and fungi in rhizosphere soil of soybean cultivated in monoculture as compared to non-rhizosphere soil, can prove little biological activity, which results in a worse phytosanitaty condition of the soil.
Przedmiotem badań przeprowadzonych w latach 1996-1998 była gleba ryzosferowa soi uprawianej w monokulturze oraz gleba pozaryzosferowa. Analiza mikrobiologiczna wykazała, że w 1 g s.m. gleby ryzosferowej średnia liczebność bakterii oraz grzybów była większa, aniżeli w glebie pozaryzosferowej. W próbach badanych gleb liczebność bakterii z rodzaju Bacillus była zbliżona. W przypadku bakterii z rodzaju Pseudomonas ogólna ich liczebność była prawie trzykrotnie większa w ryzosferze soi, aniżeli w glebie pozaryzosferowej. Badania laboratoryjne wykazały, że znacznie więcej antagonistycznych Bacillus spp., Pseudomonas spp., Gliocladium spp., Penicillium spp. i Trichoderma spp. wystąpiło w glebie pozaryzosferowej, aniżeli w ryzosferze soi. Mniejsza liczebność antagonistycznych bakterii i grzybów w ryzosferze soi uprawianej w monokulturze może świadczyć o małej aktywności biologicznej mikroorganizmów, co przyczynia się do pogorszenia fitosanitarnego stanu gleby.

In agricultural practice, there is a need to change the forms of fertilizers to be friendlier to the natural environment and human health. To this end, research has been carried out on the partial replacement of fertilisers containing nitrates or ammonium compounds with amino acid nitrogen. The present studies assessed the effect of L-glycine on growth parameters and on the content of selected nutrients in the butterhead lettuce grown in an unheated greenhouse in the spring. The treatment consisted of foliar L-glycine application at the following concentrations: 0, 20, 40, 60, 80, 100, 120, 140, 160, 180, 200, 220 and 240 mg∙L-1, with a solution dose of 50 ml∙m-2. Foliar application of L-glycine improved the butterhead lettuce growth parameters. The most favourable values of the head weight, root system weight and the number and length of leaves were recorded for the plants fed with doses ranging from 80 to 120 mg∙L-1. The weight of the head and the root system was most favourable after foliar feeding with 120 mg∙L-1 of L-glycine. Additionally, a high value of the SPAD leaf greenness index was noted as a response to the same dose, indicating a high level of leaf nitrogen nutrition. However, foliar feeding with L-glycine had no statistically significant effect on the nutritional value of lettuce, i.e. the content of dry matter, protein, sugars and ascorbic acid.

Soybean cultivation is becoming more and more popular in many countries, including Poland, due to the high nutritional value of its seeds, its resistance to diseases and pests, as well as lower propensity to lodging compared to other legumes. Under the climatic conditions of Poland, soybean yield is largely dependent on weather conditions in a particular growing season, primarily temperature and rainfall. Studies on the effect of tillage systems used for soybean cropping under the climatic conditions of Poland are scarce. This proves the need to continue such research. The aim of this study was to compare the effect of conventional tillage (CT) and no-tillage (NT) on yield, weed infestation and seed qualitative composition of the ‘Mazowia’ soybean cultivar grown under the conditions of the Lublin Upland. A field experiment was carried out in the period of 2009-2012 at the Czesławice Experimental Farm (51°18′23″N 22°16′2″E). In 2010 the soybean plantation was terminated due to adverse weather conditions and related inhibited plant emergence. The experiment was set up on loess-derived grey-brown podzolic soil as a split-block design in four replicates. The present study showed that the soybean seed yield obtained under conventional tillage conditions was higher by 14.6% compared to that obtained under no-tillage. A higher plant density after emergence and before harvest as well as a higher plant height were found in the conventional tillage treatment. The protein, oil and fibre content in soybean seed was significantly affected only by weather conditions in particular growing seasons. A higher weed weight and richer floristic composition of weeds were found under no-tillage conditions in relation to conventional tillage.
Uprawa soi w wielu krajach, jak i w Polsce staje się coraz bardziej popularna ze względu na dużą wartość odżywczą nasion, odporność na choroby i szkodniki oraz mniejszą skłonność do wylegania w porównaniu z innymi roślinami strączkowymi. Plonowanie soi w warunkach klimatycznych Polski jest w dużym stopniu uzależnione od przebiegu warunków pogodowych w danym sezonie wegetacyjnym, głównie temperatury i opadów. Badania dotyczące wpływu sposobu uprawy roli pod soję prowadzone w warunkach klimatycznych Polski są nieliczne. Świadczy to o potrzebie dalszej ich kontynuacji. Celem niniejszej pracy było porównanie wpływu konwencjonalnej uprawy roli (CT) oraz siewu bezpośredniego (NT) na plonowanie, zachwaszczenie i skład jakościowy nasion soi odmiana Mazowia, uprawianej w warunkach Wyżyny Lubelskiej. Badania polowe przeprowadzono w latach 2009-2012 w Gospodarstwie Doświadczalnym Czesławice (51°18′23″N 22°16′2″E). W 2010 roku ze względu na niesprzyjające warunki atmosferyczne i wynikające z nich utrudnione wschody plantację soi zlikwidowano. Eksperyment założono na glebie płowej wytworzonej z lessu, metodą split-block w czterech powtórzeniach. Przeprowadzone badania wykazały, że w warunkach uprawy płużnej (CT) plon nasion soi był wyższy o 14.6% w porównaniu do uzyskanego w siewie bezpośrednim (NT). Na obiekcie z uprawą płużną (CT) stwierdzono również większą obsadę roślin po wschodach i przed zbiorem oraz wysokość roślin. Na zawartości białka, tłuszczu i włókna w nasionach soi wpływał istotnie jedynie przebieg warunków pogodowych w poszczególnych sezonach wegetacyjnych. W warunkach siewu bezpośredniego soi (NT) stwierdzono znacznie większą masę chwastów oraz bogatszy skład florystyczny chwastów w porównaniu do uprawy tradycyjnej (CT).
