The fiberglass pipes were used for transportation of water in order to maintain the pressure in the oil field of Zarzaïtine (Region of In Amenas located in the south of Algeria). During the maintenance work in summer, a season well known for its extensive heat, the deposits contained in the pipe, and after its contact with the atmosphere, have caused a smoke and it was followed by a fire that ignited a portion of the pipe. To give an answer to the causes of this phenomenon, we have first incriminated the role of specific bacterial species and therefore we have made a microbiological analysis of the deposits collected from the site of the incident. The obtained results revealed the presence of a heterogeneous microbial population with a high concentration level of sulfate-reducing bacteria (SRB), methanogenic bacteria (MB), yeasts and fungi. The interaction between the different species of bacteria and the organic matter contained in the deposits has generated the formation of methane which under the influence of the great heat burnt and the fire caused the ignition of the fiberglass line.

The aim of this study was to investigate the bacteriological quality of 5 kinds of vegetables (lettuce, dill, radish, beetroot, carrot) and their rhizosphere soil, originating from conventional farms located in the Lublin Province of Eastern Poland. A total number of 35 samples of fresh vegetables (FV) taken immediately from soil, 35 samples of soil from rhizosphere of these vegetables (SR) and 35 samples of vegetables sold at retail in the markets in Lublin (VR) were examined. The samples were analysed for the content of: aerobic mesophilic bacteria (AMB) grown at 30 °C and 37 °C, Gram-negative bacteria of Enterobacteriaceae family, faecal coliform (FC) bacteria, Salmonella spp., and Clostridium perfringens. Median AMB values determined at 30 °C for FV, SR and VR were 5.27, 5.00, and 5.00 log10 CFU g-1, respectively, being significantly greater compared to those recorded at 37 °C. The exceeding of the threshold value of 6.0 log10 CFU g-1 proposed by Gelosa (1998) was noted only in 5 FV samples grown at 30 °C (14.3%), and in 3 FV samples grown at 37 °C (8.6%). The threshold value was never exceeded in SR and VR samples. Median concentrations of Enterobacteriaceae determined for FV, SR and VR were 4.03, 3.87, and 3.04 log10 CFU g-1, respectively. Eleven species of Enterobacteriaceae were identified in the FV, SR and VR samples. The percent of samples containing Escherichia coli was greatest for VR (22.9%), smaller for FV (17.1%) and smallest for SR (5.7%). The median concentrations of the faecal coliform bacteria (FC), determined by culture at 44oC, were low, amounting to 1.000 log10 CFU g-1 for FV and SR and 0.00 for VR. All examined vegetable and soil samples tested negative for the presence of Salmonella. The median concentrations of Clostridium perfringens were low, amounting to 0.00 log10 CFU g-1 for all categories of samples. This bacterium was relatively common in soil samples with the prevalence of 40.0%, but very rare in vegetable samples (occurring in 5.7% of FV and in none of VR samples). In conclusion, the results of the present study generally indicate that the microbiological quality of Polish vegetables grown on conventional farms is satisfactory and safe for consumers.

The compositions of bacterial communities and populations during aerobic stabilization of livestock manure have been investigated, focusing on how the aging of feedstock affected the bacterial diversity of the composting mass. The livestock manure was divided into two groups – aged and fresh, and then used to prepare the feedstock with additives. Composting experiments were carried out for 15 days using a pilot-scale batch reactor with vacuum-induced aeration. Two different aeration rates were applied to the batch reactor, and their effectiveness was evaluated. Changes in total heterotrophic bacteria count and moisture content were monitored. The associated changes in bacterial community compositions were characterized using 16S ribosomal ribonucleic acid (rRNA) gene sequencing. The Firmicutes in the fresh manure decreased from 48 to 13% for the first ten days, and the dominant phylum shifted to the Proteobacteria (29%), Bacteroidetes (23%), Actinobacteria (20%), and others(15%). Under the given conditions, the use of relatively fresh manure was essential to preserve thediverse bacterial populations in the feedstock and enhance the bacterial diversity during aerobic stabilization.More research should be performed to investigate the degradability of emerging contaminants (e.g., antibiotics) in livestock manure using an engineered composting system providing well-controlled environmental conditions.

Increasing interest in organic farming and consequently in the application of natural fertilisers poses a risk of modifying qualitative and quantitative composition of microorganisms’ populations living on plants fertilised with these fertilisers. Composition of epiphytic microflora on plant material intended for ensilage affects fermentation processes and stability and hygienic quality of obtained silage. Studies carried out in the years 2009-2011 were aimed at assessing the effect of fertilisation of permanent meadow with natural fertilisers (manure and liquid manure) and mineral fertilisers (NPK) on the number of selected groups of epiphytic microorganisms living on meadow sward. Plant samples were taken three times in vegetation seasons (before the 1st, 2nd and 3rd cut). In total, 207 samples of vegetation were analysed for: total number of aerobic bacteria, bacteria of the genus Enterobacteriaceae, yeasts and mould fungi. No differences in the number of studied groups of microorganisms living on meadow plants fertilised with different fertilisers applied at different doses were found. However, the number of particular groups of microorganisms depended mainly on the time of sampling.
Rosnące zainteresowanie rolnictwem ekologicznym oraz wynikające z jego zasad stosowanie nawozów naturalnych niesie ze sobą niebezpieczeństwo modyfikacji składu jakościowego i ilościowego populacji mikroorganizmów naturalnie bytujących na roślinności nawożonej tymi nawozami. Skład mikroflory epifitycznej znajdującej się w materiale roślinnym przeznaczonym do zakiszania wpływa na przebieg procesów fermentacyjnych, stabilność i jakość higieniczną uzyskanej kiszonki. Celem badań przeprowadzonych w latach 2009-2011 była ocena wpływu nawożenia łąki trwałej nawozami naturalnymi (obornikiem i gnojówką) i nawozami mineralnymi NPK na liczebność wybranych grup epifitycznych mikroorganizmów bytujących na runi łąkowej. Próbki roślin do badań pobierano trzykrotnie w trakcie sezonu wegetacyjnego (przed I, II i III pokosem). Łącznie przebadano 207 próbek zielonki. W ramach badań oceniano: ogólną liczebność bakterii tlenowych, bakterii z rodzaju Enterobacteriaceae oraz drożdży i grzybów pleśniowych. W wyniku przeprowadzonych badań nie wykazano różnic w liczebności badanych grup drobnoustrojów bytujących na roślinności łąkowej nawożonej różnymi nawozami i ich dawkami. Stwierdzono jednak, że liczebność poszczególnych grup mikroorganizmów zależała przede wszystkim od terminu poboru próby.

Opis:
One of the most important fractions of soil organic matter which has significant environmental and agricultural importance is humus. The specific humus compounds of soil include the complex of amorphous organic humic substances which include humic acids, fulvic acids and humins. The effectiveness of the natural formation of active humic substances during composting processes depends on the chemical composition of organic residues and the environmental conditions influencing the development and activity of native microorganisms. The aim of the project was to build innovative composting boxes for chicken manure that allow to effectively obtain a fertilizer with the highest possible content of humic acids, by creating perfect and controlled conditions for the rapid development of indigenous thermophilic microorganisms and by combining it with the owned bio-acceleration technology. For perfect conditions (temperature, humidity, oxygen, pH) for the controlled growth of microorganisms, an algorithm of the dependence of the box operation parameters on the course of the biological process, was built. The product obtained after composting is an easily digestible complex organic fertilizer of brown-black colour, pH 7.5-8.0 and NPK 5:3:4 and a high content of native humic acids. Its use increases the growth of green mass of plants and improves soil fertility, which has been proved by the conducted pot research.
Humus jest jedną z najważniejszych frakcji materii organicznej gleby, która ma istotne znaczenie dla środowiska i rolnictwa. Poszczególne składniki próchnicy obejmują grupę amorficznych substancji próchnicznych, w tym kwasy humusowe, fulwowe i huminy. Skuteczność naturalnego tworzenia się aktywnych substancji próchnicznych podczas procesu kompostowania zależy od składu chemicznego osadów organicznych i warunków środowiskowych, które mają wpływ na rozwój i aktywność natywnych mikroorganizmów. Celem projektu było zbudowanie innowacyjnych skrzyni kompostowych na odchody kurze pozwalające na skuteczne uzyskanie nawozu o możliwie najwyższej zwartości kwasów humusowych poprzez stworzenie idealnych i kontrolowanych warunków dla szybkiego rozwoju miejscowych mikroorganizmów termofilnych oraz poprzez połączenie ich z własną technologią bio-akceleracji. Dla uzyskania idealnych warunków (temperatura, wilgotność, tlen, pH) dla kontrolowanego wzrostu mikroorganizmów, stworzono algorytm zależności parametrów operacyjnych skrzyni. Produkt uzyskany w drodze kompostowania jest łatwo rozkładalnym złożonym nawozem organicznym o brązowo-czarnym kolorze, pH 7,5-8,0 oraz NPK 5:3:4 i wysokim poziomie natywnych kwasów humusowych. Jego zastosowanie pozytywnie wpływa na wzrost zielonej masy roślin i poprawia żyzność gleby, co zostało udowodnione przeprowadzonymi badaniami.

The paper is aimed at summarising long-term studies on the effectiveness of biological ensilage stimulators containing homo- and hetero-fermentative cultures of lactic acid bacteria used in the conservation of meadow sward intended for fodder or energy production. Study material consisted of silages from wilted meadow sward ensiled in big cylinder bales wrapped in a plastic film with the addition of four bacterial preparations made in the Institute of Agricultural and Food Biotechnology: Lactosil, Lactacel-L, Lactosil Plus and Lactosil Biogaz. Control silages were made of the same plant material without additives. Studies included the assessment of aerobic stability of silages and changes in chemical composition and microbiological profile during silage samples storage in aerobic conditions. Dry mass, pH of fresh silage and the amount of lactic, acetic and butyric acids were determined in fresh silage samples and in samples after 12 days of aerobic incubation. The number of yeasts, moulds and lactic acid bacteria was determined as well. The efficiency of applied biological additives varied during the process of ensiling meadow sward, mainly due to their different composition. Lactacel-L appeared most efficient in improving fodder quality from among all applied additives. Lactosil was the best in prolonging aerobic stability. The addition of Lactosil Biogaz helped improve aerobic stability and decreased the content of lactic acid in raw material intended for energy production.

A new strategy of enrichment of polyphosphate accumulating organisms (PAO) and denitrifying polyphosphate accumulating organisms (DPAO) at low temperature ranging from 8 °C to 11 °C was demonstrated through two lab-scale reactors operated in sequential anaerobic-aerobic (AO) or anaerobic-anoxic (AA) conditions. It was found that the AO reactor is able to achieve a good phosphorus removal performance after 40 days of operation, while a similar stable phosphorus removal can be obtained in the AA reactor after 80 days. This result suggests that the enrichment of PAO was easier than that of DP AO at low temperature. Through switching batch tests, when DPAO is exposed to aerobic conditions, it can immediately exhibit a good phosphorus removal similar to that under anoxic conditions, while PAO can only present poor phosphorus removal when exposed to anoxic conditions, suggesting that two different types of Accumulibacter were enriched both in AA and AO reactors. Microbial analysis with fluorescence in situ hybridization (FISH) and DAPI (4',6-diamidino-2 -phenylindole) staining revealed that Accumulibacter was dominant both in the two reactors, accounting for 61.6% and 79.3% of all bacteria in AA and AO reactors, respectively. Although the different amount of Accumulibacter was enriched in the two reactors, the similar microbial morphologies were observed by using scanning electron micrograph (SEM), both presenting long-rod morphology. This kind of Accumulibacter may display affinities for sodium acetate used as the carbon sources here. This strategy proposed in this study was shown to be effective in achieving a very high enrichment of Accumulibacter at low temperature by linking chemical analysis with microbial observation.
