The version of LAMS described in this paper is Version 2.1.1 . It is hoped that study of this extended paper and its preceding introductory paper (Alexander 2008a, An overview of LAMS) will be of particular value to those wishing to gain more confidence in the proficient utilisation of LAMS and to those intending to engage in debate on current LAMS issues. The paper’s thematically diverse core sections may also heighten reader awareness of a pressing issue: the need to bring together the treasures of progressively more hard-core and swiftly changing computer technologies with the beliefs of an increasingly discerning education community.

Modern language-lab management software provides an increasingly sophisticated range of functions that appear to have been designed to change or supposedly improve certain aspects of traditional classroom language teaching. However, using management software in a way that regularly puts a computer between the teacher and student may create an unnatural or unnecessary ‘electronic go-between’ in the dynamics of traditional teacher-student interaction. Human communication contains many rich visual cues that have always been an important part of the learning experience, and it is this age-old ‘human to human’ learning experience that should remain vitally important in the language lab.

Probiotic bacteria have positive impacts on a numerous of physiological functions including immunomodulation and also on prevention of various intestinal diseases. However, acidic environment of the stomach and heavy conditions of freeze-drying process decrease survival of these bacteria. There are necessary researches on cheap raw materials that increase cell protection. The aim of the present study was to determine the impact of whey and other compounds on survival Lactobacillus gasseri during the freeze-drying process and storage. There were prepared culture media for cell protection during lyophilisation, which contained a variety of mixture whey, skim milk, sucrose, starch, maltodextrin and whey protein. Lyophilisate was rubbed in aerobic and anaerobic condition with lower humidity. Survival tests were investigated directly after freeze-drying process and then periodically during storage under refrigeration. Within 6 months of storage bacterial viability was similar in the samples containing liquid whey except for media containing starch. The results of viability did not deviate than those obtained with using the reference medium, which contains skim milk and sucrose. This shows that the liquid whey may be successfully used to protect the lactic acid bacteria during lyophilisation. Moreover, the use of whey can reduce the amount of waste produced in the dairy industry.

This paper is devoted to the usage of image processing techniques in lab-on-a-chip devices for a cell detection and tracking. Advantages and disadvantages of image processing in data acquisition from lab-on-a-chip are described. A modified multiparametric object tracking method is presented. The method was tested on the lab-on-a-chip setup with living euglena and has shown plausible results for video with a low frame rate.

The authors report a new construction of a heater, which can be applied in Lab-on-a-chip systems. The heater was fabricated on a ceramic plate as a solid support. A commercially available paste DuPont 5091D with positive temperature coefficient (PTC) was used as a resistive material. Next, metallic conductors were created using gold sputtering in argon enhanced plasma environment. Finally, the gold layers were bonded with connection wires. The developed heater can be integrated with microsystems dedicated for bioanalysis, enzymatic reactions and cell cultures.

Among proposed alternatives to antibiotic growth promoters (AGPs), most effective in improving of efficiency of farming and having positive impact on animal welfare are probiotic preparations containing selected strains of lactic acid bacteria (LAB). It is known that essential oils exert ability to inhibit pathogenic bacteria. The aim of this study was to assess the pos-sibility of enrichment of probiotic preparations for animals in selected substances of vegetable origin, in order to increase the effectiveness of antimicrobial activity LAB directed against pathogenic microorganisms. The impact of three selected essential oils (thyme, sage and peppermint) on the capacity to growth, viability and antibacterial activity of probiotic or potentially probiotic strains of lactic acid bacteria (LAB) was evaluated. The assessment of the antibacterial activity of the LAB was conducted in relation to the bacteria strains pathogenic for animals (indicatory strains). The results show that the reaction of lactic acid bacteria strains for the presence of vegetable oils is very individual, depending on both the strain and the type of oil. All the tested plant’s oils inhibited the growth of strains of lactic acid bacteria. The minimum inhibitory concentration of essential oils varied. In the case of peppermint oil and thyme oil LAB growth inhibition was observed at a concentration of 0.1% (v/v) and in relation to the sage oil this effect was found with 0.2 %.(v/v) average in medium. The viability of the tested LAB was observed in media containing higher concentrations up to several per cent of essential oils. In the presence of oils in concentrations affecting growth of LAB strains antimicrobial activity against pathogenic bacterial species was not observed. The addition of essential oils did not increase the antimicrobial activity of lactic acid bacteria. It was observed that some of indicatory pathogenic bacteria strains were resistant to higher dilution of oils than LAB. There was no synergistic interaction found between LAB and essential oils what suggest the possibility of using essential oils with LAB only to increase the flavor qualities of preparation which will contribute to a better feed intake by the farm animals.
W grupie proponowanych zamienników antybiotykowych stymulatorów wzrostu (ASW) największą skutecznością działania w podwyższaniu efektywności hodowli i pozytywnym wpływaniu na dobrostan zwierzą charakteryzują się preparaty probiotyczne, zawierające wyselekcjonowane szczepy bakterii fermentacji mlekowej (LAB). Wiadomo również, że roślinne olejki hamują wzrost niektórych bakterii patogennych. Celem pracy była ocena możliwości wzbogacenia preparatów probiotycznych dla zwierząt w wybrane substancje pochodzenia roślinnego, tak aby zwiększyć skuteczność antymikrobiologicznego oddziaływania LAB, skierowaną przeciw drobnoustrojom patogennym. Zbadano wpływ trzech wybranych olejków eterycznych (tymiankowego, szałwiowego i miętowego) na wzrost, przeżywalność i aktywność antybakteryjną probiotycznych lub potencjalnie probiotycznych szczepów bakterii fermentacji mlekowej (LAB). Ocenę aktywności antybakteryjnej lab przeprowadzono w odniesieniu do panelu szczepów patogennych dla zwierząt hodowlanych. Otrzymane wyniki wskazują, że reakcja szczepów bakterii fermentacji mlekowej na obecność olejków roślinnych jest bardzo indywidualna, zależna zarówno do szczepu jak i rodzaju olejku. Wszystkie badane olejki eteryczne hamowały wzrost szczepów bakterii fermentacji mlekowej, jednakże stężenia olejków hamujące wzrost były zróżnicowane. W przypadku olejku miętowego i tymiankowego hamowanie wzrostu LAB obserwowano już przy stężeniu 0,1%, a w przypadku olejku szałwiowego taki efekt stwierdzono przy jego obecności w podłożu w stężeniu 0,2%. Natomiast przeżywalność badanych LAB obserwowano w podłożach zawierających wysokie, sięgające kilku procent stężeniach olejków eterycznych.. Aktywność antybakteryjną szczepów LAB wobec gatunków bakterii patogennych obserwowano w obecności nie wpływających na wzrost stężeń olejków. Dodatek olejków eterycznych nie powodował zwiększenia aktywności antymikrobiologicznej bakterii fermentacji mlekowej. Wykazano możliwość stosowania olejków eterycznych, w odpowiednich stężeniach, jako jednego z komponentów wieloskładnikowych preparatów probiotycznch przeznaczonych dla zwierząt. Dodatek olejku eterycznego miałby na celu uatrakcyjnienie walorów zapachowych preparatu a przez to jego lepsze pobieranie przez zwierzęta hodowlane

Opis:
The fluorometric detection in lab-on-a-chip devices for life-sciences is one of the main detection methods. Well know from “large-scale” analytical chemistry methodologies and instrumentation has been applied in lab-on-a-chip solutions. In most cases, optical instrumentation for fluorescence induction and detection is based on configuration and components borrowed from large laboratory instruments. As a result, optical instrumentation surrounding lab-on-a-chip is bulky, expensive and dedicated for operation only inside laboratory. In this paper a discussion on fluorescence detection in lab-on-a-chip is carried out. A novel image sensor – based detection instrumentation co-working with a “clever” software is described. Instrumentation is dedicated for operation in portable and low-cost devices for different life-science applications. Examples of applications of the novel method and instrumentation co-working with various lab-on-a-chips are presented on the base of author’s works.

Microfluidic devices called lab-on-a-chips utilize two kinds of characterization of a biosample, which are based on spectrofluorimetric and spectrophotometric methodologies. Lab-on-a-chips are equipped with an optical instrumentation and a software system that allow detecting of the optical signals and their processing into spectral characteristics. In the paper, technology of lab-on-a-chip (cytometer microdevice) and miniature silicon-glass field-emission light source (FELS) is described. Electrons emitted from a silicon cathode covered with carbon nanotubes excite light emission from nanocrystalline rare earth doped yttrium oxide phosphor (anode). The emitted light can be precisely tailored to the main absorption line of a fluorescent marker. The presented system has been successfully used for testing of the cow embryos as well as fluorescent-marked porcine oocytes.

Creativity is nothing but a mental process that entails the emergence of new ideas, concepts or new associations, links with existing ideas and concepts. Creative thinking is thinking that leads to original and appropriate solutions. In our dynamic and competitive world, designing a creative environment is a main goal and challenge for many companies and educational settings. Why is that so? Because it simply leads to the development of innovation and is existential for the well-being of the whole economy and civilization. One of the methods that contributes to the increase of creativity and the innovation level in education is the Project Based Learning (PBL) method. The aim of the study is to present creative methods in education based on PBL method using the example of the Smart-Up Lab (SUL) program organized by the consortium: Medical University in Lodz, Poland and EIT Health. PBL is a method of didactic work in which students implement the undertaking on the basis of assumptions established with the lecturer. At the beginning, goals and deadlines of work are set. The criteria for the assessment of individual work stages are clearly defined. The project’s task is most often performed in a group. After the end of the project, the results of students' work are presented to the public. During the implementation of the project, the lecturer acts as a coordinator and consultant - helps in making decisions regarding the division of tasks, supervises deadlines and assesses student performance by giving feedback. One of the examples using the Project Based Learning method is the Smart-Up Lab project. This project uses creativity and the PBL method to create innovative products, services and solutions to real-life challenges. This program has all features of a suitable teaching method in Science and may be a good example of the curriculum that may lead to the development of high-class specialists using creativity and innovative ideas in their everyday work.

The application value of the preparation, tested in the food industry is connected not only with therein contained strains of lactic acid bacteria but also with technological properties, determining the possibilities of obtaining the preparation in commercial form. The aim of the studies was to perform a preliminary evaluation of panel of 15 strains of lactic acid bacteria from Leuconostoc genus, being found in the collection of the Institute (IBPRS) in aspect of the possibility of their application in commercial starters for ensiling the common mushrooms A. bisporus. The conducted studies included cultures of bacteria in the laboratory conditions at temperature of 15°C and A. bisporus in MRS medium, separation of biomass from culture medium and preservation of the obtained biomass by lyophilisation method. The quantity of the obtained biomass and losses during separation process were specified, the concentration of lactic acid and acetic acid in supernatant was determined as well as survivability of bacteria during dehydration was recorded. The obtained results allowed performing the preliminary selection of the tested bacteria in aspect of their suitability for technological processes, connected with the ensiling of the mentioned mushrooms. Three LAB strains were eliminated from further studies as being technologically unsuitable whereas the remaining strains were qualified to the experiments which will be oriented to evaluation of organoleptic properties of the products, being ensiled with their participation.
Wartość zastosowanego preparatu testowanego w przemyśle spożywczym wynika nie tylko z zawartych w nim szczepów bakterii kwasu mlekowego lecz również z jego właściwości technologicznych, określających możliwości uzyskania produktu w formie handlowej. Celem badań było przeprowadzenie wstępnej oceny panelu 15 szczepów bakterii kwasu mlekowego z rodzaju Leuconostoc, będących w kolekcji Instytutu (IBPRS) w aspekcie możliwości ich zastosowania w komercyjnych starterach do zakiszania pieczarki A. bisporus. Przeprowadzone badania objęły oddzielenie biomasy z kultury medium oraz konserwację otrzymanej biomasy metodą liofilizacji z wykorzystaniem kultury bakterii w warunkach laboratoryjnych przy temperaturze 15°C z A. bisporus jako medium MRS. Określono: ilość uzyskanej biomasy, straty powstałe w trakcie procesu separacji, koncentracja kwasu mlekowego i octowego w supernatancie, przeżywalność bakterii w czasie dehydratacji. Uzyskane wyniki pozwoliły na wykonanie wstępnej selekcji badanych bakterii w aspekcie ich użyteczności w procesach technologicznych związanych z zakiszaniem wspomnianych grzybów. Trzy szczepy LAB zostały wyeliminowane z dalszych prac jako technologicznie nieodpowiednie, a pozostałe szczepy zakwalifikowano do eksperymentu nakierowanego na ocenę własności organoleptycznych produktów zakiszonych z ich udziałem.

The correct management of underground works, petroleum and gas reservoirs and geothermal appli-cations relies on the hydromechanical behaviour of rock masses. We describe a laboratory approach to measuring permeability for different types of rock specimens. A laboratory system was designed and set up using rock mechanics equipment (a servo-controlled hydraulic press, a Hoek cell, a pump for injecting water and a scale for measuring the volume of water flow). To verify the validity of the permeability measurements, tests were carried out on a reference porous rock (Corvio sandstone), with results showing good agreement with those published in the literature. Tests were subsequently carried out on artificially fissured granite specimens with different joint patterns, submitted to various confinement stresses up to 20 MPa. Results showed good agreement with traditional Klinkenberg test results. Other tests done with artificially fissured specimens are described for demonstrative purposes.

In this review we summarize stress factors that affect the lactic acid bacteria (LAB) and cause different molecular stress responses. LAB belong to a group of bacteria that is very widespread in food and beverages. They are present, and desired, in fermented products like yogurts, cheese, vegetables, meat or wine. In most of them, LAB are providing positive sensory and nutritive features. However, as harmless and desired microbes in one product, LAB can cause spoilage and a bad taste of others, especially in juices and beverages. LAB are resistant to many stress factors which allows them to survive in harsh environments. The most common stress factors they have to deal with are: heat, cold, acidity, NaCl and high hydrostatic pressure (HHP). Their ability to survive depends on their skills to cope with stress factors. Under stress conditions, LAB activate mechanisms that allow them to adjust to the new conditions, which can influence their viability and technological properties. This ability to adapt to different stress conditions may come from the cross-protection systems they have, as resistance to one factor may help them to deal with the other stress effectors. LAB are highly valuable for the food industry and that is why it is important to understand their stress response mechanisms.

Opis:
Linear alkyl benzene (LAB) is vastly utilized for the production of biodegradable detergents and emulsifiers. Predistillation unit is a part of LAB production plant in which that produced heavy linear paraffins (nC10-nC13). In this study, a mathematical model has been developed for heavy linear paraffins production in distillation columns, which has been solved using a commercial code. The models have been validated by the actual data. The effects of process parameters such as reflux rate, and reflux temperature using Gradient Search technique has been investigated. The sensitivity analysis shows that optimum reflux in columns are achieved.

Opis:
This paper describes a new contactless conductivity detector, whose electrodes are constructed of microchannels filled with solution of KCl - called pseudoelectrodes. The lab-on-a-chip microdevice was fabricated in poly(dimethylsiloxane) PDMS, using a moulding technique. The mould was made from a dry negative photoresist with a thickness of 50 µm. During the tests, the dimension and arrangement of pseudoelectrodes' microchannels were evaluated. The analyte was pumped into the microchannel using a syringe pump with a flow rate of 50 µL/min. Reproducible changes of the signal were obtained.
