Opis:
Juvenile idiopathic arthritis ( JIA ) spans several pediatric arthropathies that involve autoimmune responses. Primary genetic risk factors for JIA have been mapped to Major Histocompatibility Complex ( MHC ) class I and class II genes. Recent genome-wide association studies using SNP arrays have shown that the nonHLA genetic component of JIA involves multiple low-risk loci, reinforcing the notion that JIA is a complex genetic trait with a strong HLA component. In this work, with the goal of detailed mapping and analysis of lin kage disequilibrium ( LD ) patterns and associations with JIA in the extended MHC (x MHC ) region, we combined SNP data from Affymetrix SNP Array 6.0 and Immunochip ( IC ) platforms with high resolution typing of 8 classical HLA genes. A cohort of about 800 affected individuals and about 500 ethnically matched controls from the Cincinnati region, as well as secondary validation cohorts of affected individuals and matched controls from Germany were used to perform the analysis, and to assess reproducibility of the results across different platforms and p opulations. Accurate high resolution maps of linkage with classical HLA genes and association with individual HLA alleles were generated. High concordance of results obtained using Affymetrix SNP Array 6.0 and IC was observed, with an additional resolution and improved mapping of associat ions provided by the latter in some regions. Several new peaks of statistically signific ant and reproducible association with JIA outside the regions of strong LD with classical HLA genes were observed, including one peak in the class III region, and one in the telomeric end of x MHC . Conditional analysis provided further evidence that these associations appear to be independent of classical HLA genes studied here. The results and observed association patterns are further discussed in the context of other recent studies on autoimmune diseases, including the role of HLA DRB 1 in adult Rheumatoid Arthritis.

Coeliac disease is an autoimmune disorder with genetic predisposition. The aim was to determine the frequency of HLA-DQ2 and HLA-DQ8 in Czech and Slovak patients and the healthy population. The study included 127 patients and 66 healthy volunteers. HLA-DQ2 was identified in 85.03% patients, and 24.24% healthy individuals (P=0.0001; OR17.7632; CI=8.4347-37.4088). HLA-DQ8 was identified in 11.81% patients and 15.5% healthy individuals. HLA-DQ8 occurred more often in HLA-DQ2-negative patients compared to HLA-DQ2-positive patients (P=0.0494; OR3.5; CI 1.0428-11.7468). At least one of the studied HLA-variants was found more often in patients than in healthy individuals (P=0.0001; OR58.8; CI 7.6856-449.8602).

The human leukocyte antigen (HLA) system plays an important role in the acceptance of renal graft. Long and better graft survival has been reported in patients with HLA-identical siblings and a nonreactive cytotoxicity assay (CDC). New methods of HLA-typing and anti-HLA antibody detection techniques such as flow cytometry, solid-phase immunoassays, or antigen bead assays have further improved the outcomes of renal transplant recipients. In the present review, the explicit details of these methodologies are discussed in detail.

In this paper we present the design of a Grid HLA Management System (GHMS) supporting execution of High Level Architecture-based interactive applications in a Grid environment, while at the same time allowing for running HLA legacy codes to preserve backward compatibility with the HLA standard. In particular, we describe the following system components: an HLA-Speaking Service for multiple federates that interface the HLA application to the system, a Monitoring Service integrated with the OCMG monitoring system and HLA-based Benchmark Services informing the Broker Service of what can be expected from the application's behavior.

The HLA system is highly polymorphic and commonly used as a marker to understand core population genetical aspects such as stratification, human migration, predisposition to diseases and tissue transplantation compatibility. Although HLA alleles are generally treated as equidistant molecular units in population genetic studies, the diversity of DNA allelic frequencies among populations is also considered to be crucial to interpret the observed HLA polymorphisms. This study compares the variation of HLA DRB1 alleles across various subgroups of both Indian and African Populations, using their respective allelic frequencies. The HLA DRB1 allelic frequencies for 3,153 individuals of 32 subgroups of both Indian and African populations, were collected from the Allele Frequency Net Database (AFND). Correlation matrix and Grid diagram were compiled using the R statistical package. Allele frequencies were analyzed for the respective population subgroups using XLSTAT. The North East Indian populations were completely devoid of HLA DRB1* 16, which is fairly present in south Indian populations. The South Indian populations harbor both HLA DRB1* 10 and HLA DRB1* 15 at relatively higher levels but are very scarce of HLA DRB1* 07, HLA DRB1* 09, HLA DRB1* 12 and HLA DRB1* 13. The alleles HLA DRB1* 03, HLA DRB1* 04 and HLA DRB1* 07 could represent an epicenter for the admixture of African, North-East Indian and South Indian populations.

The article aims at reconstructing the defense of utilitarianism, a philosophical doctrine being the basis for legal positivism (which is the foundation of the constitutional concept of sources of law), undertaken by H.L.A. Hart. Hart took up this defense in the face of a significant increase in the interest of legal theorists in concepts related to natural law. Discussing the views of his master, J. Bentham, Hart also expresses his own deep doubts about the ideology of natural law, the adoption of which leads to the rejection of legal positivism deeply rooted in utilitarianism. Presented more than four decades ago, Hart’s remarks remain relevant today; modern thinkers still search an appropriate, other than referring to natural law, philosophical justification for a specific code of fundamental human rights.
Celem artykułu jest rekonstrukcja podjętej przez H.L.A. Harta obrony utylitaryzmu, doktryny filozoficznej będącej gruntem dla pozytywizmu prawniczego, który jest fundamentem konstytucyjnej koncepcji źródeł prawa. Obrony tej podjął się Hart w obliczu znaczącego wzrostu zainteresowania teoretyków prawa koncepcjami nawiązującymi do prawa natury. Omawiając poglądy swego mistrza, J. Benthama, Hart wyraża też własne, głębokie wątpliwości dotyczące ideologii praw naturalnych, której przyjęcie prowadzi do odrzucenia głęboko ugruntowanego w utylitaryzmie pozytywizmu prawniczego. Uwagi Harta sprzed ponad czterech dekad nie tracą dziś na aktualności, współcześni myśliciele wciąż szukają odpowiedniego, innego niż nawiązujące do prawa natury, filozoficznego uzasadnienia dla swoistego kodeksu podstawowych ludzkich praw.

Our previous studies showed that the nonapeptide fragment of HLA-DQ of the sequence H-Thr-Pro-Gln-Arg-Gly-Asp-Val-Tyr-Thr-OH, located in the β164-172 loop, strongly suppresses the humoral and cellular immune responses, while its shorter analogs, H-Arg-Gly-Asp-Val-OH, H-Arg-Gly-Asp-Val-Tyr-OH and H-Gln-Arg-Gly-Asp-Val-Tyr-OH show only a weak stimulatory activity in respect to the humoral immunological response. These fragments contain the Arg-Gly-Asp (RGD) sequence, known for its importance for cellular association phenomena. Based on the crystal structure of HLA-DR1, we also designed and synthesized a cyclic analog H-Cys-Arg-Gly-Asp-Val-Tyr-Cys-OH with restricted conformation, which strongly suppresses the immune response and selectively inhibits the αvβ3 integrin, suggesting that the mechanism of the immunosuppressory action of the peptide is associated with inhibition of the integrin. In this paper we present the design and synthesis of the cyclodimeric peptide, Arg-Gly-Asp-Arg-Gly-Asp, which is also known as a selective αvβ3 inhibitor. The synthesized peptide strongly suppresses both the humoral and cellular immune response. The results support our hypothesis that the immunomodulatory activity of HLA-DQ fragments may be connected with their interactions with some particular integrins on the cell surface.

A bridged peptide with the sequence: H-Thr-Pro-Gln-Arg-Gly-Asp-Val-γ-Abu-Asn-Asp-Gln-Glu-Glu-Thr-Thr-Gly-Val-Val-Ser-Thr-Pro-Leu-Ile-Arg-Asn-Gly-OH was designed to mimic the discontinuous epitope of the HLA-DQ molecule that might interact with CD4. The bridged peptide revealed distinct suppressory effect in the humoral immune response. This result supports our suggestion that the 164-172 region of the HLA-DQ molecule may enhance its interactions with coreceptors, possibly with CD4.

Introduction: We decided to examine the immunoreactivity of two antigens, B7H4 and HAL-G, within ovarian cancer cells and the macrophages that infiltrate the ovarian cancer microenvironment. It is well known that these two antigens are responsible for the inhibition of both cytotoxic T lymphocytes and NK cells. It has also been suggested that the response of the patient to applied therapy (surgery and chemotherapy) may be related to the status of the tumor microenvironment. Certainly, both B7H4 and HLA-G have been found in the ovarian cancer nest. Methods: We analyzed the immunoreactivity levels of both B7H4 and HLA-G on ovarian cancer cells with respect to the number of different chemotherapy regiments the patient underwent between the first- and second-line surgeries. Additionally, we detected the immunoreactivity of these two antigens on the macrophages present in the ovarian cancer microenvironment. The immunoreactivity analysis was performed on tissue samples derived from 17 patients. Result: In our study, we observed a statistically significantly higher amount of infiltration of B7H4-positive macrophages into the ovarian cancer relapse microenvironment in those patients who had had more than one type of chemotherapy regiment between surgical procedures compared to those who had had only one. Likewise, the HLA-G immunoreactivity level was higher in the first than in the second group. Conclusion: An immunosuppressive ovarian cancer relapse microenvironment, as indicated by the presence of suppressive macrophages, may be related to the need to supplement the primary chemotherapy.
Ekspresja antygenu B7H4 i antygenu HLA-G jest związana z ucieczką komórek raka spod nadzoru immunologicznego, ponieważ antygeny te odpowiadają za ograniczenia aktywności limfocytów cytotoksycznych T i komórek NK. Ich ekspresję wykazano zarówno w komórkach raka jajnika, jak i w obrębie błony komórkowej makrofagów znajdujących się w mikrośrodowisku raka jajnika. Ponieważ odpowiedź na zastosowane leczenie chirurgiczne i chemioterapię może zależeć od stopnia zahamowania aktywności układu odpornościowego gospodarza, postanowiliśmy zbadać immunoreaktywność tych antygenów w tkance wznowy raka jajnika u 17 chorych w zależności od odpowiedzi na zastosowane wcześniej leczenie. Metody: Oceniliśmy immunoreaktywność antygenów B7H4 oraz HLA-G w komórkach raka jajnika i w obrębie makrofagów naciekających mikrośrodowisko raka jajnika w próbkach tkankowych pochodzących od 17 chorych. Następnie przeprowadziliśmy analizę zmian ekspresji badanych antygenów w zależności od odpowiedzi na zastosowane leczenie chirurgiczne i chemioterapię. Wyniki: W naszej pracy zaobserwowaliśmy statystycznie znamiennie więcej makrofagów B7H4-dodatnich w mikrośrodowisku wznowy raka jajnika w przypadkach, w których wznowa występowała po wcześniejszym leczeniu chirurgicznym i chemioterapii I i II rzutu, w porównaniu z przypadkami wznowy po leczeniu chirurgicznym i chemioterapii I rzutu. Podobnie w pierwszej grupie obserwowano wyższą immunoreaktywność antygenu HLA-G. Wnioski: Obecność w mikrośrodowisku raka jajnika makrofagów o działaniu hamującym układ odpornościowy jest związana z powstaniem hamującego mikrośrodowiska guza i może mieć związek z odpowiedzią na leczenie.

Opis:
Decision making process in current tactical C2 systems is base on planning process of commanders and their staff. Improving tactical decision making by interfacing war game simulations with tactical C2 systems is achievable. Commander can review the results of the simulation and subsequently modify the tactical plan. Previously, the use of "training" simulations was not a viable solution to real world decision making due to the lengthy time required to input all of the combat entities, the unit organizations and personnel dispositions, the equipment configurations, status of the units and equipment, and the distribution of the available supplies. Modern C2 systems have all of this information stored in the common system databases, and this information can be used to instantiate and populate the simulation through an electronic adaptation of the data structures to match the requirements of the constructive simulation. This paper will provide description of system approach of interfacing simulation and C2 system to improve decision-making.
