Opis:
Two genes of the extreme thermophilic archaeon Thermococcus litoralis homologous to those that code for Cpn60 chaperonins were cloned and expressed in Escherichia coli. Each of the Cpn60 subunits as well as the entire Cpn60 complex crystallize in a variety of morphological forms. The best crystals diffract to 3.6 Å resolution at room temperature and belong to the space group I422 with unit cell parameters a = b = 193.5 Å, c = 204.2 Å.

In the present study, the influence of bioaugmentation strategy on the co-treatment of 13% v/v reject water and municipal wastewater at a decreasing temperature was evaluated. The experiment was performed in two identical laboratory sequencing batch reactors with the active volume of 8 L. Each one was operated using a 12-hour cycle at sludge retention time of 3 d. The SBR A was bioaugmented with a mixture of wild-living bacteria and Archaea in a dose 0.25 ml. In turn, the comparative reactor (SBR B) was non-bioaugmented, the Archaea product was replaced with an equal volume of dechlorinated tap water. The experiment was divided into 3 phases, each with a different temperature range (20, 15 and 10°C). The temperature reduction did not adversely affect the process performance in the bioaugmented and non-bioaugmented system. Significant removal efficiencies were achieved in both SBRs. The major differences were observed for the COD content in the bioaugmented SBR at a temperature of 10°C. In this case, statistically lower concentrations in the effluent were observed in the bioaugmented system than in the non-bioaugmented one. Additionally, at a temperature of 10°C, an improved process performance was observed in the Archaea presence, but the differences were of no statistical significance.

In this study, the effect of bioaugmentation on the sequencing batch reactor (SBR) performance while treating municipal wastewater and reject water under various temperature conditions was examined. Two lab-scale SBRs, each with the active volume of 8 L were used to perform this research. For bioaugmentation, a mixture of wildliving Bacteria and Archaea in a dose 0.25 mL was added to SBR A, while SBR B was a control one. Both reactors were fed with a mixture of wastewater and 13% v/v reject water. During the experiment, 5 phases with different temperature range were distinguished, each one lasted 14 d. The temperatures were investigated in 5°C increments, i.e. 20, 25, 30, 25 and 20°C. The obtained results indicated that in the bioaugmented reactor (SBR A), lower concentrations of NH4+–N, TSS, NO2-–N in effluent were observed as compared to control (SBR B). While for NH4+–N, regardless the temperature, the observed differences were statistically significant. Importantly, in both SBRs, the process was carried out in a stable way.

In the article, we presented a brief description of the Archaea, considering their structure, physiology and systematics. Based on the analysis of the literature, we bring closer the mechanism of biofilm formation, including extracellular polymeric substances as well as cellular organelles, such as archaella, pili and ‘hami’. The method of forming a biofilm depends on the type of Archaea and the environment in which it naturally lives. We are also introducing the phenomenon of quorum-sensing, as a mechanism of communication of Archaea in the environment. This phenomenon corresponds to similar molecules as bacteria, namely acylated homoserines lactones, QS peptides, autoinducer-2 and -3 and others. In the case of biofilms and the occurrence of the phenomenon of quorum sensing, it can be concluded that these phenomena are very important for the life of Archaea. The phenomena described seem to be conservative, because both in Archaea and Bacteria are regulated by the same mechanisms.
W artykule przedstawiamy opis Archaea, z uwzględnieniem ich budowy, fizjologii i systema- tyki. Na podstawie piśmiennictwa przybliżamy mechanizm tworzenia biofilmu wraz z poza-komórkowymi substancjami polimerowymi, a także typowe tylko dla archeonów organelle komórkowe, takie jak archaella, pili i „hami”. Metoda tworzenia biofilmu zależy od rodzaju Archaea i środowiska, w którym naturalnie żyje. Przybliżamy również zjawisko quorum--sensing, jako mechanizm komunikacji Archaea w środowisku. Za zjawisko to odpowiadają cząsteczki chemiczne podobne do tych u bakterii, a mianowicie acylowany lakton homoseryny, peptyd QS, autoinduktor-2 i -3 oraz inne. Opisywane zjawiska, zarówno tworzenie biofilmu jak i quorum-sensing są istotne dla życia archeonów. Opisane zjawiska wydają się konserwatywne, ponieważ zarówno u archeonów, jak i bakterii są regulowane przez te same mechanizmy.

The aim of the study was to elucidate the substantial relationship between the compositions of methanogen community that assembles in the anaerobic digester mass and link it to methane production activity. The results of the metagenomic studies were used to evaluate how the methanogen structure changes during an anaerobic digestion process under various waste retention times (21, 23, 25, 29, 33, 39, 47 and 61 days). Phylogenetically coherent populations of methanogens were assessed by 16S rRNA gene next-generation sequencing and terminal restriction fragment length polymorphism fingerprinting of a specific molecular marker, the mcrA gene. The results indicated multiple phylogenetically diverse methanogen populations associated with the various steps of anaerobic digestion. The stages of the anaerobic digestion process and waste retention times determine the microbial composition. The most dominant and acclimated microbial communities in all samples belonged to the genera Methanosaeta and Methanobacterium. The methane yield was consistent with the results of the microbial community structure, which indicated that acetotrophic Methanosaeta was the most active and most important during the methanogenic stage.

A strain of Haloarcula hispanica isolated from Tuzkoy salt mine, Turkey exhibited extracellular lipolytic activity. Important parameters such as carbon sources and salt concentration for lipase production were investigated. Optimal conditions for the enzyme production from Haloarcula hispanica 2TK2 were determined. It was observed that the lipolytic activity of Haloarcula hispanica was stimulated by some of the carbon sources. The high lipase acitivity values were obtained in the presence of 2% (v/v) walnut oil (6.16 U/ml), 1% (v/v) fish oil (5.07 U/ml), 1% (v/v) olive oil (4.52 U/ml) and 1% (w/v) stearic acid (4.88 U/ml) at 4M NaCl concentration. Lipase was partially purified by ammonium sulfate precipitation and ultrafiltration. Optimal temperature and pH values were determined as 45oC and 8.0, respectively. Lipase activity decreased with the increasing salt concentration, but 85% activity of the enzyme was maintained at 5M NaCl concentration. The enzyme preserved 41% of its relative activity at 90oC. The partially purified lipase maintained its activity in the presence of surfactants such as Triton X-100 and SDS. Therefore, the lipase which is an extremozyme may have potential applications especially in detergent industry.

This study compares the diversity of methanogenic Archaeal communities that developed during biogas production in reactors fed with different substrates. Reactor I was fed with silages of maize and of alfalfa and timothy; and Reactor II was fed with these silages plus pig slurry and glycerol as co substrates. The Archaeal community structure was studied using polymerase chain reaction––denaturing gradient gel electrophoresis based on the 16S rRNA gene. In both reactors, Methanosphaerula palustris was most abundant, and species belonging to Methanolinea, Methanoculleus, and Methanotorris were present. Only Reactor I, where the ammonia concentration was lower, had species belonging to Methanospirillum and Methanosarcina. Thus, it appears that addition of pig slurry increased the ammonia concentration, which inhibited the growth of Methanospirillum and Methanosarcina.

Anaerobic oxidation of methane (AOM) is a biochemical process that plays an important role in aquatic ecosystems, as it significantly reduces the emission of methane (CH4 ) to the atmosphere. Under anaerobic conditions, CH4 can be oxidized with electron acceptors, such as sulphates (SO42-), nitrates (NO3-) or nitrites (NO2-), iron (Fe3+), manganese (Mn4+) and humic substances. The anaerobic oxidation of methane is mainly regulated by anaerobic methanotrophic archaea (ANME) and sulphate reducing bacteria. The AOM process is crucial to understand the CH4 cycle and anticipate future emissions of the gas from water reservoirs. The process is widely described in marine environments, however very little is known about its occurrence and importance in freshwater systems. There is a great demand for this kind of the research, especially in ecosystems exposed to long-term anaerobic conditions, which may be in degraded reservoirs.

Based on a three-year field experiment under controlled condition in Ji’nan, China, the effects of peanut growth on the variation in the abundance and community structure of ammonia oxidizing bacteria (AOB) and archaea (AOA) before and after peanut growth were investigated through quantitative PCR and cluster analysis of terminal-restriction fragment length polymorphism. Our results show that the community composition of AOA and AOB was greatly affected by the peanut growth leading to the decreased abundance of AOA and increased abundance of AOB. Furthermore, AOA and AOB community structures varied before and after peanut growth. Phylogenetic analysis indicated that all AOA and AOB community sequences were clustered into the uncultured group. Altogether, the results suggested that the abundance of AOA and AOB in soil and their community compositions can be greatly affected by the peanut growth.
