Introduction:S100 calcium-binding protein B (S100B) is a biochemical marker of astroglial damage. Purpose: To assess the pathophysiological implications of S100B concentrations in blood plasma and cerebrospinal fluid of patients with neurodegenerative central nervous system disorders. Materials and Methods: In this study, we determined and compare S100B concentrations in blood plasma and cerebrospinal fluid obtained from subjects diagnosed with Alzheimer's disease (n=20), amyotrophic lateral sclerosis (n=12), multiple sclerosis (n=40) and the reference group (n=20), using enzyme-linked immunosorbent assay. Results: Concentrations of S100B in plasma collected from patients diagnosed with Alzheimer's disease (252,38±183,50 pg/mL) and multiple sclerosis (164,92±250,14 pg/mL) were above laboratory standards, but in patients with amyotrophic lateral sclerosis (53,96±56,92 pg/mL) and the reference group (2,12 pg/mL) were below laboratory norms (N>75 pg/mL). Concentrations of S100B in plasma collected from patients with Alzheimer's disease (252,38±183,50 pg/mL) were significantly higher than in patients with amyotrophic lateral sclerosis (53,96±56,92 pg/mL) (p<0,029). Concentrations of S100B in CSF collected from the reference group (546,96±236,62 pg/mL) and from patients with Alzheimer's disease (587,53±189,57 pg/mL), amyotrophic lateral sclerosis (404,41±179,56 pg/mL), multiple sclerosis (462,03±146,01 pg/mL) were very similar, and none of pairwise comparisons reached statistical significance. Conclusions: Results of our studies indicate the importance of S100B protein concentration assessment in blood in central nervous system disorders differential diagnostics.

Opis:
Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by progressive muscular paralysis reflecting degeneration of motor neurons. In recent years, in addition to several studies about genetic mechanisms leading to motor neurons damage, various epigenetic theories have been developed, involving the study of the patients’ work and lifestyle. The work aims at focusing the role of occupational exposure related to ALS by literature data analysis. Articles, selected on the basis of keywords, year of publication and topics, are related to occupational exposure, suggesting an impact on ALS onset. The literature review shows that there are still a lot of biases in the studies design, which actually do not allow to draw unequivocal conclusions.

Matrix metalloproteinases (MMPs) are implicated in the pathogenesis of motor neuron degeneration in amyotrophic lateral sclerosis (ALS) and might be potential markers of diagnosis, prognosis and monitoring treatment effects. The aim of the present study was evaluation of the MMPs significance in cerebrospinal fluid (CSF MMPs) of patients with ALS in relation to severity of the disease. Metalloproteinases MT-MMP-1, MMP-2, MMP-9 and additionally age of subjects and disease duration were analyzed. The results demonstrate that the error of differentiation between healthy subjects and ALS patients (for MMP-2 feature) as well as between mild and severe ALS states (for CSF MMPs set) equalled to 0.033. In conclusion, the pattern recognition approach may be useful for differentation of ALS progressing on the basis of CSF MMPs features.

Opis:
The two kinds of classifier based on the k-NN rule, the standard and the parallel version, were used for recognition of severity of ALS disease. In case of the second classifier version, feature selection was done separately for each pair of classes. The error rate, estimated by the leave one out method, was used as a criterion as for determination the optimum values of k's as well as for feature selection. All features selected in this manner were used in the standard and in the parallel classifier based on k-NN rule. Furthermore, only for the verification purpose, the linear classifier was applied. For this kind of classifier the error rates were calculated by use the training set also as a testing one. The linear classifier was trained by the error correction algorithm with a modified stop condition. The data set concerned with the healthy subjects and patients with amyotrophic lateral sclerosis (ALS). The set of several biomarkers such as erythropoietin, matrix metalloproteinases and their tissue inhibitors measured in serum and cerebrospinal fluid (CSF) were treated as features. It was shown that CSF biomarkers were very sensitive for the ALS progress.
