This paper describes the influence of the chain length and the functional group steric accessibility of thiols modifiers on the phase transfer process efficiency of water synthesized gold nanoparticles (AuNPs) to toluene. The following thiols were tested: 1-decanethiol, 1,1-dimethyldecanethiol, 1-dodecanethiol, 1-tetradecanethiol and 1-oktadecanethiol. Nanoparticles (NPs) synthesized in water were precisely characterized before the phase transfer process using Atomic Force Microscopy (AFM) and Transmission Electron Microscopy (TEM). The optical properties of AuNPs before and after the phase transfer were studied by the UV-Vis spectroscopy. Additionally, the particle size and size distribution before and after the phase transfer of nanoparticles were investigated using Dynamic Light Scattering (DLS). It turned out that the modification of NPs surface was not effective in the case of 1,1-dimethyldecanethiol, probably because of the difficult steric accessibility of the thiol functional group to NPs surface. Consequently, the effective phase transfer of AuNPs from water to toluene did not occur. In toluene the most stable were nanoparticles modified with 1-decanethiol, 1-dodecanethiol and 1-tetradecanethiol.

Opis:
1,1-Dimethylbiguanidium 1H,1′H-5,5′-bistetrazole-1,1′-diolate (MGBTO), a novel nitrogen-rich energetic salt, was synthesized by cation exchange. Its structure was characterized by elemental analysis, FTIR, NMR, DTA and TG-DTG. Single crystal X-ray diffraction analysis revealed that MGBTO was crystallized in the monoclinic space group C2/c. Thermal analysis demonstrated that its thermal stability extended up to 531.1 K. The nonisothermal kinetic and apparent thermodynamic parameters of the exothermic decomposition of MGBTO were determined by the Kissinger and Ozawa methods. Its detonation velocity and detonation pressure were calculated on the basis of the Kamlet-Jacobs equation and were 6342 m·s–1 and 15.78 GPa, respectively. The impact and friction sensitivities of MGBTO were quantified using standard BAM (10 kg drop hammer) procedures. The results revealed that the salt has good mechanical sensitivity (FS > 120 N, IS > 40 J), thus indicating its potential applications as an energetic material.

An important trait of tomato is the rate of fruit ripening, strongly dependent on ethylene production. The ripening-related ethylene synthesis in tomato is controlled mainly by 1-aminocyclopropane-1-carboxylate synthase LE-ACS2 and LE-ACS4 isoenzymes (Rottmann et al., 1991, J. Mol. Biol. 222: 937; Lincoln et al., 1993, J. Biol. Chem. 268: 19422; Barry et al., 2000, Plant Physiol. 123: 979). In spite of numerous reports on the LE-ACS2 and LE-ACS4 gene expression, only ones considered the genomic organisation each of these genes (Rottmann et al., 1991; Lincoln et al., 1993) reported one copy of each of these genes in tomato cv VF36. In this article we suggest that the genomic organisation of LE-ACS2 and LE-ACSS4 genes may depend on tomato cultivars and may differ from that described by the above authors. The results of Southern analyses of genomic DNAs from 17-day old seedlings (cultivars Jaga, Halicz, Betalux, New Yorker) imply that the genomic organisation of LE-ACS2 and LE-ACS4 genes in Polish cultivars differs from that reported for cv VF36.

Dysfunction of fast axonal transport, vital for motor neurons, may lead to neurodegeneration. Anterograde transport is mediated by N-kinesins (KIFs), while retrograde transport by dynein 1 and, to a minor extent, by C-kinesins. In our earlier studies we observed changes in expression of N- and C-kinesins (KIF5A, 5C, C2) in G93ASOD1-linked mouse model of motor neuron degeneration. In the present work we analyze the profile of expression of the same kinesins in mice with a dynein 1 heavy chain mutation (Dync1h1, called Cra1), presenting similar clinical symptoms, and in Cra1/SOD1 mice with milder disease progression than SOD1 transgenics. We found significantly higher levels of mRNA for KIF5A and KIF5C but not the KIFC2 in the frontal cortex of symptomatic Cra1/+ mice (aged 365 days) compared to the wild-type controls. No changes in kinesin expression were found in the spinal cord of any age group and only mild changes in the hippocampus. The expression of kinesins in the cerebellum of the presymptomatic and symptomatic mice (aged 140 and 365 days, respectively) was much lower than in age-matched controls. In Cra1/SOD1 mice the changes in KIFs expression were similar or more severe than in the Cra1/+ groups, and they also appeared in the spinal cord. Thus, in mice with the Dync1h1 mutation, which impairs dynein 1-dependent retrograde transport, expression of kinesin mRNA is affected in various structures of the CNS and the changes are similar or milder than in mice with double Dync1h1/hSOD1G93A mutations.

Opis:
The aim of this paper is to show methodology of forecasting with various horizon of prediction using grey system theory, basing on practical application to vibration condition monitoring problems. The method of forecasting was based on GM(1,1) prognostic models with various window lengths for estimating model parameters. The model GM(1,1) is very effective where we have only few data, incomplete, and with low accuracy. The moving window method applied to GM(1,1) model enables to adapt to changes in data trend. However, selecting an inappropriate window length can result in excessive forecast errors. The applied algorithm is based on tracking the current prediction error for models having various window lengths, and then eliminating the models for which the error of prediction caused by the loss of adequacy of the model to the data increases excessively.
Celem pracy jest przedstawienie metodyki prognozowania dla różnych horyzontów prognozy z wykorzystaniem teorii szarych systemów w odniesieniu do praktycznego problemu z zakresu drganiowej diagnostyki maszyn. Metoda prognozowania jest oparta o modele GM(1,1) o różnej długości okna w której wyznacza się parametry modelu. Model GM(1,1) jest bardzo efektywny gdy istnieje niewiele danych, są one niekompletne i są niskiej jakości. Metoda ruchomego okna w zastosowaniu do modelu GM(1,1) pozwala na budowanie modelu adaptacyjnego, który umożliwia dostosowanie się do zmian w trendzie krzywej życia. Niestety źle dobrana długość okna może powodować zwiększenie się błędu prognozy w porównaniu do warunków, w których długość okna byłaby optymalna. Zastosowany algorytm polega na śledzeniu aktualnego błędu prognozy modeli o różnej długości okna i eliminacji znaczenia tych modeli, które powodują nadmierny wzrost błędu prognozy związany z utratą adekwatności do danych.

Numerous evidences suggested that the hepatitis B virus (HBV) was recognized as an important factor in the development of hepatocellular carcinoma (HCC). Dickkopf-1 (DKK1) recently was reported to be involved in the progress of HCC. HBV may regulate DKK1 expression in hematoma carcinogenesis. Here, we demonstrated that HBV could regulate DKK1 promoter activity which resulted in upregulation of its mRNA and protein expression in several HBV existing cell lines, and HBx played a prominent role in this process. Transcription factor binding site search result showed that there is a SP1 site in DKK1 promoter region. Luciferase assay showed that overexpression of SP1 could increase DKK1 promoter activity in a dose dependent manner. Accordingly, siRNA inhibition of SP1 expression reduced DKK1 promoter activity and decreased the expression of DKK1 protein.

Introduction. The term ‘subarachnoid haemorrhage’ (SAH) stands for bleeding into the subarachnoid space, regardless of its source. It may be of primary character when the source of bleeding is situated within the subarachnoid space. Subarachnoid haemorrhage is often described as spontaneous bleeding, mainly in order to differentiate it from post-traumatic bleeding. Objective. The aim of the study was to evaluate the concentration of ICAM-1 and VCAM-1 in the blood of patients in the early phase following subarachnoid haemorrhage in terms of searching for markers useful in subarachnoid bleeding diagnostics and monitoring a patient’s clinical state. Materials and method. The study comprised 85 patients (47 women, 38 men), aged 29–81 (average 53±12 years), suffering from subarachnoid haemorrhage. The control group comprised 45 healthy people selected according to gender and age corresponding with the experimental group. Results. The study revealed that the concentration of ICAM-1 and VCAM-1 was significantly higher in patients suffering from subarachnoid haemorrhage. Additionally, the concentration of fibrinogen decreased, aPTT was shorter and the concentration of D-dimers increased. The studied parameters did not differ with respect to the age or gender of the patients. It was stated that according to the Hunt and Hess scale, the concentration of ICAM-1 was considerably higher in the group of patients in the most severe neurological state, compared to other patients. It was also observed that the concentration of fibrinogen was significantly higher, aPTT was shorter, and the concentration of D-dimers increased in the afore-mentioned group. Conclusions. Evaluation of the concentration of adhesion molecules, as well as values of global tests concerning the coagulation system, may serve as a useful diagnostic tool for SAH.

The objective of our study was to investigate the effect of stimulation of the cAMP-dependent pathway on the expression of an orphan nuclear receptor, SF-1/Ad4BP in mouse adrenal tumour, Y-1 cells in culture. We evaluated the temporal pattern of the effects of corticotropin (ACTH) and the adenylyl cyclase activator forskolin on the level of SF-1 mRNA, and compared the time course of induction of SF-1 with that of CYP11A1. Forskolin, corticotropin and 8-Br-cAMP significantly elevated the level of the SF-1 transcript, after 1.5 h of incubation, with a concomitant increase of SF-1 protein level, observed after 6 h. The CYP11A1 transcript increased gradually over the incubation period, and reached the maximal level after 12 to 24 h. The steady-state level of the SF-1 transcript was unaffected by forskolin when the cells were incubated with actinomycin D, indicating that stimulation of the cAMP pathway results in enhanced transcription of the gene. The effect of forskolin was augmented by cycloheximide, suggesting that an inhibitory protein, whose synthesis was inhibited by cycloheximide, could be involved in negative regulation of SF-1 expression. It is concluded that SF-1 expression is positively regulated by the cAMP pathway at the transcriptional level, and can represent the primary event in cAMP-mediated induction of steroid hormone synthesis in Y-1 cells.

Although over 85% of osteogenesis imperfecta (OI) cases are associated with mutations in the procollagen type I genes (COL1A1 or COL1A2), no hot spots for the mutations were associated with particular clinical phenotypes. Eight patients that were studied here, diagnosed with OI by clinical standards, are from the Polish population with no ethnic background indicated. Previously unpublished mutations were found in six out of those eight patients. Genotypes for polymorphisms (Sp1 - rs1800012 and PvuII - rs412777), linked to bone formation and metabolism were determined. Mutations were found in exons 2, 22, 50 and in introns 13 and 51 of the COL1A1 gene. In COL1A2, one mutation was identified in exon 22. Deletion type mutations in COL1A1 that resulted in OI type I had no effect on collagen type I secretion, nor on its intracellular accumulation. Also, a single base substitution in I13 (c.904-9 G>T) was associated with the OI type I. The OI type III was associated with a single base change in I51 of COL1A1, possibly causing an exon skipping. Also, a missense mutation in COL1A2 changing Gly→Cys in the central part of the triple helical domain of the collagen type I molecule caused OI type III. It affected secretion of the heterotrimeric form of procollagen type I. However, no intracellular accumulation of procollagen chains could be detected. Mutation in COL1A2 affected its incorporation into procollagen type I. The results obtained shall help in genetic counseling of OI patients and provide a rational support for making informed, life important decisions by them and their families.

Both ethylene and the enzymes of ethylene synthesis are subjects of intensive scientific investigation. The present review discusses structure, catalytic activity and evolutionary relationships of 1-aminocyclopropane-1-carboxylate synthase, identified for the first time in ripening tomato in 1979. This enzyme is responsible for the conversion of S-adenosyl-L-methionine to 1-aminocyclopropane-1-carboxylic acid, which is the key step of ethylene synthesis in higher plants. The role of this enzyme (especially in the fruit ripening) was demonstrated in 1991 in transgenic tomato plants, expressing 1-aminocyclopropane-1-carboxylate synthase antisense RNA. On the basis of mutagenesis and crystallization of the enzyme, new data were provided on the three-dimensional structure and amino-acid residues which are critical for catalysis. The control of ethylene production is of great interest for plant biotechnology because it can delay senescence and overmaturation. These processes are responsible for large loss of vegetables and fruit on storage. Detailed structural and biochemical data are necessary to help design 1-aminocyclopropane-1-carboxylate synthase inhibitors, whose application is expected to have immense agricultural effects.

An L(2, 1, 1)-labeling of a graph G is an assignment of non-negative integers (labels) to the vertices of G such that adjacent vertices receive labels with difference at least 2, and vertices at distance 2 or 3 receive distinct labels. The span of such a labelling is the difference between the maximum and minimum labels used, and the minimum span over all L(2, 1, 1)-labelings of G is called the L(2, 1, 1)-labeling number of G, denoted by λ2,1,1(G). It was shown by King, Ras and Zhou in [The L(h, 1, 1)-labelling problem for trees, European J. Combin. 31 (2010) 1295–1306] that every tree T has Δ2(T) − 1 ≤ λ2,1,1(T) ≤ Δ2(T), where Δ2(T) = maxuv∈E(T)(d(u) + d(v)). And they conjectured that almost all trees have the L(2, 1, 1)-labeling number attain the lower bound. This paper provides some sufficient conditions for λ2,1,1(T) = Δ2(T). Furthermore, we show that the sufficient conditions we provide are also necessary for trees with diameter at most 6.

Multiple studies showed that the cessation of TWIST1 expression is the prerequisite for osteoblasts' maturation. However, recent reports revealed that the function of TWIST1 is different in the dental pulp stem cells (DPSCs), where a high level of TWIST1 expression promoted DPSCs' differentiation. The aim of the study was to investigate the impact of TWIST1 and ID1 on the differentiation process in the human DPSCs. Methods: TWIST1 and ID1 expression in the DSPCs was modulated by lentivirus transduction. Genes expression was assessed with qRT-PCR. The proteins level was evaluated by Western blot. The DPSCs differentiation was assessed with the proliferation, alkaline phosphatase (ALP) activity, and calcium concentration assays. Results: TWIST1 silencing suppressed the expression of ID1 and both the early and late markers of odontoblasts' differentiation detected at the transcript and protein level. The forced overexpression of ID1 increased the expression of the late markers of odontoblasts differentiation but diminished the expression of the early markers. DPCSs with the silenced TWIST1 and subsequent ID1 overexpression displayed an increase in the expression of the late markers of odontoblasts differentiation. Cells with silenced TWIST1 and overexpressing ID1 had increased activity of ALP, higher calcium concentration and decreased proliferation rate. The high level of ID1 expression might be a critical factor stimulating DPSCs differentiation and it might compensate the repressed differentiation of DPSCs caused by TWIST1 silencing. Conclusion: The mutual correlation between the expression level of TWIST1 and ID1 might be a critical factor driving the process of the human odontoblasts' differentiation.

Opis:
High activity of copper based catalysts for C-O bond hydro-dehydrogenation and their poor activity for C-C bond cleavage1 have prompted an attempt to apply such catalysts in the hydrogenolysis of glycerol to 1,2- and 1,3-propanediol. In the present study the infl uence of hydrogen reduction time of the Cu/ Al2O3 and CuCr2O4 copper catalysts on glycerol conversion and selectivity of transformation to propanediols and by-products was studied. At fi rst a general comparison was made between the commercial catalysts and those prepared by the co-precipitation method. As better results were obtained in the presence of catalysts prepared by co-precipitation, they were selected for further detailed studies of the influence of reduction time. For both prepared catalysts Cu/Al2O3 and CuCr2O4 the reduction time of 8 h was optimal. In the presence of Cu/Al2O3 catalyst the conversion of glycerol was 59.0%, selectivity of transformation to 1,2-propanediol 77.4% and selectivity to 1,3-propanediol 1.9%. In the presence of CuCr2O4 the glycerol conversion was 30.3% and selectivity to 1,2-propanediol 67.3%.

Opis:
Objectives Objectives: Cigarette smoking is the major risk factor of bladder cancer via exposure to chemical carcinogens. Nicotinamide adenine dinucleotide phosphate (NADP+): quinine oxidoreductase 1 (NQO1) and sulfotransferase 1A1 (SULT1A1) have been reported to involve in the metabolism of polycyclic aromatic hydrocarbons (PAHs) and aromatic amines. Therefore, the risk of bladder cancer (BC) may be influenced by polymorphisms in the genes that modulate metabolic detoxification in particular by interacting with cigarette smoking. Considering the limited power by the individual studies with a relatively small sample size, especially when analyzing the combined effect of polymorphisms in NQO1 and SULT1A1 genes and smoking, these 2 meta-analyses have aimed to clarify the combined effects of them on BC risk by integrating related studies. Material and Methods Two meta-analyses included 1341 cases and 1346 controls concerning NQO1 Pro187Ser and smoking, and 1921 cases and 1882 controls on SULT1A1 Arg213His and smoking were performed. Odds ratios (OR) and 95% confidence intervals (CI) were used for assessing the strength of the association. Results The result has demonstrated that smokers with NQO1 Pro/Ser or Ser/Ser genotypes have a prominent association with the risk of BC as compared with non-smokers with NQO1 Pro/Pro genotype, with OR equal to 3.71 (95% CI: 2.87–4.78, $ \text{p}_\text{heterogeneity} $ = 0.376). Besides, smokers carrying SULT1A1 Arg/Arg genotypes were observed to confer 2.38 fold increased risk of BC (95% CI: 1.44–3.93, $ \text{p}_\text{heterogeneity} $ = 0.001) when compared with non-smokers with SULT1A1 Arg/Arg or His/His genotypes. Conclusions These findings have suggested that the NQO1 Pro187Ser or SULT1A1 Arg213His polymorphism combination with smoking significantly confer susceptibility to BC.
