Opis:
Induced osteogenesis of mesenchymal stem cells (MSCs) may provide an important tool for bone injuries treatment. Human umbilical cord and adipose tissue are routinely discarded as clinical waste and may be used as noncontroversial MSCs sources. It still remains to be verified which source of MSCs is the most suitable for bone regeneration. The aim of this research was to investigate the osteogenic potential of human MSCs derived from adipose tissue (AT-MSCs) and Wharton's jelly of the human umbilical cord (WJ-MSCs) differentiated under the same conditions. Osteogenic differentiation of MSCs was detected and quantified by alizarin red S (ARS) staining for calcium deposition and alkaline phosphatase (ALP) activity, osteoprotegerin (OPG), and osteocalcin (OC) secretion measurements. Under osteogenic conditions, after 21 days of differentiation, the measured ALP activity and calcium deposition were significantly higher in the AT-MSCs than in the WJ-MSCs, while the OPG and OC secretion were higher in the WJ-MSCs vs. AT-MSCs. Low concentrations of OPG and high levels of OC in AT-MSCs and WJ-MSCs, prove that these cells reached an advanced stage of the osteogenic differentiation. The levels of OC secreted by AT-MSCs were lower than by WJ-MSCs. Both cell types, AT-MSCs and WJ-MSCs possess a potential to differentiate towards the osteogenic lineage. The observed differences in the levels of osteogenic markers suggest that after 21-days of osteogenic differentiation, the AT-MSCs might have reached a more advanced stage of differentiation than WJ-MSCs.

Peroxynitrite-mediated linoleic acid oxidation and tyrosine nitration were analysed in the presence of synthetic model neuromelanins: dopamine (DA) -melanin, cysteinyldopamine (CysDA) -melanin and various DA/CysDA copolymers. The presence of melanin significantly decreased the amount of 3-nitrotyrosine formed. This inhibitory effect depended on the type and concentration of melanin polymer. It was found that incorporation of CysDA-derived units into melanin attenuated its protective effect on tyrosine nitration induced by peroxynitrite. In the presence of bicarbonate, the melanins also inhibited 3-nitrotyrosine formation in a concentration dependent manner, although the extent of inhibition was lower than in the absence of bicarbonate. The tested melanins inhibited peroxynitrite-induced formation of linoleic acid hydroperoxides, both in the absence and in the presence of bicarbonate. In the presence of bicarbonate, among the oxidation products appeared 4-hydroxynonenal (HNE). CysDA-melanin inhibited the formation of HNE, while DA-melanin did not affect the aldehyde level. The results of the presented study suggest that neuromelanin can act as a natural scavenger of peroxynitrite.

Bazując na wynikach analiz metod statystyki wielowymiarowej przeprowadzono klasyfikację grupy badanych pacjentów ze względu na grupę badanych cech. Celem analizy jest próba wyodrębnienia charakterystycznych grup czynników wśród pacjentów chorujących na raka jelita grubego w różnym stopniu zaawansowania klinicznego. Analizie poddano wybrane dane epidemiologiczne pochodzące z dokumentacji medycznej chorych z ustalonym rozpoznaniem - rak jelita grubego. Do analizy wykorzystano zmienne jakościowe: płeć, stopień zaawansowania klinicznego choroby, typ i złośliwość histopatologiczną, podział na osoby z wagą prawidłową, nadwagą i otyłością, podział ze względu na stężenie glukozy na czczo w surowicy krwi oraz współistnienie występowania innych chorób.
In this paper a classification o f examined patients was carried out based on results o f multivariate analysis using classification trees. The aim o f the analysis was to identify characteristic factors describing groups o f patients suffering from colorectal cancer with different stage o f disease. Clinical data from medical documentation o f the patients with colon cancer were analyzed. Qualitative variables such as sex, clinical stage, histopathology type o f cancer and malignancy, weight class, glucose level class and coexistence with other illnesses were used in the analysis.

Background: Head and neck neoplasms stand for 6% of all malignant neoplasms worldwide. Chemotherapy has limited use due to the biological properties of the tumor (in the majority of cases moderately and poorly differentiated squamous cell carcinoma). The fundamental molecule used in treatment is cisplatin and its derivates, that can be associated with fluorouracil. The new chemotherapeutic agents are not in common use during the treatment of head and neck malignancies. However, the use of low molecular weight complexes Pd (II) carries the potential of being more effective in therapy. Material and Methods: Fifty-one patients, 30 men and 21 women (aged 52.9 ± 12.1 years) with head and neck cancer were included in the study. Fifty-one healthy subjects, 31 men and 20 women, (aged 54.1 ± 14.7 years) years formed the control group. Antioxidant enzymes, superoxide dismutase, and catalase activities in erythrocytes were examined. Results: An increased level of antioxidant enzymes was seen in the blood samples from patients with head and neck cancer after incubation with Pd (II) complex. In the group we obtained a statistically significant result p = <0.001. Discussion: That project may contribute to the development of new, more efficient head and neck cancer treatment strategies. In our opinion, the results can be used in the future to develop a valuable prognostic marker of the disease. This is important because the initial phase of cancer is asymptomatic. The search for factors involved in pathogenesis translates into economic benefits and makes therapy more effectiveness through the reduction of treatment expenses.

Abstract: Urinary tract infections caused by wide range of pathogens including gram-negative and gram-positive bacteria as well as fungi are a severe public health problem. The predominant causative agent of both uncomplicated and complicated urinary tract infections is Escherichia coli. In an era of increasing bacterial resistance to antimicrobial agents and a high prevalence of multidrug-resistant (MDR) strains in community and hospital acquired infections, the re-evaluation of older generations of antimicrobial agents, such as nitrofuran derivatives, seems to be a reasonable approach. The aim of the study was to evaluate furazidin activity against common uropathogens in comparison to nitrofurantoin and other selected antimicrobial agents, routinely used in the treatment of urinary tract infections. Furazidin exhibited lower MICs than nitrofurantoin when tested against gram-negative and gram-positive bacteria including clinical MDR E. coli and methicillin-resistant Staphylococcus aureus. The MICs for furazidin ranged from 4 to 64 mg/L for Enterobacteriaceae strains, from 2 to 4 mg/L for gram-positive cocci, and 0.5 mg/L for anaerobic bacteria. The MICs for nitrofurantoin ranged from 16 to 64 mg/L for Enterobacteriaceae strains, from 8 to 64 mg/L for gram-positive cocci, and 4 mg/L for anaerobic bacteria. In addition, both nitrofurans displayed better activity against the tested bacterial strains than ciprofloxacin, fosfomycin, trimethoprim and co-trimoxazole. Nitrofuran derivatives displayed higher antimicrobial activity than other antimicrobial agents regardless of bacteria species or resistance mechanism.
