Opis:
The subject of the modelling work and the conducted experiments is the composite shell of a train seat. The activities carried out involved designing the geometry, planning the material structure, and selecting the materials to be used. The shell was built using polymer matrix fibrous composites (i.e. FRP – Fibre Reinforced Polymer – composites), which are lighter than steel and comply with the relevant standards for strength and safety at the same time. This was followed by creating a computational model for the shell and conducting a strength analysis in accordance with the guidelines of the relevant industry standard and strength hypotheses adopted for FRP composites. The calculations were conducted using ANSYS Composite PrepPost software based on the finite element method. The article offers a strength analysis of an optimised composite shell of a train seat. Based on the guidelines obtained as a result of the conducted modelling work, a physical prototype (validation model) of the seat was created. Hot vacuum lamination technology was applied in the production process. Th e experimental validation of the model, producing a positive result, was conducted using a test stand owned by S.Z.T.K. TAPS – Maciej Kowalski.

Purpose: The article introduces a concept of the economic imagination, describes economic public attitudes and discusses the character of the contemporary economic imagination of Poles. Methodology: Representative survey conducted on the stratifi ed random sample of 1022 adult respondents by means of CAPI (computer assisted personal interview). Statistical analysis of data. Findings: The features perceived by public as economically benefi cial to the entire economy, particular enterprises and individuals have been ranked by the representative survey and statistically grouped into more general clusters. Those considered as good for the economy are grouped into such factors as “investment and competition”, “statism (etatism)” and “progressive taxation”. The features considered as benefi cial for the enterprises are grouped into “human, social and economic capital”, “law obedience and good social relations” and “limited free market”. Personal characteristics perceived as benefi cial for individual’s economic success are grouped into “honesty, perseverance and good social relations”, “skills and diligence” and “nepotism and instrumentalism”. Intensity as well as determinants of such general value loaded convictions, and their impact on acceptance of democracy have been revealed as well. The implications: Economic imagination has to be analysed as an important factor of socio-economic system’s legitimization. Originality: Distinction and empirical defi nition of economic attitudinal syndromes at three different levels: macro-economic, enterprise and individual constitute a novelty in socio-economic research. Discussion of their determinants and wider implications constitute a new contribution to the theory of social legitimization.

Opis:
This paper presents interdisciplinary research on the transformation of Polish society and the economy. A new composite Balanced Development Index (BDI) has been constructed and applied in a ‚beyond GDP’ paradigm to analyse Poland’s socio-economic development during the 1999-2014 period and to make predictions for 2015-2017. Four groups of detailed indicators-two economic ones (external and internal) and two social ones (concerning the objective and subjective current situation, and public expectations for the future)- are used to construct four middle-level indexes, which are subsequently aggregated into the general BDI index. The congruency between the four middle-level indexes is interpreted in terms of the balance relevant for future socio-economic development. The validity of the index is proven by its high correlation with the psychological conditions of society. The results indicate a less optimistic picture of development in Poland than is provided by the GDP, which has been constantly growing.

Opis:
Potato leafroll virus is a member of the polerovirus genus. The isometric virion is formed by a coat protein encapsidating single-stranded, positive-sense, mono-partite genomic RNA with covalently attached viral protein at the 5' end. The coat protein of the virus exists in two forms: i) a 23 kDa protein, the product of the coat protein gene, and ii) a 78 kDa protein, the product of the coat protein gene and an additional open reading frame expressed by read-through of the coat protein gene stop codon. The aim of this work was the expression of potato leafroll virus coat protein-based proteins that would be able to assemble into virus-like particles in insect cells. These modified particles were tested for their ability to encapsidate nucleic acids. Two types of N-terminally His-tagged coat protein constructs were used for the expression in insect cells: one, encoding a 23 kDa protein with the C-terminal amino-acid sequence corresponding to the wild type coat protein and the second with additional clathrin binding domain at the C-terminus. The expression of these two proteins by a recombinant baculovirus was characterized by Western immunoblotting with antibodies directed against potato leafroll virus. The protection or putative encapsidation of nucleic acids by these two coat protein derivatives was shown by DNase I and RNase A protection assays.

Opis:
Translation of viral proteins from subgenomic RNAs (sgRNAs) is a common strategy among positive-stranded RNA viruses. Unlike host mRNA, sgRNA of Potato leafroll virus (PLRV) does not possess a cap at its 5' end nor a poly(A) tail at the 3' terminus, both of which are known to be crucial for translation of RNA in eukaryotic cells. Here, we demonstrate, that in wheat germ extract (WGE) truncation of the sgRNA1 5' UTR increases translation efficiency, as it has previously been observed in rabbit reticulocyte lysate (RRL), whereas removal of the 3' UTR does not affect translation. We also describe two regulatory elements located within the coding sequence of the coat protein (CP) gene and its read-through domain (RTD) and are responsible for regulation of in vitro translation of the PLRV sgRNA1. The first element is composed of the purine sequence AAAGGAAA located between the AUG codons of the CP and 17K genes. Deletion of this domain or its substitution by pyrimidines reduced by half the translation of both genes, whereas deletion of the RTD resulted in a 3.6-fold reduction in translation efficiency. This is the first report of translation regulatory elements of plant viruses located within a coding region.
