The continued lack of knowledge concerning the molecular background of adverse effects caused by microcystin-LR (MC-LR) is surprising. This toxin requires additional attention, not only for its ability to cause acute poisoning, but also for its ability to initiate cancer in acute doses, and potentially, to promote cancer via chronic exposure to low concentrations in drinking water. Our recent studies on whitefish (Coregonus lavaretus) revealed that long-term exposure to MC-LR resulted in severe liver injury, followed by regeneration of the liver and its unexpected resilience to further toxin uptake. These effects were accompanied by perturbations of hepatic microRNAs (miRNAs) that have target genes involved in cytoskeletal remodeling, cell metabolism, cell cycle regulation, and apoptosis. Among the most pronounced individual alterations, the reduction of MiR92b-3p expression was the most remarkable, and we suggest roles for the miRNA in the aberrant processes of liver cells. This project addresses potential involvement of MiR92b-3p in the as yet unknown regulatory network of MC-induced hepatotoxicity in fish. After a suite of biochemical, physiological, anatomical, and transcriptomic analyses in vitro and in vivo, we will show how MiR92b-3p works in a damaged liver and which processes it targets. Finally, the research will confirm if and how MiR92b-3p can be targeted therapeutically. We expect it to be shown effective enough to pave a way for its use as a tool for treatment of liver damage in fish. What is more, the RNA-based silencing technique that will be used should yield exciting data for our understanding of the system-level biology of vertebrates.

Eutrophication of marine and fresh waters can lead to excessive development of cyanobacterial blooms, which may contain strains that produce toxins. These toxins are secondary metabolites which can accumulate in the food chain and contaminate drinking water, thus posing a potential threat to the health of humans and aquatic organisms. These toxins include a variety of compounds with different mechanisms; this review focuses on the neurotoxicity of microcystin and other cyanotoxins. Although the hepatotoxic action of microcystins is commonly known, its neurotoxic effects have also been described, e.g. oxidative stress, cytoskeletal changes and changes in protein phosphatase activity. These effects have been partially explained by the discovery in the blood brain barrier of the same membrane transporters involved in microcystins hepatotoxic mechanisms. Additionally, this paper reviews other cyanotoxins that are known or suspected to target cholinergic synapses and voltage gated channels, including anatoxin a, anatoxin a(s), antillatoxins, cylindrospermopsin, homoanatoxin a, jamaicamide, kalkitoxin and saxitoxins. The neurotoxic and cytotoxic effects of the cyanotoxins discussed here are of particular interest because of their pharmacological potential. This review also discusses the potential of these compounds to serve as drugs for cancer and central nervous system failure.

Proximate to the environment, adipose fin of fish may be considered as a lipid storing tissue, and thus can be a target for either waterborne or dietary polycyclic aromatic compounds (PACs). We determined the effects of benzo[a]pyrene (B[a]P), a model PAC member, on CYP1A gene expression in adipose fin and compared that with the effects in gill of juvenile rainbow trout (Oncorhynchus mykiss Walbaum) using the quantitative reverse transcription polymerase chain reaction (Q-RT-PCR). The results of the study demonstrated that constitutive CYP1A mRNA was present in adipose fin of rainbow trout, but the transcripts were far less abundant than those in gill tissue. We confirmed high CYP1A gene induction potential of the gills in rainbow trout injected with benzo[a]pyrene, but also showed moderately and transiently induced CYP1A mRNA in adipose fin. The modest and transitory gene expression may preclude rainbow trout adipose fin CYP1A mRNA levels from using it as an indicator of sustained exposure of fish to the polycyclic aromatic compounds.

MicroRNAs (miRNAs) are small, highly conserved, non-coding RNAs that regulate gene expression of target mRNAs through cleavage or translational inhibition. In the field of toxicology, the relationship between toxicity and microRNA expression is poorly understood. In the present study we analyzed the abundance of 9 selected miRNAs (omy-miR-21, omy-miR-21t, omy-miR-122, omy-miR-125a, omy-miR-125b, omy-miR-125t, omy-miR-199-5a, omy-miR-295, omy-let-7a) and mRNA of 3 genes (histone H2A, ribosome protein rpl19, and Dicer which is a miRNA processing enzyme) in liver samples of whitefish exposed to Microcystin-LR (MC-LR) at a dose of 100µg*kg-1body weight for 24 or 48h. In the examined liver tissue, omymiR-122 showed the highest relative constitutive level, what is consistent with data obtained from fish and mammals. Unexpectedly, the reference H2A mRNA level was consistently up-regulated (over 20-fold; P<0.05) in fish liver after both 24 and 48h of exposure to MC-LR. The result may suggest that MC-LR acts as an initiator of specific cell-physiologic signals triggering DNA replication in fish liver cells. MC-LR treatment had no effect on the examined miRNAs levels, except for omy-miR-125a and omy-let-7a. Whereas omy-miR-125a was up-regulated (ER=2.68; S.E. 1.61-6.78; P<0.05), omy-let-7a was down-regulated (ER=0.55; S.E. 0.32-0.79; P<0.05) in whitefish liver after 48h of the treatment with MC-LR, when compared to controls. More work with the fish is essential for understanding the crosstalk of the regulatory network controlled by the two miRNAs in the context of MC-LR toxicity.

Cyanobacteria constitute a rich source of biologically active and structurally diverse compounds. The pharmacological potential of these compounds resides among others in their ability to control the proliferation and growth of cancer cell lines and potent disease-causing microbial agents. Despite recent scientific advances, the way these compounds interact with the body’s molecular structure are still unclear and science still has to discover how the cyanobacterial metabolites interact with cell structures and how cells react to them. In this project, we will study yet unexamined cyanobacterial metabolites, especially the compounds which act as chemical ligands for microRNA (miRNA) -binding sites, making them promising regulators (inhibitors) of gene networks that are involved in various diseases. We will first develop a stable cell line that constitutively expresses a unique miRNA reporter system. Then, we will conduct a screen on chemical compounds discovered in Baltic cyanobacteria to identify small molecules with inhibitory activity and specificity to MIR92b-3p, which has a significant impact on liver cell behavior in humans. We assume that a successful MIR92b-3p inhibitor will bind to the precursors of MIR92b-3p miRNA, disabling the action of either of the two processing enzymes involved in the biogenesis of any miRNA in a cell (Drosha or Dicer), thus affecting the MIR92b function. The discoveries made with these inhibitory chemical molecules could provide insight into the role of the MIR92 pathway in liver diseases and cancer, and possibly, if promising results appear, they may facilitate a strategy for treating some human diseases in the future.

Although p53, a protein of important tumor suppressive function, has been extensively studied in mammals, relatively little is known about the p53 pathways in lower vertebrates. Particularly, limited information exists on possible influences of environmental contaminants on the expression of the p53 gene in fish. In the current study, we assessed the effects of benzo[a]pyrene (B[a]P; potent tumor promoter) and cyclopenta[c]phenanthrene (CP[c]Ph; clastogenic agent) exposure on a 24h profile of p53 gene expression in head kidney of juvenile rainbow trout (Oncorhynchus mykiss). To analyze the p53 transcription rate, we developed protocol for the examination of both mRNA and heterogeneous nuclear (hn) RNA of the gene, using Real-Time RTPCR approach. The results show that both compounds are capable of suppressing p53 transcriptional activity within 12h of the treatment. Our finding supports the idea that structurally different PAHs may influence cell physiologic functions controlled by p53 in fish, in part, by down-regulating its RNA expression levels.

While estrogenic properties of mycotoxin zearalenone (ZEA) has been an extensively studied issue, little is known about molecular background of its biological responses that cannot be simply explained by the estrogenic potential. The present study describes effects of ZEA (10mg*kg-1 body weight) in binary mixtures either with benzo[a]pyrene (B[a]P), or with 17β-estradiol (E2) on ER- and AhR-dependent gene expression in juvenile rainbow trout liver, evaluated using Real-Time qPCR. The study revealed dual nature of ZEA, as the treatment with this compound alone increased mRNA levels of both ER- and AhR-mediated gene expression. However, our results did not show any synergistic or additive effect of ZEA in binary mixures with E2 or B[a]P on studied gene expression levels. Whether the intriguing potential of ZEA to elicit distinct signals was a result of binding affinity to AhR or/and ER and AhR mutual receptor interactions, should be investigated in further experiments.

Zearalenone (ZEA) is a mycoestrogen frequently found in food and animal feed materials all over the world. Despite its hydrophobic character, ZEA is also found in surface and ground waters which suggests an environmental risk for aquatic animals. Knowledge concerning mycotoxin-related mechanisms of toxicity is still incomplete, e.g. little is known about the influence of ZEA exposure on fish. The aim of this study was to investigate the effect of ZEA on selected biochemical parameters in juvenile rainbow trout after 24, 72, and 168 h of intraperitoneal exposure (10 mg/kg of body weight). The analysis showed a slight tendency towards prolonged blood clotting time and significant iron deficiency in the liver and ovary of exposed animals. However, no differences in aminotransferase (AlaAT, AspAT) activity or glucose levels in fish plasma was observed. The results of this study suggest that although trout exposed to ZEA did not exhibit any distinct symptoms of liver damage, the mycotoxin tested was able interfere with blood coagulation and iron-storage processes.

We assessed the effects of cyclopenta[c]phenanthrene (CP[c]Ph) and benzo[a]pyrene (B[a]P; positive control) on CYP1A gene expression in brain of juvenile rainbow trout (Oncorhynchus mykiss) using the quantitative reverse transcription polymerase chain reaction (Q-RT-PCR). A group of hatchery raised rainbow trout, with an average body mass of 49.4 g and total length of 15.5 cm were given an intraperitoneal injection (10 mg*kg-1) of either CP[c]Ph or B[a]P in corn oil (2 mg*mi-1 corn oil) or corn oil alone (control). After 24 and 48 h, trout brains were collected for mRNA isolation and analysis. After 24 hours of the exposure, only B[a]P-treated rainbow trout had 10-fold higher number of CYP1A transcripts (mean = 3.63*106 transcripts*µg-1 total RNA) than control fish (3.24*105 transcripts*µg-1 total RNA; Tukey test, P<0.05). After 48 hrs, significantly higher levels of CYP1A expression (Tukey test, P<0.001) were found in either CP[c]Ph- or B[a]P- induced group (1.45*106 and 6.92*106 transcriptsźµg-1 total RNA, respectively) over a control group (mean=1.41*105 transcripts*µg-1 total RNA). The finding that CYP1A in brain tissue was inducible by CP[c]Ph, a polycyclic aromatic hydrocarbon (PAH) of different than B[a]P planar characteristics, may further validate the use of rainbow trout brain CYP1A mRNA levels as a biomarker of PAH exposure.

This paper presents an influence of three different magnetic fields stimulation systems for growing strawberry fruits: permanent magnetic field generated by neodymium magnets, ”high” strengths slow varying amplitudes magnetic fields generated by coreless solenoids supplied by autotransformer and low-frequency and low-amplitude sinusoidal magnetic fields generator created for this research. The system is controlled by computer and allows for an independent control of up to 10 induction coils inducing magnetic fields with amplitudes from 0.1μT to 150μT and frequencies from 0.1Hz to 100Hz. The proposed set-up was used during the growing seasons for strawberry plants stimulation. An increase in the extract level was observed in the case of the alternating magnetic field stimulation with the parameters: frequency – from 50Hz to 100Hz and amplitude – from 50μT to 100μT.

Opis:
Identification of differentially expressed genes that could be potentially used as biomarkers of PAH exposure of common invertebrate animal (like freshwater snail) would be a valuable resource for investigators interested in toxicology and biomonitoring of aquatic environments. Therefore, the aim of this research was to investigate effects of waterborne benzo[a]pyrene (B[a]P) exposure on mRNA expression in the pond snail’s (Lymnaea stagnalis) hepatopancreatic gland. Toward this end, mature individuals of pond snail (L. stagnalis) were treated with 50µM B[a]P solution in a short 36h static exposure test. Differential Display PCR (DD-PCR) was used to generate a unique cDNA fingerprint of genes that were differentially expressed in the tissues of exposed and unexposed snails. To assess the putative identity of the isolated cDNA amplicons (ESTs), BLAST queries were performed to find similarities in their nucleotide sequence. Real-Time qPCR analysis was used to verify the DD-PCR expression profile. Finally, an additional independent exposure study, including higher dose of B[a]P (100µM), was conducted to validate the expression of selected ESTs. BLAST revealed that only 3 out of 9 isolated ESTs had meaningful information on their putative nucleotide sequence identity. The highest similarity was scored for EST-A1, identified as the transcript of UAP-like protein, found to be up-regulated after B[a]P exposure. The original expression pattern that was observed in DD-PCR step was coherent with results of the qPCR verification for 3 out of 5 analyzed ESTs. However, changes in the ESTs expression were modest and the treatment with B[a]P resulted in significant down-regulation for only 1 unidentified fragment (EST-G42, almost 2-fold; p<0.05) when compared to untreated snails. Although no significant changes were observed for EST-A1 and EST-G42 in the validation study, their expression pattern was consistent with that obtained from DD-PCR. Surprisingly, EST-C5 remained in contrast to the DD-PCR part, but it showed significant down-regulation in group of snails exposed to 100µM B[a]P (3.5-fold; p<0.05). The obtained results show that diverse genes may be involved in the molecular response of the pond snail’s hepatopancreas to treatment with B[a]P. However, further research is needed to confirm the utility of the discovered EST as PAH biomarkers in biomonitoring practices with L. stagnalis as bioindicator species.

Zearalenone (ZEN) widely contaminates animal feed of plant origin. The recommended safe concentrations of ZEN in feeds for various animal species are set mainly based on the mycotoxin’s hormonal properties (NOEL). Our growing knowledge about biologically active concentrations of ZEN, molecular mechanisms and cells/tissues targeted by ZEN indicates that the harmful effects exerted by this mycotoxin on animals may be far greater than previously believed. This experiment was performed on pre-pubertal gilts divided into a control group (n=9) and an experimental group (ZEN, n=9). The control group received placebo, whereas the experimental group was administered ZEN at a dose of 0.1 mg/kg feed (equivalent to 5 μg/kg BW/day) for 42 days. On days 14, 28 and 42 blood samples were collected from the animals to determine the concentrations of selected zearalenols, serum biochemical and haematological parameters. Conjugated ZEN was found in the blood serum of the experimental gilts. Changes in the analysed biochemical parameters included a transient increase in albumin and cholesterol levels. A statistically significant increase in the concentrations of neutrophilic and acidophilic granulocytes was observed in the white blood cell system. The results indicate that long-term per os exposure of pre-pubertal gilts to low doses of ZEN (below NOEL) has a modulatory effect on liver function and white blood cells.

Opis:
Measurement of the disc temperature of a disc brake or the rim temperature of a block brake at various simulated train brakings is one of the tests carried out at an inertia brake station. In case of temperature tests of a railway disc brake, in accordance with the guidelines presented in [1], thermocouples installed on the friction surface of the brake disc are used. Behaviour of the average temperature is defined on the basis of six thermocouples mounted in threes on each side of the friction collar. The thermocouples on each side of the disc are placed every 120 ° on three different radii of the disc. During simulated braking tests of the disc, in accordance with a test programme provided for a railway disc brake [2], the waveforms of the average temperature of the disc as a function of the braking time are recorded. This is especially important during trials of brakings from high speeds: 200 and 300 km/h with high pressures of pads to the disc and large braking masses in case of brakings of trains with goods carriages. Then the heat load limit of the brake is checked. The purpose of this paper is to analyse the results of research on behaviour of the average temperature obtained from the measurement of the railway disc brake system, in particular the disc brake, the friction pads brake and caliper (Figs. 6 and 7) using a thermographic camera in station tests. The research show that the measurements using a thermal imaging camera can substantially extend the scope of bench research on the brake disk. It is possible to determine the temperature average increment on the components of the braking system as a result of changes in the thickness of the friction pads.
: Jednym z badań prowadzonych na bezwładnościowym stanowisku hamulcowym jest pomiar temperatury tarczy dla hamulca tarczowego lub temperatury obręczy dla hamulca klockowego przy różnych symulacjach hamowań pociągu. Celem artykułu jest ocena wyników badań rozkładu średniej temperatury tarczowego układu hamulcowego, w szczególności tarcza hamulcowa-okładzina cierna wraz z obsadą hamulcową uzyskanych z pomiaru przy użyciu kamery termowizyjnej w warunkach stanowiskowych. Przeprowadzone badania termowizyjne pozwalają na ocenę zarówno jakościową jak i ilościową zmiany rozkładu średniej temperatury elementów pary ciernej hamulca spowodowaną zużyciem okładzin ciernych.

Toxic effects of polycyclic aromatic hydrocarbons (PAHs) have been extensively studied in fish, although knowledge concerning biological activities of phenanthrene and its derivatives remains still incomplete. The aim of this work was to evaluate lethal and sublethal effects of benzo(a)pyrene, phenanthrene and phenanthrene derivatives (1-methylphenanthrene, 4-methylphenanthrene, 1-phenylphenanthrene and 4-phenylphenanthrene) on zebrafish (Danio rerio) larvae. We conducted acute toxicity test, using 96h static renewal exposure to a series of the PAH concentrations (0.05, 0.50, 5.00, 50.00µmol*l-1), to determine the No Effect Concentration (NEC) value for each compound examined. The mean NEC estimates obtained in the study were 5.16۪.45µmol*l-1 (B[a]P), 4.88۪.13µmol*l-1 (Ph), 40.24䔰.93µmol*l-1 (1P-Ph), 47.92ۭ.61µmol*l-1 (1M-Ph), 24.31۱.33µmol*l-1 (4P-Ph) and 3.11۫.01µmol*l-1 (4M-Ph) and suggested the following order of PAH toxicities on Danio rerio larvae: 4M-Ph>Ph˜B[a]P>4PPhP-Ph>1M-Ph. To gain insight into possible molecular mechanisms of apparent toxicity of phenanthrene derivatives on zebrafish larvae, we examined mRNA expression of cyp1a, cyp1b1, and vtg genes in the larvae exposed for 48h to a PAH concentration of 0.50µmol*l-1. Whereas the larvae exposed to each tested PAH displayed many developmental abnormalities (i.e. pericardial and yolk sac edema, dorsal curvature, or tail malformations), no significant upregulation of cyp1a and cyp1b1 mRNA was observed, except for zebrafish exposed to B[a]P. However, significant reduction of vtg mRNA was observed in the larvae exposed to phenanthrene and its 4P- derivative. The results may contribute to the development of a new knowledge about effects of structurally diverse phenanthrene derivatives on vertebrate organisms.

Monitoring of undesirable substances by the European Union indicates a presence of natural and anthropogenic pollutants in animal feed that may be of concern for the producers, as well as the veterinary services. Although the literature concerning toxicity of DDT (an insecticide widely used in the past) is extensive, less attention has been focused on the biological properties of DDE and its interactions with other contaminants. This study reports on the concentration profile of p,p’-DDE and two other ogranochlorines (p,p’-DDT, p,p’-DDD) in different tissues of immature gilts after 14, 28, and 42 days of oral exposure to p,p’-DDE alone (0.5mg·kg-1feed·day-1) and in mixture with naturally occurring mycotoxin zearalenone, ZEN (0.5+0.1mg·kg-1feed·day-1). The treatment resulted in a time-dependent accumulation of p,p’-DDE in fat-rich tissues. Although the pesticide’s concentration found in the adipose tissue exceeded the FAO/WHO maximum residue limit (5mg·kg-1 of fat), human dietary risk is little, as it requires a substantial consumption of such contaminated pork fat. Importantly, the high concentration of p,p’-DDE found in the adrenal glands suggests a threat to the animals’ health. Finally, a difference in the accumulation of p,p’-DDE was observed between the groups treated with this compound alone or in mixture with ZEN. This is most likely related to growth performance of the animals, altered by the endocrine disrupting activity of both compounds. Further research should evaluate the effects of p,p-DDE at the observed concentrations on the pigs’ health status and enable the studies of possible interactions with other environmental pollutants.
