Bacopa monnieri (Scrophulariaceae) is a well-known plant and has been used by humans for several thousand years. In traditional Hindu medicine, it is one of the most important medicinal plants. The aim of the work was to determine the content of Fe, Mg, and Zn and selected organic compounds before and after extraction into the artificial digestive juices obtained from preparations containing B. monnieri. Commercial preparations in the form of tablets and capsules and B. monnieri shoot cultures were used in the experiment. The metal content in the considered mineralized samples was analyzed by atomic absorption spectrometry and organic compounds by RP-HPLC method. The maximum measured content of the metals released into the digestive juices was as follows: Fe – 32.85; Mg – 367.51; and Zn – 16.41 mg/100 g of the preparation. The existing research shows that metals are best released into the artificial digestive juices from the B. monnieri shoot cultures, and least efficiently from the commercial preparations available in the form of tablets. The phenolic compounds analyzed in the methanol extracts and the extracts of the artificial digestive juices were as follows: protocatechuic acid, p-hydroxybenzoic acid, neochlorogenic acid, chlorogenic acid, isochlorogenic acid, caffeic acid, ferulic acid, cynaroside, trifolin, and luteolin. Bacoside A was only determined in the extracts from the B. monnieri shoot cultures. The experimental results revealed that B. monnieri distributed in the form of tablets did not break down in the artificial digestive juices during the considered time intervals.

RESEARCH OBJECTIVE: This article aims to map the stereotypical image of Ukrainians and Russians from a cross-cultural perspective in the context of the ongoing armed conflict. RESEARCH PROBLEM AND METHODS: The research problem concerns the cultural differences that are revealed in the perception of representatives of the two nationalities that are involved in the armed conflict. Since stereotypical judgements are most often revealed in language, an adjective check list was used for the study. A sample of 92 Polish and Slovenian students took part in the study. The collected linguistic material was used to analyse and compare cross-cultural stereotypical judgements about Ukrainian and Russian citizens. THE PROCESS OF ARGUMENTATION: The argument begins with theoretical perspectives on the concept of stereotyping and the process of stereotyping. The role of language in the stereotyping process is analysed. An analysis of the results and conclusions is presented. RESEARCHRESULTS: In general, Polish students attribute fewer positive adjectives than Slovenian students to both Ukrainians and Russians. The analysis revealed significant statistical differences as well as similarities between the researched groups of Polish and Slovenian students in terms of the stereotypical image of Ukrainians and Russians. CONCLUSIONS, INNOVATIONS, RECOMMENDATIONS: Empirical evidence shows that Polish and Slovenian students have more in common in their perception of Russians, while they perceive Ukrainians quite differently. The linguistic characteristics obtained in this study provide a general insight into the perceptions of the two nationalities involved in the war. It should also be noted that these characteristics of Ukrainians and Russians may constitute socially important knowledge on the negative stereotypes about these nationalities. This knowledge may be useful in challenging counteracting or disseminating such judgements, which may be harmful and lead to prejudice or discrimination against people from the nationalities studied.  
CEL NAUKOWY: Celem artykułu jest porównanie międzykulturowej perspektywy stereotypowego obrazu Ukraińców/Ukrainek oraz Rosjan/Rosjanek w kontekście trwającego konfliktu zbrojnego. PROBLEM I METODY BADAWCZE: Problem badawczy dotyczy kulturowych różnic, jakie ujawniają się w procesie spostrzegania przedstawicieli dwóch narodowości, które są zaangażowane w konflikt zbrojny. Ponieważ stereotypowe sądy najczęściej ujawniają się w języku, dlatego też do badań wykorzystano narzędzie, które opiera się na języku (lista przymiotników), a równocześnie jest zaadoptowane do badań międzykulturowych. W badaniu wzięło udział 92 studentów z Polski i Słowenii. Zebrany materiał językowy posłużył do analizy i porównania międzykulturowych, stereotypowych sądów dotyczących obywateli Ukrainy i Rosji. PROCES WYWODU: Wywód rozpoczęto od założeń teoretycznych dotyczących pojęcia stereotypu oraz procesu stereotypizacji. Przeanalizowano rolę języka w procesie stereotypizacji, a także przedstawiono analizę wyników i wnioski. WYNIKI ANALIZY NAUKOWEJ: Ogólny wynik wskazuje, że polscy studenci przypisują mniej przymiotników pozytywnych niż słoweńscy zarówno Ukraińcom, jak i Rosjanom. Analiza wykazała istotne różnice statystyczne oraz podobieństwa między badanymi grupami studentów polskich i słoweńskich w zakresie stereotypowego obrazu Ukraińców i Rosjan. WNIOSKI, INNOWACJE, REKOMENDACJE: Wyniki przeprowadzonych badań wskazują na występowanie podobieństwa u młodych Polaków i Słoweńców w postrzeganiu Rosjan natomiast zupełnie inaczej spostrzegają oni Ukraińców. Charakterystyki językowe uzyskane w niniejszym badaniu pozwalają na ogólny wgląd w sposób spostrzegania dwóch narodowości zaangażowanych w wojnę. Należy też zauważyć, że uzyskane w badaniach charakterystyki Ukraińców i Rosjan stanowić mogą ważną społecznie wiedzę o negatywnych stereotypach, jakie funkcjonują o tych narodowościach. Wiedza ta może być przydatna w podważaniu, przeciwdziałaniu, lub rozpowszechnianiu takich sądów, które mogą być szkodliwe i prowadzić z kolei do uprzedzeń lub dyskryminacji ludzi z badanych narodowości.

Phenolic compounds, both derivatives of benzoic and cinnammic acid, possess biologically valuable properties: anti-inflammatory, antioxidant, anticarcinogenic and others. Studies of the accumulation of these compounds focused mostly on plant material. Rich sources of these compounds are representatives of Basidiomycota taxon. The aim of the study was qualitative and quantitative HPLC analysis of phenolic acids in biomass from in vitro culture of selected edible mushroom species belonging to the phylum Basidiomycota: Agaricus bisporus, Boletus badius, Cantharellus cibarius. The investigations revealed the presence of the following acids: p-hydroxybenzoic, syringic and galic acid. Both the composition and the amount of phenolic acids in biomass of A. bisporus and Boletus badius were diverse. The total amount ranged from 6.07 mg·100 g−1 DW in A. bisporus to 14.78 mg·100 g−1 DW in Boletus badius. Syryngic acid amounts fluctuated in the range of 1.75–9.66 mg·100 g−1 DW, with its maximum in Boletus badius. Gallic acid dominated in the biomass of the same species (5.12 mg·100 g−1 DW). p-Hydroxybenzoic acid was found in biomass from in vitro culture of A. bisporus at levels 0.70 mg/100 g DW. In biomass of in vitro culture of Cantharellus cibarius no phenolic compounds were found before and after hydrolysis. The results of HPLC analyses show that in vitro culture of B. badius and A. bisporus are a good dietary source of phenolic compounds with antioxidant activity.

Arthrospira platensis (Spirulina platensis) is a well-known microalga and has been utilized as a medicinal agent and foodstuff by humans since at least 16th century. The aim of this study was to determine zinc content as well as determine phenolic and indole compounds from commercial preparations containing Arthrospira platensis (lyophilizate, tablets, and capsules) before and after extraction with methanol and incubation with artificial digestive juices. The secondary aim of this study was to evaluate the quality of these preparations. The samples were incubated in artificial stomach juice and in intestinal juice. The samples were mineralized and their zinc(II) ions content was estimated using flame absorption atomic spectroscopy (F-AAS). The maximum zinc(II) ions content released into the digestive juices was found to be up to 1.6 mg 100 g-1 of the preparation. Phenolic compounds identified in the examined extracts are as follows: gallic acid; protocatechuic acid; 3,4-dihydroxyphenylacetic acid; p-hydroxybenzoic acid; syringe acid; cinnamic acid; and quercetin. Furthermore, indole compounds identified were 5-hydroxy-L-tryptophan, 5-methyl-L-tryptophan, L-tryptophan, tryptamine, and 5-methyltryptamine. Consequently, it was also found that the distributed Arthrospira platensis in the form of tablets does not disintegrate in the artificial digestive juices. Among the examined preparations, only hard capsules met the requirements of the European Pharmacopeia 8th ed.

Prematurely born babies account for nearly 7% of all infants coming into the world, and there are more and more extremely small premature babies being born. A report covering 184 countries shows that each year, 14.9 million children are born ahead of time. Maintaining life is the fi rst task faced by the medical service. The issue regarding development and the future of these children is the next stage of interest for both doctors and a number of specialists from related fields. The aim of the study was to review literature concerning premature newborns in the fi eld of sensorimotor development within the fi rst years of life, taking into account the hazards and possible forms of early support. An attempt was made to present the current state of knowledge in the scope of recommended and used methods of conduct as well as to assess the long-term effects of undertaken interventions. A review of Polish-, English- and French-language literature was conducted, which concerned sensory motor development, introduction of early rehabilitation and support programmes for premature infants. Research papers and review works from the last 10 years included in databases (PubMed and websites of Polish medical publishing houses) were analysed. The works were searched using the following keywords: premature baby, sensorimotor development. The growing group of preemies requires adapted care. To counteract the indicated threats, a therapeutic intervention should be introduced precisely and in accordance with a child’s needs. There is a need to broaden knowledge about development in this group of children and to create a proven developmental support system that would be based on long-term research. Wstęp: Noworodki urodzone przedwcześnie stanowią blisko 7% wszystkich urodzonych dzieci i przybywa dzieci urodzonych jako ekstremalnie małe wcześniaki. Raport obejmujący 184 kraje wskazuje, że każdego roku 14,9 miliona dzieci rodzi się przed czasem. Utrzymanie życia jest pierwszym zadaniem, które stoi przed służbą medyczną. Kwestia rozwoju i przyszłości tych dzieci to następny etap pozostający w kręgu zainteresowań zarówno lekarzy jak i szeregu specjalistów z pokrewnych dziedzin. Cel: Celem pracy było przeprowadzenie przeglądu piśmiennictwa dotyczącego noworodków urodzonych przedwcześnie w zakresie rozwoju sensomotorycznego w pierwszych latach życia, z uwzględnieniem występujących zagrożeń oraz możliwych form wczesnego wsparcia. Podjęto próbę przedstawienia aktualnego stanu wiedzy w zakresie zalecanych i wykorzystywanych metod postępowania, a także oceny długofalowych skutków podejmowanych interwencji. Metodyka: Przeprowadzono przegląd piśmiennictwa polskiego, anglojęzycznego i francuskojęzycznego, które dotyczyły rozwoju sensomotorycznego, wprowadzania wczesnej rehabilitacji niemowląt urodzonych przedwcześnie oraz wprowadzanych programów wspierania. Analizowano prace badawcze i przeglądowe z ostatnich 10 lat zamieszczonych w bazach danych (Pub Med i stronach polskich wydawnictw medycznych). Prace wyszukiwano posługując się słowami kluczowymi: noworodek urodzony przedwcześnie, rozwój sensomotoryczny. Wyniki i wnioski: Rosnąca grupa noworodków urodzonych przedwcześnie wymaga dostosowanej opieki. Aby przeciwdziałać wskazanym zagrożeniom, należy precyzyjnie i zgodnie z potrzebami dziecka wprowadzać interwencję terapeutyczną. Istnieje potrzeba poszerzania wiedzy na temat rozwoju tej grupy dzieci i wypracowania sprawdzonego systemu wsparcia rozwoju, który byłby oparty na długofalowych badaniach naukowych. Obecnie podejmowane wsparcie w noworodków urodzonych przed czasem jest niejednorodne, a przeprowadzone badania nie pozwalają na stworzenie jednego modelu wsparcia, albo jasno określonych standardów postępowania.

Purpose: According to the available international recommendations, at least one independent verifi cation of the calculations of number of monitor unit (MU) is required for every patient treated by teleradiotherapy. The aim of this study was to estimate the differences of dose distributions calculated with two treatment planning systems: Eclipse (Varian) and Oncentra MasterPlan (Elekta). Materials and methods: The analysis was performed for 280 three-dimensional conformal radiotherapy treatment (3D-CRT) plans with photon beams from Varian accelerators: CL 600C/D X6 MV (109 plans), CL 2300C/D X6 MV (43 plans), and CL 2300C/D X15 MV (128 plans). The mean doses in the planning target volume (PTV) and doses at the isocenter point obtained with Eclipse and Oncentra MasterPlan (OMP) were compared with Wilcoxon matched-pairs signed rank test. Additionally, the treatment planning system (TPS) calculations were compared with dosimetric measurements performed in the inhomogeneous phantom. Results: Data were analysed for 6 MV plans and for 15 MV plans separately, independently of the treatment machine. The dose values calculated in Eclipse were significantly (p <0.001) higher compared to calculations of OMP system. The average difference of the mean dose to PTV was (1.4 ± 1.0)% for X6 MV and (2.5 ± 0.6)% for X15 MV. Average dose disparities at the isocenter point were (1.3 ± 1.9)% and (2.1 ± 1.0)% for X6 MV and X15 MV beams, respectively. The largest differences were observed in lungs, air cavities, and bone structures. Moreover the variation in dosimetric measurements was less as compared to Eclipse calculations. Conclusions: OMP calculations were introduced as the independent MU verification tool with the first action level range equal to 3.5%.

The human SUV3 helicase (SUV3, hSUV3, SUPV3L1) is a DNA/RNA unwinding enzyme belonging to the class of DexH-box helicases. It localizes predominantly in the mitochondria, where it forms an RNA-degrading complex called mitochondrial degradosome with exonuclease PNP (polynucleotide phosphorylase). Association of this complex with the polyA polymerase can modulate mitochondrial polyA tails. Silencing of the SUV3 gene was shown to inhibit the cell cycle and to induce apoptosis in human cell lines. However, since small amounts of the SUV3 helicase were found in the cell nuclei, it was not clear whether the observed phenotypes of SUV3 depletion were of mitochondrial or nuclear origin. In order to answer this question we have designed gene constructs able to inhibit the SUV3 activity exclusively in the cell nuclei. The results indicate that the observed growth rate impairment upon SUV3 depletion is due to its nuclear function(s). Unexpectedly, overexpression of the nuclear-targeted wild-type copies of the SUV3 gene resulted in a higher growth rate. In addition, we demonstrate that the SUV3 helicase can be found in the HeLa cell nucleoli, but it is not detectable in the DNA-repair foci. Our results indicate that the nucleolar-associated human SUV3 protein is an important factor in regulation of the cell cycle.

In the present elaboration an attempt has been made to build the logit model which makes it possible to specify the probability of diagnosing the ovarian carcinoma in the female patients with pathological lesion in the ovary. Based on sampling of 210 patients treated and diagnosed at the Teaching Hospital of Operative Gynaecology and Gynaecological Oncology of Women and Girls of the Pomeranian Medical University, the evaluations of the parameters of two logit models were determined and the estimation of the quality of obtained models was made. The obtained results may contribute to supporting of the ovarian cancer diagnostics.

Introduction and objectives. The aim of the study was to asses the impact of long-term therapy with the second generation antiepileptic drug levetiracetam (LEV) with arachidonyl-2’-chloroethylamide (ACEA), a highly selective cannabinoid CB1 receptor agoniston the process of neurogenesis in a mouse pilocarpine model of epilepsy (PILO). Additionally, a relationship was established between the treatment with ACEA in combination with LEV, and hippocampal neurogenesis in mouse PILO brain. Materials and method. All experiments were performed on adolescent male CB57/BL mice injected i.p. with LEV (10 mg/kg), ACEA (10 mg/kg) and PMSF (30 mg/kg) (phenylmethylsulfonyl fluoride — a substance protecting ACEA against degradation by the fatty-acid amidohydrolase), pilocarpine (PILO, a single dose 290 mg/kg) and methylscopolamine (30 min before PILO to stop the peripheral cholinergic effects of the pilocarpine, 1 mg/kg). The process of neurogenesis was evaluated after10 days treatment with LEV and ACEA. Results. Obtained results indicated that the combinations of ACEA+PMSF+LEV and ACEA +PMSF increased the total number of total newborn cells compared to the control. Moreover, ACEA+PMSF administered alone and in combination with LEV had a significant impact on neurogenesis increasing the total number of newborn neurons compared to the control group. Neither LEV nor PMSF had a significant impact on the number of proliferating cells and newborn neurons when compared to the control PILO group. In turn, LEV administered alone decreased significantly the number of astrocytes. However, ACEA+PMSF has demonstrated significant increase of astrocytes compare to control mice. Conclusions. These data provide substantial evidence that the combination of LEV+ACEA significantly increases the level of newborn neurons in the PILO dentate subgranular zone.

Opis:
Hemagglutinin (HA), as a major surface antigen of influenza virus, is widely used as a target for production of neutralizing antibodies. Monoclonal antibody, mAb6-9-1, directed against HA of highly pathogenic avian influenza virus A/swan/Poland/305-135V08/2006(H5N1) was purified from mouse hybridoma cells culture and characterized. The antigenic specificity of mAb6-9-1 was verified by testing its cross-reactivity with several variants of HA. The mimotopes recognized by mAb6-9-1 were selected from two types of phage display peptide libraries. The comparative structural model of the HA variant used for antibody generation was developed to further facilitate epitope mapping. Based on the sequences of the affinity- selected polypeptides and the structural model of HA the epitope was located to the region near the receptor binding site (RBS). Such localization of the epitope recognized by mAb6-9-1 is in concordance with its moderate hemagglutination inhibiting activity and its antigenic specificity. Additionally, total RNA isolated from the hybridoma cell line secreting mAb6-9-1 was used for obtaining two variants of cDNA encoding recombinant single-chain variable fragment (scFv) antibody. To ensure high production level and solubility in bacterial expression system, the scFv fragments were produced as chimeric proteins in fusion with thioredoxin or displayed on a phage surface after cloning into the phagemid vector. Specificity and affinity of the recombinant soluble and phage-bound scFv were assayed by suitable variants of ELISA test. The observed differences in specificity were discussed.

Opis:
Gram-positive and nonpathogenic lactic acid bacteria (LAB) are considered to be promising candidates for the development of new, safe systems of heterologous protein expression. Recombinant LAB has been shown to induce specific local and systemic immune response against selected pathogens, and could be a good alternative to classical attenuated carriers. The main goal of our study was to express the avian influenza haemagglutinin (H5) and chicken interleukin 2 (chIL-2) in Lactococcus lactis. Results of this study were anticipated to lead to construction of lactococcal strain(s) with potential vaccine properties against the avian influenza A (H5N1) virus. Expression of the cloned H5 gene, its His-tagged variant and chIL-2 gene, under the control of the ptcB gene promoter was attested by RT-PCR on transcriptional level and Western or dot blot analysis on translational level, demonstrating that system can be an attractive solution for production of heterologous proteins. The results of the preliminary animal trial conducted in mice are a promising step toward development of a vaccine against avian bird flu using Lactococcus lactis cells as antigen carriers.
