The studies were carried out on 12 UV-mutants of B. subtilis B-3 which synthesise from 18 to 105% more hydrolytic enzymes (proteinases, amylases, cellulases, and lipases) than the wild strain. After five years' storage of the mutants, a 26-fold increase in amylase biosynthesis was observed for a G7 UV-mutant. This mutant showed also other phenotypic differences. The increased ability of mutant G7 to synthesise amylases was controlled by the maltose-substrate induction.
W pracy oceniano stabilność biosyntezy pozakomórkowych hydrolaz (amylaz, proteinaz, celulaz i lipaz) przez 12 UV mutantów bakterii B.subtilis B-3, po 5-letnim przechowywaniu szczepów. Wykazano, że w przewadze mutanty zachowały zdolność do zwiększonej syntezy enzymów. Jednak do dalszych badań wybrano mutanta G7 który wyróżniał się spośród pozostałych tym, że obecnie syntetyzował 26-krotnie więcej amylaz niż szczep naturalny(rys. 2). Ponadto szczep ten syntetyzował też 3-krotnie więcej proteinaz (rys. 1), śladowe ilości celulaz (rys. 4), oraz utracił całkowicie zdolność syntezy lipaz (rys. 3). Między mutantami G7 a szczepem naturalnym B.subtilis B-3 wykazano też inne różnice fenotypowe w postaci zwiększonych rozmiarów komórek, w kinetyce wzrostu i wytwarzaniu barwników (tab. 2). Regulacja syntezy amylaz przez UV mutanta G7 miała charakter indukcyjny (tab. 1).

Opis:
The purpose of this paper was testing suitability of the time-series analysis for quality control of the continuous steel casting process in production conditions. The analysis was carried out on industrial data collected in one of Polish steel plants. The production data concerned defective fractions of billets obtained in the process. The procedure of the industrial data preparation is presented. The computations for the time-series analysis were carried out in two ways, both using the authors’ own software. The first one, applied to the real numbers type of the data has a wide range of capabilities, including not only prediction of the future values but also detection of important periodicity in data. In the second approach the data were assumed in a binary (categorical) form, i.e. the every heat(melt) was labeled as ‘Good’ or ‘Defective’. The naïve Bayesian classifier was used for predicting the successive values. The most interesting results of the analysis include good prediction accuracies obtained by both methodologies, the crucial influence of the last preceding point on the predicted result for the real data time-series analysis as well as obtaining an information about the type of misclassification for binary data. The possibility of prediction of the future values can be used by engineering or operational staff with an expert knowledge to decrease fraction of defective products by taking appropriate action when the forthcoming period is identified as critical.

Opis:
Statistical Process Control (SPC) based on the well known Shewhart control charts, is widely used in contemporary manufacturing industry, including many foundries. However, the classic SPC methods require that the measured quantities, e.g. process or product parameters, are not auto-correlated, i.e. their current values do not depend on the preceding ones. For the processes which do not obey this assumption the Special Cause Control (SCC) charts were proposed, utilizing the residual data obtained from the time-series analysis. In the present paper the results of application of SCC charts to a green sand processing system are presented. The tests, made on real industrial data collected in a big iron foundry, were aimed at the comparison of occurrences of out-of-control signals detected in the original data with those appeared in the residual data. It was found that application of the SCC charts reduces numbers of the signals in almost all cases It is concluded that it can be helpful in avoiding false signals, i.e. resulting from predictable factors.

Opis:
The aim of the paper was an attempt at applying the time-series analysis to the control of the melting process of grey cast iron in production conditions. The production data were collected in one of Polish foundries in the form of spectrometer printouts. The quality of the alloy was controlled by its chemical composition in about 0.5 hour time intervals. The procedure of preparation of the industrial data is presented, including OCR-based method of transformation to the electronic numerical format as well as generation of records related to particular weekdays. The computations for time-series analysis were made using the author's own software having a wide range of capabilities, including detection of important periodicity in data as well as regression modeling of the residual data, i.e. the values obtained after subtraction of general trend, trend of variability amplitude and the periodical component. The most interesting results of the analysis include: significant 2-measurements periodicity of percentages of all components, significance 7-day periodicity of silicon content measured at the end of a day and the relatively good prediction accuracy obtained without modeling of residual data for various types of expected values. Some practical conclusions have been formulated, related to possible improvements in the melting process control procedures as well as more general tips concerning applications of time-series analysis in foundry production.

The purpose of the study was to evaluate the effectiveness of preservation of two bacteria strains: Bacillus subtilis B-3 and Bacillus poly my xa В-20 (proteinase and amylase producers) in the form of dried spores on glass beads. In our study we have found that the survival level of the two strains after 15 months of storage was 100%. The viability of B. subtilis B-3 was 20% lower than that of the control culture maintained by subculturing on agar slants and stored at 4°C (µ = 1.8 h-1 and µ = 2.16 h-1), whereas the viability of B. polymyxa B-20 was similar to that of the control culture (µ = 1.3 h-1). The biosynthetic capability of B. polymyxa B-20 remained at unchanged levels of 303 FSU and 2.5 AU, but only when the spores had been stored on glass beads with starch as the protective agent. The biosynthetic activity of B. subtilis B-3 was 76% lower in the case of proteinases and 60% lower in the case of amylases. The method of storage of bacteria in the form of spores dried on glass beads, described in this paper, can be recommended for B. polymyxa B-20 strain, but it is not suitable for B. subtilis B-3 since the bacteria have lost, to a large extent, their biosynthetic ability.
W pracy oceniano skuteczność metody przechowywania w formie przetrwalników na perełkach szklanych, dwóch szczepów B.subtilis B-3 i B.polymyxa B-20, producentów proteinaz i amylaz. Wykazano, że po 15 miesiącach przechowywania badane szczepy przeżywały w 100% (Tabela 1,2). Żywotność szczepu B.subtilis B-3 po tym czasie byla niższa o 20% w stosunku do kontroli którą stanowiła kultura okresowo przeszczepiana i przechowywana na skosach w temperaturze 4°C (µ = 1,8 godz-1 i µ = 2,16 godz-1), natomiast szczepu B.polymyxa B-20 była zbliżona do kontroli (µ = 1,3 godz-1). Szczep B.polymyxa B-20 zachowywał zdolność biosyntezy enzymów na niezmienionym poziomie (303 JFS i 2,5 JA) ale tylko podczas przechowywania przetrwalników na perełkach ze skrobią (Rys. 2). Aktywność biosyntetyczna szczepu B.subtilis B-3 była niższa w przypadku proteinaz o 76% zaś amylaz o 60% (Rys. 1). Opisaną metodę przechowywania bakterii w postaci wysuszonych przetrwalników na perełkach szklanych można polecić dla szczepu B.polymyxa B-20 i nie jest ona odpowiednia dla szczepu B.subtilis B-3, gdyż bakterie traciły w znacznym stopniu, uzdolnienia biosyntetyczne.

Opis:
Presence of plant species, associated with ancient forests (continuous existence >200 years) can be used for assessing woodland biodiversity. The aim of this study is to specify which plant species, included in the ancient forests plant species list, can indicate old woodlands in parks and what plant features are characteristic for certain habitat types. Richest in ancient forest plant species are those fragments of woodlands, which have existed continuously for at least 120 years, and their fragments suffered least from human pressure during that time, whereas recent woodlands are characterized by a small share of ancient forest indicator species and domination of nitrophilous species. Investigated plants usually propagate vegetatively, their seeds are of a small size and they have regularly, densely set leaves. Plants associated with ancient woodlands are good indicators of woodland continuity in parks so they can be used for age assessment in these areas. Accumulation of such plants can be a useful criterion in parks evaluation. Proposed idea of sustainable management of a park space, including selection of areas of a great ecological value, can be a chance to preserve woodland biodiversity.

This study investigates the feasibility of using an organic substrate in the form of acetic acid to enable wastewater denitrification in a Sequencing Batch Biofilm Reactor (SBBR). The impact of nitrates presence on the yield of biological dephosphatation was determined as well. The experiment included 296 cycles and was divided into 4 series differing in the load of nitrates. The N:(C and P) ratios were: 7:(140 and 7); 35:(140 and 7); 70:(140 and 7) and 140:(140 and 7). The hydraulic retention time in the reactor was 12h (6h of mixing – dissolved oxygen concentration below 0.1 and 6h of aeration – concentration of dissolved oxygen 3.0±0.8 mgO2·dm-3). The study demonstrated that the 30-day adaptation period (60 cycles) was sufficient for the development of a stable biofilm. The C:N ratio of 2 ensured the total nitrogen concentration in the effluent below 1 mgN·dm-3. The mean efficiency of biological dephosphatation reached 7.0, 17.4, 18.7, and 30.3% in series 1–4, respectively. In the case of series 2 and 3, no significant differences were demonstrated in the total phosphorus concentration in the effluent. In the other series, the differences turned out to be significant.
