Triterpenes are group of biologically active compounds which can be found in higher plants. Their main source are plants bark, leaves, twigs, fruits, resins or oils. The biological activity of triterpens is very diversified and many studies have already confirmed the following therapeutic effects: anti-inflammatory, antimicrobial, antiviral, antifungial, immunomodulatory, and hepatoprotective. Synthesis of triterpenes derivates can be performed by chemical or enzymatic reactions, however biotransformation is more specific and eliminates the side products and the molecule alterations. These processes use isolated enzymes or microorganisms. Cell culture in vitro eliminates problems like extract variability as well as instability of the compounds being obtained during the extraction process. What is more, it ensures high reproducibility and optimal regio- and enantioselectivity. The most widely used technique is a classical screening of a series of microbial strains. Studies on triterpene biotransformation give a lot of information about new biologically active compounds and let predict the metabolism of biological compounds. This review presents most important advancements in the metabolic engineering of microorganisms for the production of triterpenoids. Moreover, the review highlights general strategies to obtain rich biochemical diversity of plants by employing the biocatalysts produced by microorganisms or tissue cultures.

Opis:
In the study, the possibility of an application of Ajuga reptans leaf and root extracts in antipollution cosmetics was investigated. The influence of Ajuga extracts on the skin condition was also evaluated. Both leaf and root Ajuga ethanolic extracts were obtained and added to the developed cosmetic formulations. Two types of emulsion W/O and O/W, washing gels and eye serum, containing as an active substance Ajuga extracts were prepared. For the stable formulations physicochemical and user properties were studied. The obtained results show that cosmetic products, containing the Ajuga reptans extracts, positively affect the skin condition: causing an improvement in the degree of skin hydration and elasticity, reducing the skin pores size and skin hyperpigmentation, and reducing the wrinkles depth.

During reaction to stress caused by viral infection, RNA granules are formed in order to protect mRNA. Stress granules (SG) and processing bodies (PB) provide cell homeostasis and mRNA stability. They are formed, for example, during polio virus and MRV (mammalian orthoreovirus) infections. Some viruses, such as influenza virus and HTLV-1 (Human T-lymphotropic virus 1), block the formation of granules. In addition, there are viruses like West Nile Virus, Hepatitis C Virus (HCV) or human Herpes viruses, which influence the functioning of the granules.

Opis:
Trans-Resveratrol, a natural polyphenol found in various plant species, has gained significant attention due to its potential health-promoting properties. This article reviews the biological activities attributed to trans-Resveratrol, its dietary sources, and the analytical methods employed for its quantification. The compound antioxidant, anti-inflammatory, anticancer, cardioprotective, and neuroprotective activities are discussed in detail. Furthermore, the sources of trans-Resveratrol, including grapes, berries, and certain nuts, are examined in terms of their content variability and factors influencing production. Various chromatographic, spectroscopic, and immunoassay methods for the analysis of trans-Resveratrol in different matrices are also explored. This comprehensive overview underscores the significance of trans-Resveratrol as a potential molecule for enhancing human health and longevity.

Non-homologous RNA recombination is a process enabling the exchange of genetic material between various (related or unrelated) RNA-based viruses. Despite extensive investigations its molecular mechanism remains unclear. Studies on genetic recombination in brome mosaic virus (BMV) have shown that local hybridization between genomic RNAs induces frequent non-homologous crossovers. A detailed analysis of recombinant structures suggested that local complementary regions might be involved in two types of non-homologous recombination in BMV: site-specific and heteroduplex-mediated. To verify the above hypothesis and better recognize the mechanism of the phenomenon studied we have tested how the putative types of recombination are affected by a specific mutation in the BMV polymerase gene or by changes in RNA structure. The experiments undertaken revealed substantial differences between site-specific and heteroduplex-mediated recombination, indicating that they occur according to different mechanisms. The former can be classified as homology-assisted, and the latter as homology-independent. In addition to local RNA/RNA hybridization, short regions of homology are required for site-specific crossovers to occur. They are most efficiently mediated if one homologous sequence is located at the beginning of and the second just before a double-stranded region. At present it is difficult to state what is the mechanism of heteroduplex-mediated recombination. Earlier it was postulated that strong RNA/RNA interaction enforces template switching by the viral replicase. There are, however, several observations questioning this model and indicating that some other factors, which are still unknown, may influence heteroduplex-mediated crossovers.

Coprinus comatus strains (CCMs) originating from Poland were identified using ITS region sequencing. Based on the sequences obtained, the genetic relationship between the CCM strains was determined and a clear separation of all strains into two main clusters was obtained. The Coprinus strains were also genetically characterized for the first time by the AFLP technique. The analysis showed that the CCMs separated into four main clusters and a high complication of a UPGMA-based dendrogram was achieved. C. comatus strains included in the analysis displayed an AFLP profile similarity level in the range from 44 to 66%. The highest similarity coefficient, 0.490, was found between CCM12 and CCM13, and the lowest (0.202) between the CCM2 and CCM5 isolates. Biolog FF MicroPlates were applied to obtain data on utilization of 95 carbon sources and mycelial growth. The analysis allowed comparison of the functional diversity of the CCM strains and revealed a broad variability within the analyzed Coprinus species based on substrate utilization profiles. Significant differences (2-48) have been shown in the substrate richness values. The Biolog experiments proved to be a good profiling technology for studying the diversity in shaggy manes due to metabolic differences and demonstrated that all the strains might be considered individually. It is evident that the strain metabolic grouping does not correlate with the grouping based on the ITS sequences and AFLP profiles, however, some similarities may be observed.

Cytotoxicity of laronidase (Aldurazyme®), employed in enzyme replacement therapy (ERT) for mucopolysaccharidosis type I (MPS I) and various siRNAs, tested previously in studies on substrate reduction therapy (SRT) for mucopolysaccharidoses, was tested. The enzyme did not cause any cytotoxic effects, and the siRNAs did not inhibit growth of most investigated cell lines. However, some cytotoxic effects of some tested siRNAs were observed in one MPS IIIA cell line. The efficacy of a combination of enzyme replacement therapy and siRNA-based substrate deprivation therapy was tested on three MPS I cell lines. Surprisingly, different results were obtained for different cell lines. The decrease of glycosaminoglycan storage in cells treated simultaneously with both methods was: (i) less pronounced than obtained with either of those methods used alone in one cell line, (ii) similar to that observed for enzyme replacement therapy in another cell line, and (iii) stronger than that obtained with either of the methods used alone in the third cell line. Therefore, it appears that the effects of various therapeutic methods may strongly depend on the features of the MPS cell line.
