The work describes purification and biochemical characterization of two inducible antimicrobial peptides from the hemolymph of Galleria mellonella. The peptides were isolated by a sequence of reversed-phase chromatography steps from the hemolymph of larvae immunized with viable bacteria. The first peptide is a member of the cecropin family while the second one is rich in proline residues and has a unique sequence.

Opis:
Oxidation of proteins is a common phenomenon in the inflammatory process mediated by highly reactive agents such as hypochlorite (HOCl/OCl-) produced by activated neutrophils. For instance, in rheumatoid arthritis hypochlorite plays an important role in joint destruction. One of the major targets for HOCl/OCl- is collagen type II (CII) - the primary cartilage protein. In our study, HOCl/OCl- mediated collagen II modifications were tested using various methods: circular dichroism (CD), HPLC, ELISA, dynamic light scattering (DLS), fluorimetry and spectrophotometry. It was shown that hypochlorite action causes deamination with consecutive carbonyl group formation and transformation of tyrosine residues to dichlorotyrosine. Moreover, it was shown that ammonium chloramine (NH2Cl) formed in the reaction mixture reacts with CII. However, in this case the yield of carbonyl groups and dichlorotyrosine is lower than that observed for HOCl/OCl- by 50%. CD data revealed that collagen II exists as a random coil in the samples and that chlorination is followed by CII fragmentation. In the range of low HOCl/OCl- concentrations (up to 1 mM) 10-90 kDa peptides are predominant whereas massive production of shorter peptides was observed for high (5 mM) hypochlorite concentration. DLS measurements showed that chlorination with HOCl/OCl- decreases the radius of collagen II aggregates from 30 to 6.8 nm. Taking into account the fact that chlorinated collagen is partially degraded, the DLS results suggest that smaller micelles are formed of the 10-90 kDa peptide fraction. Moreover, collagen chlorination results in epitope modification which affects CII recognition by anti-CII antibodies. Finally, since in the synovial fluid the plausible hypochlorite concentration is smaller than that used in the model the change of size of molecular aggregates seems to be the best marker of hypochlorite-mediated collagen oxidation.

Zinc accumulation and the growth of Saccharomyces cerevisiae were investigated in a culture with zinc sulfate-supplemented medium. The cultivations were performed on Sabouraud dextrose broth medium in aerobic conditions, without the addition of zinc (control culture) and with the addition of zinc sulfate (5, 10, 15, 30 and 60 mg ZnSO l -1 medium) at 28 ° C for 72 hours. The results showed similar trends of yeast growth rates at 24, 48, and 72-hour interval, with concentrations above 10 mg l -1 ZnSO in the nutritional medium significantly decreasing the yeast growth rate and the biomass yield ( P <0.05). Substantial differences between the initial ZnSO concentrations in the growth medium were demonstrated in the overall adsorption of Zn ions (Zn 2+ ) in yeast cells by a colorimetric assay ( P <0.05). Similarly, the content of total accumulated zinc, as well as the fractions of Zn present in cells depended mainly on the zinc concentration in the medium, as the total Zn accumulation and organically bound Zn fractions were increased by elevating the ZnSO supplementation in the culture medium up to 30 mg l -1 , but gradually reduced by any further addition of ZnSO determined by an ICP-MASS assay ( P <0.05). In the presence of 30 mg l -1 ZnSO , the Zn content in the biomass increased by 24-fold, to 4132.34 m g g -1 in comparison to 171.9 m g g -1 achieved in the basal medium. Thus, the ability of S. cerevisiae to accumulate zinc can be used for production of a zinc-rich ingredient for functional food products.

Opis:
Background: The medical device industry needs to be sustainable and should consider the safest, trusted quality and accessibility for use when they are required for patient diagnostic procedures. This study is conducted to identify the factors influencing sustainable development of medical devices. Methods: The analytical hierarchy process (AHP) is being adopted to prioritize the influencing factors. Based on the classified criteria, the alternative factors are evaluated and compared with each other using AHP to make an optimal selection. Results: The findings show that there are six main factors and seventeen sub-factors in this study that can support the development of sustainable medical devices. This study provides useful information for the medical device supplier to improve their current and future product design toward sustainable medical device development. Conclusions: This study adds to the understanding of sustainable medical device development and its consequences on the intention to use from the consumer's perspective.

Kininogens are multifunctional proteins involved in a variety of regulatory processes including the kinin-formation cascade, blood coagulation, fibrynolysis, inhibition of cysteine proteinases etc. A working hypothesis of this work was that the properties of kininogens may be altered by oxidation of their methionine residues by reactive oxygen species that are released at the inflammatory foci during phagocytosis of pathogen particles by recruited neutrophil cells. Two methionine-specific oxidizing reagents, N-chlorosuccinimide (NCS) and chloramine-T (CT), were used to oxidize the high molecular mass (HK) and low molecular mass (LK) forms of human kininogen. A nearly complete conversion of methionine residues to methionine sulfoxide residues in the modified proteins was determined by amino acid analysis. Production of kinins from oxidized kininogens by plasma and tissue kallikreins was significantly lower (by at least 70%) than that from native kininogens. This quenching effect on kinin release could primarily be assigned to the modification of the critical Met-361 residue adjacent to the internal kinin sequence in kininogen. However, virtually no kinin could be formed by human plasma kallikrein from NCS-modified HK. This observation suggests involvement of other structural effects detrimental for kinin production. Indeed, NCS-oxidized HK was unable to bind (pre)kallikrein, probably due to the modification of methionine and/or tryptophan residues at the region on the kininogen molecule responsible for the (pro)enzyme binding. Tests on papain inhibition by native and oxidized kininogens indicated that the inhibitory activity of kininogens against cysteine proteinases is essentially insensitive to oxidation.

Among Legionella species, which are recognized to be pathogenic for humans, L. gormanii is the second prevalent causative agent of community-acquired pneumonia after L. pneumophila. Anti-L. gormanii activity of Galleria mellonella hemolymph extract and apolipophorin III (apoLp-III) was examined. The extract and apoLp-III at the concentration 0.025 mg/ml caused 75% and 10% decrease of the bacteria survival rate, respectively. The apoLp-III-induced changes of the bacteria cell surface were analyzed for the first time by atomic force microscopy. Our studies demonstrated the powerful anti-Legionella effects of the insect defence polypeptides, which could be exploited in drugs design against these pathogens.

The tumor associated antigen RCAS1 (receptor-binding cancer antigen expressed on SiSo cells) expressed with high frequency in various cancer and trophoblast cells, inhibits growth of estrogen receptor-expressing cells and induces apoptosis. Because previous reports demonstrated RCAS1 presence only by non-quantitative immunocytochemistry methods, we decided to use a Western blotting with anti-RCAS1 monoclonal antibodies for estimation of the relative content of the tumor-associated antigen. One hundred tissue samples were assayed (neoplasms, chronic inflammatory diseases, healthy tissues, trophoblasts and placentas at term). RCAS1 was present in all neoplastic, placental and trophoblast tissue samples and its level in malignant samples was statistically significantly higher than in benign neoplasms. The amount of RCAS1 in chronic inflammations was also significantly increased in immune mediated diseases, like allergic nasal polyps and sarcoidosis. The RCAS1 protein was not revealed in healthy mucous membrane and in muscle tissues. The presented results suggest that RCAS1 might play an important role in tumor escape from host immunological surveillance and carry weight in the down regulation of the maternal immune response, thereby maintaining pregnancy.

A small "precursor" of resonance is observed before the main resonance peak in the current-voltage characteristic of double-barrier resonant-tunnelling devices. The competition between the precursor and main-peak current is examined within the temperature range 4.2-400 K. The precursor is interpreted as 3DEG contribution to the resonant tunnelling dominated by a 2DEG injection from a triangular well formed under bias in the emitter spacer layer.

Introduction. Schizophrenic patients present cognitive dysfunctions which are regarded to be one of endophenotypical markers predisposing to schizophrenia. Currently, schizophrenia can be treated as a neurodegenerative and neurodeveloping disease with genetic background. Objective. Assessment of the possible positive effect of neuropsychological rehabilitation in schizophrenia, in patients presenting cognitive dysfunctions. An additional aim was to verify the hypothesis that some genetic polymorphisms can be a prognostic factor for success in neuropsychological rehabilitation. Material and methods. 41 participants and 40 control subjects were randomly selected. Both groups had the diagnosis of paranoid schizophrenia. Cognitive functions were checked with the Wisconsin Card Sorting Test, Trail Making Test, and Stroop Test at the beginning and end of the experiment. In the research group, each patient trained with the rehabilitation programme RehaCom, whereas the control group did not receive such training. Genes COMT rs4680 and BDNF rs6265 were analysed in the genetic part of study. Results. RehaCom procedures appear to be useful in the neuropsychological rehabilitation of cognitive dysfunctions in patients diagnosed with schizophrenia. The research group showed a moderate improvement in the training programmes. Analysis of parameters obtained in the neuropsychological tests showed a slight improvement in both groups. At the present time, analysis of the polymorphisms of genes cannot be treated as a prognostic factor for the success of neuropsychological rehabilitation because statistical analyses showed few dependences with little statistical significance. Conclusions. Cognitive rehabilitation produces moderate improvement in cognitive functioning.

Anisoptera scaphula (Roxb.) Kurz, the tallest tree species in Bangladesh, is critically endangered worldwide due to its poor natural regeneration. This study describes the appropriate regeneration technique for A. scaphula through seed germination and clonal propagation by stem cutting. The fastest and highest germination percentage (65) was observed in dewinged seeds sown inverted orientation with half buried position (T10) followed by dewinged seeds (62.5%) sown horizontally in full buried (T7) and delayed germination with lowest percentage (15) was in control (T0). Optimum initial growth was also shown when seedlings were developed from the seeds under T7. Both winged and dewinged seeds sown vertically in full buried or half buried position produced curved seedlings. However, dewinged seeds sown in horizontal or inverted (winged side down) position produced otherwise i.e., straight seedlings. The highest rooting percentages (63%) and maximum number of root (4.8) per cutting were obtained when one node cuttings treated with 0.8% IBA solution followed by 0.4% IBA in turn affecting for better survival and growth of rooted cuttings in the nursery conditions.
