Opis:
The study involved the use of a bacterial strain isolated from environmental samples which produce the biopolymer in the form of pellets in the submerged culture. This material (bacterial exopolysaccharide) is produced by bacteria of the Komogateibacter xylinus which are prevalent in the environment. The aim of this study was to characterize bacterial exopolysaccharides and commercial dextran-based “microcarriers” in terms of their roughness and cell culture effects, including the morphology and viability of the human hybridoma vascular endothelial cell line EA.hy926. The pellets were characterized using scanning electron microscopy (SEM) and atomic for¬ce microscopy (AFM). The resulting structures were used for cell culture of adherent cells (anchorage¬-dependent cells). At the same time, the cultures with commercial, dextran-based “microcarriers” were carried out for comparative purposes. After com¬pletion of the cell culture (24 hours of culture), the cellulose and commercial “carriers” were analyzed using SEM and AFM. Finally, the obtained cell dens¬ities (fluorescence labelling) and their morphological characteristics (SEM) were compared. The obtained results strongly support the applicability of bacterial exopolysaccharide (EPS) in tissue engineering to build innovative 3D scaffolds for cell culture, the more so that it is technologically possible to produce EPS as spatially complex structure

Nowadays nanostructures are more and more often designed as carriers for drug delivery, especially to improve the drug pharmacokinetics and pharmaco-dynamics. Numerous kinds of nanostructures are considered a good prospect for medical applications thanks to their small size, acceptable biocompatibility and toxicity. Due to the fact that nanotechnology is a new field of science, every nano-scale product must be thoroughly examined regarding its toxicity to the human body. This study provides new insights into effects of exposing endothelial cells to the selected nanostructures. Dendrimers of the fourth generation (PAMAMs), multi-walled carbon nanotubes (MWCNTs) and silver nanoparticles (SNPs) were used to evaluate nanostructures influence on endothelial cells in vitro. The nanostructures were evaluated via transmission electron microscopy and dynamic light scattering technique. The cells previously exposed to the nanostructures were observed and analyzed via the atomic force microscopy and scanning electron microscopy to obtain a quantitative evaluation of the cells morphology. The presence of multi-walled carbon nanotubes and silver nanoparticles on the cells surface was confirmed by the scanning electron microscopy. Our results confirm that the surface association and/or uptake of nanostructures by the cells resulting from physicochemical and biological processes, affect the cells morphology. Morphological changes can be induced by the membrane proteins interaction with nanomaterials, which trigger a sequence of intracel-lular biological processes.

Zasadniczym celem autorów publikacji jest przeprowadzenie analizy i dokonanie oceny podejścia do zarządzania przewozami ładunków wielkogabarytowych w pojazdach nienormatywnych. Analizy dokonano na przykładzie przewozów ponadgabarytowych w Polsce na tle barier i stymulatorów przewozu ładunków ponadnormatywnych występujących w Unii Europejskiej. Tok wywodów ma także na celu zweryfikowanie hipotezy badawczej: „Optymalizacja zarządzania przewozami ładunków wielkogabarytowych w pojazdach nienormatywnych jest możliwa dzięki kompleksowej platformie rejestracji przejazdów, aplikowania o pozwolenie na przejazd, wyznaczania trasy przejazdu, monitorowania przewoźników, a także w drodze permanentnej poprawy komunikacji pomiędzy jednostkami administracji publicznej”. W publikacji: zdefiniowano ładunki wielkogabarytowe i pojazdy nienormatywne oraz dokonano ich uproszczonej klasyfikacji; zaprezentowano miejsce przewozów wielkogabarytowych w transporcie oraz wskazano wymagania związane z poruszaniem się pojazdów nienormatywnych; scharakteryzowano podejście do zarządzania przewozami ładunków wielkogabarytowych w pojazdach nienormatywnych w Polsce; zdiagnozowano bariery oraz stymulatory rozwoju rynku przewozów ponadgabarytowych; zaproponowano modyfikacje w zakresie przewozu ładunków wielkogabarytowych w pojazdach nienormatywnych w Polsce.
The main goal of the authors of this publication is to analyse and evaluate the approach to the management of oversize loads in special transport vehicles. The analysis was made with regard to heavy and oversized transportation in Poland against the background of barriers and stimulants for heavy and oversized transportation in the European Union. The reasoning of the arguments is also to verify the research hypothesis: "Optimization of the management of oversize loads in special transport vehicles is possible thanks to a comprehensive platform for registering such shipments, applying for a permission to operate on roads, determining the route, monitoring carriers, as well as through permanent improvement of communication between public administration units". In this publication: oversize loads and special transport vehicles were defined and roughly classified; the place of heavy and oversized transportation was presented and requirements related to the movement of special transport vehicles were indicated; the approach to managing oversize loads in special transport vehicles in Poland was characterized; barriers and stimulants for the development of heavy and oversized transportation were diagnosed; as for heavy and oversized transportation in special transport vehicles in Poland, modifications were proposed.

Opis:
Benzo[a]pyrene is a polycyclic aromatic hydrocarbon of well proved toxic effect on animal cells and tissues. We used chicken in ovo developmental model to verify its influence on selected parameters of the heart rhythm and on the antioxidative defense in the heart tissue. We determined that the dose of 1 mg/kg weight of eggs of benzo[a]pyrene strongly activates the glutathione-dependent antioxidative system, but it do not significantly affect the heart conducting system of the chicken embryo. We postulate that further study on the benzo[a]pyrene action during embryonic development of birds is recommended.

Opis:
Thin and lightweight achromatic focusing elements with F-number close to 1 are desirable in many practical applications. We present the idea to use diffractive structures designed to work for the substantially increased THz frequency range. The paper analyses mono- and multi-focal lenses forming point-like foci as well as axicon and light sword optical elements focusing THz radiation into line segments located along the optical axis. We consider diffractive elements in a form of the first and the second order kinoforms having various thicknesses. Designed and fabricated elements were numerically and experimentally examined to verify their achromatic functioning. We present point spread functions (XY scans) and 2D energy maps (XZ scans) for different THz frequencies. Moreover, a diagram of chromatic aberration is created by registering energy distribution along the optical axis for different frequencies. The distance corresponding to the highest energy is chosen for each frequency. Therefore, we can compare broadband working of designed structures. The spherical lens coded as kinoform of the second order provides the best broadband functioning, however it is two times thicker than structures providing extended depth of focus (light sword and axicon) working with slightly smaller efficiency but being much thinner.

A complex view of the prehistory in southern Jordan emerges from the excavations of the Jagiellonian University team, which carried out in 2018 its second season of fieldwork at the sites of Munqata’a and Faysaliyya, even as analyses of finds from the previous season were underway. Human communities living here in the Neolithic and Early Bronze Age practiced both sedentary and mobile lifestyles. The changing landscape around them, caused by natural erosion processes and periodical climate change, is also taken into consideration while interpreting the explored relics.

The HLC (Heritage–Landscape–Community) archaeological metaproject, carried out since 2016 by the Jagiellonian University in cooperation with the Department of Antiquities, Ministry of Antiquities and Tourism, Hashemite Kingdom of Jordan, targets the archaeological heritage of southern Jordan (Tafila region), focusing currently on remains of the Early Bronze Age and earlier cultures that were found in the region. The project has already identified and verified several previously undocumented or poorly documented sites. Its main objective is to establish chronological phasing of human activity in this microregion, particularly during the Early Bronze Age, and to assess the scale and nature of human presence in that period. Two sites, Faysaliyya and Munqata’a, were excavated within the frame of the project. The article presents the preliminary results of this work. An important side issue is the protection of Jordanian heritage in the Tafila region through the identification of natural and human agents that may damage or destroy it.
