Globally, tree-holes are important ecological component of forest and woodlands. Numerous microorganisms rely on cavities, both natural and those excavated by primary cavity nesting birds, mainly by woodpeckers, for their survival and reproduction. However, the fungi occurring in cavities are not well characterized. Specifically, very little is known about the fungal communities inhabiting the woodpecker nest cavities. Therefore, in this study, we investigated the fungal diversity of cavities in southern Poland. The samples were collected from freshly excavated woodpecker nest cavities using a nondestructive method (ND). The spatial distribution of fungal communities within the cavities was evaluated by sampling different parts of a single cavity using a destructive method (D). We detected 598 fungal isolates that included 64 species in three phyla and 16 orders using the ND method. Most of the fungi isolated from the cavities represented the phylum Ascomycota (73.9% of the isolates) with 11 orders, and Microascales was the predominant order (30% of the isolates). The most common species detected was Petriella musispora, which was isolated from 65% of the cavities. A total of 150 isolates (25%) were members of Basidiomycota, with Hymenochaetales being the dominant order (16% of the isolates). The basidiomycetous fungi were isolated from 55% of the cavities. Several taxa closely related to the pathogenic fungi and associated with secondary animal infections were detected in the wood of cavities. We identified different fungal communities in the three cavity parts using the D method. The cavity entrance had more number of species than the middle and bottom parts. The results of this study advanced our current knowledge on the mycobiota in woodpecker nest cavities and provided preliminary evidence for tree cavities being the hotspot for fungal diversity.

Opis:
Comparative statistical analysis of the infiuence of processing parameters, for electrospinning (ES) and solution blow spinning (SBS) processes, on nanofibrous poly(L-lactic acid) (PLLA) material morphology and average fiber diameter was conducted in order to identify the key processing parameter for tailoring the product properties. Further, a comparative preliminary biocompatibility evaluation was performed. Based on Design of Experiment (DOE) principles, analysis of standard effects of voltage, air pressure, solution feed rate and concentration, on nanofibers average diameter was performed with the Pareto’s charts and the best fitted surface charts. Nanofibers were analyzed by scanning electron microscopy (SEM). The preliminary biocompatibility comparative tests were performed based on SEM microphotographs of CP5 cells cultured on materials derived from ES and SBS. Polymer solution concentration was identified as the key parameter infiuencing morphology and dimensions of nanofibrous mat produced from both techniques. In both cases, when polymer concentration increases the average fiber diameter increase. The preliminary biocompatibility test suggests that nanofibers produced by ES as well as SBS are suitable as the biomedical engineering scaffold material.

Opis:
A method of manufacturing hydrogel coatings designed to increase the hydrophilicity of polyurethanes (PU) is presented. Coatings were obtained from polyvinylpyrrolidone (PVP) by free radicalpolymerisation. The authors proposed a mechanism of a two-step grafting – crosslinking process and investigated the influence of reagent concentration on the coating’s physical properties hydrogel ratio (HG) and equilibrium swelling ratio (ESR). A surface analysis of freeze-dried coatings using scanning electron microscopy (SEM) showed a highly porous structure. The presented technology can be used to produce biocompatible surfaces with limited protein and cell adhesive properties and can be applied in fabrication of number of biomedical devices, e.g. catheters, vascular grafts and heart prosthesis.

Dextran-polycaprolactone nanoparticles were prepared and the influence of catalyst on particle diameter was investigated. Nanoparticles were synthesized by the simple one-pot reaction of spontaneous dextran modification with ɛ-caprolactone. Afterwards, nanoparticles were purified by dialysis, spray-dried and checked for cytotoxicity by the MTT assay. Prepared particles are a promising carrier for anticancer drug due to their biocompatibility and a narrow size distribution.
Otrzymano nanocząstki dekstranowe z użyciem polikaprolaktonu jako czynnika zwijającego oraz zbadano wpływ ilości katalizatora na ich średnicę. Synteza składa się z prostej jednostopniowej reakcji, w czasie której zachodzi polimeryzacja ε-kaprolaktonu na łańcuchu dekstranowym, a następnie spontaniczne zwinięcie łańcucha w cząstkę. Nanocząstki oczyszczono poprzez dializę, suszono rozpyłowo i przebadano pod kątem cytotoksyczności testem MTT. Otrzymane nanocząstki są obiecującym nośnikiem dla leków przeciwnowotworowych ze względu na wąski rozkład średnic oraz biokompatybilność.

Polyurethanes (PU) are nowadays one of the most widely used types of polymers for biomedical applications. The segmented nature of these materials enables to obtain both flexibility and mechanical strength within the same material. However, there is a need to modify a surface of PU in order to produce highly hemocompatible materials that can be used in fabrication of permanent blood-contacting devices such as vascular implants, valves or hearts. Among many types of modifications, hydrogel coating appears to be very promising. Such modification increases the hydrophilicity of PU which in turn causes an increase in the bio- and hemocompatibility of the material. The aim of presented study was to create a polyvinylpyrrolidone (PVP) hydrogel coating onto a surface of polyurethane. A possibility of using a free radical polymerization reaction was investigated. The obtained materials were analyzed in terms of durability, swelling ratio and contact angle. The FTIR-ATR analysis confirmed that polyurethane surfaces have been successfully coated with PVP.

The phenomenon of blood coagulation on the surfaces of implantable devices such as artifi cial blood vessels, valves or hearts is the major problem that has to be taken into consideration during the construction of such devices. Since the main factor that has an infl uence on clotting of blood is the surface of the material the device is made of, numerous attempts has already been undertaken to modify surfaces of existing materials in order to provide necessary protection against clot formation, very often by coating the materials with bio-active substances. The aim of presented study was to create a coating of phosphatidylcholine that would mimic natural cell outer membrane on the surface of polyurethane — a material already in use in construction of cardiovascular implants. The obtained coated materials have been evaluated with the use of FT-IR/ATR spectroscopy. The change in contact angle of modifi ed material was also investigated. Finally in vitro cytotoxicity tests were performed in order to examine biocompatibility of obtained material.

Nonwoven fibrous mats were produced in the process of solution electrospinning. Polymeric fibres generated in this process consist of poly(L-lactic) acid (PLLA), biodegradable and biocompatible polymer. Produced fibrous mats were examined by scanning electron microscopy and additionally degradation rate of fibrous material was investigated. Obtained fibres exhibit porous surface and fibre diameter varied from 200 nm to 1,2 ım, depending on the process parameters. Low degradation rate of scaffold material was designed for long-term scaffold usage. The influence of solvent type and solution concentration as well as the solution flow rate, applied voltage and the setup geometry on the fibres morphology and diameter were examined and presented. The influence of polymer concentration on the solution viscosity was also evaluated. Further, the degradation rate of obtained fibres was investigated, as well as the influence of degradation process on surrounding environment. Materials produced in electrospinning process have potential application as long-term biodegradable scaffold for tissue engineering, especially in bone tissue, vascular tissue or cartilage tissue engineering.

Because of excellent properties, similar to natural bone minerals, and variety of possible biomedical applications, hydroxyapatite (HAp) is a valuable compound among the calcium phosphate salts. A number of synthesis routes for producing HAp powders have been reported. Despite this fact, it is important to develop new methods providing precise control over the reaction and having potential to scale-up. The main motivation for the current paper is a view of continuous synthesis methods toward medical application of produced hydroxyapatite, especially in the form of nanoparticles.

W prezentowanych badaniach użyto nanocząstek hydroksyapatytu (nanoHAP) do przygotowania nanowłóknistego kompozytu poli-L-kwasu mlekowego/hydroksyapatytu za pomocą elektroprzędzenia. Zbadano średni rozmiar i strukturę nanoHAP, średnią średnicę nanowłókien i ich strukturę. Dodatkowo, zbadano skład chemiczny wszystkich przygotowanych materiałów. Wytworzono nanowłókna PLLA/nanoHAP o średniej średnicy włókna 202 ± 6 nm z jednorodnie rozproszonymi cząstkami nanoHAP w strukturze włóknistej.
In present study, nanoparticles of hydroxyapatite (nanoHAP) were used to prepare nanofibrous composite of poly-L-lactic acid/hydroxyapatite by electrospinning. An average size and structure of nanoHAP, nanofibers average diameter and structure of nanofibrous materials were investigated. Additionally, the chemical composition of all materials was investigated. Nanofibers of PLLA/nanoHAP having the average diameter of 202 ± 6 nm with uniformly dispersed nanoHAP in the fibrous structure were produced.

The main objective of the work was to design and fabricate an injectable biomaterial with osteoconductive properties that can be used in dental applications in peri-implant therapy concerning guided bone regeneration. For that purpose, a self-setting biomaterial consisting of chitosan/tricalcium phosphate microparticles and sodium alginate was formulated. The obtained material was characterized regarding microsphere and formed agglomerates morphology and microstructure. Physical properties, relating to setting time and mechanical properties, were also investigated. Finally, in vivo response to implanted biomaterial was studied on a rat model and compared with commercially available alloplastic material. The results showed that designed injectable biomaterial fulfi lled main requirements for guided bone regeneration application.

Polylactide (PLLA) containing β-TCP is biodegradable composite and an attractive biomaterial for bone tissue engineering, however, hydrophobicity of PLLA based composites is major limitation for their use as scaffolds for cell culture. In our study lecithin was used to improve hydrophilicity and cytocompatibility of PLLA/ β-TCP composite. Thin films of PLLA, PLLA/ β-TCP and PLLA/β-TCP/lecithin were manufactured by solvent-casting technique. Comparative analysis of all types of films was performed. Addition of β-TCP did not change hydrophilicity of PLLA. The hydrophilicity of PLLA/β-TCP/lecithin increased in comparison to PLLA and PLLA/β-TCP. Degradation of PLLA/β-TCP composite surpassed the degradation of PLLA while addition of lecithin diminished the degradation of composite. The cytocompatibility of composites were studied in 7 day long in vitro assay. Human bone derived cells were seeded on all tested surfaces. Cell viability was estimated by Live/Dead fluorescent staining and Alamar Blue test. Surprisingly, although lecithin addition improved hydrophilicity of the PLLA-based composite, adhesion and proliferation of human bone derived cells were markedly hampered on PLLA/β-TCP/lecithin in comparison to PLLA and PLLA/β-TCP. Despite positive effect we found of lecithin addition on hydrophilicity and stability of PLLA-based composite, its effect on cell attachment and proliferation is negative. Hence, incorporation of lecithin did not improve properties of PLLA/β-TCP/lecithin composite intended for bone tissue regeneration.
