The aim of this study was to assess the sensitivity of Escherichia coli isolates originating from a municipal waste landfill to the selected heavy metals. The analyses were conducted using environmental strains, isolated from surface water – a stream flowing along the landfill and from leachate and the observations were compared to the reaction of a reference strain EC ATCC 25922. The growth rate of bacterial cultures was evaluated in the liquid medium supplemented with 0.02; 0.1 and 0.5 mg dm–3 of heavy metal salts: chromium, zinc, cadmium, copper, lead and mercury. The bacterial growth was examined turbidimetrically every 24 hours for 5 days. The performed study showed differences between the examined isolates in their response to the addition of the heavy metals in the liquid medium. Additionally, varied intensity of the heavy metals’ effect on bacterial growth was observed, with the weakest growth inhibition being recorded in the case of lead, while chromium and mercury causing the greatest growth inhibition of bacterial strains.
Celem pracy była ocena wrażliwości izolatów Escherichia coli, pochodzących ze składowiska odpadów komunalnych na działanie wybranych metali ciężkich. Badaniu poddano izolaty środowiskowe, pochodzące z wody powierzchniowej – strumienia płynącego wzdłuż składowiska oraz z odcieków, a także szczep wzorcowy EC ATCC 25922. Ocenie poddano tempo wzrostu kultur bakteryjnych w podłożu płynnym z dodatkiem 0,02; 0,1 oraz 0,5 mg dm–3 soli metali ciężkich: chromu, cynku, kadmu, miedzi, ołowiu i rtęci. The Effect of Heavy Metals on the Growth of Waterborne Escherichia coli... 183 Wzrost bakterii badano turbidymetrycznie w odstępach 24-godzinnych przez okres 5 dni. Na podstawie przeprowadzonych badań stwierdzono różnice pomiędzy badanymi izolatami w ich reakcji na obecność badanych metali ciężkich w podłożu. Zaobserwowano także różną intensywność działania metali, przy czym najsłabsze zahamowanie wzrostu bakterii stwierdzono w przypadku ołowiu, natomiast najsilniejszy efekt hamujący miały chrom i rtęć.

The study investigates the genetic differentiation among two subspecies of Allium ursinum L., namely A. ursinum subsp. ursinum and subsp. ucrainicum as well as their putative hybrid that is represented by individuals with intermediate morphology. Inter-Simple Sequence Repeats (ISSR) were applied to determine the status of intermediate morphotypes in terms of their genetic pattern and to assess the level of genetic variability within and between various populations of A. ursinum. The study comprises 144 specimens from nine populations along the east-west transect in Poland, which includes localities of both subspecies and their putative hybrid. Among the examined populations, 48 bands were amplified, of which 45 were found to be polymorphic. The principal coordinate analysis (PCoA), the neighbour-net analysis and Mantel test showed a strong correlation between genetic variability and geographic distance. Analysis of molecular variance (AMOVA) revealed that a greater proportion of total genetic variation resided within populations rather than among them. The Structure Bayesian clustering analysis revealed the presence of three distinct genetic groups within studied populations, where 'eastern' genotypes correspond to A. ursinum subsp. ucrainicum, and 'western' to subsp. ursinum; whereas the third genetic group has the largest share in the individuals occurring at the border of the distribution ranges of both subspecies. The emergence of the third genetic group is probably an effect of hybridization events occurring within the secondary contact zone. Typical morphologically intermediate populations occur only in a relatively narrow geographical zone, but the hybrid zone revealed by molecular markers is actually much wider than it is suggested by the morphological pattern of individuals. The current distribution pattern of both subspecies of A. ursinum and their hybrid zone is related to the two main directions of postglacial migration of Fagus sylvatica to the area of Poland. The hybrid zone arose as an effect of the secondary contact of two divergent lineages of A. ursinum.

This study aimed to assess microbiological and physicochemical quality of landfill leachate and water in the landfill’s vicinity. Nine samples consisting of groundwater, surface water and leachate were collected within and in the vicinity of a municipal landfill site in Tarnow. Physicochemical analyses of groundwater and leachate included: determination of pH, electric conductivity, heavy metal concentration (Pb, Cd, Cu, Zn, Cr VI and Hg) and sum of policyclic aromatic hydrocarbons. Surface water analyzes were complemented with the determination of Ni, As, Ag, V, volatile phenols concentration, content of total nitrogen and phosphorus, as well as CODCr and BOD5 Microbiological analyzes consisted of determination of total mesophilic and psychrophilic bacteria, Staphylococcus spp., Salmonella spp., the number of coliforms and fecal coliforms, Enterococcus faecalis, Clostridium perfringens, yeast and mold fungi. No threshold values concerning the examined chemical contaminants were exceeded in any of the water or leachate samples. However, the landfill vicinity resulted in severe microbiological contamination of surface and groundwater, which contained high concentrations of not only mesophilic bacteria but also fecal and pathogenic microorganisms. The results of this study revealed that waste and leachates are a major source of microbial emissions. Therefore, there is a need to conduct both physico-chemical and microbiological analyzes of the landfill-surrounding environment and this necessity should be legally enforced.
Celem badań była ocena mikrobiologicznej i fizykochemicznej jakości odcieków oraz wód w rejonie składowiska odpadów komunalnych. Przebadano 9 próbek wód podziemnych, wód powierzchniowych i odcieków zebranych na terenie oraz w rejonie składowiska odpadów komunalnych w Tarnowie. Analizy fizykochemiczne wód podziemnych i odcieków obejmowały: oznaczenie pH, przewodności elektrolitycznej, stężenia metali ciężkich (Pb, Cd, Cu, Zn, Cr VI i Hg) oraz sumy wielopierścieniowych węglowodorów aromatycznych. Analizy wód powierzchniowych uzupełniono o oznaczenie stężeń Ni, As, Ag, V, lotnych fenoli, całkowitego azotu i fosforu, a także ChZTr i BZT5 Wykonano również analizy mikrobiologiczne: oznaczenie ogólnej liczby bakterii mezofilnych i psychrofilnych, Staphylococcus spp., Salmonella spp., bakterii grupy coli, bakterii grupy coli typu kałowego, Enterococcus faecalis, Clostridium perfringens, drożdży i pleśni. Za równo w próbkach wód, jak i odcieków nie stwierdzono przekroczenia wartości granicznych dla żadnych z oznaczanych składników chemicznych. Zauważono jednak, że bliskie sąsiedztwo składowiska przyczynia się do znacznego skażenia mikrobiologicznego wód powierzchniowych i podziemnych, w których stwierdzono duże ilości nie tylko bakterii mezofilnych, lecz również wskaźników zanieczyszczenia kałowego, a także bakterii chorobotwórczych. Wyniki niniejszych badań wykazały, że odpady i odcieki ze składowisk są istotnym źródłem emisji drobnoustrojów do środowiska. W związku z tym ważne jest, aby na terenie i w bliskim sąsiedztwie składowisk prowadzone były nie tylko analizy fizykochemiczne, lecz również oznaczenia mikrobiologiczne, a także aby wprowadzić odpowiednie przepisy, regulujące konieczność i częstotliwość wykonywania wymienionych badań.

This paper shows the possibility of the use of IR thermography for testing the operational parameters of wood stoves. The study showed differences in the operation parameters of the plate steel wood stove, plate steel wood stove with accumulation and accumulative wood stove. There were compared temperature distribution on the external surfaces of tested units and the amount of energy transferred to heated rooms. Moreover, there were also tested two types of units dedicated to heat recovery from flue gas: flue gas to air steel heat exchanger and accumulative ceramic heat exchanger. The use of IR thermography allowed to simplify measurements and gave approximate results in comparison to more detailed tests.

The bird’s-eye primrose (Primula farinosa L.) is an endangered species in Poland. The sole remaining, and critically endangered, population of approximately 300 flowering plants is located in the Beskid Sądecki Mountains (Jaworki, Poland). The genetic investigation was performed using highly variable microsatellite markers on a total of four populations: the Polish population and its three nearest known Slovak counterparts. We hypothesize that the Polish population is a relic of the previously much wider Central European mountain/lowland range and is thus genetically distinct from the rest of the Slovak stock. Clarification of this issue is needed before active protection interventions such as artificial supplementation can be carried out. Our results, particularly those from STRUCTURE Bayesian clustering, showed clear population structure within the dataset: all three Slovak populations were dominated by one genetic group, while the Polish population comprised mostly individuals dominated by the second genetic group. Only limited gene flow was observed between the Polish and Slovak populations. This demonstrates the distinct genetic makeup of the Jaworki population, which is probably the result of prolonged isolation from the rest of the P. farinosa range and admixture of genes from various lineages. Their origin could be determined by including plant material from the rest of the P. farinosa range, i.e., Scandinavian, Baltic, and Alpine/Carpathian locations, in a future study. The immediate practical application of our results is the recommendation that all supplementation interventions to the Jaworki population must be limited to plants produced from locally collected seeds.

We analyzed occupational exposure to potentially infectious body fluids among health care workers (HCWs). Nurses were the most common exposed category of HCWs. In 73.6% cases needle sticks had been the reason of exposure. Recapping a needle was the cause of exposure in 6.9% accidents. Among 189 registered HCWs, 66 (34.9%) performed invasive procedures without any personal protective equipment. Prophylaxis with antiretroviral drugs was necessary in 43 (22.8%) cases. As many as 60.3% of exposure incidents to potentially infectious material result from non-compliance with the relevant recommendations. Continuous education and training is critically needed to prevent occupational exposure to blood-borne infections among health care workers.

The paper presents the cellular automaton (CA) model for tracking the development of dendritic structure in non-equilibrium solidification conditions of binary alloy. Thermal, diffusion and surface phenomena have been included in the mathematical description of solidification. The methodology for calculating growth velocity of the liquid-solid interface based on solute balance, considering the distribution of the alloy component in the neighborhood of moving interface has been proposed. The influence of solidification front curvature on the equilibrium temperature was determined by applying the Gibbs Thomson approach. Solute and heat transfer equations were solved using the finite difference method assuming periodic boundary conditions and Newton cooling boundary condition at the edges of the system. The solutal field in the calculation domain was obtained separately for solid and liquid phase. Numerical simulations were carried out for the Al-4 wt.% Cu alloy at two cooling rates 15 K/s and 50 K/s. Microstructure images generated on the basis of calculations were compared with actual structures of castings. It was found that the results of the calculations are agreement in qualitative terms with the results of experimental research. The developed model can reproduce many morphological features of the dendritic structure and in particular: generating dendritic front and primary arms, creating, extension and coarsening of secondary branches, interface inhibition, branch fusion, considering the coupled motion and growth interaction of crystals.

Aconitum lasiocarpum (Carpathian endemic) and A. variegatum (European endemic) occur sympatrically in the Polish Western Carpathians. Here their taxonomic hybrid A. ×pawlowskii occurs. The aim of this study was to determine the relationship between the taxonomic (Linnaean approach) and genetic structure (PCR-ISSR analysis) of the populations and individuals in two allopatric and four sympatric populations. We determined 309 individuals (OTUs) to species, subspecies and nothospecies using the Linnaean system of classification, and then genetically fingerprinted 39 randomly chosen OTUs. Comparison of the Nei and Li distances obtained from ISSR and morphological matrices using the Mantel test indicated a significant correlation (n = 39, r = 0.53, P = 0.001). Genetic analysis (NEWHYBRIDS) pointed to 7 OTUs as being later-generation hybrids (B1 introgessants) in the sympatric area. Five of them belong to A. variegatum, indicating cryptic introgression, and two belong to A. ×pawlowskii. Nonmetric multidimensional scaling (NDMS) showed gene flow between A. lasiocarpum and A. ×pawlowskii. Allopatric, morphologically pure A. lasiocarpum and A. variegatum populations differed significantly in their ISSR profiles (Fischer's R×C test, P < 0.0001). Expected heterozygosity (Hj) was significantly (p=0.05) lower in allopatric (0.1261–0.1268) than in sympatric populations (0.1348–0.1509), indicating a genetic melting pot in sympatry. The results support the existence of a natural interspecific hybrid swarm zone in the sympatric area of occurrence of Aconitum, and the taxonomic circumscription of the nothospecies within the Linnaean taxonomic system.

Phylogenetic relations within Aconitum subgen. Aconitum (Ranunculaceae) in Europe are still unclear. To infer the phylogeny of the nuclear (ITS) region and chloroplast intergenic spacer trnL(UAG)-ndhF of the chloroplast DNA (cpDNA), we analyzed 64 accessions within this taxon, 58 from Europe and six from the Caucasus Mts. Nuclear ITS sequences were identical in 51 European and two Caucasian accessions, whereas the remaining sequences were unique. cpDNA sequences could be categorized into five haplotypes, i.e., A–E, including a European-Caucasian Aconitum haplotype B. Ten cpDNA sequences were unique. A 5-bp indel distinguished the diploids from the tetraploids. None of the extant European diploids were basal to the tetraploid local group. A phylogenetic tree based on combined ITS and cpDNA sequences (bayesian inference, maximum likelihood, minimal parsimony) placed Aconitum burnatii (Maritime Alps, Massif Central) and A. nevadense (Sierra Nevada, Pyrenees) in a sister group to all other European species. A Bayesian relaxed clock model estimated the earliest split of the Caucasian species during the Late Miocene [ca. 7 million years ago (Mya)], and the divergence of A. burnatii and A. nevadense from the European genetic stock during the Miocene/Pliocene (ca. 4.4 Mya). Diploids in Europe are likely to be descendants of the Miocene European-Caucasian flora linked with the ancient Asian (arctiotertiary) genetic stock. The origins of the tetraploids remain unclear, and it is possible that some tetraploids originated from local, now extinct diploids. Both the diploids and tetraploids underwent rapid differentiation in the Late Pliocene – Quaternary period.

The genetic diversity of two Aconitum species endemic to the Carpathian Mountains and Sudetes was studied. A reticulate evolution between them was earlier postulated as an effect of secondary contact. The genetic diversity at the individual and taxonomic levels was examined across the entire geographical ranges of the taxa in 11 populations based on 247 AFLP markers found in 112 individuals in the Sudetes and Western Carpathians. The overall genetic differentiation was greater within the Sudetic A. plicatum (Fst = 0.139, P < 0.001) than within the Carpathian A. firmum (Fst = 0.062, P < 0.001), presumably due to the long-lasting geographic isolation between the Giant Mts and Praděd (Sudetes) populations of the species. Interestingly, relatively distant and presently isolated populations of A. plicatum and A. f. subsp. maninense share a part of their genomes. It could be an effect of their common evolutionary history, including past and present reticulations. The introgression among infraspecific taxa of Aconitum is common, probably as a result of seed dispersal within a distance of ca. 20 km (Mantel's r = 0.36, P = 0.01). Aconitum f. subsp. maninense had the highest genetic diversity indices: Nei's h and rarefied FAr, and divergence index DW (P < 0.05), pointing to its presumably ancient age and long-term isolation.

Aconitum bucovinense, a high-mountain species endemic to the Eastern and Southern Carpathians, including the Apuseni Mountains, is legally protected and classified in the Polish Red Data Book of Plants. It attains its NW geographical range in two peripheral populations in the Western Bieszczady Mountains (Polish Eastern Carpathians), isolated by a distance of 13.1 km. PCR-ISSR analysis has been used to elucidate the within- and among-populational levels of species genetic diversity. A UPGMA and block clustering showed discreteness of the populations and subpopulations based on ISSR banding pattern. Analysis of Molecular Variance (AMOVA) revealed significant divergence (P = 0.024) of the two marginal populations and highly significant (P < 0.001) differentiation of subpopulations within populations. The theta index calculated for the two marginal populations and the core population in the Carpathians was 0.131 ±0.030 S.D. Most of the population-genetic diversity indices of the marginal populations were not different from those in the core area but the Shannon’s and rarity indices were lower in the marginal populations. It seems that founder effect and subsequent genetic bottleneck resulted in a fine-scale population genetic structure. The marginal populations under study need a relevant recovery program to maintain their genetic diversity.

Opis:
Currently existing solutions rarely protect message integrity, authenticity and user anonymity without burdening the user with details of key management. To address this problem, we present Aldeon—a protocol for anonymous group conversations in a peer-to-peer system. The efficiency of Aldeon is based on a novel tree synchronization algorithm, which is proposed and discussed in this paper. By using this algorithm, a significant reduction in the number of exchanged messages is achieved. In the paper, the formal definition of the proposed hash branch function and the proof of its efficiency are presented.

Opis:
The aim of our study were qualitative and quantitative analyses of two polyphenolic acids: chlorogenic and gallic acids. These compounds were determined in two species of Rhodiola: R. kirilowii and R. rosea. After collecting plants, aqueous and hydroalcoholic extracts were prepared. In order to identify analysed polyphenolic compounds ultra performance liquid chromatography - tandem mass spectrometry (UPLC-MS/MS, Waters) was used. Gallic acid is commonly found in the roots of these plants. Aqueous extract in both species is a rich source of gallic acid. The UPLC-MS/MS studies allow to use this analytical method for determination of polyphenolic acids accordance with the requirements of ICH. Chromatographic method developed by our team is more precise then previously published.
W Instytucie Włókien Naturalnych i Roślin Zielarskich podjęto badania mające na celu opracowanie metody detekcji kwasu chlorogenowego oraz galusowego za pomocą ultrasprawnej chromatografii cieczowej sprzężonej z tandemowym spektrometrem mas (UPLCMS/ MS, Waters). Badaniom poddano dwa gatunki różnica: Rhodiola kirilowii oraz R. rosea. Rośliny zostały wyhodowane w uprawie gruntowej w Instytucie. Przeprowadzona walidacja metody pozwoliła na jej wykorzystanie w ocenie zawartości kwasu chlorogenowego oraz galusowego w badanych roślinach, ponieważ zawartość analizowanych związków zależna jest zarówno od gatunku jak i warunków uprawy.

The aim of the study was the identification and quantitative analysis of phenylpropanoid compounds in the roots of Rhodiola species. Rosavin, rosarin and rosin were determined in the roots of R. kirilowii and R. rosea from the field cultivation, Institute of Natural Fibres and Medicinal Plants. For the quantitative analysis, the ultra performance liquid chromatography - tandem mass spectrometry (UPLC-ESI MS/MS, Waters) was used. The results showed differences in the quantitative and qualitative assessments of these two species. In the root of R. kirilowii the presence of phenylpropanoids was not confirmed. In R. rosea the most common phenylpropanoid was rosavin (0.022%). The UPLC-MS/MS studies allowed to use this analytical method for determination of phenylpropanoids in the accordance with the requirements of ICH.
Celem przeprowadzonych badań w ramach projektu badawczego było opracowanie metody analitycznej pozwalającej na oznaczenie trzech związków fenylopropanoidów w dwóch gatunkach różeńca. Do detekcji rozawiny, rozyny i rozaryny wykorzystano wysokosprawny chromatograf cieczowy sprzężony z tandemowym spektrometrem mas (UPLC-MS/MS). Obydwa gatunki różeńca Rhodiola kirilowii oraz Rhodiola rosea zostały zebrane z upraw prowadzonych w Instytucie Włókien Naturalnych i Roślin Zielarskich w Poznaniu w 2009 r. Dodatkowo z tych surowców przygotowano po dwa wyciągi: wyciąg wodny oraz wodnoalkoholowy (50% etanol), które następnie przebadano pod względem zawartości fenylopropanoidów. Wszystkie przeprowadzone analizy potwierdziły możliwość wykorzystania tej metody do oznaczenia zawartości fenylopropanoidów w rodzaju Rhodiola.

Purpose: To evaluate chosen mucins (mucin 1, 4, 5AC) expression in pancreatic intraepithelial neoplasia, which is a precursor lesion of pancreatic ductal adenocarcinoma. Materials and methods: The study group included 70 patients operated on due to inflammation, cysts and pancreatic ductal adenocarcinoma with pancreatic intraepithelial neoplasia revealed additionally. Mucin 1, 4 and 5AC expression was assessed by immunohistochemical method using polyclonal antibodies. Results: Statistical analysis proved a positive correlation between the expression of mucin 1, 4 and 5AC proteins and the presence and staging of pancreatic intraepithelial neoplasia (p<0.001). Statistically significant correlations were determined between mucin 1, 4 and 5AC and the location of PanIN lesion in the pancreas. Positive correlations were found between mucin 5AC expression and the type of a basic disease (p=0.014). Differences in the expression of MUC 1, 4 and 5AC proteins between healthy pancreatic ducts and various stages of pancreatic intraepithelial neoplasia were statistically significant (p<0.001). Conclusions: Overexpression of mucin 1, 4 and 5AC is related to the presence of pancreatic intraepithelial neoplasia. This suggests that overproduction of mucus is a phenomenon occurring early in the process of carcinogenesis in the pancreas and has its beginning in precancerous lesions of an early stage.
