Opis:
Chitosan (Ch) is an attractive biopolymer with multiple reactive groups. However it is poorly soluble at neutral pH. Quaternization improves its solubility and permits the development of various positively charged drug delivery systems. The aim of this work was to study the solubility, toxicity, cell binding, and penetration of 20 kDa chitosan with 9, 40, 58 and 98% of quaternary ammonium group substitution (ChQ1 to ChQ4 accordingly). We showed that ChQ with substitution degree >40% was soluble in a wide pH range. Unexpectedly ChQ2 and ChQ3 were more toxic to cells than Ch, ChQ1 and ChQ4. Higher toxicity of ChQ was found against macrophage like cell line RAW264.7 than against epithelial cells MiaPaCa-2. All ChQ, in contrast to unmodified Ch, easily bound and penetrated the cells with the highest uptake by ChQ4. Thus, quaternized chitosan derivatives can be used for biomedical applications.

This paper presents the results of chitosan acylation with linoleic (LA) and dilinoleic acid (DLA). The chemical structures of the new derivatives with different degrees of substitution of LA and DLA were assessed by Fourier transform infrared spectroscopy. Polymer films were prepared by solution casting and drying. The resistance of films to bacterial degradation was tested according ISO 846 standard: “Plastic-Evaluation of the Action of Microorganisms” and the antimicrobial properties were assessed using Gram-negative bacterium Escherichia coli and the fungi Candida albicans in an agar diffusion test. Bioassays showed an antimicrobial effect in direct contact with the material.

This article provides data on the analysis of the properties of chitosan derivatives as a functional ingredient used in food technologies as a structure-forming agent, immunomodulator, enterosorbent of xantobiotics, anti-sclerotic and anti-arthritic factor, and as a regulator of the acidity of gastric juice. The expediency of the use of N-methylallyl-chitosan (chitosan derivative) as a substance that improves the functional-technological (moisture and fat-retaining power) and rheological properties of meat-and-vegetable emulsions is shown. The modes of micronising the emulsion depending on the chitosan derivative mass concentration, the speed of rotation of the knives of the microniser and the processing time were studied. Dependences on the change of moisture and fat-retaining power (MRP, FRP) and effective viscosity (η) on the above-mentioned parameters were investigated. The parameters of micronising were validated: the speed of rotation of the knives (υ) was 2500 rpm, the processing time (τ) was 60 s, υ was 3000 rpm, and τ was 50 s; also, the mass fraction of chitosan derivatives was 0.06%, which ensures the production of meat-and-vegetable emulsions of high quality.

Chitosan and its derivatives are renewable biopolymers characterized by high biocompatibility; therefore, they are harmless to humans and allow immune tolerance and improved hydrophilicity. Moreover, chitosan has been the most studied of all polysaccharides used in biomedical applications during the last decade. Combinations of chitosan and porphyrinoid compounds in hybrid materials have revealed many potential applications for biomedical sciences. The main advantage of such materials is an increase in the solubility of porphyrinoids in body fluids and therefore greater release of singlet oxygen to the treated tissue. Chitosan-based drug delivery systems can improve the targeting of porphyrinoids and their release at predetermined locations and finally achieve desired therapeutic effects with minimal side effects. Hence, porphyrinoid-chitosan materials can be applied in drug delivery systems, cancer theranostics and magnetic resonance imaging. The combination of chitosan and porphyrinoids also appears useful in the healing and repairing of damaged organs, tissue engineering, regenerative medicine, as well as dressing materials. Huge benefits are related to the treatment of wounds, which has been presented for self-healing hydrogels based on chitosan and porphyrinoids. Furthermore, the chitosan/porphyrinoid combinations have revealed enormous benefits for antimicrobial photodynamic therapy.

Chitosan derivatives with a series of fatty acids (FA) have been developed using simultaneous N- and O-acylation reaction by the combination of two ways of conducting the reaction, i.e. a carbodiimide catalysis and ionic amino group protection. The chemical structure of chitosan derivatives as well as the characterization of the FA substitution degree were done by the IR spectra analysis. The correlation between the substitution of the chitosan functional groups as well as the saturation of FA and the changes of structural and thermal properties of the derivatives has been presented.

Opis:
Chitosan is one of the most promising polymers for biomedical applications due to its unique properties, such as its biocompatibility, low toxicity, biodegradation, and the presence of reactive amino and hydroxyl groups. Analysis of physicochemical properties of chitosan and its derivatives is a time-consuming process and requires expensive equipment and large amounts of the sample. This paper proposes a method for express analysis of the molecular weight (MW), the degree of deacetylation (DD), the substitution degree (SD), and the charge of chitosan and its derivatives using agarose gel electrophoresis under acidic and neutral conditions with Coomassie staining. Positively charged chitosan samples required acidic Tris Acetate-EDTA (TAE) buffer to move in the gel. The electrophoretic mobility of chitosan depended on MW, DD, SD, and the chitosan charge. Based on the dependences obtained by the proposed method, the MW and DD of commercial chitosan samples were determined. Express analysis of chitosan and its derivatives in agarose gel can be used to monitor the reactions of chitosan modification and to analyse samples with unknown characteristics.

In modern medicine, many diseases can be treated using gene therapy, which requires a DNA delivery system to prevent DNA from degradation and to transport it to the cells. Liposomal reagents are expensive for such a therapy and new inexpensive biodegradable DNA carriers are required. Chitosan (Ch) is a cationic polymer with promising potency for gene delivery. Some Ch derivatives have been shown to efficiently transfect mammalian cells in vitro. However, there are many inconsistencies in the literature concerning the effectiveness of Ch systems for in vivo gene transfer. The aim of this work was to develop a Ch-based vector for in vivo delivery of a large-size DNA fragment coding for the far-red fluorescent protein (RFP). Among several Ch derivatives, hexanoyl-Ch (HCh) with a molecular weight of 20 kDa effectively transfected cells in vitro. Intratumoural injection of polyplexes in colon and lung tumours resulted in local expression of RFP in tumours.

Biodegradable nanoparticulated carriers have important potential applications for administration of therapeutic proteins and peptides. Chitosan based nanoparticles have attracted attention due to their biological properties such as biodegrability and biocompatibility. However, chitosan nanoparticles are not stable in colloid form. Therefore, to increase the stability of these particles is an important problem. The aim of this work was to obtain stable nanoparticles formed by N-acyl derivatives of chitosan by ionic gelation and to investigate their ability to interact with acidic, neutral and basic proteins.

An increase in the number and volume of surgical interventions leads to an increase in the frequency of postoperative adhesions. The development of the adhesion process in the abdominal cavity causes pain, a decrease in the quality of life of patients, a violation of the reproductive function of women as well as acute adhesion intestinal obstruction. Recently, polymer biomaterials, including those based on chitosan, have been widely used for the prevention of adhesions. Due to their biocompatibility and biodegradation ability, they do not require repeated operations to extract the material. It is believed that these materials act as barriers, physically separating the damaged surfaces. The molecular mechanism of their action is still poorly understood. In this review, the main mechanisms of adhesion formation, as well as ways to prevent them with the help of materials based on chitosan and its derivatives, are discussed

This article presents the results of the development process for the preparation of micro and nano-sized polymer-colloid complexes (РСС) on the basis of water-soluble natural polymer chitosan (СTZ) and the sodium salt of chitosan succinylamid (SСTZ) with silver halide sols in aqueous media. Results of research of СTZ, sodium salt of SСTZ solutions and PСС of CTZ and SСTZ with colloidal particles of silver iodide influence on structurally-functional properties of erythrocytes’ membranes on model of acidic hemolisis are presented in the article. Their influence on the nature of erythrocytes distribution by degree of their stability and on kinetic parameters (the beginning, intensity and completion of process of their destruction) under the influence of the damaging agent (HCl) is shown. The comparative analysis of results convinces that СTZ, SСTZ solutions and disperse systems on the basis of PСС of СTZ and SСTZ with colloidal particles of the silver iodide are capable of modulating variously matrix properties of erythrocytes of blood.

This review considers articles on the formation of hydrogels based on chitosan as well as succinylated and quaternized chitosan derivatives. They are synthesized using low toxicity reagents, under ordinary conditions (low production costs). Chitosan derivatives are soluble in an extended range of pH values and characterized by mucoadhesiveness, bioavailability and biodegradability, which extends the potential of their medical applications. One of the most important properties of chitosan and its derivatives is the ability to form hydrogels. Depending on the nature of the bonds that occur during formation, hydrogels are divided into chemically or physically crosslinked, or a mixture of the two. Chemically crosslinked gels have covalent bonds, while physically crosslinked gels are formed by noncovalent interactions, for example, ionic. Mixed hydrogels have both types of crosslinking.
