In this study, 50 samples of air particulates collected from different places in- and outside the Al-Tuwaitha nuclear site, south of Baghdad were used to measure daily gross alpha and beta activity concentrations (AAC and BAC) for the period from 28 January 2015 to 13 April 2017. At the same time, several meteorological factors such as air temperature, wind speed, wind direction, air pressure, relative humidity, and solar radiation, were also measured. Air stability classes were also derived from wind speed and solar radiation. AAC/BAC variations in the surface air layer were discussed in relation to these factors. The results show that there are inverse relations between AAC/BAC and wind speed and temperature, linear relations between AAC/ABC and air pressure and weak relations between AAC/BAC and relative humidity and solar radiation. Lastly, AAC/BAC measurements in unstable air are as large as in neutral air.

In this study, after the pulse shape calibration of a liquid scintillation counting (LSC) spectrometer (Quantulus 1220), the efficiency was determined depending on sample quenching parameters. Then, gross alpha and beta activities in two spiked water samples obtained from International Atomic Energy Agency (IAEA) were used for the validation of the ASTM D7283-06 method, which is a standard test method for alpha and beta activity in water by LSC. Later, the drinking water samples (35 tap water and 9 bottled water) obtained from different districts of Ankara, Turkey, were measured. The maximum gross alpha activities are measured to be 0.08 Bq/L for tap waters and 0.13 Bq/L for bottled waters, whereas the maximum gross beta activities are found to be 0.18 Bq/L for tap waters and 0.16 Bq/L for bottled waters. These results indicate that these drinking water samples are below the required limits, which are 0.1 Bq/L for alpha emitting radionuclides and 1 Bq/L for beta emitting radionuclides. As a result, gross alpha and beta activities in drinking water of Ankara were determined accurately by this validated LSC method. It is also worth noting that LSC is a rapid and accurate method for the determination of gross alpha and beta activities without requiring a tedious sample preparation.

Knowledge about the spatio-temporal variability of soil enzymatic properties is crucial in evaluating their structure-function relationship and their impact on ecosystem functions. The aim of this study was to assess the spatio-temporal variability of soil β-glucosidase (BG) activity against selected physico-chemical properties at the arable fild scale. A grid soil sampling (10×10 m) was used to measure the spatial variation of soil properties across a 0.4-ha fild. The BG activity was analyzed in soil samples that were collected from the upper 20 cm of Luvisols at 50 locations in April and August 2007 and 2008. Additionally, total organic carbon (CORG), total nitrogen (NTOT) concentrations, soil pHKCl as well as texture and water content were determined. The dataset was analyzed using classical statistical and geostatistical methods. Based on the analysis of variance, it was found that the BG activity, CORG, and water content showed signifiant differences between the four sampling dates. The BG activity showed a high contribution of nugget effect in sill (over 50%) and revealed a moderate spatial structure. The range of spatial autocorrelation calculated for the BG activity was between 15.0 and 61.0 m. Less intensive sampling grid should be recommended for soil BG activity in further studies concerning spatial variability in arable fild scale. In turn, more frequent sampling must be included in the sampling strategy in order to better understand whether the BG activity always shows permanent spatial patterns in soil or whether it is more randomized.

A pot experiment was carried out to determine the effect of soil (loamy sand and sandy loam) contamination with copper doses of 0, 150, 450 mg Cu·kg-1 d.m. soil on the activity of β-glucosidase (EC 3.2.1.21), acid phosphatase (EC 3.1.3.2), alkaline phosphatase (EC 3.1.3.1) and arylsulfatase (EC 3.1.6.1) in soil. The resistance of these enzymes to copper pollution was also estimated. Soil samples were contaminated with copper chloride. The experiment was carried out in five replications, in two series. The first series was performed on uncropped soil and the second one — on cropped soil. The experimental plants were oat, spring rape and yellow lupine. The activity of soil enzymes was determined in the analyzed samples on the 25th and the 50th day of the experiment. The results of the experiment showed that copper contamination in doses of 150 mg to 450 mg·kg-1 soil significantly inhibits soil’s biochemical activity. The sensitivity of the tested enzymes to copper was determined in the following order: alkaline phosphatase > arylsulfatase > acid phosphatase > β-glucosidase. The resistance of the above enzymes to copper depended on the cultivated plant spe- cies, soil type and the type of soil use and management. In samples of sandy loam, copper induced the smallest change in the activity of acid phosphatase and alkaline phosphatase, and in loamy sand — in the activity of arylsulfatase and acid phosphatase. In uncropped soil, copper was the least effective in changing the activity of arylsulfatase and acid phosphatase. All of the tested enzymes were less resistant to copper contamination in cropped than in uncropped soil. In soil planted with oat, β-glucosidase was the most resistant and arylsulfatase was the least resistant enzyme to copper contamination. In samples sown with spring rape, the analogous enzymes were arylsulfatase and alkaline phosphatase. In yellow lupine treatments, alkaline phosphatase was the most and β-glucosidase was the least resistant enzyme.
W doświadczeniu wazonowym badano wpływ zanieczyszczenia gleby (piasku gliniastego oraz gliny piaszczystej) miedzią w dawkach: 0, 150, 450 mg Cu·kg-1 s.m. gleby na aktywność: β-glukozydazy (EC 3.2.1.21), fosfatazy kwaśnej (EC 3.1.3.2), fosfatazy alkalicznej (EC 3.1.3.1) i arylosulfatazy (EC 3.1.6.1) w glebie. Określono także odporność tych enzymów na zanieczyszczenie miedzią. Glebę zanieczyszczano chlorkiem miedzi. Badania prowadzono w 5 powtórzeniach, w dwóch seriach. W pierwszej serii doświadczenia gleba była nieobsiana roślinami, w drugiej — obsiana. Roślinami doświadczalnymi były: owies, rzepak jary i łubin żółty. W 25. i 50. dniu trwania eksperymentu oznaczono w próbkach glebowych aktywność enzymów glebowych. Stwierdzono, że zanieczyszczenie gleby miedzią w zakresie od 150 mg do 450 mg·kg-1 gleby istotnie hamuje jej aktywność biochemiczną. Testowane enzymy, pod względem wrażliwości na miedź, można uszeregować następująco: fosfataza alkaliczna > arylosulfataza > fosfataza kwaśna > β-glukozydaza. Odporność enzymów na działanie miedzi zależała od gatunku uprawianej rośliny, rodzaju gleby i sposobu jej użytkowania. W glinie piaszczystej miedź wywołała najmniejsze zmiany w aktywności fosfatazy kwaśnej i fosfatazy alkalicznej, natomiast w piasku gliniastym — β-glukozydazy oraz arylosulfatazy. W glebie nieobsianej roślinami miedź wywoływała najmniejsze zakłócenia w aktywności arylosulfatazy i fosfatazy kwaśnej. W glebie obsianej roślinami wszystkie testowane enzymy były mniej odporne na zanieczyszczenie miedzią niż w glebie obsianej. Najbardziej odpornym enzymem na działanie miedzi pod uprawą owsa była β-glukozydaza, a najmniej — arylosulfataza, pod uprawą rzepaku jarego, odpowiednio — arylosulfataza i fosfataza alkaliczna, natomiast pod uprawą łubinu żółtego najbardziej odporna była fosfataza alkaliczna, a najmniej β-glukozydaza.

Increased ROS generation by the overload by metabolic substrates mitochondria paralleled by decrease of antioxidant activity are typical events found in metabolic syndrome and diabetes type 2. Metabolites of beta-carotene (BC) such as retinoic acid (RA), as well as low concentration of reactive oxygen species (ROS) modify the mitochondrial bioenergetic function. The aim of the study was to investigate the effect of beta-carotene on mitochondrial activity in human preadipocytes. BC used in concentrations, 10 or 30 µM, decreased mitochondrial membrane potential, inhibited mitochondrial respiration and decreased cellular ATP content. We conclude, that BC, the known antioxidant may decrease oxidative phosphorylation capacity of mitochondria.

Four populations of J. communis L. subsp. communis of natural localities in forested regions of the Bory Tucholskie (Poland) were analysed in respect of phenolic compounds concentration and –glucosidase activity. The variability noted in needles was confirmed by adaptation changes; water-logged and poor insolated localities (deciduous mixed fresh forest, dmff – no 3 and coniferous mixed bog forest, cmbf – no 4) stimulated the phenolics accumulation and enzyme activity. On the contrary, the level of phenolics in unripe berry-like cones was almost two fold lower under disadvantageous juniper growth and fructification conditions, whereas the mature berries from poor vigour plants were characterized by higher concentrations of these metabolites. Obtained results show that morphological variation of juniper population was accompanied by variability in phenolic metabolism.

The effect of copper stress on betacyanin accumulation and guaiacol peroxidase (GPOD) activity in leaves of different age was evaluated in red beet (Beta vulgaris L. var. Crosby Egyptian) plants. In hydroponic culture, plants were treated with 0.3 μM (control), 50 μM, 100 μM, and 250 μM of CuSO4 for 6 days. Copper was taken up and accumulated in old roots but was not translocated to leaves. However in young leaves, the increase of lipid peroxidation and reduction of growth were evident from day 3 of copper exposure; whereas in old leaves, the lipid peroxidation and growth were the same from either copper-treated or control plants. In response to copper exposure, the betacyanin accumulation was evident in young leaves by day 3, and continued to increase until day 6. Betacyanin only were accumulated in old leaves until day 6, but the contents were from 4 to 5 times lower than those observed in young leaves at the same copper concentrations. GPOD activity increased 3.3- and 1.4-fold in young and old leaves from day 3 of copper treatment respectively, but only in the young leaves was sustained at the same level until day 6. Old roots shown betacyanin in the control plants, but the betacyanin level and growth were reduced with the copper exposure. In contrast, young roots emerged by copper effect also accumulated copper and showed the highest betacyanin content of all plant parts assayed. These results indicate that betacyanin accumulation and GPOD activity are defense responses to copper stress in actively growing organs.

Amaranthus caudatus L. seed germination was studied under different levels of water deficit induced by PEG 6000 in laboratory conditions. PEG at osmotic potentials -0.2 to -0.3 MPa at 24°C in darkness delayed germination and reduced final germination percentage. PEG solutions at osmotic potential lower than -0.3 MPa almost totally blocked seed germination. Ethephon was much more effective than GA3 in reversing PEG-caused inhibition of A. caudatus seed germination. PEG decreased α-amylase activity after 14 h incubation. It decreased β-amylase activity after 14 and 20 h, and caused an increase in total dehydrogenase activity only after 20 h of incubation. Unlike GA3, ethephon increased α-amylase activity in seeds after 12 and 14 h of incubation under water deficit. After 20 h of incubation there was no difference in α-amylase activity in any of the treatments. Neither ethephon nor GA3 affected the activity of β-amylase and dehydrogenase.

β-glucosidase (E.C. 3.2.1.21), an enzyme involved in cellulose degradation, plays an important role in the soil organic carbon cycle. Cellulose is the most abundant organic compound in the biosphere so a product of its enzymatic hydrolysis is important as an energy source for soil microorganisms. Since β-glucosidase is very sensitive to different factors, determination of its activity might be helpful in soil quality monitoring. The objective of the study was to assess the effect of various doses of farmyard manure (FYM) and mineral nitrogen on β-glucosidase activity in soil samples taken under winter wheat cultivated in crop rotation systems depleting soil from organic matter (A) and enriching soil in organic matter (B). Soil samples were taken in 2002 from a two-factor fertilization experiment carried out as randomized sub-blocks cropped with winter wheat cultivated on lessivé soil. The experiment was located at the Experimental Station of the Institute of Tillage and Soil Science in Grabowo on the Vistula River. All fertilization combinations included FYM (0, 20, 40, 60 and 80 t.ha–1) and nitrogen fertilization (0, 40, 80 and 120 kg ha–1). The activity of β-glucosidase was determined according to Eivazi, Tabatabai (1988). The enzyme activity ranged 3.604-7.041 mM pNP g–1 h–1 in soil samples taken from crop rotation A and between 4.931-7.445 mM pNP g–1 h–1 in those collected from the crop rotation enriching the soil in organic matter. These data were closely related to the applied FYM and nitrogen fertilization doses. Moreover, β-glucosidase activity depended significantly on sampling dates. Enzyme activity was closely connected with soil organic carbon and total nitrogen content, which was confirmed by highly significant correlation coefficients between these parameters (r=0.611-0.770 for Corg, and r=0.844-0.912 for Nog; p<0.01 and p<0.001).
Biorąca udział w rozkładzie celulozy β-glukozydaza (E.C. 3.2.1.21) odgrywa ważną rolę w obiegu węgla w glebie. Jak wiadomo, celuloza to jeden z najobficiej występujących związków organicznych w biosferze, a produkt jego hydrolizy enzymatycznej stanowi cenne źródło energii dla mikroorganizmów glebowych. Ponieważ β-glukozydaza jest bardzo czuła na działanie różnorodnych czynników, oznaczanie jej aktywności może być pomocne w monitorowaniu jakości gleby. Celem pracy było określenie wpływu zróżnicowanych dawek obornika i nawożenia azotowego na aktywność β-glukozydazy w próbkach gleby spod pszenicy ozimej uprawianej w zamianowaniu zubożającym (A) i wzbogacającym (B) glebę w materię organiczną. Próbki gleby do badań pobrano w 2002 r. spod pszenicy ozimej uprawianej na glebie płowej w dwuczynnikowym doświadczeniu nawozowym założonym metodą losowanych podbloków. Doświadczenie zlokalizowano w RZD w Grabowie nad Wisłą. W każdym z bloków zastosowano kombinacje nawozowe obornika (0, 20, 40, 60 i 80 t ha–1) oraz nawożenia azotowego (0, 40, 80 i 120 kg ha–1). Aktywność β-glukozydazy oznaczono wg metody EIVAZI, TABATABAI (1988). Kształtowała się ona w zakresie 3,604-7,041 mM pNP g–1 h–1 w próbkach gleby z doświadczenia A oraz 4,931-7,445 mM pNP g–1 h–1 w próbkach z doświadczenia B i pozostawała w ścisłej zależności od zastosowanych dawek obornika i azotu mineralnego. Aktywność β-glukozydazy zależała także istotnie od terminu pobrania próbek glebowych. Ponadto aktywność oznaczanego enzymu była ściśle powiązana z zawartością Corg i Nog, o czym świadczą uzyskane wysokie współczynniki korelacji (r = 0,611-0,770 – dla Corg, oraz r = 0,844-0,912 – dla Nog; p<0,01 i p<0,001).

Introduction: Ischaemia-reperfusion injury (IRI) is a factor leading to the damages of the transplanted kidney, what affects mainly the proximal tubules. Early monitoring of tubule damage can be an efficient tool to predict the allograft dysfunction. Present in proximal tubules, N-acetyl-beta-glucosaminidase (NAG) is a lysosomal enzyme whose excretion rises as a result of IRI or acute rejection. The aim of this study was to monitor the NAG urine activity to evaluate the early proximal tubule damage, and to try to predict the long-term function of the transplanted kidney. Material and methods: The study enrolled 87 Caucasian renal transplant recipients (61.7% males, 38.3% females, mean age 45.56±14.34 years). Urine samples were collected for NAG and creatinine analysis on the 1st day after transplantation, and then in the 3rd and 12th month. Protocol biopsies were performed in the 3rd and 12th month. Results: A significant positive correlation between NAG urine activity in the 3rd month after transplantation and creatinine concentration on the 14th (p=0.004) and 30th day (p=0.05), in the 3rd month (p=0.009) and after the 1st (p=0.005) and 2nd year (p=0.003) was observed. A statistically significantly higher urinary NAG activity in samples collected in the first 3 days and in the 3rd month after transplantation among patients with DGF (p=0.006 and p=0.03 respectively) was found. There was a significant positive correlation between NAG urine activity in the 3rd month and the grade of tubular atrophy in specimens collected in the 3rd (p=0.03) and 12th (p=0.04) month. Conclusions: Monitoring of NAG urine activity is useful in the evaluation of early proximal tubule damage and predicting the long-term function of the transplanted kidneys.

The objective of this study was to determine the effect of ß-l,3/l,6-D-glucan (Biolex-Beta HP) on the phagocytic activity and oxidative metabolism of peripheral blood granulocytes and monocytes in rats intoxicated by cyclophosphamide. The experimental material comprised 40 adult Wistar rats aged 14 weeks, divided into two equal groups, a control group and an experimental group, each of 20 adult rats, including 10 males and 10 females. In the course of 3 successive days, 20 rats from the experimental group were administered cyclophosphamide intramuscularly at a dose of 50 mg/kg body weight/day. On the 8th day of the experiment, 10 control group (K) rats and 10 experimental group (C) rats were sacrificed. Arterial blood samples were collected and diluted with heparin to determine and compare the phagocytic activity (Phatogest) and oxidative metabolism (Bursttest) of peripheral blood granulocytes and monocytes by flow cytometry. Starting on the 8lh day of the experiment, the feed of the remaining 10 rats from the experimental group (C+G) and 10 rats from the control group (G) was supplemented with Biolex-Beta HP at a dose of 50 mg/kg body weight/day for 14 consecutive days. On day 22, arterial blood samples were collected from all (C+G) and (C) group rats, diluted with heparin to determine and compare the phagocytic activity and oxidative metabolism of peripheral blood granulocytes and monocytes by flow cytometry. The results showed that Biolex-Beta HP modulated the phagocytic activity and oxidative metabolism of blood neutrophils and monocytes suppressed by cyclophosphamide in rats. The immunorestoring activity of Biolex-Beta HP was observed in this study.

It was determined that activity of β-fructofuranozydase in baker's yeast put through a uniform, constant, constant but pulsating, and alternating magnetic field with induction levels between 0 and 200 Gs at a linear speed of 2.5X 10⁻³ to 2.5 m/sec. was inhibited by 7.4% to 23% regardless of type of the field. Growth rate of aerobic cultivated baker's yeast was not effected.
Na podstawie statystycznego modelu losowej, dwuczynnikowej analizy wariancji badano wpływ liniowej szybkości przepuszczania komórek drożdży przez pole magnetyczne o różnej wartości indukcji magnetycznej na aktywność β-fruktofuranozydazy drożdży piekarskich. W tym celu wodne zawiesiny jednodobowych drożdży Saccharomyces cerevisiae Mautner przepuszczano z prędkością liniową 0,0025 do 2,5 m/s przez jednorodne stałe, pulsujące z częstotliwością 4 Hz i zmienne pole magnetyczne o indukcji 0 do 200 Gs. Stwierdzono, że niezależnie od charakteru pola magnetycznego aktywność β-fruktofuranozydazy była hamowana odpowiednio o 18,9% w stałym, 7,4% w stałym pulsującym i 23,0% w zmiennym polu magnetycznym. Za pomocą analizy wariancji wykazano ponadto, że w takich samych warunkach liniowa szybkość przepuszczania komórek drożdży przez pole magnetyczne nie wpływała na aktywność β-fruktofuranozydazy. Ponadto stwierdzono, że oddziaływanie pola magnetycznego o indukcji 100 Gs na komórki drożdży w czasie ich tlenowego wzrostu nie wpłynęło na szybkość przyswajania przez nie sacharozy z pożywki melasowej oraz właściwą szybkość wzrostu.

In recent years, many herbal plants and their active components have been tested in different models of neurodegenerative diseases. Some studies are focused rather on studies of chemical compounds of plant origin than on plant extracts. Several natural polyphenols (i.e. flawonoids) are known to exhibit wide spectrum of beneficial effects on brain functioning and to protect against neurodegenerative processes [1, 2]. It seems that influence on β-amyloid is a promising point of pharmacological action of these plant components, because this protein is a major biological risk factor contributing to Alzheimer’s disease (AD)-associated cascade including severe neuronal loss in the brain regions key for memory. In this review the attention is paid to studies on interesting natural chemical compounds of flavonoids (i.g. luteolin, myricetin, icariin) which are a promising study material for research of the potential neuroprotective effects by decreasing the activity of β-secretase (BACE -1) leading to diminish the generation and deposition of β-amyloid (Aβ) in the central nervous system. This range is even more interesting because plant polyphenols can be included in healthy diet and in multi-target drug therapy of neurodegererative diseases.

Influence of beta2- and beta3-adrenoceptor agonists on contractile activity of the porcine myometrium in the luteal phase and the first days of pregnancy

This study analysed the relaxant properties of salbutamol (β₂₋adrenoceptors agonist) and BRL 37344 (β₃₋adrenoceptors agonist) regarding the contractility of porcine myometrium on days 10-14 of the oestrous cycle (cyclic group; n = 10) and on days 3-5 of pregnancy (early pregnant group; n = 6). The activity of myometrial strips (tension, frequency and amplitude) was recorded under isometric conditions using force transducers. The contractility was assessed further following the administration of increasing concentrations of the agonists (10⁻⁹-10⁻⁴ M), both with and without β-adrenoceptor antagonists (butaxamine – a selective β₂₋ adrenoceptor antagonist, propranolol- a non-selective β₁₋and β₂₋adrenoceptor antagonist and bupranolol – a non-selective β₁₋, β₂₋ and β₃₋adrenoceptor antagonist) at a concentration of 10⁻⁴ M. Although neither salbutamol nor BRL 37344 caused changes in the tension, at the highest concentrations they decreased the frequency and amplitude of contractions. These changes were more evident after salbutamol treatment and in the early pregnant group. Antagonists given alone did not cause changes in the parameters examined but changed some activity of the agonists. Butoxamine reduced the decrease in frequency and amplitude induced by salbutamol and produced a decrease in the tension after BRL 37344 treatment in the early pregnant group. Propranolol reduced the decrease in frequency and amplitude induced by salbutamol in both examined groups and did not cause significant changes in BRL 37344 activity. The administration of bupranolol before salbutamol treatment caused an increase in the tension and reduced the decrease in the frequency in the cyclic group. Moreover, bupranolol eliminated a decrease in frequency and induced an increase in amplitude caused by BRL 37344 in both groups and these changes were more evident in the early pregnant group. The data indicates that both β₂₋ and β₃₋adenoreceptors are involved in the regulation of the contractility in both groups, but the changes after agonists and antagonists treatment are more evident in the early pregnant myometrium.
