Flavonoids, polyphenolic compounds present in many food products, affect growth of different bacterial species when tested as purified or synthetic substances. They can also influence gene expression in human cells, like fibroblasts. Here, we asked if soy isoflavone extracts, commonly used in many products sold as anti-menopausal dietary supplements, influence bacterial growth similarly to a synthetic isoflavone, genistein. Four commercially available products were tested in amounts corresponding to genistein concentrations causing inhibition of growth of Vibrio harveyi (a model bacterium sensitive to this isoflavone) and Escherichia coli (a model bacterium resistant to genistein). Differential effects of various extracts on V. harveyi and E. coli growth, from stimulation, to no changes, to inhibition, were observed. Moreover, contrary to genistein, the tested extracts caused a decrease (to different extent) in viability of human dermal fibroblasts. These results indicate that effects of various soy isoflavone extracts on bacterial growth and viability of human cells are different, despite similar declared composition of the commercially available products.

Opis:
Transposition of insertion sequences (IS) is an enzyme-mediated process that only occurs in a minority of cells within a bacterial culture. Transposition is thus a rare event, but transposition frequency may vary depending on experimental conditions. For instance in a rich broth, IS elements are known to transpose during stationary phase but not during exponential growth. Using a reporter system which involves the activation of the cryptic bgl operon in Escherichia coli, we show that the frequency of IS1 transposition is a function of glucose concentration in the growth medium, it is increased by streptomycin amounts that are below minimum inhibitory concentration (sub-MIC) and is inhibited in an rpsL150 strain with high translation accuracy. Since starved cells are known to enhance ribosome frameshifting, our data suggests that growth conditions applied in this study could affect IS1 transposition by increasing translation infidelity.

Heterocyclic aromatic amines (HCAs) ingested with heat-processed meat can contribute to colon cancer development. Those compounds interact with human intestinal microbiota. Under the research study, the effect was analyzed of IQ, MeIQx, or PhIP amines (each of them at 5 and 25 μg/ml concentration rates) on the growth (24 h cultivation in MRS broth) and survival (incubation for max. 120 h in a phosphate buffer) of four probiotic strains of Lactobacillus. It was found that no concentration level of the HCAs analyzed impacted the growth of bacteria. A higher concentration level of IQ (25 μg/ml) decreased the survival of Lb. casei 0900 after a 24 h period of incubation. MeIQx and PhIP also decreased the survival of Lb. paracasei 0919 after a 24 h period of incubation. Totally, the count of living cells decreased from 109 cfu/ml to 108 cfu/ml. Three strains were absolutely resistant to PhIP during a 120 h period of incubation. The results obtained prove that the probiotic bacteria studied maintain their high viability in the presence of the amines tested and, probably, they can bind together with them in human colon and, then, the aggregated particles are removed from the human body.
Heterocykliczne aminy aromatyczne (HCA) spożywane wraz z termicznie przetworzonym mięsem mogą przyczyniać się do rozwoju raka jelita grubego. W pracy badano wpływ amin IQ, MeIQx oraz PhIP (każda w stężeniu 5 i 25 μg/ml) na wzrost (24-godzinna hodowla w pożywce MRS) i przeżywalność (do 120 h inkubacji w buforze fosforanowym) czterech szczepów probiotycznych z rodzaju Lactobacillus. Zaobserwowano, że żadne z badanych stężeń HCA nie wpływało na wzrost bakterii. Wyższe stężenie IQ (25 μg/ml) obniżało żywotność szczepu Lb. casei 0900 od 24 h inkubacji. Aminy MeIQx i PhIP obniżały żywotność Lb. paracasei 0919 również od 24 h inkubacji. Generalnie liczba żywych komórek obniżyła się z 109 jtk/ml do 108 jtk/ml. Trzy szczepy były całkowicie oporne na obecność PhIP podczas 120 h inkubacji. Uzyskane rezultaty wskazują, że badane szczepy probiotyczne zachowują wysoką żywotność w obecności testowanych amin i prawdopodobnie mogą wiązać się z nimi w jelicie grubym, po czym te zagregowane cząstki usuwane są z organizmu.

The objective of the present study was to evaluate heterotrophic bacteria capable of growth in the presence of different antibiotics and their mixture in such dynamic ecosystem as marine beach. Mixture antibiotics had the strongest inhibitory effect on the growth of bacteria inhabiting sand of studied beach. Culturable bacteria were more resistant to ampicillin than to novobiocin and tetracycline. Inhibitory influence antibiotics on growth bacteria inhabiting studied beach were in the following order: mixture antibiotics > novobiocin > tetracycline > ampicillin.
W pracy przedstawiono wyniki badań dotyczących określenia hamującego wpływu rożnych antybiotyków i ich mieszaniny na wzrost heterotroficznych bakterii zasiedlających piasek plaży morskiej zlokalizowanej na terenie Słowińskiego Parku Narodowego w rejonie Czołpina. Proby piasku na tej plaży pobierano w profilu horyzontalnym z czterech stanowisk (morze, strefa brzegowa, środkowa część plaży, wydma), a w profilu wertykalnym na każdym stanowisku z dwóch (0-5 cm, 10-15 cm) głębokości. Badania te wykazały, że mieszanina antybiotyków w podłożu hodowlanym wywierała bardziej hamujący wpływ na wzrost bakterii zasiedlających badaną plażę niż pojedyncze antybiotyki. Wśród testowanych antybiotyków neomycyna i tetracyklina wykazywały znacznie większy niż ampicylina hamujący wpływ na wzrost bakterii. Wykazano, że hamujący wpływ antybiotyków i ich mieszaniny na wzrost bakterii zasiedlających powierzchniowe i podpowierzchniowe warstwy piasku był podobny. Stwierdzono, że testowane antybiotyki i ich mieszanina miały wpływ na wzrost chromogennych i achromogennych bakterii zasiedlających piasek badanej plaży morskiej.

Chlorpyrifos-methyl (CM) is a broad-spectrum organophosphate insecticide, which is widely used in pest control. In this research, the isolation, and biochemical and molecular identification of bacterial strains obtained from three soils located in northeastern Algeria were carried out, as well as the evaluation of their ability to grow in the presence of CM. Out of 48 bacterial isolates between Gram-negative and Gram-positive identified, several were able to grow on mineral agar with at least 25 mg·l–1 of CM. Four bacteria showed the best growth capacity, were identified as Bacillus sp. H1-80, Brevibacterium frigoritolerans strain WJB99 and two Bacillus sp. strains GL5. The strains were tested for their ability to grow on liquid media with CM as the sole energy and carbon source. In general, these strains showed slow but significant growth visualized by the 600 nm turbidity control, suggesting that they could be used for bioremediation applications of CM polluted soils.

The paper describes application of the fuzzy c-regression models algorithm in bacterial population dynamics modelling. Such a well-known algorithm, which provides simultaneous estimates of the parameters of c-regression models, together with a fuzzy partitioning of data, was used to determine bacterial growth curve given by the piecewise continuous function. The real as well as artificial data were used and promising results were obtained even for data with significant errors resulting from the measurement inaccuracy.

Animal manures are routinely applied to agricultural lands to improve crop yield, but the possibility to spread bacterial phytopathogens through field fertilization has not been considered yet. We monitored 49 cattle, horse, swine, sheep or chicken manure samples collected in 14 Polish voivodeships for the most important plant pathogenic bacteria - Ralstonia solanacearum (Rsol), Xanthomonas campestris pv. campestris (Xcc), Pectobacterium carotovorum subsp. carotovorum (Pcc), Pectobacterium atrosepticum (Pba), Erwinia amylovora (Eam), Clavibacter michiganensis subsp. sepedonicus (Cms) and Dickeya sp. (Dsp). All of the tested animal fertilizers were free of these pathogens. Subsequently, the growth dynamics of Pba, Pcc, Rsol, and Xcc in cattle, horse, swine, sheep and chicken manures sterilized either by autoclaving or filtration was evaluated. The investigated phytopathogens did not exhibit any growth in the poultry manure. However, the manure filtrates originating from other animals were suitable for microbial growth, which resulted in the optical density change of 0.03-0.22 reached within 26 h (48 h Rsol, 120 h Xcc), depending on bacterial species and the manure source. Pcc and Pba multiplied most efficiently in the cattle manure filtrate. These bacteria grew faster than Rsol and Xcc in all the tested manure samples, both the filtrates and the autoclaved semi-solid ones. Though the growth dynamics of investigated strains in different animal fertilizers was unequal, all of the tested bacterial plant pathogens were proven to use cattle, horse, swine and sheep manures as the sources of nutrients. These findings may contribute to further research on the alternative routes of spread of bacterial phytopathogens, especially because of the fact that the control of pectionolytic bacteria is only based on preventive methods.

Iron-regulated outer membrane proteins (IROMPs) of P. multocida serotype A3, which function as receptors for complexes containing iron ions, are induced by iron deficiency in the bacterial growth environment. Analysis of an electrophoresis image of proteins isolated from bacteria grown on medium supplemented with 2,2'-dipyridyl revealed expression of 16 new proteins that were not noted in the case of the bacteria grown in standard conditions, with molecular weights from 30 to 160 kDa. Induction of IROMP expression occurred within 30 minutes after restricted iron conditions were established. In immunoblotting, distinct reactions were noted for proteins of molecular weight ranges of 25-49 kDa, 61-95 kDa, and 108-214 kDa. Proteins of the molecular weight of 68, 75 and 86 kDa were analysed using mass spectrometry and matched with the highest probability to proteins in the NCBI data base. Several dozen different proteins with similar amino acid sequences were matched to each sample.

Endophytes produce a wide range of compounds with high application potential, mainly in medicine and agriculture. In this study, we test the hypothesis that endophytic bacteria produce indole-3-acetic acid (IAA), have positive influence on plant root development and are possible to application as plant-growth promoters. Endophytic bacteria were isolated from 3 native growing plant species: Chelidonium majus L., Elymus repens L., Solidago gigantea L. All endophytic strains produced IAA and the highest levels of IAA were observed for Pseudomonas azotoformans P3 strain. Triticale seed bacterization did not affect the seed germination, but had significant influence on root length and the longest roots were obtained after seed treatment with Pseudomonas sp. strains. Triticale roots were longer only in seedlings grown from seeds treated with endophytic strains producing high IAA levels (more than 22 µg ml–1). Our results suggest that endophytic Pseudomonas sp. strains isolated from Elymus repens L. can be used as plant-growth promoter.

The genus Proteus belongs to the Enterobacteriaceae family, where it is placed in the tribe Proteeae, together with the genera Morganella and Providencia. Currently, the genus Proteus consists of five species: P. mirabilis, P. vulgaris, P. penneri, P. hauseri and P. myxofaciens, as well as three unnamed Proteus genomospecies. The most defining characteristic of Proteus bacteria is a swarming phenomenon, a multicellular differentiation process of short rods to elongated swarmer cells. It allows population of bacteria to migrate on solid surface. Proteus bacteria inhabit the environment and are also present in the intestines of humans and animals. These microorganisms under favorable conditions cause a number of infections including urinary tract infections (UTIs), wound infections, meningitis in neonates or infants and rheumatoid arthritis. Therefore, Proteus is known as a bacterial opportunistic pathogen. It causes complicated UTIs with a higher frequency, compared to other uropathogens. Proteus infections are accompanied by a formation of urinary stones, containing struvite and carbonate apatite. The virulence of Proteus rods has been related to several factors including fimbriae, flagella, enzymes (urease - hydrolyzing urea to CO2 and NH3, proteases degrading antibodies, tissue matrix proteins and proteins of the complement system), iron acqusition systems and toxins: hemolysins, Proteus toxin agglutinin (Pta), as well as an endotoxin - lipopolysaccharide (LPS). Proteus rods form biofilm, particularly on the surface of urinary catheters, which can lead to serious consequences for patients. In this review we present factors involved in the regulation of swarming phenomenon, discuss the role of particular pathogenic features of Proteus spp., and characterize biofilm formation by these bacteria.

The article presents results of research into the influence of bacterial preparations (rizobacterin and sapronit) and plant regulators of growth (epin and homobrassinolid) on the productivity and quality of grain of oat, spring wheat and lupine in the separate and mixed crops depending on level of a nitrogen nutrition. We have established that in the conditions of sod-podzolic soil of average degree cultivation the optimal dose of nitrogen fertilizer for oat, wheat and mixed crops is 40 kg of acting substance per hectare, for lupine – 10 kg of acting substance per hectare (at level P60K90). Inoculation of seeds bacterial preparations (rizobacterin and sapronit) and application of growth regulators (epin and homobrassinolid) in technology of cultivation of oats, spring wheat, lupine and their mixed crops allows to reduce doses of mineral nitrogen to 30 kg of acting substance per hectare and to improve quality indicators of grain of studied cultures.
W publikacji zostały przedstawione wyniki badań wpływu bakteryjnych preparatów (rizobakterin i sapronit) i regulatorów wzrostu roślin (epin i homobrassinolid) na plon i jakość ziarna owsa, pszenicy jarej i łubinu w jednogatunkowych i mieszanych siewach w zależności od poziomu nawożenia azotowego. Ustalono, że w warunkach gleb darniowo-bielicowych o oerednim stopniu wzrostu bakterii, optymalna dawką azotu dla owsa, pszenicy jarej i mieszanek tych kultur na poziomie P60K90 była 40 kg ha–1, dla łubinu – 10 kg ha–1. Inokulacja nasion bakteryjnymi preparatami (rizobakterin i sapronit) i zastosowanie regulatorów wzrostu (epin i homobrassinolid) w technologii uprawy owsa, pszenicy jarej, łubinu i mieszanek tych kultur pozwala zmniejszyć dawki azotu mineralnego do 30 kg ha–1 i poprawić jakościowe charakterystyki ziarna badanych kultur.

Globally, more than 5.2 billion hectares of farming fields are damaged through erosion, salinity, and soil deterioration. Salt stress-tolerant bacteria have plant growth-promoting (PGP) characteristics that can be used to overcome environmental stresses. Isolation and screening of salt-tolerant endophytes from Bougainvillea glabra were achieved through surface sterilization of leaves, followed by their cultivation on 0.5% NaCl-supplemented media. The performance of isolates for indole acetic acid (IAA) production, phosphate solubilization, ACC deaminase activity, ammonia production, siderophore production, and stress tolerance was determined. In the present study, 5 morphologically distinct salt-tolerant endophytic bacteria were cultured. Out of 5 isolates, 60% salt-tolerant endophytes showed phosphate solubilization, 100% IAA production, 40% ACC-deaminase activity, 100% siderophore production, 40% ammonia production and 60% HCN production. Dendrogram generated based on stress tolerance showed three clusters: clusters 1 and 2 containing two isolates and cluster 3 containing 1 isolate. Based on the highest PGP activities, BoGl17 and BoGl21 isolates were chosen and further tested for wheat and corn growth promotion. The isolates increased wheat shoot and root dry weight by 40% and 80%, and by 69% and 126% for BoGl17 and BoGl21, respectively. The bacterial isolates identified as Pseudomonas reidholzensis BoGl17 had 95.59% identity with Pseudomonas reidholzensis strain ID3, while the isolate Bacillus aerius BoGl21 had 90.96% identity with Bacillus aerius strain 24K based on phenotypic features and 16S rRNA gene sequencing results. Both isolates significantly improved plant growth as compared to uninoculated plants.
