Butyrylcholinesterase variants
Opis:
Butyrylcholinesterase (BChE) is synthesized in the liver and found in high concentrations in blood plasma, liver, heart, pancreas, vascular endothelium, skin, brain white matter, smooth muscle cells and adipocytes. BChE is a non specific enzyme that hydrolyzes different choline esters (succinylcholine, mivacurium) and many other drugs such as aspirin, cocaine and procaine. The enzyme is also considered as a bioscavenger due to its ability to neutralize the toxic effects of organophosphorus compounds (nervous system fs agents) such as soman. BChE displays several polymorphisms that influence its serum activity; therefore they could determine the individual sensitivity to chemical nerve agents. In this study, we investigated the correlation between BChE variants and the degree of enzyme inhibition and reactivation after soman application on blood samples of 726 individuals. The blood samples of individuals expressing abnormal variants, were more sensitive to soman compared to variants of homozygotes and heterozygotes for U-allele. We found significant differences in the degree of enzyme reactivation between different variants (with and without U-presence).

Opis:
This study aimed to evaluate the antidotal potency of tenocyclidine (TCP) that probably might protect acetylcholinesterase (AChE) in the case of organophosphate poisoning. TCP was tested alone as a pretreatment or in combination with atropine as a therapy in rats poisoned with ¼ and ½ of LD50 of soman. Possible genotoxic effects of TCP in white blood cells and brain tissue were also studied. Results were compared with previous findings on the adamantyl tenocyclidine derivative TAMORF. TCP given alone as pretreatment, 5 min before soman, seems to be superior in the protection of cholinesterase (ChE) catalytic activity in the plasma than in brain, especially after administration of the lower dose of soman. Plasma activities of the enzyme after a joint treatment with TCP and soman were significantly increased at 30 min (P < 0.001) and 24 h (P = 0.0043), as compared to soman alone. TCP and atropine, given as therapy, were more effective than TCP administered alone as a pretreatment. The above therapy significantly increased activities of the enzyme at 30 min (P = 0.046) and 24 h (P < 0.001), as compared to controls treated with ¼ LD50 of soman alone. Using the alkaline comet assay, acceptable genotoxicity of TCP was observed. However, the controversial role of TCP in brain protection of soman-poisoned rats should be studied further.

Opis:
Due to the multifold growth in demands of multimedia services and mobile data, the request for increased channel capacity in mobile and wireless communication has been quickly increasing. Developing a wireless system with more spectral efficiency under varying channel condition is a key challenge to provide more bit rates with limited spectrum. Multiple Input Multiple Output(MIMO) system with Orthogonal Frequency Division Multiplexing (OFDM) gives higher gain by using the direct and the reflected signals, thus facilitating the transmission at high data rate. An integration of Spatial Modulation (SM) with OFDM (SM OFDM) is a newly evolved transmission technique and has been suggested as a replacement for MIMO -OFDM transmission. In practical scenarios, channel estimation is significant for detecting transmitted data coherently. This paper proposes pilot based, Minimum Mean Square Error (MMSE) channel estimation for the SM OFDM communication system. We have focused on analyzing Symbol Error Rate (SER) and Mean Square error (MSE) under Rayleigh channel employing International Telecommunication Union (ITU) specified Vehicular model of Pilot based MMSE channel estimator using windowed Discrete Fourier Transform (DFT) and MMSE weighting function. Simulation output shows that proposed estimator’s SER performance lies close to that of the MMSE optimal estimator in minimizing aliasing error and suppressing channel noise by using frequency domain data windowing and time domain weighting function. Usage of the Hanning window eliminates error floor and has a compact side lobe level compared to Hamming window and Rectangular window. Hanning window has a larger MSE at low Signal to Noise Ratio (SNR) values and decreases with high SNR values. It is concluded that data windowing technique can minimize the side lobe level and accordingly minimize channel estimation error when interpolation is done. MMSE weighting suppresses channel noise and improves estimation performance. Since Inverse Discrete Fourier Transform(IDFT)/DFT transforms can be implemented with fast algorithms Inverse Fast Fourier Transform( IFFT)/Fast Fourier Transform(FFT) computational complexity can be remarkably reduced.

Opis:
This work concentrates on Structural Health Monitoring (SHM) of a wind turbine tower. The paper investigates the use of a decision level data fusion based on bi-axial tracking of change in the neutral axis (NA) position for damage detection in wind turbine towers. A discrete Kalman Filter (KF) is employed for the estimation of the NA in the presence of measurement noise from the strain sensors. The KF allows data fusion from the strain sensors and the yaw mechanism for the accurate estimation of the NA. Any change in the NA position may be used for detecting and locating the damage. The tan inverse of the ratio of the change in the NA along two perpendicular axes is taken and used for the localization. The study was carried out on a simulated finite element (FE) model of a wind turbine tower with a surface crack. The sensitivity studies carried out on the structure in terms of different crack sizes, crack locations and crack orientations indicate that the methodology is robust enough to detect the crack under different operational loading conditions.

Opis:
Glycidyl Azide Polymer (GAP) is one of the most potential energetic binders for rocket propellants and gas generator compositions. In the present paper GAP of molecular weight (Mn) ~2000 was cured with a mixture of di- and tri-isocyanates without a cross linker. The curing profile and time of curing was recorded using a rheometer. The minimum curing time was observed for samples cured with Desmodour N-100 alone, whereas the maximum curing time was observed for samples cured with a mixture of Desmodour N-100 and Isophorone Diisocyanate (IPDI) (1:1 w/w). It was observed that all of the samples cured well and were void or bubble free. The mechanical properties data showed that the tensile strength (TS) of GAP cured with Desmodour N-100 alone was 1.19 kgf/cm2, which is a minimum, while the maximum TS (3.66 kgf/cm2) was achieved with a mixture of N-100 and 4,4’methylenebis(phenylisocynate) (MDI). The percent elongation for a sample cured with Desmodour N-100 was 160, and was reduced to 64.27 when a mixture of MDI and N-100 was used. In order to study the curing of GAP without an isocyanate, GAP diol was cured with hexanediol di-acrylate. GAP was also cured with an alkyne-based curing agent i.e. bis-propargyl succinate (BPS), which showed improved curing. Comparative thermal studies of GAP cured with isocyanate and acrylate was carried out. Differential Scanning Calorimetry (DSC) and Simultaneous Thermal Analysis (STA) curves for all of the cured samples were recorded in order to study and compare the thermal decomposition behaviour of the cured GAP. Isocyanate cured GAP exhibited a single stage decomposition, with larger heat output. Acrylate cured GAP exhibited a two stage decomposition. Finally, a mixture of IPDI and Desmodour N-100 was selected for curing of GAP. Accordingly, curing was carried out and was tested in a small ballistic evaluation motor (BEM) to observe the combustion behaviour and burn rate. From the pressure-time profile it was found that this composition gave smooth burning with a pressure of ~3 kg/sec2 for 7 seconds of burn.

Opis:
Friction stir welding is a solid state innovative joining technique, widely being used for joining aluminium alloys in aerospace, marine automotive and many other applications of commercial importance. The welding parameters and tool pin profile play a major role in deciding the weld quality. In this paper, an attempt has been made to understand the influences of welding speed and pin profile of the tool on friction stir welded joints of AA6082-T6 alloy. Three different tool pin profiles (tapered cylindrical four flutes, triangular and hexagonal) have been used to fabricate the joints at different welding speeds in the range of 30 to 74 mm/min. Microhardness (HV) and tensile tests performed at room temperature were used to evaluate the mechanical properties of the joints. In order to analyse the microstructural evolution of the material, the weld’s cross-sections were observed optically and SEM observations were made of the fracture surfaces. From this investigation it is found that the hexagonal tool pin profile produces mechanically sound and metallurgically defect free welds compared to other tool pin profiles.
Parametry spawania i profil trzpienia narzędzia grają główną rolę gdy chodzi o jakość spawu.
