Opis:
AMP-deaminase from hen stomach smooth muscle was isolated and physico-chemical properties of the purified enzyme were investigated. The enzyme had an activity optimum at pH 6.5, and poorly deaminated the substrate analogues tested. At optimum pH (6.5), in the absence of regulatory ligands (control conditions), the enzyme manifested hyperbolic substrate-saturation kinetics with half-saturation constant (S0.5) of about 4.5 mM. Additions of adenine nucleotide effectors (ATP, ADP) activated the enzyme strongly at all the concentrations tested, diminishing significantly the value of S0.5 constant. In contrast, the regulatory effect of orthophosphate was variable, and depended on the orthophosphate concentration used. The molecular mass of the enzyme subunit determined in SDS/PAG electrophoresis was about of 37 kDa. The obtained results suggest that in different types of hen muscle, similarly as in humans and rats, expression of AMP-deaminase is under the control of independent genes.

Opis:
Measurements of boiling heat transfer coefficients in water, methanol and refrigerant R141b are reported for the bundles of smooth tubes that represent a portion of a flooded-type evaporator. Each bundle contained 19 instrumented, electrically heated tubes in a staggered triangular-pitch layout. The effect of heat flux, tube pitch and operating pressure is studied in the paper. Bundle factor and bundle effect are discussed as well. A correlation for prediction of a bundle average heat transfer coefficient is proposed.

Previously we have shown that aldolase (ALD; EC 4.1.2.13) is present in cardiomyocyte nuclei. Now, we focused our attention on ALD localization in smooth muscle cells. Immunocytochemical methods were used to study the subcellular localization of ALD. Aldolase was localized in the cytoplasm as well as in the nuclei. Within the nuclei ALD was located in the heterochromatin region. Native polyacrylamide gel electrophoresis followed by aldolase activity staining in gel was used to study the ALD isoenzyme pattern in porcine smooth muscle cells. Two ALD isoenzymes, A and C, were found in these cells but in the nuclei only the muscle isoenzyme was detected. To support the nuclear localization of ALD, measurement of aldolase activity in the smooth muscle cell nuclei isolated from porcine stomach was performed. The ALD activity in the isolated nuclei was detectable only after preincubation of the nuclear fraction with Triton X-100 and high concentration of KCl.

Complex microstructure analysis of TiN/Ti/a-C:H multilayer coatings, subjected to mechanical and biological tests, were performed by means of transmission electron microscopy (TEM) and confocal scanning laser microscopy (CSLM), respectively. Influence of interface numbers and phase ratios on coating properties was studied. Thin films were fabricated by a hybrid PLD technique (PLD supported by magnetron sputtering). The a-C:H phase was characterized by an amorphous structure, while TiN was built of columnar crystallites. Multilayer coatings contained sequentially deposited TiN and a-C:H layers with thin metallic Ti inter- layers deposited at each interface. Mechanisms of mechanical wear of analyzed systems were presented focusing on the cracking propagation revealed in the scratch test. Biological tests were done basing on smooth muscle cells adhesion to coating surfaces. An increase of TiN phase in the coating led to improvement of mechanical properties, while the carbon phase improved the biological behavior. Coatings comprising a higher rate of the carbon (a-C:H) revealed brittleness and were prone to delaminating. Optimal properties from the mechanical and biological point of view were stated for 8xTiN/Ti/a-C:H coating with 1:1 phase ratio (TiN to a-C:H).

Opis:
Free damped oscillations of a double tracked road vehicle with a nonline- ar response of the suspension are considered. A 7-DOF nonlinear model is used to analyze dynamics of the suspension with smooth characteristics. Vibration modes of the system and the corresponding skeleton curves are obtained. Nonlinear vibrations of the quarter-car model are considered for the case of non-smooth characteristics of the shock absorber.
W pracy rozważono tłumione i swobodne drgania dwuśladowego pojazdu drogowego o nieliniowej charakterystyce zawieszenia. Pojazd zamodelowano nieliniowym układem o 7 stopniach swobody, a w analizie jego dynamiki przyjęto gładką charak- terystykę zawieszenia. Znaleziono postacie drgań takiego układu i wyznaczono odpowiednie krzywe szkieletowe. Zagadnienie drgań nieliniowych modelu ”ćwiartkowego” (o 2 stopniach swobody) rozważono dla przypadku zawieszenia z niegładką charakterystyką amortyzatora.

The mechanical link coupling cytoskeletal and contractile proteins to the sarcolemma of smooth muscle cells is essential for transmitting tension from the cell's interior to exterior. In addition to the well-characterized actin-integrin associations present in adhaerens junctions, our recent work has postulated the existence of a reversible annexin-dependent membrane-cytoskeleton complex, forged in response to a rise in intracellular Ca2+ concentration following smooth muscle cell stimulation (Babiychuk et al., J. Biol Chem. 1999, 274, 35191-35195). Detailed biochemical characterization of the interactions responsible for the formation of this complex revealed that annexins II and VI interact with actomyosin, or detergent-insoluble glycosphingolipid-enriched membrane domains (rafts) purified from smooth muscle, in a concentration- and Ca2+-dependent manner. Annexin II interacted with lipid rafts with high Ca2+-sensitivity, while for annexin VI this interaction required non-physiologically high concentrations of free Ca2+. However, the Ca2+-sensitivity of the latter interaction strongly increased in the presence of purified smooth muscle actomyosin. The detailed biochemical analysis of the interactions occurring between annexin II, annexin VI, actomyosin and rafts suggests that annexins regulate sarcolemmal organization during smooth muscle cell contraction.

Twenty seven entries (10 cultivars and 17 breeding strains) of smooth-stalked meadowgrass (Poa pratensis L.) were tested during 2002 – 2004 for seed yield and turf quality. Seed yield potential was estimated on the basis of: seed yield per plot, seed yield per panicle, seed heads per 1 m2, panicle length, 1000 seed weight, plant height, leaf width and heading time. Turf quality was estimated on the basis of: visual merit, shoot density, leaf fineness and colour. Tested entries were significantly different for all traits measured. No significant correlations were calculated for seed yield and any from turf quality traits. None of top quality turf varieties (BARCELONA, LIMUSINE and CONNI) yielded as high as the highest yielding entries (BALIN, BARON etc.)...

Following the study of the arc structure of singularities, initiated by J. Nash, we give criteria for the existence of smooth curves on a surface singularity (S,O) and of smooth branches of its generic hypersurface section. The main applications are the following: the existence of a natural partition of the set of smooth curves on (S,O) into families, a description of each of them by means of chains of infinitely near points and their associated maximal cycle and the existence of smooth curves on any sandwiched surface singularity. A wedge centered at a smooth curve on (S,O) is essentially a one-parameter deformation of the parametrization of the curve. We show that there is no wedge centered at smooth curves of two different families.

Advancements in computer technology that have occurred in recent decades have enabled an intensive development in cartographic methods for direct representation of phenomena dynamics. Even with the appearance of ever more advanced technical solutions, the theoretical basis still needs supplementing. The previous cartographic literature emphasises the importance of congruence and isomorphism principles preservation that aims at increasing the effectiveness of dynamic displays. Nevertheless, it is frequently the case that discontinuous phenomena are depicted with the use of smooth transitions. For this reason, it is vital that experimental research should lead to defining which representation methods are appropriate for a given type of content. Our study was focused on the cartographic design of scene transitions in animated maps. Two main conclusions of the research indicate that 1) mode of transition influences the interpretation of the content of cartographic animation depicting discrete changes, 2) maps executed in a smooth mode demonstrate lower effectiveness when compared with animations using an abrupt and abrupt with decay effect transitions.

Analysis of the geometric features and determination of the condition of the surfaces machined during an abrasive process is of crucial importance for evaluation of their wear properties. Numerous parameters can be used for assessing topography of technical surfaces. Many of them contain information that is general or poorly correlated with the wear features. The group of parameters used for such purpose should include those that do not cause information redundancy and provide distinct information. Evaluation of the surface geometric structure after precision machining is a difficult process due to the limited scope of unevenness heights that fit within the range of a few micrometers to a few nanometers. In this paper the synthetic indexes of the evaluation of extra smooth surfaces were created, taking into consideration numerous elementary parameters. The value of the synthetic index was determined as the geometric average of the partial indexes which, when compared to the arithmetic average, is more dependent on the data believed to be disadvantageous.

This paper examines the nonlinear relationship between inflation and government spending using quarterly data over the period of 1990-2013, by using Smooth Transition Regression Model. Results suggested a two regime model by using inflation, government expenditure growth, GDP growth and liquidity growth. Lag of liquidity was recognized as transition variable. This study showed that in regime of tight money or low growth of liquidity, government expenditure is not inflationary. In regime of low growth of liquidity, this variable has low inflationary impact and probably stimulates economic growth. Inflationary expectations in first regime are more effective in causing short run inflation. In expansionary regime, increase of money supply has more effects on inflation rather than production. So monetary and fiscal policies could be used to control inflation and stimulate aggregate demand in low regime. Also in easy money regime, monetary and fiscal discipline can be useful for inflation decrease.

So-called Engineered Grinding Tools (EGT) have been developed in order to satisfy the industry's demand for more efficient and cost-effective grinding tools, which differ from classic grinding tools by placing and brazing single abrasive grains in a predefined pattern onto the grinding tool body. Single grain cutting simulations form an important part towards an advanced understanding of the engineered grinding process. Cutting simulations with FE Methods encounter problems arising from large deformation and material separation. These can be overcome using meshless methods. In this work, a Smooth Particle Hydrodynamics (SPH) Method is used to model the single diamond grain cutting, which can be used as the basis process for physical simulation of the grinding process. As results the influence of the grain geometry, grain orientation and grain placement on the cutting forces, the burr generation and the chip removal rate are presented. It is shown that the cutting forces for a given grain geometry as well as the burr generation are heavily influenced by the orientation of the minor and the main cutting faces. Also cutting in material being work hardened by preceding grains, is simulated in order to be able to synthesize the grinding process from single grain cutting.
