In vivo and in vitro self-pollination of whole pistils of some clones of Salix viminalis enabled to obtain mature seeds containing cotyledonary embryos which after the transfer to MS medium developed into wholly formed seedlings. Pollination in vitro of placentae led to abundant pollen germination and formation of tubes which occasionally they were entering the ovules through micropyle. Fertilized ovules normally developed into germinable seeds. Distant pollination of stigmas in vivo and in vitro with pollen grains of Populus tremula, P.tomentosa, P. lasiocarpa showed the ability of pollen to germinate and to form tubes several hours after pollination. Some tubes penetrated the styles but did not enter into the placenta. When placentae were directly pollinated than pollen germinated abundantly and occasionally pollen tubes were found entering the micropyle. Embryological analysis of those ovules performed 3-5 days after pollination demonstrated the presence of globular embryos with several endosperm nuclei. The technique of in vitro placental pollination works well for Salix viminalis and it could probably be applied to other Salix species.

Experiments on somatic embryogenesis in selected spruce, fir and larch species were performed to determine if this method of micropropagation enables production of quality seedlings for forest nurseries. High frequencies of embryogenic callus in spruce (23-31%) and fir (29%) were achieved when mature zygotic embryos were used as explants, while in larches (36%) only when megagametophytes (endosperms with immature embryos) were used. The possibility of somatic embryogenesis initiation also from somatic embryos (resulting in secondary and third generation of embryogenic callus) indicate high efficiency of this method of micropropagation. The best results at all stages of somatic embryogenesis (good proliferation of embryogenic callus, high rate of embryo regeneration and their survival) were obtained for Norway spruce, European larch and some hybrid larches.

Embryo development of Lupinus albus L. and L. mutabilis Sweet. after pollination with L. angustifolius L. was studied. Observations of ovules revealed non-typical embryo development. From the 5th day after pollination, high number of degenerated ovules was found; the highest number was observed on 15 and 20 days after pollination. The period between the 10th and 15th day after pollination was the most suitable for isolation of embryos in all the examined combinations, with exception L. albus x L. angustifolius where optimal isolation time was between 15th and 20 th day after pollination. Embryos were obtained in each cross-pollination. Some of them developed in vitro into plantlets and grew in an artificial medium for some time, but most of them died due to non-typical development of the root system, stem, or both. The hybrid character of the survived plants requires confirmation.

In order to estimate the best germination conditions of Encyclia aff. oncidioides seeds, five different media (Fast, Knudson C modified by Vajrabhaya, Murashige and Skoog, PB2 and modified Vacin and Went) with different concentrations of plant growth regulators such as benzyladenine (BA), naphthaleneacetic acid (NAA) and gibberellic acid (GA3) were tested. No beneficial effect was observed when BA and NAA were applied to the germination medium and GA3 inhibited germination. The effect of light, activated charcoal, coconut water and casein hydrolysate on seed germination was also studied. The growth rate of seedlings on three different media supplemented with activated charcoal and plant growth regulators was checked. The applied plant growth regulators had no beneficial effect on the further growth of seedlings. Fast and PB2 media with 0.2% activated charcoal proved to be the best for E. aff. oncidioides seed germination, seedling development and plantlet propagation.

The influence of melatonin (MEL) in comparison with IAA, IBA (all at 5.71 µ M), and auxin-free (0) medium on development of highbush blueberry (Vaccinium corymbosum hort. non L.) ‘Herbert’ in vitro shoot cultures was examined. Depending on the kind of hormone in vitro cultures consisted of various number of axillary (AX) and adventitious (AD) shoots. The influence of melatonin on in vitro cultures was intermediate but more similar to IAA than IBA. Production of axillary shoots on media supplemented with MEL and IAA was comparable and higher than on medium with IBA. In contrast to IAA melatonin reduced development of adventitious shoots. Contrary to IBA-obtained cultures AX shoots grown on ‘IAA’, ‘0’, and ‘MEL’ media resembled AD shoots.
W pracy badano wpływ melatoniny (MEL 5.71 µ M) na rozwój pędowych kultur in vitro borówki wysokiej (Vaccinium corymbosum hort.  non L. ‘Herbert’). Odniesienie stanowiły pożywki zawierające IAA, IBA (5.71 µ M) lub wolne (0) od auksyn. W zależności od rodzaju hormonu kultury in vitro składały się z różnej liczby pędów kątowych (AX) i przybyszowych (AD). Wpływ melatoniny na kultury in vitro był pośredni, choć bardziej podobny do wpływu IAA niż IBA. Wytwarzanie pędów kątowych na pożywkach z dodatkiem melatoniny i IAA było zbliżone i większe niż na pożywce z IBA. W przeciwieństwie do IAA melatonina osłabiała rozwój pędów przybyszowych. Pędy kątowe otrzymane przy użyciu IBA wyraźnie różniły się od pędów przybyszowych. Na pozostałych pożywkach (‘IAA’, ‘0’ i ‘MEL’) wygląd pędów kątowych i przybyszowych był podobny.

Opis:
Adventitious bud cultures were established by using buds of a selected birch clone (Betula pendula Roth.) resistant to industrial pollution. The Murashige and Skoog medium (1/2 and 1/4 MS) was used for multiplication and rooting of shoots. Aluminium was added to the medium, in the form of aluminium sulphate (50–100 mg Al dm–3), and birch culture was continued in vitro for over 12 months. The shoots developed on media with aluminium (Al+) proved to be more tolerant to aluminium and copper (added to the medium as nitrates or sulphates, at a concentration of 0.05–2.0 mM) during multiplication and rooting than those developed on media without aluminium (Al–). Rooted birch microcuttings obtained from cultures on media with aluminium (Al+) grew better in the soil from an unpolluted area (Zwierzyniec, Z) and from an area polluted by a phosphate fertilise factory (Luboń, L) than those from media without aluminium (Al–).

Different nutrient media (MS [Murashige and Skoog 1962]; QL [Quoirin and Lepoivre 1977] and WPM [Lloyd and McCown 1980]); plant growth regulators BA (benzil adenin), GA3 (gibberellic acid), IBA (indole-3-butyric-acid), NAA (naftalen acedic acid); and sucrose concentrations were studied to determine the in vitro culture effects on healthier and faster seedling development from mature lentisk (Pistacia lentiscus L.) seeds. After 28 days of culture, the percentage of germinated seeds was the highest (70%) in the full-strength MS medium. The cytokinin BA was superior to other tested treatments in terms of its ability to promote germination of lentisk seeds. When tested at different concentrations, sucrose gave the best results obtained at concentrations of 1–4%, whereas high concentrations (6 and 8%) mainly decreased germination rate and there was no a regular pattern for elongation of the aerial parts of plants. With this described protocol, on average 76.67% seeds germinated 4 weeks after culture. Developed seedlings were satisfactorily acclimatized in sterilized peat, soil and perlite containing compost, with high percentage survival viability was obtained 9 months after transfer to in vivo conditions (93.33%). The results obtained showed that the enriched full-strength MS medium supplemented with 1 mg L–1 BA and 3% sucrose induced homogeneous and healthy seedling development in a period of 4 to 8 weeks of culture.

Opis:
Mandevilla sanderi is an important commercial ornamental pot plant. Traditional vegetative propagation is limited due to the low rate, therefore there is a need to develop an alternative, more efficient method. There is an interest in development of micropropagation technology for the species, as it allows to obtain a lot of offsprings in a relatively short time. The aim of the present work was to estimate an influence of explants type and position on regeneration of Mandevilla sanderi in tissue culture. Four different types of explants (leafy shoot tips, decapitated leafy shoot tips, defoliated shoot tips, decapitated and defoliated shoot tips) were used in the experiment, which were placed on the media vertically, while defoliated shoot tips were placed horizontally or vertically upside down. The explants were cultivated on a Murashige and Skoog (MS) medium supplemented with 1 mg·dm–3 benzyladenine (BA) and 0.5 mg·dm–3 indole-3-butyric acid (IBA). It was noted that both explants orientation and positioning, influenced the multiplication rate. Defoliated shoot tips placed horizontally were characterized by higher multiplication rate (6.8) in comparison to upside down vertical positioning (3.2). It was also observed that removal of shoot apex improved axillary branching, while defoliation of shoots placed in a normal position reduced multiplication rate.

Opis:
Observations of Japanese pagoda trees indicate that they undergo a full cycle of vegetative and generative development without self-renovation. The aim of this study was to obtain a successive media protocol propagation of Japanese pagoda tree by in vitro cultures. The effects of growth regulators were studied with reference to primary regeneration, shoot multiplication and rooting. As explants source were used the part of shoot of 90-year-oldtree. Explants were placed on MS (Murashige and Skoog 1962) basal medium with the addition of 6-benzylaminopurine – BAP (1.0–2.0 mg dm-3), thidiazuron – TDZ (0.1–0.3 mg dm-3) and indole-3-acetic acid – IAA (0.5–1.0 mg dm-3). BAP was the growth regulator which significantly increased shoot regeneration on initial explants. TDZ in turn, inhibited the formation of adventitious shoots and caused the explants which had been placed on the medium to die. The multiplication of the Japanese pagoda tree by in vitro cultures should be conducted on MS media with the addition of 0.5 mg dm-3 BAP, and they should be rooted on media with the addition of 0.3 mg dm-3 indole-3-butyric acid – IBA. It seems, that devising an efficient method of Japanese pagoda tree micropropagation is realistic.
