Opis:
To understand the molecular mechanism controlling in vitro plant morphogenesis, a culture system enabling induction of alternative morphogenic pathways (somatic embryogenesis, SE; shoot organogenesis, ORG) in a well defined population of somatic cells is needed. Arabidopsis is the most useful model plant for genomic studies, but a system in which SE or ORG can be induced alternatively in the same type of explant has not been proposed. Immature zygotic embryos (IZEs) of Arabidopsis provide the only explants with embryogenic potential, and have been recommended for studying mechanisms of SE induced in vitro. This study was aimed at defining culture conditions promoting induction of alternative morphogenic pathways: shoot ORG in IZE explants. The established protocol involves pretreatment of IZE explants with liquid auxin-rich callus induction (CIM) medium, followed by subculture on solid cytokinin-rich shoot induction medium (SIM). The method enables efficient shoot induction in Columbia (Col-0) and Wassilewskija (Ws), genotypes commonly used in molecular studies. During 3 weeks of culture up to 90% of Col-0 and 70% of Ws explants regenerated shoots via an indirect morphogenic pathway. We analyzed the qRT-PCR expression patterns of the LEC (LEC1, LEC2 and FUS3) genes, the key regulators of Arabidopsis embryogenesis, in the IZE explants induced to promote shoot ORG. The sharp decline of LEC expression on SIM medium confirmed that culture of Arabidopsis IZE explants enables experimental manipulation of the morphogenic response of somatic cells. A scheme illustrating various in vitro morphogenic responses of IZEs in relation to hormonal treatment is presented.

Opis:
The soaring prices of nitrogen fertilizer production, low use efficiency of nitrogen compounds, and environmental threats resulting from soil nitrogen leaching and gaseous nitrogen losses are the main reasons for conducting research on increasing nitrogen use efficiency (NUE). The aim of the study was to assess the effect of foliar application of the new generation of N fertilizers in the second and third dose on the yield of winter triticale and grain protein content, and to evaluate the indicators of nutrient efficiency: agronomic efficiency and apparent nitrogen recovery, relative to fertilizer type and method of application. The experiment was set up as a single factor one with a complete randomized block design. The experimental factor was the varying dose of mineral nitrogen fertilization against the constant level of phosphorus and potassium application. According to the experimental design, two liquid fertilizers (A1N and A2N) applied to foliage were the source of nitrogen in the second and third doses of fertilization, except for the control treatment (AN). The use of the hybrid technique of nitrogen application in the triticale fertilization technology at the same time reduced N fertilizer doses by 20 and 25% and significantly enhanced the yield of grain. Moreover, the treatment with the tested fertilizers increased significantly the nitrogen use efficiency from 45% to 75%. Considering the average protein contents from four experiments, the reduction in nitrogen doses, irrespective of the liquid fertilizer used, resulted in a decline in the grain protein content compared to the control. The results demonstrate the possibility of meeting the legal requirements for reducing the nitrogen dose while ensuring the yield at the same or even an increased level.

We report both decrease and increase in the 2D carrier gas density in a simple (Cd,Mn)Te/(Cd,Mg)Te heterostructure with (Cd,Mn)Te quantum well. The two effects were achieved by light with different photon energies. The quantum wells were 10 nm wide with 2D hole gas supplied by surface states. For the sample with 25 nm cap layer thickness, it was possible to tune the hole gas concentration from almost empty well (hole density below 1×10$\text{}^{10}$ cm$\text{}^{-2}$) to 45×10$\text{}^{10}$ cm$\text{}^{-2}$. The illumination with 425 nm wavelength almost doubled the hole gas density from the initial 24×10$\text{}^{10}$ cm$\text{}^{-2}$. The depletion mechanism was most effective for illumination with the orange (575 nm) light.

Photoluminescence of p-type modulation doped (Cd,Mn)Te quantum wells is studied with carrier density up to 5×10$\text{}^{11}$ cm$\text{}^{-2}$ at various spin splittings. This splitting can be made larger than the characteristic energies of the system thanks to the giant Zeeman effect. At small spin splitting and regardless of the carrier density, the photoluminescence exhibits a single line, which corresponds to the charged exciton in the singlet state. Above a certain spin splitting, the charged exciton is destabilized in favor of the exciton at vanishing hole density, and in favor of a double line at higher carrier density. It is found here that the charged exciton destabilization energy hardly depends on the carrier density. The double line is found to be band-to-band like, with the same initial state - where the holes have the same spin orientation - and final states that differ by some excitation of the 2D hole gas. In addition, the spin splitting needed to fully polarize the hole gas is twice smaller than expected from the single particle image and gives a unique insight into many-body effects in the hole gas.

New structures aiming at controlling the ferromagnetic properties of diluted magnetic semiconductor quantum wells are presented. The carrier density is controlled by applying a voltage across a p-i-n diode. A new method, creating a 2D hole gas by adjusting the distance between the quantum well and surface, offers opportunities for a broader range of structures.
