ETHNOGRAPHIC LABORATORY ASSOCIATION – ETHNOGRAPHY IN THIRD SECTOR The Ethnographic Laboratory Association was created in 2006 by students and graduates from the Institute of Ethnology and Cultural Anthropology at the University of Warsaw. The organization has undertaken steps aimed at the protection and promotion of cultural heritage, encouragement and support of creative local initiatives, social communication, promotion and raising awareness of the perception of cultural phenomena and integration of the environments of young researchers of culture. In 2009 we completed two major projects: “Open-air museums in Poland in Internet” and “ARAA – Academy of Development of Animation Activities.” Thanks to their implementation and success, the Ethnographic Department became recognizable and highly valued among the NGOs, as well as among the cultural and scientific institutions.

Smart specialisation constitutes an important framework for regional innovation policy making in the EU. According to the EU documents, the process of smart specialisation can be applied in all types of regions: the most developed ones as well as the least innovative ones. It is thus of great importance for regions with a low level of innovation performance due to numerous innovation challenges faced by these regions (such a situation can be observed in the case of four least innovative regions in Poland). The main objective of the paper is to assess the advancement of the least innovative Polish regions in the process of smart specialisation. In order to achieve this objective, the following detailed objectives are expected to be met: 1. presentation of innovation challenges for the least developed regions in Europe; 2. selection of the least innovative Polish regions on the basis of four indicators; 3. assessment of the advancement in the smart specialisation process in selected regions with reference to their economic, social and innovation potential. As research methods, the authors used descriptive analysis, analysis of strategic documents, case studies analysis and statistical analysis. As results from the analysis, the approach to the identification process of smart specialisations in the least innovative Polish regions was diverse. Depending on the maturity level of work on updating Regional Innovation Strategies, awareness of competitive advantages at the sectoral and technological level, used methods, different concepts of their identification have been adopted.

In this report we describe cloning and expression of human Rho GTPase activating protein (ARHGAP6) isoform 4 in Escherichia coli cells as a fusion protein with 6xHis. We cloned the ARHGAP6 cDNA into the bacterial expression vector pPROEX-1. Induction of the 6xHis-ARHGAP6 protein in BL21(DE3) and DH5α cells caused lysis of the cells irrespective of the kind of culture medium used. Successful expression of the fusion protein was obtained in the MC4100Δibp mutant strain lacking the small heat-shock proteins IbpA and IbpB. Reasonable yield was obtained when the cells were cultured in Terrific Broth + 1% glucose medium at 22°C for 16 h. The optimal cell density for expression of soluble 6xHis-ARHGAP6 protein was at A_600 about 0.5. Under these conditions over 90% of the fusion protein was present in a soluble form. The 6xHis-ARHGAP6 protein was purified to near homogeneity by a two step procedure comprising chromatography on Ni-nitrilotriacetate and cation exchange columns. The expression system and purification procedure employed made it possible to obtain 1-2 mg of pure 6xHis-ARHGAP6 protein from 300 ml (1.5 g of cells) of E. coli culture.

Objectives The aims of this study were: to evaluate the prevalence of abdominal wall defects in the Polish population, to analyze temporal trends in the prevalence, to identify areas (clusters) of high risk of abdominal wall defects, and to characterize, with respect to epidemiology, children with abdominal wall defects and their mothers in the area defined as a cluster. Material and Methods We used isolated congenital malformations (gastroschisis Q79.3 and omphalocele Q79.2 according to the International Statistical Classification of Diseases and Related Health Problems, 10th revision (ICD-10, the extended version)) data reported to the Polish Registry of Congenital Malformations (PRCM) over the years 1998– 2008 based on the population of 2 362 502 live births. We analyzed 11 administrative regions of Poland with complete epidemiologic data. Results Of 11 regions, 2 had a significantly higher standardized prevalence of isolated gastroschisis: Dolnośląskie (1.7/10 000 live births, p = 0.0052) and Śląskie (1.9/10 000 live births, p < 0.0001). Furthermore, within the region of Dolnośląskie, we defined a clear prevalence of the isolated gastroschisis cluster (p = 0.023). We comprehensively examined demographic and socio-economic risk factors for abdominal wall defects in this area, and we found that these factors failed to account for the cluster. Conclusions We identified a distinct prevalence cluster for isolated gastroschisis, although a precise reason for the disease clustering in this region remains unknown. Cluster identification enables more focused research aimed at identification of specific factors with teratogenic effects.
