A simple and coumarin-based fluorescence probe has been designed and synthesized with silyl group as recognition group of fluoride ions (F−) in this study. The results showed that the fluorescence intensity of the probe displayed prominent enhancement with addition of F− at 445 nm with incubation of 1 min. There was an excellent linear relationship between fluorescence intensity and fluoride concentration from 0 to 30 μM (0~0.57 ppm), which offered the important condition for the quantitative analysis. In addition, the highly selective response to fluorion, the low detection limit with 28 nM (0.532 ppb), low toxicity and bioimaging afforded an advantage for practical application and detecting fluoride in biological systerms.

Equid herpesvirus type 2 (EHV-2) together with equid herpesvirus type 5 are members of Gammaherpesvirinae subfamily, genus Rhadinovirus. EHV-2 is one of major agents causing diseases of horses common worldwide. A possible role of EHV-2 in reactivating latent equid herpesvirus type-1 has been suggested, because reactivation of latent EHV-1 was always accompanied by EHV-2 replication. Variety techniques, including cell culture, PCR and its modifications, have been used to diagnose EHV-2 infections. The aim of this study was to develop, optimize and determine specificity of real-time PCR (qPCR) for EHV-2 DNA detection using HybProbesR chemistry and to evaluate clinical samples with this method. The analytical sensitivity of assay was tested using serial dilutions of viral DNA in range between 70 and 7x105 copies/ml. The limit of detection (LOD) was calculated using probit analysis and was determined as 56 copies/ml. In further studies 20 different clinical samples were tested for the presence of EHV-2. Described in-house qPCR method detected viral DNA in 5 of 20 specimens used. The results of this work show that developed HybProbes-based real-time PCR assay is very reliable and valuable for detection and quantification of equid herpesvirus type 2 DNA in different clinical samples. The high level of sensitivity, accuracy and rapidity provided by the LightCycler 2.0 instrument are favorable for the use of this system in the detection of EHV-2 DNA in veterinary virology.

The purpose of this investigation was to compare Rapid Hicoliform Agar® (RHA, Himedia, India) with the conventional m-Endo Agar (DIFCO, Michigan, USA) for detection and enumeration of Total Coliforms (TC) and with the conventional Seven Hour Fecal Coliforms Agar (7hFC) for detection and enumeration of Escherichia coli using Membrane Filtration (MF) in River Nile water, Egypt. Sixty samples were collected (five samples per month) from five regions (Kafr El-Elw, El-Maasara, El-Giza, Embaba and El-Galatma) during one year (December 2005-November 2006). The results showed that total coliforms counts ranged from 103 to 104 CFU*100ml-1 using RHA but using m-Endo Agar the counts were around 104 CFU*100ml-1. Escherichia coli counts ranged from 103 to 104 CFU*100ml-1 using 7hFC Agar, while using RHA the counts ranged from 103 to 104 CFU*100ml-1. Statistical analysis showed that there was no significant difference (p=0.423) between total coliforms counts using m-Endo Agar and RHA media, also, there was no significant difference (p=0.546) between fecal coliforms (E. coli) counts using 7hFC Agar and RHA media. We conclude that RHA is a potential alternative medium for detection and enumeration of total coliforms and E. coli at the same time and the same Petri dish within 24h. Also, RHA medium was a very sensitive medium, which did not require any confirmatory tests and secured rapid recovery of total coliforms or E. coli.

The study was carried out in seven reproductive herds of pigs. In three of them reproductive disorders were observed. Three herds consisted of 10-50 and four consisted of 120-500 adult sows and they were called small and medium, respectively. Fifty-seven adult sows were randomly selected from herds. Serum samples were tested using the complement fixation test and swabs from both eyes and from the vaginal vestibule were examined using real-time PCR. All serum samples were negative. Infected sows were present in each of the study herds. In total, there were 28 positive samples (53%, 28/48) in real-time PCR in sows with reproductive disorders and 35 (53%, 35/66) in sows selected from herds without problems in reproduction. One isolate proved to be Chlamydophila pecorum, whereas all the remaining were Chamydia suis.

A loop-mediated isothermal amplification (LAMP) assay was developed to detect the sexuallytransmitted parasite, Trichomonas vaginalis in vaginal swabs. The presence of T. vaginalis was detected from 121 female sex workers attending a social hygiene clinic in Balibago, Angeles City, Pampanga, Philippines using culture, polymerase chain reaction (PCR), and the developed LAMP assay. The high analytical sensitivity of LAMP detected a higher prevalence of T. vaginalis (42.06%) compared to culture (8.26%) and PCR (7.44%). Additionally, this assay did not cross-react with DNAs of other trichomonads that can infect humans such as Trichomonas tenax and Pentatrichomonas hominis as well as the pathogens, Candida albicans and Staphylococcus aureus. The LAMP assay developed had a limit of detection (0.036 ng/μl) lower than that of PCR using the primers TvK3 and TvK7 (0.36 ng/μl). Prevalence of T. vaginalis in female sex workers in this area of the Philippines may be higher than previously estimated. Discordant results of PCR and LAMP may be due to different reactions to different kinds of inhibitors in the vaginal swabs.

The aim of the study was to evaluate a novel diagnostic scheme which combines quantitative PCR and High-Resolution Melting (qPCR-HRM) curve analysis for rapid differentiation based on E5 partial CDS of bovine papillomavirus type 1 or 2 (BPV-1 or BPV-2), and to perform a phylogenetic analysis of the complete CDS of the E5 gene of BPV detected in equine sarcoids. Samples of 38 skin lesions obtained from 27 horses were collected for molecular examinations. All lesions were clinically diagnosed as sarcoids, but results of histopathological examinations did not always corroborate the clinical diagnosis. Although all the samples were positive for the presence of BPV DNA, after qPCR-HRM analysis 6 (16%) specimens were recognized as BPV-1 “wild”, 24 (63%) as BPV-1 “European” and 8 (21%) as a “variant” of BPV E5 ORF partial CDS. Phylogenetic analysis based on nucleotide sequences of E2 ORF partial CDS and E5 ORF complete CDS was conducted on 7 specimens, whose sequences were published in GenBank and recognized as: 2PL (Accession Number - Acc. No. KC684939) - “variant” BPV-1, 7aPL (Acc. No. KC684940) - “European” BPV-1, 10PL (Acc. No. KC693480) - “variant” BPV-1, 16PL (Acc. No. KC693484) - “variant” BPV-2, 17PL (Acc. No. KC693481) - “variant” BPV-1, 20aPL (Acc. No. KC693482) - “European” BPV-1 and 20cPL (Acc. No. KC693483) - “wild” BPV-1. Amino acid (aa) sequences of E5 ORF complete CDS were also analyzed. The E5 variant of aa sequences found in isolate 10PL (protein identification - ID: AGM 20700) is a novel variant of E5 ORF complete CDS of BPV-1 detected in equine sarcoid in Poland.

In this survey the use of OptiMAL test for rapid diagnosis of malaria was evaluated. It was proved that this test allowed to diagnose the Plasmodium sp. antigen in 72% of examined blood specimens, 82% for P. falciparum infection and 69% for P. vivax, whereas P. ovale was not detected at all. The test sensitivity depended on the parasitemia level. It showed a sensitivity of 100% for parasitemia density exceeded 1%, 95.4% with the parasitemia ranging from 0.1-0.99%. For lower parasite density, the test’s sensitivity was of 32 and 60%. The OptiMAL test showed a 99.1% specificity thus it revealed to be significantly high.

Clostridium perfringens (MTCC 1349) is a Gram-positive, anaerobic, endospore forming, and rod-shaped bacterium. This bacterium produces a variety of toxins under strict anaerobic environment. C. perfringens can grow at temperatures ranging between 20°C and 50°C. It is the major causetive agent for gas gangrene, cellulitis, septicemia, necrotic enteritis and food poisoning, which are common toxin induced conditions noted in human and animals. C. perfringens can produce produce four major types of toxins that are used for the classification of strains, classified under type A-E. Across the globe many countries, including the United States, are affected by C. perfringens food poisonings where it is ranked as one of the most common causes of food borne infections. To date, no direct one step assay for the detection of C. perfringens has been developed and only few methods are known for accurate detection of C. perfringens. Long detection and incubation time is the major consideration of these reporter assays. The prensent study proposes a rapid and reliable colorimetric assay for the detection of C. perfringens. In principale, this assay detects the para nitrophenyl (yellow colour end product) liberated due to the hydrolysis of paranitrophenyl phosphetidyl choline (PNPC) through phospholipase C (lecithinase). Constitutive secretion of phospholipase C is a charactristic feature of C. perfringens. This assay detects the presence of the extracellular lecithinse through the PNPC impragnated impregnated probe. The probe is impregnated with peranitrophenyl phosphotidyl choline ester, which is colourless substrate used by lecithinase. The designed assay is specific towards PNPC and detectes very small quantites of lecithinase under conditions used. The reaction is substrate specific, no cross reaction was observed upon incubation with other substrates. In addition, this assay gave negative results with other clostridium strains, no cross reactions were observed with other experimental strains like C. tetani, C. botulinum, C. acetobutyricum, Bacillus subtilis, and Escherichia coli. This assay is extramly rapid and provides reliable and reproducible results within one hour of incubation at 37°C.

An improved harmonic detection method based on average arithmetic is proposed. According to the research results, the designed solution uses an LPF (low-pass-filter) and a mean value module connected in series instead of the conventional mean value module, and simultaneously, a three-phase voltage phase-locked module instead of commonly used PLL (phase lock loop) module is applied in order to reduce the influence caused by three-phase distorted voltage and rapid variation of load. The experimental results show that the application of this solution leads to increase in the accuracy of harmonics detection for distorted three-phase voltage and rapid variation of load.

Three different rapid strip tests: TRU RSV, BinaxNOW RSV and RSV Respi-strip were compared with RT-PCR and ELISA BRSV Ag for the ability to detect bovine respiratory syncytial virus (BRSV) in nasal swabs collected from calves experimentally vaccinated with live vaccine Rispoval RS-PI3. The reference strains of BRSV (375 and A51908) were detected by ELISA BRSV Ag whereas the strains of human respiratory syncytial virus (HRSV) and bovine parainfluenza virus type 3 (BPIV-3) were not. All rapid strip tests as well as RT-PCR reacted positively both to HRSV and BRSV reference strains and negatively to BPIV-3. The detection limit for RT-PCR was 39.1 TCID50 (strain 375 of BRSV), whereas for each of the rapid tests it was approximately 156 TCID50 and 312 TCID50 for antigen ELISA. Diagnostic sensitivity in detecting BRSV in nasal swabs for TRU RSV and RSV Respi-strip tests was 33% and 50% for BinaxNOW RSV. Diagnostic specificity of TRU RSV was 100%, whereas for both BinaxNOW and Respi-strip it was 87%. We concluded that TRU RSV could be used as a supportive rapid test for BRSV screening in nasal swabs taken directly on a farm. However, due to the small group of animals used in the experiment, the results should be regarded as preliminary and the study should be repeated on a larger number of animals.

Background. The key tool used in the European Union in order to eliminate the risks associated with the consumption of potentially hazardous food is RASFF - Rapid Alert System for Food and Feed Safety. The RASFF was established to increase accountability and strengthening cooperation between states of the European Union in the field of food safety control. Objective. The aim of this study was to explore the trends and temporal and spatial distribution of notifications on food safety hazards between January 2011 and December 2015 with a special emphasis on meat and meat products on the basis of notification from RASFF. Material and methods. The study analyzed notifications on the annual reports of the RASFF published by the European Commission and requests added to the portal RASFF in the period 01.01.2011 - 31.12.2015 on the category of “meat and meat products (other than poultry) and “poultry meat and poultry meat products”. Analysis included detailed information on each notification, such as the classification and date, hazard category, notifying country, country origin. Results. The most common classifications of notification were ‘alert’ and ‘border rejection’. Generally, basis of this notifications were ‘company’s own check’ and ‘official control on the market’. Pathogenic microorganisms were the most often hazard of category in which the higher number of notifications concerned with Salmonella spp. Conclusion. Alert notification which is the most dangerous for consumers were the most common type of classification for notifications on ‘meat and meat product’ category. The most of notifications in category ‘poultry meat and poultry meat products’ were the result of border control. Pathogenic microorganisms were the reason for the huge number of notifications in studied product categories. Many of notifications were associated with products which origin countries were outside RASFF member states.
Wprowadzenie. Kluczowym narzędziem stosowanym w Unii Europejskiej w celu wyeliminowania ryzyka związanego ze spożywaniem potencjalnie niebezpiecznej żywności jest RASFF - System Wczesnego Ostrzegania o Niebezpiecznej Żywności i Paszach. Stworzenie Systemu miało na celu zwiększenie odpowiedzialności i wzmocnienie współpracy pomiędzy państwami Unii Europejskiej w zakresie kontroli bezpieczeństwa żywności. Cel. Celem niniejszego opracowanie była analiza powiadomień zgłoszonych do systemu RASFF w latach 2011 - 2015 dotyczących mięsa i produktów mięsnych. Materiał i metody. W niniejszym opracowaniu przeprowadzono analizę powiadomień dostępnych w rocznych sprawozdaniach RASFF i znajdujących się w portalu RASFF opublikowanych przez Komisję Europejską w okresie 01.01.2011 - 31.12.2015. Analizowane zgłoszenia należały do kategorii „mięso i produkty mięsne (inne niż drób) i „mięso i produkty drobiowe”. Badania opierały się na informacjach zawartych w raportach rocznych systemu RASFF i platformie internetowej Systemu RASFF. Wyniki. Najczęściej zgłoszenia klasyfikowane były jako powiadomienia alarmowe lub odrzucenia na granicy. Podstawą zgłoszeń były głównie kontrole wewnętrzne lub urzędowe kontrole. Najczęściej pojawiającym się zagrożeniem były patogenne mikroorganizmy, które najczęściej spowodowane były obecnością Salmonella spp. Krajami, które najczęściej wysyłały zgłoszenia do Systemu RASFF były Włochy w przypadku kategorii ‘mięsa i produktów mięsnych’ oraz Holandia w przypadku ‘drobiu i produktów z drobiu’. Wnioski. Powiadomienia alarmowe, które stanowią największe zagrożenie dla konsumenta były najczęściej pojawiającym się typem zgłoszeń w przypadku mięsa i produktów mięsnych. Najwięcej powiadomień w kategorii ‘drób i produkty drobiowe’ było wynikiem kontroli granicznej. Patogenne organizmy były powodem ogromnej liczby zgłoszeń w badanych kategoriach produktów. Duża liczba zgłoszeń związana była z produktami, które pochodziły z krajów spoza UE, nie objętych systemem RASFF.
