In this study, wheat (Triticum aestivum L.) roots were treated with hypoxic water. The staining of cell preparations with DAPI revealed morphological changes of the cells such as nuclear condensation, deformation and fragmentation. Under TEM, cellular membrane shrinkage and breakage, chromatin condensation and apoptotic- like bodies were displayed. The number of mitochondria increased dramatically; their cristae were damaged; the interior became a cavitation and only some flocculent materials were distributed. Indirect immunofluorescence staining indicated that cytochrome C diffused from mitochondria to nucleoplasm and cytoplasm. TUNEL positive nuclei indicated double strand breaks of DNA. DAB staining was used for the identification of hydrogen peroxide and examination showed that the longer the treating time, the darker the staining of the meristematic zones of the roots which suggested the increased accumulation of these Reactive Oxygen Species (ROS). The elevation of hydrogen peroxide production was paralleled with the increase of SOD and POD activities. A negative correlation between the exposure time under hypoxia and the contents of soluble proteins was found. No obvious effect of hypoxia on MDA was established. The obtained results demonstrate that hypoxia causes programmed cell death in the root-tip meristematic cells of Triticum aestivum L. which is most probably attributed to the accumulation of large amounts of ROS.

Opis:
Earliest deadline first (EDF) and fixed priority (FP) are the most commonly used and studied scheduling algorithms for real-time systems. This paper focuses on combining the EDF and FP strategies in one system. We provide a new sufficient schedulability analysis for real-time hybrid task systems which are scheduled by EDF and FP. The proposed analysis has a polynomial time complexity and no restrictions on task parameters, where the relative deadline of each task could be less than, equal to, or greater than its period. By extensive experiments, we show that our proposed analysis significantly improves the acceptance ratio compared with the existing results of the sufficient schedulability test for hybrid scheduling systems.

Opis:
Accumulation of LaCl3 , a well-known Ca 2+ -channel blocker, can inhibit plant growth. However, the current understanding of its effects on gene expression is limited. In this paper, different concentrations of LaCl 3 (0, 0.5, 1.0, 1.5, 2.0 mM) were used to treat germinated wheat (Triticum aestivum L.) seeds for 24 h. The degree of root growth inhibition gradually increased with increasing LaCl 3 concentration. qRT-PCR analysis revealed that the expression of several key genes related to the cell cycle process, such as pcna, mcm2, rdr and cyclin B, were significantly down-regulated. Further analysis of genomic DNA instability using Random Amplified Polymorphic DNA (RAPD) and methylation levels by Coupled Restriction Enzyme Digestion-Random Amplification (CRED-RA) analysis indicated a significant increase in genomic DNA polymorphisms and methylation levels. The results of this study verified that the reasons why LaCl3 treatment can inhibit the growth of wheat roots are as follows: interference in the normal progression of the cell cycle, induction of genomic DNA instability and increase in DNA methylation levels.

Opis:
Electrospinning is an effective technique for fabricating submicron to nanoscale fibers from synthetic polymer as well as natural proteins. In this study, multiwalled carbon nanotubes (MWNTs) were embedded via electrospinning by adding MWNTs into the spinning dope, and found to be well aligned along the fiber axis in the silk fibroin nanofibers. The morphology and microstructure of the electrospun nanofibers were characterised using a field emission scanning electron microscope (FESEM) and Transmission electron microscopy (TEM). X-ray diffraction (XRD) and TG-DTA were used to study the crystal structure of the silk/MWNTs composite nanofibres, carried out to alter the strength, toughness and electrical conductivity of silk nanofibers by adding a small amount of MWNTs. The electrospun random silk mats with 1% MWNTs had a Young’s modulus, ultimate tensile strength and strain of 107.46 ± 9.15MPa, 9.94 ± 1.2MPa and 9.25 ± 1.5%, respectively, and electrical conductivity increased to 1.2×10-4S/cm. The silk/MWNTs composite nanofibres could potentially be applied in nerve repair materials owing to their excellent mechanical properties and electrical conductivity.
Za pomocą elektroprzędzenia wytworzono jedwabne nanowłókna z dodatkiem nanorurek węglowych. Zbadano wpływ nanorurek węglowych na morfologię, strukturę, właściwości mechaniczne i przewodność elektryczną j nanowłókien. Morfologię i mikrostrukturę otrzymanych nanowłókien scharakteryzowano za pomocą skaningowego mikroskopu elektronowego (FESEM) i transmisyjnej mikroskopii elektronowej (TEM). Wyniki badań rentgenowskich i cieplnych wykazały, że dodanie nanorurek nie wywierało istotnego wpływu na strukturę nanowłókien w porównaniu do niemodyfikowanych nanowłókien fibroinowych. Poprzez dodanie nanorurek uzyskano polepszenie właściwości mechanicznych, została również polepszona przewodność elektryczna nanowłókien. Na podstawie otrzymanych wyników stwierdzono, że jedwabne nanowłókna z dodatkiem nanorurek węglowych mogą być stosowane w materiałach do naprawy nerwów dzięki ich doskonałym właściwościom mechanicznym i przewodności elektrycznej.

In order to properly understand the effect of freezing-thawing circle (FTC) to mechanical behavior of costal soft soil (CSS), unconfined compressive test is conducted. Six kind FTC times are designed from zero to five. The tested data show that: (1) unconfined compressive strength of CCS decreases nonlinearly with more FTC, and the strength after five FTC times becomes about 22% of its original strength without any freezing-thawing experience; (2) stressstrain curves of all unconfined compressive samples can be well fitted by three-parameter hyperbolic model; (3) and relationship between two parameters and FTC times can be fitted by exponent function, while another parameter can be considered as 0.95. Consequently, one composite hyperbolic- exponent empirical formula is established in order to describe freezing-thawing-dependent stress-strain behavior of CSS. Finally, good agreements have been found between tested dada and simulated results.

Based on crystal- and ligand-field theory, double-spin-orbital coupling approach was used to analyze the crystal-field energy levels and spin-Hamiltonian parameters of $Ni^{2+}$ ion at trigonal site in $CdBr_2$. The local lattice distortion (Δ R and $τ_{Ni^{2+}}$) is estimated from the crystal field parameters; the crystal field energy Hamiltonian was diagonalized in the full basis consisting of 45 wave functions of the $Ni^{2+}$ ion. Results of calculations are in good agreement with experimental data. The reasonableness of the theoretical results is discussed.

In this study, when germinated Triticum aestivum L. seeds were treated with 0, 2, 4 and 6 mM ethyl glycol tetraacetic acid (EGTA), root growth was suppressed and the mitotic index decreased. These inhibitory effects were positively correlated with EGTA concentration. RT-PCR analysis revealed that the expression of several gene markers related to the G1/S transition of the cell cycle were significantly downregulated. Confocal microscopy of Fluo-3/AM-stained roots showed chelation of nearly all of the Ca2+ within the root meristematic regions. Both random amplified polymorphic DNA (RAPD) and coupled restriction enzyme digestion-random amplification (CRED-RA) techniques showed significant increases in the levels of genomic DNA polymorphisms and degree of DNA methylation. The study provides information concerning the impact of Ca²+) chelator, EGTA, on the growth, expression of cell cycle transition marker genes, and changes in DNA structure and methylation in the wheat roots.
