Opis:
The digital holographic phase microscopy (DHPM) technique which has been proposed for cellular morphology and dynamic analysis yielded highly desirable results. However, for nucleated cells (especially white blood cells (WBCs)), their submicroscopic structure has not yet been deconstructed through a phase unwrapping method due to the heterogeneity of an internal phase. By analyzing the phase heterogeneity of subclasses of WBCs, the typical phase models of them are built first in this paper; using the simulation method, the wrapped phase distributions of these models are obtained. However, by optimizing the wrapped phase maps and analyzing the relationships between them and typical blood cells, their features are selected and extracted. Then the models built are sorted out from each other successfully without unwrapping via analyzing these extracted features, which provides a valuable approach and technological base for the classification and identification of blood cells.

Fruit biomechanics is needed for quality determination, multiscale modelling and engineering design of fruit processes and equipments. However, these determined fruit biomechanics data often have obvious differences for the same fruit or tissue. In order to investigate it, the fruit biomechanics based on anatomy was reviewed in this paper. First, the anatomical characteristics of fruit biomaterials were described at the macroscopic ‘tissue’ level and microscopic ‘cellular’ level. Subsequently, the factors affecting fruit biomechanics based on anatomy and the relationships between fruit biomechanics, texture and mechanical damage were summarised according to the published literature. Fruit biomechanics is mainly affected by size, number and arrangement of cells, quantity and volume of intracellular spaces, structure, thickness, chemical composition and permeability of cell walls, and pectin degradation level and turgor pressure within cells based on microanatomy. Four test methods and partial determined results of fruit biomechanics were listed and reviewed. The determined mechanical properties data of fruit are only approximate values by using the existing four test methods, owing to the fruit biomaterials being non-homogeneous and living. Lastly, further aspects for research on fruit biomechanics were proposed for the future.

In this paper, a new lifting wavelet domain audio watermarking algorithm based on the statistical characteristics of sub-band coefficients is proposed. First of all, an original audio signal was segmented and each segment was divided into two sections. Then, the Barker code was used for synchronization, the LWT (lifting wavelet transform) was performed on each section, a synchronization code and a watermark were embedded into the first section and the second section, respectively, by modifying the statistical average value of the sub-band coefficients. The embed strength was determined adaptively according to the auditory masking property. Experiments show that the embedded watermark has better robustness against common signal processing attacks than present algorithms based on LWT and can resist random cropping in particular.

To provide a theoretical and practical foundation for ramie genetic analysis, simple sequence repeats (SSRs) were identified in the ramie genome and employed in this study. From the 115 369 sequences of a specific-locus amplified fragment library, a type of reduced representation library obtained by high-throughput sequencing, we identified 4774 sequences containing 5064 SSR motifs. SSRs of ramie included repeat motifs with lengths of 1 to 6 nucleotides, and the abundance of each motif type varied greatly. We found that mononucleotide, dinucleotide, and trinucleotide repeat motifs were the most prevalent (95.91%). A total of 98 distinct motif types were detected in the genomic-SSRs of ramie. Of them, The A/T mononucleotide motif was the most abundant, accounting for 41.45% of motifs, followed by AT/TA, accounting for 20.30%. The number of alleles per locus in 31 polymorphic microsatellite loci ranged from 2 to 7, and observed and expected heterozygosities ranged from 0.04 to 1.00 and 0.04 to 0.83, respectively. Furthermore, molecular identity cards (IDs) of the germplasms were constructed employing the ID Analysis 3.0 software. In the current study, the 26 germplasms of ramie can be distinguished by a combination of five SSR primers including Ibg5-5, Ibg3-210, Ibg1-11, Ibg6-468, and Ibg6-481. The allele polymorphisms produced by all SSR primers were used to analyze genetic relationships among the germplasms. The similarity coefficients ranged from 0.41 to 0.88. We found that these 26 germplasms were clustered into five categories using UPGMA, with poor correlation between germplasm and geographical distribution. Our study is the first large-scale SSR identification from ramie genomic sequences. We have further studied the SSR distribution pattern in the ramie genome, and proposed that it is possible to develop SSR loci from genomic data for population genetics studies, linkage mapping, quantitative trait locus mapping, cultivar fingerprinting, and as genetic diversity studies.
