Purpose: Enterococcus faecium strains have been reported worldwide as etiologic factors of many nosocomial infections, which are difficult to manage because of the constantly increasing resistance of these microorganisms to antibiotics and the ability to form biofilm. The aim of this study was to analyze the ability to produce a biofilm in E. faecium strains, depending on the patient’s clinical material. Materials and methods: Sixty-six E. faecium strains were investigated. Identification and susceptibility testing were conducted by the VITEK2 system. The ability to form biofilm was assessed by phenotypic methods. The presence of selected virulence genes was established by PCR followed by gel electrophoresis and sequencing. Results: Among the tested E. faecium isolates, 72.7% were biofilm-positive (BIO+) and 27.3% biofilm-negative (BIO-). Strains were collected mostly from rectal swabs (30.4%) and blood (18.3%). BIO+ strains from infections constituted 31.8% (52.4% isolated from blood) and from colonization 40.9% (48.2% from rectal swabs). 91.7% of the Blood Group strains and 68.5% of the Other Group strains produced biofilm. Strains from the Colonization Group produced biofilm in a proportion similar to the Infection Group (about 75%). There were no statistically significant differences in virulence and resistance, except for vancomycin (more resistant BIO+ Other than the BIO+ Blood Group, and more resistant BIO+ Colonization than BIO+ Infection Group) and teicoplanin (more resistant BIO+ Colonization than the BIO+ Infection Group). Conclusion: The majority of E. faecium isolates carries high levels of resistance to many antimicrobials, is well equipped with virulence genes, and possesses the ability to form biofilm.

Opis:
Purpose: Today, Enterococcus species are one of the most frequent etiological agents in nosocomial infections. The aim of this study was to determine the susceptibility to antibiotics and the prevalence of high-level aminoglycoside resistance (HLAR) among Enterococcus strains. Materials and methods: The susceptibility of 85 isolates of Enterococcus (47 E. faecalis and 38 E. faecium) was determined using the disk diffusion method. The results were interpreted according to European Committee on Antimicrobial Suscepti-bility Testing (EUCAST) guidelines. PASW Statistics 17.0 was used for statistical analysis. Results: E. faecalis strains showed the highest susceptibility to ampicillin, tigecycline, vanco-mycin, imipenem, and linezolid and E. faecium to linezolid, tigecycline, and quinupristin/dalfopristin. Among all tested strains, high-level gentamicin resistance (HLGR) was found in 4% of E. faecalis and 8% of E. faecium strains, high-level strepto-mycin resistance (HLSR) in 45% and 42%, and HLAR in 50% and 32% of strains, respectively. HLGR was detected only in vancomycin-resistant Enterococcus (VRE)− strains (12%), while HLSR in 76.9% of VRE+ and 24% of VRE− strains, and HLAR in 23.1% of VRE+ and 64% of VRE− strains. The tested strains were also divided into two groups: HLSR+ and HLAR+. In both groups, statistically significant susceptibility differences (p<0.05) were found for ampicillin, imipenem and trimethoprim/sulfamethoxazole. The most frequent antibiotic resistance profile among E. faecalis strains was SR (resistance phenotype to strepto-mycin), and among E. faecium, AMPR, IMPR, CNR, SR, SXTR (ampicillin, imipenem, gentamicin, streptomycin, trimethoprim/sulfamethoxazole). Conclusions: This study showed the slowly increasing prevalence of HLAR and resistance to newer antibiotics (linezolid and tigecycline) among Enterococcus strains. It is necessary to search for new directions in the treatment of enterococcal infections.

W niniejszej pracy przedstawiono doświadczalne wyniki analizy spalin silników benzynowych o zasilaniu gaźnikowym Renault 19 Chamade 1.4 i wtryskowym Volkswagen Polo 6N 1.4 eksploatowanych w układach: bez aktywatora paliwa, z trójpolowym aktywatorem paliwa Power oraz z jednopolowymi aktywatorami paliwa Dynamic Fuel GTM i Dynamic Fuel Reference. Wykazały one dwie różne zależności. Dla przypadku silnika gaźnikowego Renault 19 Chamade 1.4 stwierdzono wzrost emisji toksycznych gazów, natomiast dla przypadku silnika wtryskowego Volkswagena Polo 6N 1.4 zaobserwowano spadek zawartości szkodliwych gazów. W przypadku tego drugiego silnika, najkorzystniejszymi własnościami w zakresie zmniejszenia toksyczności spalin wykazał się jednopolowy aktywator paliwa Dynamic Fuel Reference.
At present work the experimental analyses of combustion products gasoline engines about carburettor admission Renault 19 Chamade 1.4 and injection Volkswagen Polo 6N 1.4 worked in configuration: without fuel activator, with three magnetic fields Power and one magnetic fields activators Dynamic Fuel GTM and Dynamic Fuel Reference was introduced. Different dependences were demonstrated. For carburattor engine Renault 19 Chamade 1.4 was affirmed the increase of release of toxic gases, however for case the injection's engine Volkswagen Polo 6N 1.4 was observed the decrease of content of harmful combustion products. In case of the second engine the most advantageous properties in range of decrease of toxicity of combustion products the fuel activator Dynamic Fuel Reference was displayed.

Over the last years, systemic fungal infections have dramatically increased in hospitalized patients. The Candida is the main pathogen caused nosocomial fungal infections. The aim of the study: The aim was to analyze frequency of occurrence of the yeast-like fungi in different biological materials isolated from the patients of an Intensive Care Unit of the University Hospital of Lodz in the period of 2000-2003. Material and methods: 123 strains of fungi were analyzed with the use of API 20 C AUX® test (bioMarieux). Results: Among all the investigated Candida strains C. albicans accounted for 52.0%. Samples from respiratory system and urine most often contained the strains of C. albicans (56.3 and 60.5%, respectively); blood samples contained C. parapsilosis (44.8%). In patients who were untreated by bacterial antibiotics C. albicans was the most frequent species, whereas in patients who were ordered bacterial antibiotics it was C. parapsilosis that dominated. Conclusions: (1) Candida is the most frequent cause of fungal infections in patients hospitalized in an intensive care unit. (2) C. parapsilosis is the main pathogen caused bloodstream infections. This species is also more frequent in patients who were ordered antibacterial antibiotics over five days. (3). Species other than C. albicans become more and more frequent and dangerous.

Nowadays, in positron emission tomography (PET) systems, a time of fl ight (TOF) information is used to improve the image reconstruction process. In TOF-PET, fast detectors are able to measure the difference in the arrival time of the two gamma rays, with the precision enabling to shorten signifi cantly a range along the line-of-response (LOR) where the annihilation occurred. In the new concept, called J-PET scanner, gamma rays are detected in plastic scintillators. In a single strip of J-PET system, time values are obtained by probing signals in the amplitude domain. Owing to compressive sensing (CS) theory, information about the shape and amplitude of the signals is recovered. In this paper, we demonstrate that based on the acquired signals parameters, a better signal normalization may be provided in order to improve the TOF resolution. The procedure was tested using large sample of data registered by a dedicated detection setup enabling sampling of signals with 50-ps intervals. Experimental setup provided irradiation of a chosen position in the plastic scintillator strip with annihilation gamma quanta.

Opis:
In this article we present a novel method of hit time and hit position reconstruction in long scintillator detectors. We take advantage of the fact that for this kind of detectors amplitude and shape of registered signals depend strongly on the position where particle hits the detector. The reconstruction is based on determination of the degree of similarity between measured and averaged signals stored in a library for a set of well-defined positions along the scintillator. Preliminary results of validation of the introduced method with experimental data obtained by means of the double strip prototype of the J-PET detector are presented.

Opis:
J-PET analysis framework is a flexible, lightweight, ROOT-based software package which provides the tools to develop reconstruction and calibration procedures for PET tomography. In this article we present the implementation of the full data-processing chain in the J-PET framework which is used for the data analysis of the J-PET tomography scanner. The framework incorporates automated handling of PET setup parameters' database as well as high level tools for building data reconstruction procedures. Each of these components is briefly discussed.

Opis:
For the first time a molecule of 2-(4-styrylphenyl)benzoxazole containing benzoxazole and stilbene groups is applied as a scintillator dopant acting as a wavelength shifter. In this article a light yield of the plastic scintillator, prepared from styrene doped with 2 wt% of 2,5-diphenylbenzoxazole and 0.03 wt% of 2-(4-styrylphenyl)benzoxazole, is determined to be as large as 60% ± 2% of the anthracene light output. There is a potential to improve this value in the future by the optimization of the additives concentrations.

The J-PET detector being developed at the Jagiellonian University is a positron emission tomograph composed of the long strips of polymer scintillators. At the same time, it is a detector system that will be used for studies of the decays of positronium atoms. The shape of photomultiplier signals depends on the hit time and hit position of the gamma quantum. In order to take advantage of this fact, a dedicated sampling front-end electronics that enables to sample signals in voltage domain with the time precision of about 20 ps and novel reconstruction method based on the comparison of examined signal with the model signals stored in the library has been developed. As a measure of the similarity, we use the Mahalanobis distance. The achievable position and time resolution depend on the number and values of the threshold levels at which the signal is sampled. A reconstruction method as well as preliminary results are presented and discussed.

In this paper, we present prospects for using the Jagiellonian positron emission tomograph (J-PET) detector to search for discrete symmetries violations in a purely leptonic system of the positronium atom. We discuss tests of CP and CPT symmetries by means of ortho-positronium decays into three photons. No zero expectation values for chosen correlations between ortho-positronium spin and momentum vectors of photons would imply the existence of physics phenomena beyond the standard model. Previous measurements resulted in violation amplitude parameters for CP and CPT symmetries consistent with zero, with an uncertainty of about 10−3. The J-PET detector allows to determine those values with better precision, thanks to the unique time and angular resolution combined with a high geometrical acceptance. Achieving the aforementioned is possible because of the application of polymer scintillators instead of crystals as detectors of annihilation quanta.

Opis:
Results of positron annihilation lifetime spectroscopy (PALS) and microscopic studies on simple microorganisms, brewing yeasts, are presented. Lifetime of ortho-positronium (o-Ps) were found to change from 2.4 to 2.9 ns (longer-lived component) for lyophilized and aqueous yeasts, respectively. Also hygroscopicity of yeasts in time was examined, allowing to check how water – the main component of the cell – affects PALS parameters, thus lifetime of o-Ps were found to change from 1.2 to 1.4 ns (shorter-lived component) for the dried yeasts. The time sufficient to hydrate the cells was found below 10 hours. In the presence of liquid water, an indication of reorganization of yeast in the molecular scale was observed. Microscopic images of the lyophilized, dried, and wet yeasts with best possible resolution were obtained using inverted microscopy (IM) and environmental scanning electron microscopy (ESEM) methods. As a result, visible changes to the surface of the cell me mbrane were observed in ESEM images.
