Beech decline phenomenon has been observed in Poland since the 1980s. The reasons are still unclear but the discovery of pathogens genus Phytophthora shed a new light on it. Stems as well as roots of beech trees were severely affected by P. citricola and P. cambivora. In order to find out the genetic background of infection, three loci of chloroplast microsatellite DNA were investigated in the genetic differentiation study of beeches growing in the Siewierz Forest District in Poland. Parameters of genetic diversity (h Nei) and differentiation (GST) in the chloroplast genome were estimated and compared between healthy and damaged trees. Healthy beeches were more heterozygous (h=0.243) than the damaged ones (h=0.113), and the distribution of cpDNA alleles was different between these two groups. Healthy trees were nearly 50% more genetically diversed than declining ones.

Black Alder Alnus glutinosa (L.) Gaertn. is an important tree commonly growing in Poland. Alders are actinorhizal plants that play an important ecological role in riparian ecosystems through atmospheric nitrogen fixation, filtration and purification of waterlogged soils as well as providing a refuge for terrestrial and aquatic organisms thus helping to stabilize stream banks. Black Alder used to be considered a very pest and disease resistant species but, the situation changed in 2000, when an unprecedented decline of Alders was observed in Poland. In the Białowieża Forest District, this decline has been observed on wet meadow habitats and along rivers or watercourses. Currently, there are several hypotheses explaining Alder dieback, among them climatic changes and Phytophthora infections. In terms of climate, Black Alder requires a high atmospheric humidity during all phases of its reproductive cycle. It tolerates neither long-term summer flooding nor a significant decrease in the groundwater level. In terms of pests, oomycete pathogens of the genus Phytophthora are the most destructive plant pathogens known and many of them are present in forests and nurseries all over Europe. The aim of this study was to evaluate the health of Black Alder along the Narewka River in the Białowieża Forest District. Selected areas were monitored in 2012 and 2018, but no relationship between drought and alder health was found. A preliminary analysis of soil and water samples by real time PCR revealed the presence of two Phytophthora species: P. alni and P. cactorum. Further and more detailed research is required to elucidate the role of these pathogens in Alder dieback.

Due to the deterioration in the health condition of Norway spruce on the IPTNS-IUFRO 1964/68 observation plots in Krynica (Poland), there was an urgent need to determine the cause of the decline in order to design preventive measures. For this purpose, a health inventory was carried out in the years 2002 and 2003. This included an assessment of the condition of crowns in 144 trees and a thorough examination of 29 felled trees. It was found that individual trees or groups of trees have declined in some blocks of the experiment. The tops of some trees were dying and the crowns were getting transparent. The discolouration (turning brown) of needles was quite common and even green needles were shed. Some of the roots showed symptoms of necrosis and died. Seven taxa of pathogens were isolated from the diseased roots and trunk parts of spruces, among them Phytophthora citrophthora, Fusarium avenaceum, F. solani and Trichoderma. The number of diseased trees was related to provenance. The provenances Babenhausen (Germany), Frantiskovy Lazne (Czech Republic), Traunstein 1/4 D, 6 A, B, 7 A (Germany), Wundsiedel-Weissenstadt (Germany), Mestwinowo (Poland), and Magland (France) proved to be more susceptible than others to fungal pathogen attack.

In the situation of limited number of pesticides being approved for use in forest nurseries it is necessary to examine new products available on European market that stimulate growth and improve resilience and vitality of seedlings as a part of intgrated plant protection program. Paper presents test of fertilizer Actifos used in seven Polish nurseries in order to stimulate the growth of seedlings. In 64% of cases, Actifos stimulated increament of shoots (significantly for oaks, beeches, pines, spruces and alders) as well as for roots (significantly only for alders and oaks).

Opis:
Traditional detection methods such as baiting or direct isolation take a long time and are incapable to handling large volume of material to be tested. The real−time PCR−based techniques are faster, more sensitive, more easily automated, and do not require post−amplification procedures. Species−specific primers for Phytophthora were designed based on the internal transcribed spacer regions (ITS) of rDNA collected from the NCBI DNA database. Primers and probes were designed using the Allele ID 7 at default search criteria. Specific probes were labeled with the reporter dyes JOE (6−carboxy−4,5−dichloro−2,7−dimethoxyfluorescein) at the 5' end and HBQ1 quencher at the 3' end (Sigma−Aldrich). The specificity of primers and fluorogenic probes was tested against genomic DNA of P. alni subsp. multiformis, P. lacustris and P. taxon hungarica. The real−time PCR reactions with the specific probes and primers yielded positive results with five concentrations of standards obtained by standard PCR reaction for corresponding Phytophthora species. The negative control (lack of DNA pathogens) yielded no amplification products. Standard curves showed a linear correlation between input DNA and cycle threshold (Ct) values with R² from 0.994 (P. alni) to 0.998 (P. taxon hungarica). The amplification efficiency of target DNA varied from 94.6% (P. alni) to 100% (P. taxon hungarica). The validation of the primers and probes designed for analysed Phytophthora species was performed on pure cultures, on soil samples from the forest nursery and declining oak stands. The designed probes displayed the high specificity of the detection of investigated species in pure cultures. The presented new molecular TaqMan probes can fully assist the integrated pest management as a powerful tool for a quick detection of above pathogenic organisms in forest nurseries. The molecular detection of harmful phytophthoras and in consequences diminishing of fungicides use for their control in forestry fully support European Union directives as well as the ‘Good plant protection practice measures' elaborated by European and Mediterranean Organisation of Plant Protection.

The genus Phytophthora plays an important role not only in agriculture but also in forest ecosystems. As the number of known Phytophthora species continues to grow, identifying new isolates in this genus has become increasingly challenging even by DNA sequencing. Therefore, the development of proper techniques for detection and identification is crucial for monitoring and control of these pathogens in the forestry sector. In recent years, new molecular methods using innovative approaches have indeed been developed. However, the majority of these methods was designed to detect single Phytophthora species. Techniques that are able to target multiple species would offer advantages, especially for the assessment of Phytophthora diversity in the environment. This paper describes a multiplex assay for the identification of eight Phytophthora isolates, down to the species level, based on a Fluidigm platform employing pyrosequencing. The obtained results showed that for an accurate determination of the species, it is sufficient to know the sequence of two markers, ITS and COX1. The sensitivity of this test is sufficient to identify Phytophthora in a pure culture. Unfortunately, analysis based on a pyrosequencing platform does not provide enough data to simultaneous identify multiple Phytophthora species in samples collected in the field. This problem could be resolved in the future by sequencing using more efficient platforms like Illumina or IonTorrent.

Phytophthora species are best known as pathogens of agricultural crops (e.g. P. infestans), but there are also invasive pathogens destroying forest atands (e.g. P. ramorum in the USA) or even whole forest ecosystems (e.g. P. cinnamomi in Australia). Still, little is known about indiginous species, especially in wild ecosystems. Rhododendrons are well known as a reservuar for oomycetes’ development. The main objectives of the present study were to develop the new tool for the identification of pathogenic Phytophthora and Phytium based on DNA sequence analysis of the ITS1, 5.8S gene and ITS2 region. Rhododendrons leaves served as specific plant baits. In order to reach the goal the real time PCR, the nested PCR and the DNA sequencing of the rDNA ITS region were carried out. The genomic DNA was extracted form the symptomatic rhododendron leaves. Three distinct Oomycetes species: Phytophthora cactorum, Pythium mercuriale and em>Pythium recalcitrans were detected in rhododendron leaves and registered in GenBank.

Opis:
The study was conducted in 2015 in six spruce stands situated in different forest districts administratively belonging to the Regional Directorate of Forests State in Krosno. Each spruce population was represented by 30 trees and assessed in terms of their current health status. Genetic analyses were performed based on shoot samples from each tree using nine nuclear DNA markers and one mitochondrial DNA marker (nad1). The health status of the trees was described according to the classification developed by Szczepkowski and Tarasiuk (2005) and the correlation between health classes and the level of genetic variability was computed with STATISTICA (α = 0.05). Nuclear DNA analyses revealed a low level of genetic variability among spruce populations (only 3% of the total genetic variation (FST = 0.028) and a high variability within populations (97%). The total heterozygosity in all stands (HT) was calculated as 0.646. Based on UPGMA analysis, the most genetically similar populations are spruce stands in the Bieszczady National Park and the Ustrzyki Dolne Forest District, which have the smallest genetic divergence of all populations (DN = 0.0165). Our analysis of the mitochondrial gene nad1 revealed the presence of six different haplotypes “a”, “a1”, “b”, “c”, “d” and “d1”. Comprising 56% of all haplotypes, “a” was the most common showing a predominant impact on spruce migration from the Carpathian area. The analysis based on mitochondrial markers (by Nei) revealed a heterozygosity of 0.525. Based on the observations of disease symptoms, 29% of the trees belong to health class 1,30% to class 2,28% to class 3 and class 4 contains 13% of trees. The comparison between health status and the level of genetic variation in the analyzed stands showed a positive correlation. Spruce stands with better health were also characterized by a greater degree of genetic variability. Since most of the investigated spruce populations shared the mitochondrial haplotype “a”, we have ascertained their Hercynian- Carpathian origin. Only one stand (Cisna) had a high frequency (43.3%) of the Nordic haplotype “c” suggesting that this provenance is derived from the Baltic post-glacial refugium of P. abies in Europe.
