Opis:
We have constructed and tested a device for incubation of individual eggs of fish. We measured the diameter of fertilized and water hardened eggs of vendace (Coregonus albula). Each egg developed in a separate incubator, which was a modified Pasteur pipette. Incubation of 315 eggs resulted in 249 hatched embryos (eleutheroembryos). The technique enabled identification of each eleutheroembryo hatching from a given egg. Example of relationship between external diameter of egg and the length of eleutheroembryo showed potential applications of the technique in studies on fish reproduction biology.

Opis:
Highly variable microsatellite DNA loci show high levels of variation per locus and provide molecular markers for species and for populations of small effective size. In this study we applied microsatellite markers to identify specimens belonging to several sturgeon species and their interspecific hybrids. Nine microsatellite loci (Afu-19, Afu-22, Afu-34, Afu-39, Afu-54, Afu-57, Afu-58, Afu-68, Afu-69) were analysed for five species (beluga, Siberian sturgeon, Russian sturgeon, sterlet and paddlefish) and for their three interspecific hybrids: Siberian sturgeon x Russian sturgeon, beluga x sterlet (called bester) and for beluga x bester. Certain alleles at five loci: Afu-22, Afu-39, Afu-54, Afu-57 and Afu-68 were diagnostic for the examined sturgeon species and their interspecific hybrids.

Opis:
Gynogenetic northern pike (Esox lucius L.) were produced using UV irradiated sperm and heat shock applied to inseminated eggs shortly after gamete activation. Milt was diluted in immobilizing solution (1:9) and UV irradiated (6.4 W*m-2) for 2-20 min, with dosage in the range of 768-7680 J*m-2. Genetic inactivation of spermatozoa was most efficient when milt was irradiated for 8 min (3072 J*m-2). Insemination of eggs with irradiated milt yielded 100% haploid larvae with hatching rate at 72.1۪.8% (mean±SD), expressed as a percentage of inseminated eggs. Haploid embryo developed and most of them hatched (showing "haploid syndrome") but all haploid larvae died within 48 hours after hatching. After insemination with irradiated sperm the eggs were exposed to a thermal shock of 34°C or 34.5°C, lasting 3 or 5 min, applied 11-16 min after gamete activation. The efficiency of heat shock and survival in experimental groups significantly depended on the source (individual female effect) and quality of eggs. The time of application and duration of the heat shock also affected the survival of embryo and percentage of gynogenetic larvae. Gynogenetic larvae were obtained in all experimental groups with hatching rate as high as 24.2ۭ.6%.

The mutagenicity of two, newly synthesized amino-derivatives of cyclopenta[c]phenanthrene (CP[c]Ph) was investigated in the Ames test using TA98 and TA100 histidine dependent Salmonella typhimurium strains. Neither of the examined compounds showed mutagenic activity without metabolic activation. The incorporation of an activation system caused high mutagenicity of CP[c]Ph derivatives in both S. typhimurium strains. The results are compared with the previous data on the mutagenic activity of unsubstituted CP[c]Ph and possible mechanisms of activation are discussed.

Opis:
Coregonid fishes (coregonids) (Salmonidae, Coregoninae) of Siberia show immense morphological and ecological variability, what is reflected in their complex interspecific and intraspecific structure: i.e. occurrence of numerous species, forms and endemic species. Studies of the coregonids of Siberia is crucially important for understanding of their origin, taxonomy and evolution. We have analysed electrophoretically some 13 enzyme systems from over 20 samples of East-Siberian coregonids. This paper was intended as an introduction to a number of primary papers that are planned that will describe the biological and genetic characteristics of studied populations.

American paddlefish (Polyodon spathula) is a new species in Polish aquaculture, its broodstocks are few and small, and it is possible that all mature fish originated from only a few spawners. Studies on polymorphism of highly variable microsatellite DNA allow revealing genetic characteristics of individual spawners as well as estimation of genetic variation within and divergence between broodstocks. This paper describes optimised protocols for isolation of DNA from fin tissues, amplification of nine microsatellite loci using PCR technique, and for fish genotyping using automatic capillary DNA sequencer. Our technique was tested towards the fin samples taken from all paddlefish reared in Poland and approaching their sexual maturity; the study included also samples taken from 47 fish of the Ukrainian breeding center (Gorny Tykich).

The aim of this study was to compare physiological responses in Siberian sturgeon (Acipenser baeri) induced by propofol and CO2 anaesthesia. Two procedures were applied during the experiment. In procedure I, blood samples were collected immediately after exposure (1, 2, 5, 10 min) to the anaesthetic. In procedure II, fish were exposed to the anaesthetic for 10 minutes and then were moved to anaesthetic free water. Blood was sampled after 5, 10, 20 or 30 min of recovery time. Gasometrical and biochemical analyses were performed on collected blood. In CO2 anaesthetized fish strong hypercapnic acidosis was revealed. The drop of the HCO3-/CO2 ratio, from 28:1 in control fish up to 4:1 in CO2 anaesthetized ones, proved that the compensation mechanism is not capable of preventing acidosis during CO2 anaesthesia in Siberian sturgeon. In contrast, only moderate, respiratory acidosis occurred in sturgeons anaesthetised with propofol. Hypercapnic acidosis during CO2 anaesthesia was followed by a fourfold increase of ammonia level in the blood. Glucose level, increasing only during recovery time, indicates that a secondary stress response occurred when awareness of anaesthetized fish had been restored.
