Currently much attention is paid to the use of stem cells in transplantation. The reason for this interest are the characteristic regenerative capabilities of stem cells and their ability to regenerate and differentiate. Stem cells are becoming the basic unit in regenerative medicine and are increasingly used in transplantation. Currently used innovative methods during cell culture are also a field of activity for biotechnology, which deals with the improvement of methods and looks for new applications of stem cells. The work aims to familiarize the properties and applications of stem cells and present selected techniques for their culture for transplantation. Based on the collected data, it can be concluded that new sources of stem cells should be constantly sought. The improve and create new diagnostic tools and culture media, as well as construct systems that would allow stem cells to be cultured with right orientation.

Diabetic retinopathy as a complication of diabetes is one of the most common causes of vision loss. It is a dangerous disease, which affects more and more people. It is important to know its different causes, background and to generate new, effective therapeutic threatments. One of the possible causes of diabetic retinopathy is the accumulation of advanced glycation products and over-expression of AGEs receptors (RAGE) leading to tissue damage, oxidative stress formation, and the promotion of other changes and disorders. There are many genetic variants that encode the AGEs receptor (RAGE). Some polymorphisms (Gly82Ser, 1704G/T, 2184A/G, -429T/C, -374T/A) may be associated with diabetic retinopathy. This paper aims to systematize knowledge in this field and to show the relationship between these compounds and diabetic retinopathy.

Cataract is one of the main causes of vision loss. So far, it has been found that one of the causes of cataract formation is the accumulation of heavy metals in ocular tissues and environmental pollution. Research material consisted of blood from healthy volunteers (n = 81; Mogilno Lakeland) and those suffering from cataract (n = 90) from Ophthalmology Clinic, Bydgoszcz (2013, 2016). Samples were prepared in order to obtain plasma, which was used to analyze Fe, Cu, Zn, Mn, Pb, Cd, and Hg concentration (ICP-MS). The purpose of this study is to show the interaction of cataract with cadmium and lead during initial pro-antioxidative reactions and to study element-element interactions. To this end, we analyzed physiological mechanisms that aim to stop the destructive effects of toxic metals. We have shown that the action of zinc, copper and manganese is antioxidant. Concentration of these elements is higher (α = 0. 05, p < 0.05) in the cataract group than in the control. We found interactions Fe-Cu-Zn-Mn-Pb-Cd-Hg. Cd and Pb show a negative effect on the eye tissue and generate pathophysiological changes leading to lens opacity and influence the destabilization of pro-antioxidant reactions. Correlations of elements in the control (Cd-Zn: R = 0.240; Cd-Cu: R = 0.316) and in the sick group (Cd-Cu; R = 0.329) were significant. On the basis of our research we found that there are numerous relationships between the concentrations of chemical elements in the patients with cataracts and the controls. Significant role of antagonisms with cadmium and lead has been demonstrated; these elements contribute to the gradual development of cataract. A correlation between elements exhibiting pro-antioxidant activity in the interactions with cadmium and lead (p=0.000) can be a source of cataract changes. The differences in Cd and Pb concentration and their interactions with elements between the control and sick groups can be considered as a factor for the development of cataract.

Biotechnology is a relatively young interdisciplinary discipline of science. Thanks to her techniques, we can currently do a lot of research. Biotechnologists in cooperation with other scientists are trying to develop new standards that would become the future of treatment for many diseases, including cataracts. Today, cataracts are increasingly recognized as a civilization disease of the eyes. Unfortunately, if left untreated, it can lead to complete blindness. The etiology of this disease is complex and not fully understood. At present, its foundations are being sought to be able to diagnose her faster and to introduce appropriate treatment early. Literature data currently indicate that it can arise as a result of: age- related changes, genetic (congenital) conditions, inflammation, metabolic disorders, the effects of certain drugs, and complications of eye diseases. From the latest reports we learn that antioxidation can have positive effects when dissolving the protein directly affecting the formation of cataracts. Biotechnology has amazing opportunities to conduct research that can help bring closer the etiology of cataracts and start to treat the disease faster by which so many people in the world lose sight.

Currently, much attention is paid to the search for the etiology of many eye diseases. It has been shown that environmental pollution with heavy metals (cadmium, lead, mercury) is of significant importance. One of these diseases is cataract, i.e. clouding of the lens of the eye. It turns out that heavy metals are deposited in the tissues of the eyes, damaging them and accelerating the disease. This work is aimed at presenting the problem of the influence of heavy metals on cataracts. The study compares the concentration of heavy metals between the control group (81 healthy volunteers) and the group of cataract patients (72 sick). Plasma was used in the research and the ICP-MS method was used.

Opis:
The main element of genomic molecular analysis is isolation of DNA from biological material. DNA isolation is an important process of PCR methods. Efficency of PCR processes depends of purness and concentration of DNA. Many methods allows to izolate good quality DNA. There are many Purification Kits and methods of extraction genomic material. We show available techniques and methods of increasing efficiency of the process in our studies. Previous data shows that operating with kits needs optimization, whilst protocols do not have all information about isolation process. We concentrate on Epicentre MasterPure DNA Purification Kit, which is cheap, fast and allows to purification nucleic acids from various materials. We show detailed advantages and disadvantages of this set. Our main aim is to develop protocol to get optimal concentration (50-100 ng/μL) and pureness (A 260/280; 1.6-1.9) of DNA. We also should optimize the process of DNA isolation to obtain efficiency of about 100%. Optimized methodology of DNA isolation, with using MasterPure DNA Purification Kit allows to get good quality material in laboratory without specialistic equipment. This survey contains our assumptions of ways how we can influent on purification process according to previous data.
