It has been found that alpha-chymotrypsin inhibitor isolated from Ascaris lumbricoides suum acts embryotoxically on the Leghorn chicken embryo. An increase in embryo mortality with increasing dose of the inhibitor has been observed after its administration into the yolk sac on day 4, 8 or 13 of incubation. There is a linear interrelationship between the logarithm of the dose of alpha-chymotrypsin inhibitor and the mortality of chickens. The LD50 values for alpha-chymotrypsin inhibitor increased for injections performed at later stages of embryo development. A significant decrease of mean mass of chicks injected with alpha-chymotrypsin inhibitor in comparison with control groups was observed. There was a more frequent occurrence of developmental abnormalities and pathological changes in groups of hatched chicks which received the Ascaris inhibitor.

The objective of the study was the phenotypic characteristics of thymocytes and T-lymphocytes and the evaluation of T cell function in the course of experimental trichinellosis in mice. It was found that the cortical part of the thymus was significantly diminished and the expression of Thy, Ly1 and Ly2 antigens were decreased during the intestinal phase of the parasitic invasion. Simultaneously, the increase of Thy⁺ bearing cells was observed in peripheral lymphatic tissue (spleen and lymph nodes) during this and the next early muscular phase of the invasion of T. spiralis. The expression of L3T4 and Ly2 antigens was also higher than or at least the same as in the control animals. The results of the evaluation of T lymphocytes reactivity show that the parasitic invasion results in changes in cellular immunity as was demonstrated by GvH reactions. These changes are dependent on both the intensity of the parasitic invasion and the phase of infection. The possible mechanisms of the modulation (stimulation or inhibition) of host immunity were discussed.

Ascaris suum
Opis:
The development of Ascaris suum eggs in the culture containing prostaglandins (PGA, PGB, PGE and PGF) or inhibitors of their synthesis (indomethacin and acetylsalicylic acid) was studied. Exposure of eggs to indomethacin (0.2 and 0.5%) or acetylosalicylic acid (1 %) delayed the development. 0.5% acetylsalicylic acid didn't exert any effect. The development was also delayed by PGB, PGE, PGF and higher concentration of PGA. But lower concentration of PGA accelerated embryonic development.

Opis:
The risk factors of Toxocara can is infestation were assessed among population of patients from the Lublin region. Sera from 151 patients suspected of toxocarosis were tested on the presence of the specific antibodies by ELISA (Biokema-Bordier, Switzerland). Sera from 74 women and 77 men were collected (mean: 24 years; range: 0-74 years). Women sera were found positive in 27 cases (36.5%) and men in 34 cases (44.2%). Positive results were found in the following age groups: young people up to 15 years - 34 positive from 71, people from 16 to 30 years old - 7 from 33, people from 31 to 45 years old - 9 from 24 and people older than 45 years - 11 from 23. Patients suspected of ocular larva migrans were found serologically positive in 37cases from 98. Among patients with eosinophilia 8 positive cases were found from 17 (47.1%). Rural inhabitants were found positive in 41 cases from 73, whereas inhabitants of small towns in 17 cases from 55 and inhabitants of Lublin city (population nearly 400,000) in 3 cases from 23. This relationship was statistically significant (p<0.01). The difference between the group of living in a house (52% positive cases) compared to those living in flats (18% positive cases) was also statistically significant (p<0.001).

In the paper are presented entomopathogenic nematodes (EPN) as a vector of bacteria killing insects. Many abiotic and biotic factors influence the survival and the virulence of mentioned organisms. These biological agents of control are used in practice over twenty years. Knowledge about the relationships between EPN-insect-bacteria allow to increase efficacy of treatment used in order to limiting populations of pests. The paper presents a short review of literature data of titled subjects.

ELISA can be used to measure produced antibodies or Trichinella spp. antigens in the samples. They are detected with antibodies linked to an enzyme that reacts with a substrate and generate a colour reaction. The optical density (OD) of the reaction is measured spectrophotometrically. ELISA assays can be done in several different procedures called „direct”, "indirect", "sandwich", and "competition" ELISA. Since the 1970s, the studies have been done on improving or replacing direct methods of Trichinella diagnosis with serological methods based on the ELISA. When somatic antigens of L1 T. spiralis were used, the specificity of the ELISA was poor due to a high probability of cross-reactions with other pathogens. During the 1980s the specificity of the ELISA was improved by excretory-secretory (E/S) antigens obtained during Trichinella muscle larvae incubation in vitro. Recently a synthetic glycan antigen has been developed and the increasing of ELISA specificity and sensitivity was noticed. The sensitivity of the ELISA using an E/S antigen ranging from 93.1 to 99.2% but the specificity from 90.6 to 99.4%. The ELISA method is relatively simple to apply, reliable, readily standardized and provides an acceptable balance of sensitivity and specificity. But all modified procedures should be validated. In Poland, the studies on the usefulness of ELISA for antibodies detection against T. spiralis in pigs and wild animals are limited. Own ELISA procedure was prepared in Pathophysiology Lab. in W. Stefański Institute of Parasitology of PAS. ELISA was used to examine IgG level against L1 T. spiralis in pigs and wild boars serum samples. Of 1474 pig samples, only 12 were positive. Of 1880 wild boar samples only 14 were positive. The results of this study are comparable with performance obtained using commercial sets. The results showed the usefulness of ELISA for T. spiralis diagnosis in pigs and wild boars and confirmed the possibility of use the ELISA test for application in the slaughterhouse.

Background. Trichinellosis is a parasitic zoonosis transmitted to humans by consumption of raw or undercooked meat from animals infected by worms of the Trichinella genus. Every year seropositive cases are found among the human population and thus trichinellosis still remains an epidemiologically important disease in Poland. The usefulness of ELISA for anti-T. spiralis IgG detection in pigs is still limited by the nature of antigen. The objective in the present study was to compare the usefulness of excretory-secretory antigens of L1 T. spiralis for the serological detection of IgG antibodies in pigs. Material and methods. The antigens were prepared in different laboratories: Ag ES L1 T. spiralis (N) in Germany, Ag ES L1 T. spiralis (W) in Italy and Ag ES L1 T. spiralis in Poland. Conventional, Iberian pigs were infected with 200, 1000 and 20 000 muscle larvae of T. spiralis. Serum samples were obtained at 5 and 1 dbi (day before infection), and 5, 10, 15, 20, 25, 30, 40, 50, 60 dpi (day post infection) and screened for specific IgG antibodies to excretory-secretory L1 T. spiralis antigens. Serum samples were obtained from the EU project TRICHIPORSE. The cut-off value of ELISA was determined on serum samples from 248 Trichinella-free pigs from Poznań and Boża Wola, that were examined by artificial digestion. Results. In pigs infected with 200 L1 T. spiralis larvae, specific IgG were detectable from 50 dpi, when the Ag ES L1 T. spiralis (N) was used, whereas when Ag ES L1T. spiralis (W) and Ag ES L1 T. spiralis were used, the specific IgG were detectable from 40 dpi. In pigs infected with 1000 L1 T. spiralis larvae, specific IgG was observed from 30 dpi when Ag ES L1 T. spiralis (W) and Ag ES L1 T. spiralis were used, but when Ag ES L1 T. spiralis (N) was used specific IgG were detectable from 40 dpi. In the group infected with the highest dose of T. spiralis larvae, specific IgG were detectable from 30 dpi when Ag ES L1 T. spiralis (N) and Ag ES L1 T. spiralis (W) were used, whereas when Ag ES L1 T. spiralis was used specific IgG were detectable from 20 dpi. The results strongly indicated that in the examined pigs, the specific IgG response against T. spiralis infection is dose dependent. Furthermore, it was shown that the high infectious dose induced earlier increasing of specific IgG response. Statistical analysis revealed a significant positive correlation between OD values obtained in procedures based on the three antigens. The results were statistically repeatable for procedures and for single pigs (P<0.01).

Recent epidemiological reports have shown an increasing incidence of Echinococcus multilocularis infection among red foxes and inhabitants from Pomorskie, Warmińsko−Mazurskie and Podkarpackie Districts of Poland. The study describes the actual recommendations for a proper diagnosis and optimal treatment in patients infected with E. multilocularis in order to standardize diagnostic and therapeutic procedures for this severe parasitic disease by reference centres. Liver lesions caused by the parasite were visualised by ultrasonography and computerized tomography scan. The clinical diagnosis based on typical imaging findings was completed by Em2plus ELISA and Em16 and Em18 Western blot immunodiagnostic tests using specific antigens, and confirmed by positive histopathological and/or molecular examinations. Since 1992, forty−five cases of alveolar echinococcosis were registered in Poland. Professions related to forestry or picking−up mushrooms and blueberries were potential risk factors for an occurrence of the infection, especially in the northern region of the country. In the imaging techniques, the liver lesions were characterized by heterogenous, irregular masses with necrotic cavities, and clausters of calcifications located in the peripheral parts. The final diagnosis of alveococcosis was based on PAS−positive staining of lesions in histopathology and/or the detection of the parasite's DNA in liver sections by PCR. The patients were treated by radical surgery with concomitant long−term intensive chemotherapy with albendazole. Conclusions: Collaboration with physicians of various medical specialities is crucial for an early and more effective detection of alveococcosis in Poland. E. multilocularis infection should always be considered in the differential diagnosis of space−occupying lesions in the liver characterized by irregular heterogenous masses, suggesting slow tumor growth.
