Opis:
The aim of the study was to determine whether treatment with recommended doses of meloxicam or flunixin had an effect on the apoptosis of peripheral blood T lymphocytes in calves. The study was carried out on 4-5 months old calves (n = 24, 8 per group). Experimental animals were injected subcutaneously with a single dose of 0.5 mg . kg-1 of meloxicam or intravenously with 3 doses of 2.2 mg . kg-1 day-1 of flunixin. The non-treatment animals served as control. Blood samples were taken at day 0 and at days 1, 2, 3, 5, 7 and 14 after the first NSAIDs injection. Apoptosis was determined by flow cytometry using Annexin V-PE/7-AAD staining. The kinetic analysis of apoptosis in the total lymphocyte population, as well as in the CD4+ and CD8+ subsets did not reveal significant differences in the frequency of early apoptotic cells between control and experimental groups throughout the period studied. Although, 24 h after administration of the first dose of NSAIDs, late-stage apoptosis/necrosis was significantly increased in the total lymphocyte population (the meloxicam group), as well as in the CD4+ (the meloxicam group and the flunixin group) and CD8+ (the flunixin group) subsets of T cells. However, this disturbance was transient, relatively poorly expressed and, thus, unlikely to be of clinical significance. Our results indicate that the use of meloxicam or flunixin in accordance with the recommended dosage regimen in cattle do not have a clinically significant influence on apoptosis of peripheral blood T cells.

The influence of the some immunomodulators (PHA-P, TFX and dexamethasone) on the process of apoptosis, occurring in the course of trichinellosis in mice, has been studied. It has been found that PHA-P activates this process in the jejunum mucosa and prolongs it in the muscular inflammatory infiltration, whereas TFX has no influence and dexamethasone distinctly decreases the level of the apoptotic cells. The number of the intestinal trichinae on the successive days of infection was similar in all groups of animals, however, the number of the muscular larvae in the groups receiving immunostimulators was much lower and in the group treated with dexamethasone - a little higher than that in control. As in mice receiving PHA-P and TFX, the cellular inflammatory infiltration in the muscles was larger than that in control, and in the group to which dexamethasone was administrated - smaller, the authors think that it was extensiveness of the infiltration and not the level of the apoptotic cells that influenced the number of the outliving larvae.

Opis:
Photodynamic therapy (PDT) is a promising treatment method for non-oncological and oncological diseases. PDT requires the use of photosensitizer, light, and cell oxygen. A selective, cytotoxic effects can be achieved in cancer cells, thanks to formation of radical oxygen species (ROS). Recent data indicate, that PDT with aminolevulinic acid (ALA) is successful in treatment of colorectal cancer. However, anti-cancer effects PDT in vivo, can vary in PDT in vitro, due to low level of oxygenation of tumor cells. Experimental research confirms the role of cholesterol in oncogenesis and indicates, that in colorectal cancer the cholesterol synthesis is disrupted; among others lovastatin belong to inhibitors 3-hydroxy-3-methyl-glutharyl-coenzyme A (HMG-CoA) enzyme of cholesterol synthesis pathway. Anti-tumor potential of statins: antiproliferative and pro-apoptotic, were demonstrated in various tumors in vitro and in vivo. Colorectal cancer, which often has genes mutations that are related to programmed death cells, shows resistance to chemotherapy. The objective of the research was to investigate the effect of antiproliferative and cytotoxic lovastatin and ALA-PDT (in low dose) working separately and in combination method (lovastatin and PDT) − for apoptosis induction, in metastatic colon cell line SW620. Our finding showed, that lovastatin inhibits the growth of cancer cells, depending on the dose and incubation time. ALA-PDT in low dose works on cell cytotoxic, yet do not initiate apoptosis. Whereas, preincubation of cells with lovastatin (IC50) increases the cytotoxic effect after PDT and induces apoptosis in cancer cells SW620, as demonstrated using an assay of Annexin V for flow cytometry. Our research demonstrates for the first time, the effectiveness of ALA-PDT connection with an inhibitor of cholesterol synthesis pathway – lovastatin, in increasing the cytotoxicity and induction of apoptosis in colon cancer cells, that are resistant to chemotherapy.
Terapia fotodynamiczna (ang. photodynamic therapy, PDT) jest obiecującą metodą w leczeniu nowotworowych i nienowotworowych schorzeń. Działanie PDT wymaga użycia fotouczulacza, światła i tlenu komórkowego, dla uzyskania wybiórczego cytotoksycznego efektu w komórkach patologicznych. Efekt ten jest wynikiem powstania wolnych rodników (ROS). Badania pokazują, że PDT z kwasem aminolewulinowym (ALA) jest skuteczna w leczeniu raka jelita grubego. Jednak efekty PDT in vivo, ze względu na niski poziom oksygenacji guza, mogą odbiegać od działania PDT in vitro. Dane eksperymentalne potwierdzają rolę cholesterolu w procesie onkogenezy i wskazują na zaburzony szlak syntezy cholesterolu, m.in. w raku jelita grubego. Statyny, np. lowastatyna, należą do inhibitorów 3-hydroksy-3-metylo-glutarylo-koenzymu A (HMG-CoA) − enzymu szlaku syntezy cholesterolu. Potencjał antynowotworowy statyn, antyproliferacyjny i proapoptotyczny, wykazano w nowotworach in vitro i in vivo. Rak jelita grubego, cechujący się mutacjami genów związanych z apoptozą, wykazuje oporność na standardowe chemioterapeutyki. Celem pracy było zbadanie efektu antyproliferacyjnego i cytotoksycznego lowastatyny i ALA-PDT (w niskiej dawce) działających osobno, oraz w metodzie połączonej (lowastatyna i PDT) – na indukcję apoptozy w przerzutowej linii raka jelita grubego SW620. Wyniki pokazały, że lowastatyna hamuje wzrost komórek w związku zależnym od dawki i czasu inkubacji. ALA-PDT w niskiej dawce działa cytotoksycznie, ale nie inicjuje apoptozy. Natomiast preinkubacja komórek raka z lowastatyną (IC50) zwiększa efekt cytotoksyczny po PDT i indukuje apoptozę w komórkach, co wykazano w teście z aneksyną V na cytometrze przepływowym. Nasze wyniki po raz pierwszy demonstrują skuteczność metody łączonej: ALA-PDT i inhibitora syntezy szlaku cholesterolu – lowastatyny, w zwiększeniu cytotoksyczności i indukcji apoptozy w komórkach raka jelita grubego, opornego na chemioterapię.

Blood platelet proteome of hemodialyzed uremic patients exhibits significant difference in comparison to the blood platelet proteome of healthy subjects. This alteration is manifested by the presence of high concentrations of low molecular peptides within the whole range of pI. Increased platelet apoptosis has been put forward as a possible cause of this phenomenon (1). The aim of the present research was to assess whether blood platelet populations from hemodialyzed uremic patients exhibit more binding sites for Annexin V (a marker of apoptosis) than control samples from healthy donors. Blood was obtained from uremic patients immediately before and after hemodialysis. At the same time samples from control healthy donors were also collected. Blood was anticoagulated with sodium citrate and was immediately exposed to propidium iodide, fluorescent labeled Annexin V and CD61 antibodies. The samples were incubated for 10 minutes in the dark and next the labeled samples were processed in a BectonDickinson FACScan flow cytofluorymeter. Our preliminary study was performed for 12 hemodialyzed patients, 13nondialyzed uremic patientsand 12 controls. It was found that the blood platelet population of hemodialyzed patients exhibited significantly higher level of fluorescence intensity attributed to Annexin V. Furthermore, this intensity was comparable before and after hemodialysis and was independent on patient age. The results support the hypothesis that blood platelet contact with artificial surfaces during the process of hemodialysys may be partially responsible for triggering blood platelet apoptosis.

Endoplasmic Reticulum (ER) is an organelle that is vital for cell growth and maintenance of homeostasis. Recent studies have reported that numerous human diseases, including cancer, are strictly connected to disruption of ER homeostasis. In order to counteract adverse intracellular conditions, cancer cells induce protein kinase R (PKR)-like endoplasmic reticulum kinase (PERK)-dependent, pro-adaptive unfolded protein response (UPR) signaling branches. If ER stress is severe or prolonged, pro-adaptive signaling networks are insufficient, resulting in apoptotic cell death of cancer cells. The main aim: of the study was to evaluate the biological activity of a small-molecule PERK inhibitor GSK2606414 in two cancer cell lines - human neuroblastoma (SH-SY5Y) and human colorectal adenocarcinoma (HT-29) cell lines. We analyzed the level of phosphorylation of the eukaryotic initiation factor 2 (eIF2), which is the main substrate of PERK and a subsequent activator of UPR, which under long-term ER stress may evoke apoptotic death of cancer cells. Material and methods: In the study, we utilized commercially available cell lines of human colorectal adenocarcinoma HT-29 and human neuroblastoma SH-SY5Y. Cells were exposed to the tested PERK-dependent signaling inhibitor GSK2606414 in suitable culture media with addition of thapsigargin (500 nM) to induce ER stress. To identify the protein, Western blot with specific antibodies was used. Detection of immune complexes was performed using chemiluminescence. Results: We found a complete inhibition of p-eIF2α expression due to the GSK2606414 inhibitor in both cell lines, SH-SY5Y and HT-29. Conclusions: Currently available cancer treatments are insufficient and cause various side effects. It has been assumed that utilization of small-molecule inhibitors of the PERK-dependent signaling pathway, like GSK2606414, may switch the pro-adaptive branch of UPR to its pro-apoptotic branch. It is believed that the tested inhibitor GSK2606414 may become a promising treatment for many cancer types.

Introduction: Despite the progress in gaining knowledge about carcinogenesis, it is still unclear what processes are directly responsible for the differentiation of gastric cancer into its intestinal or diffuse form. Dividing of these two forms is based on one of the oldest, yet still commonly used classifications – the classification of Lauren. There are many factors that may influence the formation of gastric tumors of various aggressiveness. Purpose: To evaluate the expression of proteins: fragile histidine triad (FHIT), E-cadherin, α-catenin, γ-catenin, cathepsin B, epidermal growth factor (EGF), HER-2, MMP-9, MCM-2, Bak, Bax, BID, Bcl-XL, p53, FasL, Bcl-2, caspase-8, procaspase-3 in gastric cancer cells, depending on the type of tumor by Lauren classification. Materials and methods: Study group consisted of 91 patients treated surgically for gastric cancer in the Second Department of General and Gastroenterological Surgery, Medical University of Bialystok in years between 2000 and 2006. Results: It is shown, that the expression of E-cadherin was significantly higher in the Lauren I gastric cancer cells than in Lauren II. In case of caspase-8 there has been significantly less frequent expression of this protein in Lauren I gastric cancer cells compared to Lauren II. The authors describe no statistically significant differences in the expression of other proteins taken into consideration. Conclusions: These results suggest the role of adhesion and apoptosis-related proteins in the development of two different types of gastric cancer according to Lauren’s classification.
