Opis:
The obligate oomycete Sclerospora graminicola (Sacc.) Schroet, is the incitant of downy mildew disease, which is the main constraint in pearl millet production worldwide. Different elicitors from Trichoderma hamatum UOM 13, e.g. mycelial extract and cell wall glucans, were assessed for their resistance elicitation efficiency and the possible underlying mechanisms. Both mycelial extract and cell wall glucans of T. hamatum UOM 13 positively influenced seed quality parameters of pearl millet, significantly enhanced seed germination and seedling vigor in comparison to the untreated control. Seed priming with cell wall glucan elicitors of T. hamatum UOM 13 suppressed downy mildew on susceptible pearl millet seedlings under greenhouse conditions by induction of systemic host resistance. Of the different elicitor delivery methods tested, transplant root dip was more effective than seed treatment and foliar spray. A combination of transplant root dip + seed treatment + foliar spray was significantly more effective than the single delivery methods. The induced resistance corresponded to up regulation of genes of important defense proteins upon pathogen inoculation. Transcripts of genes of defense enzymes glucanase, phenylalanine ammonia lyase, peroxidase and polyphenoloxidase were significantly increased due to the T. hamatum UOM elicitor effect. Expression of hydroxyproline-rich glycoprotein genes, known to play an important role in cell wall cross-linking, were also up regulated in response to T. hamatum UOM cell wall glucan treatment. This study emphasizes the role of T. hamatum UOM as a potential elicitor of downy mildew resistance in pearl millet and presents novel insights into the involvement of important defense proteins mediating such as resistance trigger.

Opis:
Fusarium graminearum and F. culmorum are the causal agents of Fusarium head blight (FHB) in cereal crops worldwide. Application of resistant cultivars is the most effective and economic method for management of FHB and reducing mycotoxin production in wheat. Understanding the physiological and biochemical mechanisms involved in basal resistance of wheat to FHB disease is limited. In this research, after screening resistance levels of eighteen wheat cultivars planted in Iran, Gaskozhen and Falat were identified as partially resistant and susceptible wheat cultivars against Fusarium spp., respectively. Also, we investigated the role of hydroxyl radical (OH−), nitric oxide (NO), callose deposition, lipid peroxidation and protein content in basal resistance of wheat to the hemi-biotrophic and necrotrophic Fusarium species causing FHB. Nitric oxide as a signaling molecule may be involved in physiological and defensive processes in plants. Our results showed that NO generation increased in seedlings and spikes of wheat cultivars after inoculation with Fusarium species. We observed earlier and stronger callose deposition at early time points after infection by Fusarium spp. isolates than in non-infected plants, which was positively related to the resistance levels in wheat cultivars. Higher levels of OH− and malondialdehyde (MDA) accumulation (as a marker of lipid peroxidation) were observed in the Falat than in the Gaskozhen cultivar, under non-infected and infected conditions. So, estimation of lipid peroxidation could be useful to evaluate cultivars’ susceptibility. These findings can provide novel insights for better recognition of physiological and biochemical markers of FHB resistance, which could be used for rapid screening of resistance levels in wheat cultivars against this destructive fungal disease.

Opis:
Lupine diseases caused by pathogenic fungi constitute a serious problem in agriculture. They lead to partial yield loss and deterioration of crop quality through the changes in biochemical composition of seeds or their contamination with mycotoxins. Some of common lupine diseases are fusarioses caused by Fusarium oxyspomm f. sp. lupini. Morphometric and metabolic changes were investigated in yellow lupine seedlings infected with F. oxyspomm f. sp. lupini. It was found that infection caused temporary inhibition of seedling growth, overcome at later development, and activation of ß-glycosidase and peroxidases. The changes in enzymes activity indicate the induction of defense mechanism against F. oxyspomm f. sp. lupini and inhibition of pathogen spread.
Choroby łubinu powodowane przez grzyby chorobotwórcze stanowią poważny problem w rolnictwie. Prowadzą do zniszczenia części plonu oraz pogorszenia jego jakości, poprzez zmiany w składzie biochemicznym nasion lub zakażenie ich mykotoksynami. Do powszechnie występujących chorób łubinu zaliczane są fuzariozy powodowane przez Fusarium oxysporum f. sp. lupini. Badano zmiany morfologiczne oraz metaboliczne w siewkach łubinu żółtego infekowanych F. oxysporum f. sp. lupini. Stwierdzono, że infekcja spowodowała okresowe zahamowanie wzrostu siewek, przełamane w dalszej fazie ich rozwoju oraz aktywację ß-glikozydazy oraz peroksydaz. Poinfekcyjne zmiany aktywności enzymów wskazują na uruchamianie mechanizmu obronnego względem F. oxysporum f. sp. lupini i zahamowanie rozprzestrzeniania się patogena.
