In this study we report the usefulness of nested PCR for screening of the persistent B. microti infections in rodent hosts. Female BALB/c mice were inoculated with 100 μl of donor blood infected with B. microti. Infections were detected using microscopic examination of Giemsa-stained blood smears. To determine whether B. microti DNA was present in blood and/or spleen tissue, nested PCR was performed targeting a specifi c fragment of the gene encoding the 18S rRNA. Blood was sampled every 10 days post-infection (dpi) until day 30, after which mice were sampled every 30 days until the end of experiment at 360 dpi. The most extensive parasitaemia (39% of infected erythrocytes) was observed at 10 dpi. Between 20-60 dpi, less then 1% of infected erythrocytes were detected in blood smears, and from 90 dpi onwards, infected erythrocytes were no longer observed. B. microti DNA was successfully amplifi ed from the blood of mice from 10 dpi until 180 dpi, as well as from spleens of infected mice at 10 and 20 dpi. The presented results show that nested PCR is the method of choice for monitoring infections of B. microti in the blood of rodent hosts, and could therefore be a tool for environmental monitoring of naturally infected rodents which are the predominant source of infection for tick vectors.

Opis:
Administration intraperitoneally of the Ascaris α-chymotrypsin inhibitor (40-300 mg/kg/day.) at a late stage of organogenesis (8-12 days of gestation) disturbed course of mouse pregnancy. The low doses of α-chymotrypsin inhibitor (40-80 mg/kg/day) significantly decreased the number of live fetuses per litter, increased the number of fetal resorptions. The symptoms of matemal toxicity that occurred after administration of the highest doses of the inhibitor (80-300 mg/kg/day) to pregnant mice included: decreased body weight gain as compared to control, vaginal hemorrhage, intrauterine resorption of litters, abortions, altered behaviour of animals immediately after injection and death. There is a linear interrelationship between the logarithm of the doses of the inhibitor and mortality of pregnant mice. The DL50 value of the inhibitor for female was 116 mg/kg /day (confidence interval: 95.5-140.0 mg/kg/day).

The influence of the some immunomodulators (PHA-P, TFX and dexamethasone) on the process of apoptosis, occurring in the course of trichinellosis in mice, has been studied. It has been found that PHA-P activates this process in the jejunum mucosa and prolongs it in the muscular inflammatory infiltration, whereas TFX has no influence and dexamethasone distinctly decreases the level of the apoptotic cells. The number of the intestinal trichinae on the successive days of infection was similar in all groups of animals, however, the number of the muscular larvae in the groups receiving immunostimulators was much lower and in the group treated with dexamethasone - a little higher than that in control. As in mice receiving PHA-P and TFX, the cellular inflammatory infiltration in the muscles was larger than that in control, and in the group to which dexamethasone was administrated - smaller, the authors think that it was extensiveness of the infiltration and not the level of the apoptotic cells that influenced the number of the outliving larvae.

Rodents, as mice and rats are the most common laboratory animals used in research and testing. They are seldom investigated for autochthonous ecto- and endoparasites prior their utilization in the experiments. Helminth parasites can alter the interpretation of final results. Pinworms commonly infecting laboratory rodents include mainly the mice pinworms Syphacia obvelata and Aspiculuris tetraptera, and in rats Syphacia muris. The fact that many laboratory rodent colonies were found to be parasite contaminated suggests a need for eradication and improvment of the quality of laboratory rodents. This review reports the data on the presence of helminth parasites in laboratory rodents colonies, and suggests to pay special attention on controlling the sanitary conditions of animal houses.

Studies were undertaken to evaluate by parasitological techniques the course of intestinal and muscular invasions in mice infected with Trichinella spiralis and treated with TFX-Thymomodulin (TFX-Th). Studies were conducted on 120 mice of BALB/c and SWISS strains, infected each with a mean invasive dose of 300 T. spiralis larvae. TFX-Th (Thymoorgan GmbH Pharmazie Co. KG, Vienenburg, Germany) was administered subcutaneously at 30 mg/kg body weight. Depending upon duration of TFX-Th administration and stages of intestinal and muscular invasions, three experiments were distinguished in the study. The results indicated that TFX-Th promoted eradication of larvae from muscles, whether administered at the early of the late stage of T. spiralis invasion.

Opis:
The effect of adriamycin on the behaviour of CD4⁺ and CD8⁺ cells in the course of trichinellosis in mice has been studied. The animals infected with 200 larvae per mouse were administered intraperitoneally adriamycin (Adriblastin from Farmitalia) at 1st and 28th day post infection (dpi), in a dosis of 0.2 mg. The mice were killed weakly for 6 weeks and then at the 60th dpi. The examinations were made on histologic sections from the spleen, mesenteric lymph node, jejunum and masseter muscle using immunofluorescent and immunoenzymatic methods with monoclonal antibody. The mice receiving adriamycin exhibited more CD8⁺ cells in the intestinal mucosa and by the end of the experiment also in the muscles in comparison with the control animals, which, however, did not affect the course of the infection.

Opis:
A variety of Acanthamoeba spp. pathogenic strains were isolated from water pools in the area of Poznań. In many cases amoebae invaded lugs, kidney heart, liver and diaphragm causing significant changes in these Organs. The presence of amoeba outside of CNS was found mainly in the early periods of infection. Amoeba isolated from different organs of dead animals did not exhibit any specificity in relation to the organs in following infections. Infection with both low-and high-virulent strains resulted in pathomorphological changes of similar character. However, changes induced by low-virulent parasites developed later and were less extensive.
