The objectives of this study were to isolate and characterize endophytic bacteria from sugar beet roots focusing on their ability to produce indole acetic acid (IAA). In order to isolate endophytic bacteria the sugar beet roots were used. To determine the amounts of IAA produced by endophytic Bacillus strains (B. amyloliquefaciens, B. megaterium and B. subtilis), a colorimetric technique was applied with Salkowski reagent. The isolates were grown in Laurin Broth medium supplemented with L-tryptophan over the concentration range of 100–10000 g/cm3 and incubated at 30 oC for 7 days. The highest concentration of IAA was recorded after 4 days of culturing in the supernatant obtained from the media containing 10000 g/cm3 of tryptophan. For the strain B. subtilis and B. megaterium the concentration of IAA marked in the post –culturing liquid amounted about 82.00 g/cm3, and for the B. amyloliquefaciens strain it amounted over 121.28 g/cm3. The strains Bacillus under study produced IAA in a different amount in the presence of L-tryptophan and in its absence.
Celem pracy było wyizolowanie i scharakteryzowanie bakterii endofitycznych korzeni buraka cukrowego pod kątem ich zdolności do produkcji kwasu indolilooctowego (IAA). Materiał doświadczalny do izolacji bakterii endofitycznych stanowiły korzenie burak cukrowego. Ilość wytworzonego IAA przez endofityczne szczepy Bacillus (B. amyloliquefaciens, B. megaterium, B. subtilis) oznaczono metodą kolorymetryczną z odczynnikiem Salkowskiego. Szczepy hodowano w pożywce LB (Laurin Broth) suplementowanej L-tryptofanem w stężeniach 100–10000 g/cm3 i inkubowano w 30 oC przez 7 dni. Najwyższe stężenie IAA odnotowano po 4 dniach w hodowlach zawierających 10000 g/cm3 L-tryptofanu. Dla szczepu B. subtilis i B. megaterium stężenie IAA wynosiło około 82,00 g/cm3, a dla szczepu B. amyloliquefaciens 127,28 g/cm3. Badane szczepy Bacillus spp. wytwarzały IAA na różnym poziomie zarówno w obecności L-tryptofanu jak i przy jego braku.

Tuberculosis is one of the common causes of death worldwide, and according to the World Health Organization, in 2015, about 10.4 mln people globally is suffering from that disease. Despite great development in medicine and discovery of many antimycobacterial drugs, tuberculosis remains an unsolved problem. The populations of countries in Africa, Asia and South America have difficult access to hygiene, medical care and medicines, and they therefore resort to consulting traditional practitioners who mainly use drugs from natural sources which they prepare themselves. Plants are the biggest source of such medicines and are used as extracts, decoctions or powders, often without any evidence of their actual activity. Isolates from plants sometimes contain unknown substances or some second metabolites of microorganisms. In vitro studies have shown that some of these medicines have effective antimycobacterial properties, even against clinical, resistant strains. This is hopeful in the situation where more and more cases of tuberculosis prove to be resistant to standard drugs. Extracts of medicinal plants are often synergistic with synthetic drugs. Substances with insufficient MIC can be a basis for derivatives which have better properties. This thesis summarizes the current international knowledge of the use of plant substances used in the treatment of tuberculosis that could be potential new antituberculous drugs.

Introduction and aim. Viral infections stand to be among the most devastating diseases globally. Though significant efforts have been made in research and drug development against viral infections, the search for safe, affordable and effective vaccines against the current ravaging severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is still on. This is because already approved vaccines still need improvement. This review draws the attention of researchers on the potentials of bioactive substances from endophytes against the novel coronaviruses. Material and methods. This assessment was made using references of articles published in English peer reviewed journals indexed in PubMed and Google Scholars databases up to June, 2022. The following key words were used; ‘coronaviruses’, ‘Endophytes’, ‘Endophytes and viral infections, ‘Endophytes and COVID-19, ‘SARS-CoV’. Analysis of the literature. In-silico, in-vitro and in-vivo studies revealed that natural compounds from endophytes showed antiviral activities against various human coronavirus, including HCoV 229E and a norovirus surrogate, the feline coronavirus FCV F9, COVID-19, Coronavirus 2 (SARS-CoV-2), SARSCoV-2 Mpro, among others. Conclusion. This finding calls for researchers to also focus on endophytes, as part of drugs development in the bid to finding possible solution in combating the devastating COVID-19, an emerging situation.

Opis:
Endophytes are organisms that live within the plant tissue without usually causing any symptoms. In plants of natural ecosystems, endophytic fungi are in fact ubiquitous. This review summarizes research carried out on their biology emphasizing their functionality in terms of the host range, the colonization extent, the way of transmission between hosts and their influence on host fitness. The main focus will be on two classes of fungal endophytes, class 2 and 4 (Dark Septate Fungi), due to their potential for practical application in forestry. Raising awareness of the potential of endophytes to enhance the host’s resistance to pathogens, insects and anthropogenic disturbances is a key factor in developing applications for forest management.

The objective of this study was to explore the possible improvement of plant growth using the activity of the bacterial enzyme ACC (1-aminocyclopropane-1-carboxylate) deaminase (endophytes and rhizobacteria). The beneficial effect of ACC deaminase activity was tested on the plants growing under stress conditions (high concentrations of heavy metals: cadmium, lead, zinc in the soil). The bacteria were isolated from three plants species: Festuca rubra L., Agrostis capillaris L., Arabidopsis thaliana L. Heynh, acquired from the area contaminated with heavy metals. The strains with the highest ACC deaminase activity were used to prepare a bacterial consortium and inoculate the plants. It has been shown that inoculation of plants with ACC producing bacteria has a positive effect on their growth under stress conditions. The bacterial entophytes strains showed a higher activity of ACC deaminase, which resulted in a higher biomass growth of inoculated plants. The PGPB bacteria may limit the toxicity of harmful ions and thus the increase the adaptive properties of plants. Moreover, it was discovered that the bacteria mainly belonging to genus Bacillus and Pseudomonas had the highest ACC deaminase activity in the environment contaminated with multiple heavy metals. The use of selected microorganisms and plants will provide results in an increasing efficiency of phytoremediation.

Endophytes are an important constituent of sustainable agriculture because of their ability to produce a large number of agriculturally important metabolites. A salt-tolerant fluorescent green pigment-producing endophytic bacterium was isolated on 2.5% NaCl-supplemented nutrient agar from the leaf samples of Zanthoxylum alatum Roxb. The isolate Z1B4 was identified as Pseudomonas fluorescens based on morphological features, fatty acid methyl ester analysis, biochemical tests, and 16S rRNA gene sequencing. P. fluorescens Z1B4 showed positive results for tricalcium phosphate solubilization; 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity; and production of auxins, siderophores, hydrogen cyanide, and ammonia. P. fluorescens Z1B4 also showed strong antagonistic activity against Curvularia lunata (MTCC 283), Fusarium verticillioides (MTCC 3322), and Alternaria alternata (MTCC 1362) and exhibited stress tolerance to a wide range of temperature and pH and concentrations of NaCl and calcium salts. Under natural conditions, following inoculation with the isolate Z1B4, a significant increase in the growth of pea and maize test plants in pots was observed compared to that of uninoculated control plants. The rifampicin-resistant mutant Z1B4Rif was recovered from the roots, shoots, and leaves of the test plants, indicating that the isolated endophytic bacterium can grow well within different plant tissues. The present study indicated that the endophytic bacterium P. fluorescens Z1B4 can be used as a bacterial inoculant in stressed environments for sustainable agriculture.
