Opis:
DNA microarray technology is a powerful tool in functional genomics study of many organisms, the forest−trees included. This method allow to examine simultaneously the changes in expression of thousands of genes and it is based on specific hybridization of cDNA probes from an organism with the DNA library immobilized in an array. The power of this method consists in miniaturization, automation and parallel study of large−scale genome from multiple samples. In forest science, the microarray technology has already been applied in some study of gene−expression in Populus, Pinus and Picea species and the number of new reports is still increasing every year. Since far, some gene−expression have been studied among woody plants in regard of development and growth processes (xylem, adventious−root and zygotic embryo formation, flowering, ripening, shooting of leaves), resistance mechanisms against biotic (fungi, viruses) and abiotic factors (drought, NaCl, elevated CO2 and O3 concentrations). Many practical applications of the microarray technique may concern the early selection in nursery of trees for morphologically valuable traits, the sustainable forest regeneration and the production of genetically transformed species for the chosen trait.

Beech decline phenomenon has been observed in Poland since the 1980s. The reasons are still unclear but the discovery of pathogens genus Phytophthora shed a new light on it. Stems as well as roots of beech trees were severely affected by P. citricola and P. cambivora. In order to find out the genetic background of infection, three loci of chloroplast microsatellite DNA were investigated in the genetic differentiation study of beeches growing in the Siewierz Forest District in Poland. Parameters of genetic diversity (h Nei) and differentiation (GST) in the chloroplast genome were estimated and compared between healthy and damaged trees. Healthy beeches were more heterozygous (h=0.243) than the damaged ones (h=0.113), and the distribution of cpDNA alleles was different between these two groups. Healthy trees were nearly 50% more genetically diversed than declining ones.

Opis:
The genetic variability and biodiversity of tree populations ensure the stability and sustainability of forest ecosystems. New research tools based on molecular DNA markers enable precise characterisation of forest genetic resources, i.e. detection of different allele frequencies in mature trees and progeny populations. The paper describes the genetic structure of mature stands of Scots pine (Pinus sylvestris L.) in Oława Forest District and Norway spruce (Picea abies L. Karst.) in Jawor Forest District and stands of their respective progeny. In the Scots pine stand, there was a slight increase (0.6%) in heterozygosity level and a larger increase (4.9%) in the inbreeding coefficient of progeny trees. In the Norway spruce stand, despite a small reduction (0.9%) in heterozygosity, a similar increase (4.6%) in the inbreeding coefficient of progeny was revealed. In both stands, allele richness and the partition probability of basic clustering were high. Both pine and spruce adults and progeny trees were characterised by high levels of genetic similarity (96% and 79%, respectively). Gene flow between the mature and progeny populations was high (Nm > 1) for both Scots pine and Norway spruce. Conservation of the gene pool within forest tree stands requires an increase in the proportion of natural regeneration. To estimate the extent to which genes are transmitted between adult trees and their progeny, more studies are needed, especially taking into account the influence of silviculture measures, like selective tree cutting, on the genetic variability of the younger generation. These results confirm that the gene pool was conserved when transmitted between the stands studied, as well as highlight the usefulness of such a study for silvicultural purposes.

The genetic variability of sessile oak (Quercus petraea L.) mature stand and its natural progeny was investigated. Comparison between genetic structure of parental and progeny trees was based on frequencies of nuclear microsatellite (SSR) alleles occurring in three DNA loci. A slight (4%) increase of gene pool between oak mature and progeny trees was revealed by heterozygosity level estimation, maintaining 86.3% of genetic similarity between generations. Also allele richness, partition probability of basic clustering and inbreed coefficient proved the high genetic similarity between parental and progeny of investigated oak trees. The gene flow occurred within the stands as far as rare alleles were transmitted or new ones appeared in the progenies. The results highlight the necessity of such a study for silvicultural measures taken in order to proceed natural or artificial regeneration in forest tree stand.

The study showed the genetic structure of nine Schrenk spruce stands, which represented altitude and geographical variants in the Tien−Shan mountains in Kyrgyzstan. Comparison between genetic structure of stands was based on frequencies of nuclear microsatellite (SSR) alleles occurring in three DNA loci. The total genetic differentiation of Schrenk spruce populations was low (FST=0.0651). Eight main groups of populations were distinguished in the dendrogram defined by Nei's genetic distances based on microsatellite markers.

Opis:
In forestry management, artificially produced planting material is mainly used for renewal the tree population. Seedlings are cultivated in two systems: in the ground (the bare−root seedlings) and in controlled conditions (container seedlings). The aim of the study was to analyse the microsatellite markers of nuclear and chloroplast DNA, in terms of the number and frequency of rare, private, low frequency and common alleles in the planting material of Scots pine. The rare alleles included alleles occurring with less than 1% in analyzed group of seedlings and low frequency alleles occurred with a frequency of less than 25%. The private alleles were detected only in one group of seedlings. Genetic pools of seedlings from traditional (soil) and container production were compared. Planting material came from nurseries in the Olsztynek (N Poland) and the Oleszyce (S Poland) forest district. With the similar number of observed nDNA and cpDNA alleles in both analyzed locations, a higher number of rare, low frequency and private alleles was found within container seedlings. Most private alleles were a rare allele. Rare and private alleles are supposed to be responsible for adaptation to changing climatic conditions and a stressful environment. It seems reasonable to continue research on the meaning of rare and private alleles under conditions of strong selective pressure.

Opis:
Oak decline phenomenon has been observed in Poland in the 1980s. Especially, Krotoszyn Forest District was affected in the years 1982−1984, when oaks were dying in a mass extend causing economic and ecological problems. Twenty five years later many Phytophthora species were isolated from rhizosphere soil suggesting their important role in fine root damage. This research was focused on application of fertilisers increasing oak resistance to soil−borne pathogens. In total, 60 soil samples were collected c.a. 1 meter from trunks of oaks representing control and treated variant with Actifos® and Kalex® fertilisers. The fine root parameters (smaller than 2 mm) were evaluated with the water scanner and WinRhizo software. The same soil samples were analysed for the presence of macro− (N, P, K, Ca, Mg) and microelements (Al, Cu, Fe, Mn, Na, Zn). The use of ammonium and potassium phosphites in the form of foliar spray had a beneficial effect on the health of oak stands (Quercus robur L.) in the Krotoszyn Forest District. Three parameters: defoliation (ICP Forest), vitality according to Roloff method and synthetic index of damage were applied in order to evaluate the status of oaks health. Two aforementioned crown parameters were obtained from visual assessment from the ground, while the synthetic index of damage was calculated according to the formula developed by Dmyterko and Bruchwald. The parameters of fine roots after treatments, especially their number, length and surface improved. Trees with better quality fine roots had more chance to survive in unfavourable environmental conditions like drought. Phosphite treatments of oak stand positively influenced the chemical properties of soil, decreased acidity, but increased availability of Zn for plants, and probably reduced root damage by toxic aluminium ions. An improvement in the health of fine roots (that were saved by phosphites from damage caused by pathogenic species of Phytophthora genus present in the soil) will have a positive effect on the reconstruction of crown architecture (shoots development from dormant buds).

The study was conducted in 2015 in six spruce stands situated in different forest districts administratively belonging to the Regional Directorate of Forests State in Krosno. Each spruce population was represented by 30 trees and assessed in terms of their current health status. Genetic analyses were performed based on shoot samples from each tree using nine nuclear DNA markers and one mitochondrial DNA marker (nad1). The health status of the trees was described according to the classification developed by Szczepkowski and Tarasiuk (2005) and the correlation between health classes and the level of genetic variability was computed with STATISTICA (α = 0.05). Nuclear DNA analyses revealed a low level of genetic variability among spruce populations (only 3% of the total genetic variation (FST = 0.028) and a high variability within populations (97%). The total heterozygosity in all stands (HT) was calculated as 0.646. Based on UPGMA analysis, the most genetically similar populations are spruce stands in the Bieszczady National Park and the Ustrzyki Dolne Forest District, which have the smallest genetic divergence of all populations (DN = 0.0165). Our analysis of the mitochondrial gene nad1 revealed the presence of six different haplotypes “a”, “a1”, “b”, “c”, “d” and “d1”. Comprising 56% of all haplotypes, “a” was the most common showing a predominant impact on spruce migration from the Carpathian area. The analysis based on mitochondrial markers (by Nei) revealed a heterozygosity of 0.525. Based on the observations of disease symptoms, 29% of the trees belong to health class 1,30% to class 2,28% to class 3 and class 4 contains 13% of trees. The comparison between health status and the level of genetic variation in the analyzed stands showed a positive correlation. Spruce stands with better health were also characterized by a greater degree of genetic variability. Since most of the investigated spruce populations shared the mitochondrial haplotype “a”, we have ascertained their Hercynian- Carpathian origin. Only one stand (Cisna) had a high frequency (43.3%) of the Nordic haplotype “c” suggesting that this provenance is derived from the Baltic post-glacial refugium of P. abies in Europe.

Opis:
Considered to be the most natural of all other Polish stands Picea abies in Białowieża Forest has suffered severely of bark beetle damages for decades. In order to find out some historical events in native Norway spruce stands diversity at regional level, we carried out the study on poly− morphism with DNA markers and performed Principal Coordinate Analysis (PCoA) on the basis of genetic distance matrix. Two ranges (i.e. Baltico−Nordic and Hercyno−Carpathian) of Norway spruce cover north−eastern and southern Poland, respectively. The spruceless zone separates the these two ranges in lowland Poland. From genetic point of view, the Baltico−Nordic P. abies populations mainly harbor mitochondrial haplotype ‘c', whereas the spruces from the Hercyno− −Carpathian range – haplotype ‘a'. Until recently, the Białowieża Forest has been considered to belong to the northern range of spruce occurrence in Europe. Seven populations from Biało− wieża Forest were studied (two of them from the Strict Reserve in Białowieski National Park) according to mitochondrial DNA diversity, and compared with genetic diversity found in 24 other populations located in Białystok Regional Directorate of the State Forests (north−eastern Poland). All studied spruce populations were more than 100 years old and were of natural origin. As result, the spruces from the Białowieża Forest harbored three mitochondrial haplotypes of the nad1 gene (‘c', ‘a' and ‘a1'), with the haplotype ‘a' being more frequently present (>51%) than the hap− lotype ‘c'. The spruce stands from Białowieża Forest have the highest gene diversity (h Nei=0.527) comparing to the other stands from the Białystok RDSF. The PCoA proved the particularity of the spruces from Białowieża Forest grouping them into one cluster of genetic similarity. Our data demonstrated that most of Norway spruces populations from Białystok RDSF harbor haplotype ‘c', which confirms their historical relationship with the Baltico−Nordic range of P. abies in Europe, while most spruces from Białowieża Forest have different historic origin, because they share high frequency of the southern haplotype ‘a'. It also turned out that Norway spruce pop− ulations from two different European ranges met in lowland Poland after the last glacial period, as proved by the presence of two mitochondrial haplotypes ‘a' and ‘c'. It can be assumed, that the spruceless zone was created by human activity in the past millennia. The results of all molecular analyses confirmed the unique character of spruces of Białowieża Forest, which is distinguished by the greater richness of the gene pool in comparison to the region of Białystok RDSF as well as good adaptation to local environmental conditions.
